Aim: In this study antioxidant and anti-inflammatory effect of ethanolic extract of Quisqualis indica leaves was evaluated. Study Design: In-vitro analysis of Quisqualis indica leaf extract. Place and Duration of Study: Molecular Biology laboratory, Department of Biotechnology, G.B Pant Engineering College, Pauri, between July 2015 and July 2016. Methods: Non-enzymatic and enzymatic assays such as DPPH (1, 1diphenyl-2-picryl hydrazyl), FRAP assay, superoxide dismutase SOD (EC 1.15.1.1), catalase (EC 1.11.1.6), for radical scavenging activity of ethanolic extracts of Quisqualis indica Linn. plant leaves had done. For estimation of anti-inflammatory action, two methods were employed: protein denaturation method and membrane stabilization method. Results: Ethanolic extract of leaves on higher concentration had better antioxidant potential when compared with reference standard ascorbic acid. They exhibited strong antioxidant radical scavenging activity values for ethanolic extract of leaves. Results of anti-inflammatory method suggested better potential values for ethanolic extract and compared with standard drug diclofenac sodium respectively. A significant relationship between antioxidant, anti-inflammatory capacity and total phenolic content was examined, indicating that phenolic compounds are the major contributors for the antioxidant and anti-inflammatory properties of this plant. Conclusion: Ethanolic extract of Q. indica exhibited strong anti-inflammatory and antioxidant activity and this can be used for designing novel drug inhibitors with better efficacy.

Euphorbia hirta L (Euphorbiaceae) also called asthma herb has long been prescribed in traditional medicine because it exhibits diverse pharmacological actions due to the presence of alkaloids, flavonoids, polyphenols, triterpenoids, and saponins. The present study is aimed at the study of phytochemical and antioxidant activity and anti-inflammatory screening of E. hirta. Extraction of dried powder was performed followed by phytochemical screening using color reactions. Total phenolic content (TPC) and total flavonoid content (TFC) of the extracts were estimated by Folin-Ciocalteu and Aluminum chloride method respectively. The antioxidant activity was studied by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging method.  Anti-inflammatory activity was studied by using protein denaturation in vitro bioassay. Phytochemical screening showed the presence of flavonoids, alkaloids, and phenolic compounds. TPC, TFC and antioxidant activity (IC50) of the extract were found as 288.10 mg gallic acid equivalent per gram (GAE/g), 29.36 mg quercetin equivalent per gram (QE/g) and 32.23 µg/mL (p<0.05) respectively. Diclofenac sodium and E. hirta extract showed the maximum inhibition of 91.28% and 68.20% respectively at the concentration of 1000 µg/mL compared with control (p>0.05). The phenolic compounds and flavonoids exert antioxidant and anti-inflammatory activities because of their scavenging ability. The demonstrated antioxidant and anti-inflammatory activities may be the rationale behind some of its folkloric uses and also may be responsible for some of its pharmacological effects. Thus, E. hirta can be considered a good source of antioxidants and anti-inflammatory actions, which might be beneficial for combating oxidative stress.

Garcinia mangostana L. is medicinal plant. Its fruit, the mangosteen contains many bioactive xanthones. This study investigates the potential of organic leaf extracts of G. mangostana grown in Ivory Coast. We tested two organic leaf extracts: G. mangostana aqueous ethanolic leaf extract (ethanol: water, 80:20 v/v) (GMLE) and dichloromethane leaf extract (GMLD). We measured total phenolic and total Flavonoids. We analysed the in vitro anti-inflammatory, anti-radical and anti-proliferative activities of leaf extracts. Ethanol leaf extract showed a considerable amount of phenolic content (328.78±34.32 mg GAE/g) and moderate flavonoids content (43.60±1.48 mg QE/g), Dichloromethane extract had low values of phenolic (70.31±4.55 mg GAE/g) and flavonoids (8.49±0.69 mg QE/g). However, GMLD extract gave a significant anti-inflammatory activity (IC50=152.79±3.34 µg/mL), comparable to the standard drug diclofenac sodium (IC50=142.30±1.22 µg/mL), contrary to GMLE extract (IC50=652.33±12.23 µg/mL). The radical scavenging assay showed a very significant ability of ethanol leaf extract to reduce the DPPH radical (IC50=33.40±0.67 µg/mL) compared to references molecules such as Trolox (IC50=43.72±0.31 µg/mL) and acid ascorbic (IC50=27.20±0.17 µg/mL), dichloromethane extract results showed lowest activity (IC50 = 580.00±23.03 µg/mL). All the organic leaf extracts of G. mangostana had moderate anti-proliferative activity on L20B, RD and VS cell lines studied with IC50 values ranging from 110.89 ±4.82 µg/mL to 860.60±25.78 µg/mL). Our results prove the high potential of the G. mangostana leave extracts as anti-inflammatory and anti-oxidative stress drugs. However, further studies are to determine and validate all the medicinal properties of G. mangostana leaves extracts.

Aims: The present study evaluates the anti-inflammatory and antioxidant activity of the crude dichloromethane (CDCME), ethyl acetate (CEAE), and methanol (CMeE) extracts from the plant Oldenlandia corymbosa L. Background:Oldenlandia species have been popular among the people of the Indian subcontinent to treat several types of internal and external inflammation for a long time. Plant decoctions have been used to battle inflammation in cases of tonsilitis, pneumonia and cholecystitis, among others. Objective: The present work designed to demonstrate the properties of the previously mentioned plant extracts to prevent inflammation both in vivo and in vitro. This work is the first investigation of such extracts from this species and their relationship with anti-inflammatory activity. Method: The anti-inflammatory properties of the Oldenlandia corymbosa L. extracts were evaluated in vitro with the Red Blood Cell (RBC) membrane stabilization method and the protein denaturation method and in vivo with the carrageenan-induced paw oedema method. Furthermore, the free radical scavenging activity of the extracts was carried out with the 1,1-diphenyl-2- picrylhydrazyl (DPPH) radical oxidation, total antioxidant capacity and iron reduction assay. Result: Both in vivo and in vitro studies showed that CDCME had the most predominant effects with the value of 80.5% for RBC membrane stabilization, 60% for inhibition of protein denaturation at the concentration of 1000 µg/mL and 63.28% (after 3 h, * p < 0.05) for inhibition of paw oedema (300 mg/kg bwt) compared to carrageenan-induced mice. The free radical scavenging activity was studied by DPPH, total antioxidant and reducing activity assay. CDCME showed scavenging activity in all the methods and an IC50 value of 473.86 µg/mL for DPPH method. Conclusions: The findings of the study remarked that CDCME of the plant has strong anti-inflammatory and antioxidant effects that validate the traditional use of the plant to get remedy from pain. Other: The plants Oldenlandiacorymbosa Linn were provided by the Bangladesh Council of Scientific and Industrial Research Laboratory campus, Rajshahi, Bangladesh. Experiments on animals were conducted by ethical permission of Institute of Biological Sciences, University of Rajshahi, Bangladesh (license no: 225/320-IAMEBBC/IBSc).

Objectives: The study was undertaken to evaluate the in vitro anti-inflammatory and anti-arthritic activity of the ethanolic extract of leaves of Pyrenacantha volubilis (EEPV) using human red blood cells (HRBCs) membrane stabilization and protein denaturation methods. Methods: In the present study, the in vitro anti-inflammatory and anti-arthritic activity of EEPV was carried out using HRBC membrane stabilization by hypotonicity-induced hemolysis and protein denaturation using egg albumin methods at various concentrations (100, 200, 400, 800, and 1000) of EEPV. Diclofenac sodium was used as reference standard. Results: P. volubilis was effective in inhibiting HRBC membrane stabilization and protein denaturation in a dose-dependent manner and was comparable to the standard drug diclofenac sodium. Conclusion: The study suggests that P. volubilis has potential anti-inflammatory and anti-arthritic activity.

Background & Introduction : Antioxidants are known to prevent oxidative stress-induced damage to the biomolecules and thus, delay the onset of cancers and many age-related diseases. Therefore, the development of novel and potent antioxidants is justified. Method: During this study, we synthesized symmetrical bis-Schiff bases of carbohydrazide 1-27, and evaluated their in vitro antioxidative activity and cytotoxic activity. Results: Among synthesized compounds, six compounds 20 (IC50 = 12.89 ± 0.02 µM), 16 (IC50 = 14.32 ± 0.43 µM), 17 (IC50 = 18.52 ± 0.83 µM), 19 (IC50 = 22.84 ± 0.62 µM), 24 (IC50 = 35.1 ± 0.82 µM) and 15 (IC50 = 40.03 ± 1.06 µM) showed an excellent 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, better than the standard butylatedhydroxyanisole (BHA) (IC50 = 44.6 ± 0.6 µM). Likewise, two compounds 16 (IC50 = 4.3 ± 1.3 µM) and 20 (IC50 = 6.6 ± 1.6 µM) showed oxidative burst scavenging activity better than the standard drug ibuprofen (IC50 = 11.2 ± 1.9 µM). Some synthesized compounds showed good to moderate toxicity against prostate cancer (PC-3) cell lines. Conclusion: This study has identified potent antioxidants and good cytotoxic agents with the potential to further investigate.

Objective: To investigate the antioxidant activity of ethanolic extract of Terminalia bellerica fruit pulp (TBFP), seed (TBS) and bark (TBB), and identification of phytochemical constituents of said extracts.Methods: The antioxidant potential of T. bellerica (TB) parts was evaluated by free radical scavenging activity (FRSA), superoxide anion radical scavenging activity (SARSA), hydroxyl radical scavenging activity (HRSA) and compared with reference standard quercetin. Lipid peroxidation (LPO), ferric thiocyanate activity (FTC) and reducing power (RP) of the plant extracts were also examined. The phytochemical constituents of said extracts have been quantified by gas chromatography-mass spectrometer (GC-MS) method.Results: Ethanolic extract of TBFP exhibited high phenolic content (254.33) followed by TBS (227.33) and TBB (185) mg/g of GAE. TBFP showed lowest IC50 for FRSA (62 µg/ml), SARSA (39 µg/ml) and HRSA (27 µg/ml) and highest RP (3.39 ascorbic acid equivalents/ml). TBB showed lowest IC50 for LPO (48 µg/ml) whereas TBS showed lowest IC50 for FTC (109 µg/ml). Predominant phyto-compounds present in TB extracts were quinic acid, gallic acid, ethyl galate, 9, 12 octadecadienoic acid and glucopyranose in varying concentrations as analyzed by GC-MS.Conclusion: On the basis of correlation of antioxidant studies with total phenolic content (TPC) and GC-MS analysis of different part of TB, it may be concluded that high concentration of total phenolics and other bioactive phytochemicals of TB extracts make it a potential source of nutraceutical antioxidants.

In the present study, the antioxidant and analgesic potential of the ethanolic extract of the leaves of Derris trifoliata was evaluated. The free radical scavenging activity of the crude extract on the stable radical 1, 1-diphenyl-2-picrylhydrazyl (DPPH) was determined by comparing the DPPH inhibitory capacity of the extract. In the quantitative assay, Derris trifoliata extract displayed a free radical scavenging activity in the DPPH assay (IC50 = 19 ?g/ml) which is comparable to that of ascorbic acid (IC50 = 7.80 ?g/ml), a well-known standard antioxidant. The analgesic responses of the given samples of extracts were evaluated using the Tail immersion method. In the analgesic activity test, extract at dose of 200 mg/kg and 400 mg/kg exhibited significant (P<0.05 and P<0.001 respectively) inhibition of pain by 166.82 and 184.95 after 120 and 180 minutes respectively while the standard drug Diclofenac Na inhibition was found to be 217.67 after 180 minutes at a dose of 25 mg/kg body weight DOI: http://dx.doi.org/10.3329/ijpls.v1i2.12951 International Journal of Pharmaceutical and Life Sciences Vol.1(2) 2012

Honey and propolis are natural substances produced by Apis mellifera that contain flavonoids, phenolic acids, and several other phytochemicals. The aim of this study was to phytochemically characterize three different types of honey and propolis, both separately and mixed, and to evaluate their wound-healing activity. Total phenolic compounds and flavonoids were determined using the Folin–Ciocalteu’s and aluminum chloride colorimetric methods, respectively. The antioxidant activity was evaluated by both the DPPH free radical scavenging assay and β-carotene bleaching test, and the anti-inflammatory activity was determined by a protein denaturation method. To evaluate the wound-healing activity of the samples, NHDF cells were subjected to a wound scratch assay. The obtained results showed that dark-brown honey presents a higher concentration of phenolic compounds and flavonoids, as well as higher antioxidant and anti-inflammatory activities. Propolis samples had the highest concentrations in bioactive compounds. Examining the microscopic images, it was possible to verify that the samples promote cell migration, demonstrating the wound-healing potential of honey and propolis.
