Autophagy is an evolutionarily conserved cellular-degradation mechanism implicated in antiviral defense in plants. Studies have shown that autophagy suppresses virus accumulation in cells; however, it has not been reported to specifically inhibit viral spread in plants. This study demonstrated that infection with citrus leaf blotch virus (CLBV; genus Citrivirus, family Betaflexiviridae) activated autophagy in Nicotiana benthamiana plants as indicated by the increase of autophagosome formation. Impairment of autophagy through silencing of N. benthamiana autophagy-related gene 5 (NbATG5) and NbATG7 enhanced cell-to-cell and systemic movement of CLBV; however, it did not affect CLBV accumulation when the systemic infection had been fully established. Treatment using an autophagy inhibitor or silencing of NbATG5 and NbATG7 revealed that transiently expressed movement protein (MP), but not coat protein, of CLBV was targeted by selective autophagy for degradation. Moreover, we identified that CLBV MP directly interacted with NbATG8C1 and NbATG8i, the isoforms of autophagy-related protein 8 (ATG8), which are key factors that usually bind cargo receptors for selective autophagy. Our results present a novel example in which autophagy specifically targets a viral MP to limit the intercellular spread of the virus in plants.

Culex quinquefasciatus mosquitoes are a vector of transmission of several types of diseases, such as filariasis, Japanese encephalitis, and dirofilariasis. Larval control is the key strategy of disease control programs caused by vectors around the world because it can prevent larvae to enter the adult stage. Control of mosquito larvae that are often used is chemically controlled. Citrus hystrix is a natural plant and contains chemical compounds that have biological activity, such as flavonoids, carotenoids, and limonoids. The present study aimed to determine the effect of citrus leaf extract (Citrus hystrix) on the mortality of the larva Culex quinquefasciatus. The method used in the current study was a laboratory experimental study method with the experimental design using a completely randomized design. The research plot carried out was the rearing larvae of Culex quinquefasciatus, the manufacture of citrus leaf extract, the manufacture of larvicidal material, and the observation phase after treatment. Extract N-hexane omplet name hystrix leaf has high toxicity because it contains essential oils, flavonoids, alkaloids, terpenoids, saponins, and Limonoids. The Optimal concentration of N-hexane extracts of citrus leaves that lead to larval mortality of Culex quinquefasciatus in vitro amounted to 93.33% on 4 hours of observation with a concentration of 4000 ppm. Extract N-hexane lime leaf has a larvicidal effect on the larva Culex quinquefasciatus in vitro.

ABSTRAKPermasalahan utama yang dihadapi oleh pihak kelurahan Sungai Pinang Luar Samarinda adalah penggunaan minyak jelantah yang berulang kali oleh ibu rumah tangga untuk menggoreng makanan dan membuang minyak jelantah tersebut sembarangan. Pengabdiaan ini bertujuan untuk memberikan pemahaman kepada ibu-ibu PKK kelurahan Sungai Pinang Luar Samarinda tentang bahayanya penggunaan minyak jelantah yang digunakan berulangkali bagi kesehatan dan pembuangan minyak jelantah ke lingkungan yang akan mengganggu stabilitas ekologi pada lingkungan tersebut, sehingga dibentuk sebuah upaya tindakan melalui pelatihan pengolahan minyak jelantah menjadi sesuatu yang bernilai ekologis dan ekonomis. Metode yang digunakan dalam pengabdian ini adalah pelatihan langsung kepada ibu-ibu PKK di kelurahan Sungai Pinang Luar dalam pembuatan lilin aroma daun jeruk dari minyak jelantah. Hasil pengabdian ini terlihat antusiasme peserta untuk mencoba membuat sendiri lilin aroma daun jeruk dari minyak jelantah yang digunakan. Kata Kunci: pelatihan; minyak jelantah; lilin aroma daun jeruk. ABSTRACTThe main problem faced by the Sungai Pinang Luar village Samarinda is the repeated use of used cooking oil by housewives to fry food and throw the oil carelessly. This service aims to provide an understanding to PKK women in Sungai Pinang Village, Luar Samarinda about the dangers of using used cooking oil repeatedly for health and the disposal of used cooking oil into the environment which will disrupt the ecological stability of the environment, so that an action effort is formed through oil processing training, waste cooking becomes something of ecological and economic value. The method used in this service is direct training to PKK women in Sungai Pinang Luar village in making orange leaf scented candles from used cooking oil. The results of this service were seen by the enthusiasm of the participants to try to make their own orange leaf scented candles from the used cooking oil.Keywords: training; cooking oil; citrus leaf scented candle.

Xanthomonas citri pv. citri (Xcc) is the causative agent of citrus canker, one of the most devastating disease threatening global citrus production. Here, we present the genome sequence of Xcc strain GD82 from a formalin-fixed citrus leaf specimen showing canker lesions collected at 1982 in Guangdong Province, China. The GD82 genome comprised of 5,197,217 bp with G+C content of 64.8 %, along with four circular plasmids, pXAC33 (32,377 bp), pXAC64 (63,972 bp), pXAC47 (47,810 bp) and pGD82.1 (219,560 bp). This is the oldest Xcc genome from historical citrus canker specimens in China, which will enrich the current Xcc genome database and facilitate genomic evolution research of Xcc.
