Abstract Background Sandwich ELISA is an ideal antigen detection and quantification assay. Recently, it was used as the basic concept for high technology diagnostics. The specificity of the assay depends on the exclusion of detection cross-reactivity which arises from using two antibodies developed in different species. Since mice and rats are the common laboratory animals used to develop antigen specific antibodies. Therefore, the questions we addressed here were (1) can one use antigen-specific antibodies raised in mice and rats in the same assay to specifically detect/quantify antigens? and (2) which antibodies of the two rodents should be placed for capturing and for detection in the antigen-detection sandwich? Results Direct ELISA assay was used to assess for the specific reaction of the HRP-conjugated antibody to the target serum. First reaction was to compare between either conjugate anti-rat IgG (homologous) or anti-mouse IgG (heterologous) for the detection of rat sera IgG. Following the dilution factor optimization, the O.D. were 0.744±0.051 and 0.604±0.05, respectively (p= .004). The difference in mean O.D. of 0.14 reflected an unaccepted non-specific reaction. The second reaction was to compare between either conjugate anti-mouse IgG (homologous) and anti-rat IgG (heterologous) for the detection of mouse sera IgG. The recorded O.D. were 0.9414±0.14 and 0.317 ±0.141, respectively (p= .0002). The improved difference in mean O.D. of 0.624 reflecting a minimized cross-reaction. Conclusions Our results suggest that it is possible to use both Swiss albino mice and albino rats in a single sandwich ELISA, given that the captured antibody species to be from the Swiss albino mice and the detection antibody to be from the albino rat. The described working details are limited to the source of the antibodies used in the study. However, the approach stresses on the importance of such optimization steps before making any interpretations based on the antigen detection. To our knowledge, this study is the first to cover the optimal order of the capturing and the detection antibodies in a sandwich ELISA assay. In addition to addressing the possible interfering cross-reactivity that result from using mouse and rat serum antibodies in a single assay. Graphical abstract

: An overview of the current uptake of continuous flow techniques for various functional group interconversion reactions is presented. Besides highlighting a variety of prominent examples and their main features, this review provides insights into specific reaction classes, such as oxidations, reductions, rearrangements as well as different C-H functionalization processes. While this review can only include key examples from the last decade, the reader will find a solid foundation of important transformations along with further references to inform and appreciate the opportunities arising from modern synthesis technologies such as flow synthesis.

AbstractMemories are thought to undergo an episodic-to-semantic transformation in the course of their consolidation. We here test if repeated recall induces a similar semanticisation, and if the resulting qualitative changes in memories can be measured using simple feature-specific reaction time probes. Participants studied associations between verbs and object images, and then repeatedly recalled the objects when cued with the verb, immediately and after a two-day delay. Reaction times during immediate recall demonstrate that conceptual features are accessed faster than perceptual features. Consistent with a semanticisation process, this perceptual-conceptual gap significantly increases across the delay. A significantly smaller perceptual-conceptual gap is found in the delayed recall data of a control group who repeatedly studied the verb-object pairings on the first day, instead of actively recalling them. Our findings suggest that wake recall and offline consolidation interact to transform memories over time, strengthening meaningful semantic information over perceptual detail.

В статье рассмотрен вопрос идентифицируемости математической модели кинетики химической реакции. В процессе решения обратной задачи по оценке параметров модели, характеризующих процесс, нередко возникает вопрос неединственности решения. На примере конкретной реакции продемонстрирована необходимость проводить анализ идентифицируемости модели перед проведением численных расчетов по определению параметров модели химической реакции. The identifiability of the mathematical model of the kinetics of a chemical reaction is investigated in the article. In the process of solving the inverse problem of estimating the parameters of the model, the question arises of the non-uniqueness of the solution. On the example of a specific reaction, the need to analyze the identifiability of the model before carrying out numerical calculations to determine the parameters of the reaction model was demonstrated.

Carbonylation reactions with carbon monoxide (CO) provide efficient and attractive routes for the synthesis of bulk and fine chemicals. However, the practice of using a large excess of an inflammable, lethal and greenhouse CO gas is always a concern in this chemistry. The development of CO surrogates has gained substantial interest and become a green alternative to gaseous CO. Many of the recent studies have focused on the development of other benign and safe reagents to work as a CO source in carbonylation reactions, and the assortment of feasible CO surrogates for specific reaction can be accomplished by the literature data. This review describes the recent developments in palladium-catalyzed carbonyl insertions without the direct use of gaseous CO.

The selenophene derivatives are an important class of selenium-based heterocyclics. These compounds play an important role in prospecting new drugs, as well as in the development of new light-emitting materials. During the last years, several methods have been emerging to access the selenophene scaffold, employing a diversity of cyclization-based synthetic strategies, involving specific reaction partners and particularities. This review presents a comprehensive discussion on the recent advances in the synthesis of selenophene-based compounds, starting from different precursors, highlighting the main differences, the advantages, and limitations among them.
