Background. We have previously shown that nitrogenous metabolites have immunomodulatory effects in healthy rats and humans as well as in patients with encephalopatia. The purpose of this study is their immunotropic activity in patients with chronic pyelonephritis. Materials and Methods. The object of observation were 24 men (aged 23-76 years) with chronic pyelonephritis in remission. The plasma levels and urinary excretion of uric acid, urea and creatinine and parameters of immunity twice (on admission and after 10 days of balneotherapy at the Truskavets’ Spa) was performed. Results. Judging by the multiple correlation coefficient uricemia exhibits maximal immunotropic activity (R=0,772), followed by creatininemia (R=0,643), urea plasma (R=0,584) and creatinineuria (R=0,506) instead, urea and uric acid excretion correlate with immune parameters insignificantly (R=0,327 and 0,262 respectively). Nitrogenous metabolites together upregulate most parameters of phagocytosis by neutrophils Staph. aureus and E. coli, the level in the blood of CD8 T-lymphocytes, CIC, IgM, componentes of leukocytogram as well as entropy of leukocytogram and immunocytogram. Instead, they downregulate the relative level of lymphocytes in general and of CD4 T-lymphocytes in particular. Conclusion. Nitrogenous metabolites exhibit immunotropic activity in both healthy humans and in patients with chronic pyelonephritis in remission. Both common and distinctive features of immunomodulation were revealed.

Background. We have previously shown that nitrogenous metabolites have immunomodulatory effects, both in healthy rats and in humans exposed to pathogenic influences. The purpose of this study is their immunotropic activity in clinically healthy people. Materials and Methods. The object of observation were 27 men (aged 24-63 ys) and 14 women (33-62 ys). The plasma levels of the nitrogenous metabolites and parameters of immunity twice with an interval of 5 days was performed. Results. Judging by the multiple correlation coefficient uric acid exhibits maximal immunotropic activity (R=0,665), followed by creatinine (R=0,596) and urea (R=0,541), and closes the constellation of metabolites bilirubin, with the activity of conjugated bilirubin predominating over that of unconjugated (0,539 vs 0,484). Nitrogenous metabolites together upregulate the level in the blood of B-lymphocytes, CIC, IgG, IL-1, eosinophils and monocytes, as well as most parameters of phagocytosis by neutrophils Staph. aureus and E. coli. Instead, they downregulate the phagocytosis activity of Staph. aureus, the relative content of rod-shaped neutrophils, lymphocytes in general and NK-, T active and 0-Lymphocytes in particular. Downregulation of 0-Lymphocytes reflects upregulation of receptor expression, apparently CD22. Conclusion. Nitrogenous metabolites exhibit immunotropic activity in both healthy rats and humans, as well as in patients.

The immunotropic activity of polyelectrolyte complexes (PEC) of κ-carrageenan (κ-CGN) and chitosan (CH) of various compositions was assessed in comparison with the initial polysaccharides in comparable doses. For this, two soluble forms of PEC, with an excess of CH (CH:CGN mass ratios of 10:1) and with an excess of CGN (CH: CGN mass ratios of 1:10) were prepared. The ability of PEC to scavenge NO depended on the content of the κ-CGN in the PEC. The ability of the PEC to induce the synthesis of pro-inflammatory (tumor necrosis factor-α (TNF-α)) and anti-inflammatory (interleukine-10 (IL-10)) cytokines in peripheral blood mononuclear cell was determined by the activity of the initial κ-CGN, regardless of their composition. The anti-inflammatory activity of PEC and the initial compounds was studied using test of histamine-, concanavalin A-, and sheep erythrocyte immunization-induced inflammation in mice. The highest activity of PEC, as well as the initial polysaccharides κ-CGN and CH, was observed in a histamine-induced exudative inflammation, directly related to the activation of phagocytic cells, i.e., macrophages and neutrophils.

In recent years, there have been negative developments such as deforestation, overgrazing, and loss of plant species due to the mining and pasture misuse, and global warming. Due to this, the immunity of Mongolian livestock has been decreasing. The uses of immunosuppressive drugs is limited in the veterinary practice. The aim of this study, the technology of 3 types of drugs prepared from the different type of medical plants and their immunotropic activity was tested in the laboratory animals. In the result, the immunotropic activity of 1th group preparation /SL (50), AS (30) and OPG (20)/ was higher than control group and also other group preparation.    Дархлаа тэтгэх үйлдэлтэй эмийн бэлдмэлийн иммунотроп үйлдлийг судалсан дүн Сүүлийн жилүүдэд уул уурхайн олборлолт, бэлчээрийг зөв зохистой ашиглахгүй байх, дэлхийн цаг агаарын дулаарал зэрэгтэй холбоотойгоор бэлчээр хомсдох, талхлагдах, ургамлын төрөл, зүйлийн тоо хомсдох зэрэг сөрөг үзэгдэл ажиглагдах боллоо. Antibacterial drugs are routinely used in therapy of periodontal diseases. However, increasing incidence of antibiotics resistance necessitates development of novel therapeutic approaches for oral diseases. Currently, newly designed antibacterial agents based on natural, semi-synthetic and synthetic peptides is the most promising approach in dentistry. Among them is a product containing synthetic peptide (ZP2) replicating active site of granulocyte-macrophage colonystimulating factor (GM-CSF) as the main active ingredient in Atsegram-spray (manufactured by Academic Innovation Research Center, Chelyabinsk). Our study was aimed at assessing efficacy of a combination therapy of inflammatory periodontal diseases by introducing Atsegram-spray as well as examining potential relationship between peptide ZP2-related biological properties, physicochemical properties of the spray and mechanism of antibacterial and immunotropic action for substantiating its application on oral mucosa. During the first stage experiments, it was found that the peptide ZP2 was able to trigger lymphocyte blast transformation in vitro indicating that it might influence cell proliferation and exhibit marked immunotropic activity. Next, we assessed potential effects of the peptide ZP2 on biofilm formation mediated by staphylococcal clinical isolates. It was shown that peptide ZP2 inhibited biofilm formation in 75.0±9.0% of S. aureus and 50.0±15.1% of S. epidermidis strains, with mean inhibition index of biofilm formation reaching 25.1±3.8 and 50.4±6.0%, respectively. However, peptide ZP2 in 8.3–25.0% of staphylococcal clinical isolates was found to stimulate/lack effect on biofilm formation by 14.9–48.5 and 16.7–25.0% cultures, respectively. Thus, the synthetic peptide ZP2 exerts divergent, but mainly inhibitory effects on biofilm formation with staphylococcal clinical strains, which are characterized by inter- and intraspecific (strain) variability. Use of a synthetic peptide-based spray in antibacterial therapy of mild chronic generalized periodontitis (main group) one month after the onset was found to improve oral hygiene by 28.5% as well as decrease PMA index and gum bleeding index by 82.8 and 100%, respectively. In contrast, such parameters in patients receiving basic therapy (comparison group) were lower on average by 2-fold. While analyzing physicochemical properties of the spray such as pH, buffer capacity and solution osmotic pressure, it was found that they were related to the antibacterial mechanism of drug activity and efficacy in treatment of inflammatory periodontal diseases. Thus, assessing peptide ZP2-related biological properties and physicochemical parameters of the spray allows to evaluate their role in mechanism of previously unknown antibacterial and immunotropic activity. These findings confirm feasibility and efficacy of using Atsegram-spray in dentistry as an alternative means to antimicrobial agents, such as antibacterial drugs.
