Background: In cattle, many serotypes of Salmonella enterica are responsible for a wide variety of clinical manifestations, which can cause considerable economic loss. Some serotypes can cause cows to abort sporadically, such as the Dublin serotype. This study was carried out on different cattle farms in the Algiers region to determine the prevalence of Salmonella Dublin using bacteriological and immunological methods.Methodology: The prevalence of Salmonella was determined by bacteriological analysis in accordance with the reference method AFNOR NF U 47-100 on faecal samples collected from 184 cattle belonging to 19 different farms, and serotyping for S. Dublin. Immunological analysis by enzyme-linked immunosorbent assay (ELISA) for S. Dublin was carried out on milk samples collected from 91 cattle. A survey of case (n=5) and control (n=14) farms for comparative analysis was performed to demonstrate a link between abortion in cows and prevalence of S. Dublin with both bacteriological and immunological methods. Sensitivity, specificity, Cohen Kappa coefficient, McNemar test odds ratios, and confidence intervals were calculated using Winepiscope 2.0 and StatA 9.1 software,and p<0.05 was considered as statistically significant.Results: The bacteriological results showed a prevalence of 7.6% (95%CI: 3-10), for Salmonella and serotyping revealed a prevalence for S. Dublin of 2.7%. The immunological analysis of milk by the ELISA technique revealed a prevalence of 13.2% (95%CI: 5-20) for S. Dublin. The comparative study between immunological results from milk and bacteriological results from faeces for detecting S. Dublin  showed poor agreement between the two tests (k=0.25), with enzyme immunoassay being significantly more sensitive than the bacteriological test (p<0.05). The results of the survey did not demonstrate a clear association between bacteriological detection of S. Dublin in faeces and abortion in cows (OR=8.66, 95%CI: 0.58-130.12). However, with the immunological analysis of milk for S. Dublin, there was a significant positive association (OR=62.33, 95%CI: 2.13-18.22) between a positive antibody response to S. Dublin in milk and the presence of abortions on the farm.Conclusion: In view of these results, we can conclude that Salmonella infections should systematically feature in the differential diagnosis of abortions in dairy cattle in Algeria. Undirected modifications between food proteins and secondary plant metabolites can occur during food processing. The results of covalent interactions can alter the functional and biological properties of the proteins. The present work studied the extent of which covalent conjugation of the bioactive metabolite benzyl isothiocyanate (BITC; a glucosinolate breakdown product) to the whey protein α-lactalbumin affects the protein’s allergenicity. Additional to the immunological analysis of native untreated and BITC-modified α-lactalbumin, the analysis of antigenic properties of proteolytically digested protein derivatives was also performed by high performance thin layer chromatography and immunostaining. As a result of the chemical modifications, structural changes in the protein molecule affected the allergenic properties. In this process, epitopes are destroyed or inactivated, but at the same time, buried epitopes can be exposed or newly formed, so that the net effect was an increase in allergenicity, in this case. Results from the tryptic hydrolysis suggest that BITC conjugation sterically hindered the cleavage sites for the enzyme, resulting in reduced digestibility and allergenicity. Residual antigenicity can be still present as short peptide fragments that provide epitopes. The desire to make food safer for allergy sufferers and to protect sensitized individuals from an allergenic reaction makes it clear that the detection of food antigens is mandatory; especially by considering protein interactions.

This is the first report using an anti-tumor drug called Pazopanib for a case of solitary fibrous tumor that occurred in the ear canal. Pazopanib will be useful for clinicians involved in this case in the future, we have reported the detailed progress and immunological analysis.

Hepatocellular carcinoma (HCC) is the fourth leading cause of cancer death worldwide with a minority of patients being diagnosed early enough for curative-intent interventions. We report the first use of preoperative cabozantinib plus nivolumab to successfully downstage what presented as unresectable HCC as part of an ongoing phase 1b study. Preoperative treatment with cabozantinib and nivolumab led to >99% reduction in alpha-fetoprotein, −37.3% radiographic reduction by RECIST 1.1 and a near complete pathologic response (80% to 100% necrosis). An integrated immunological analysis was performed on the post-treatment surgical tumor sample and matched pre-treatment and post-treatment peripheral blood samples with high-dimensional imaging and cytometry techniques. Bayesian non-negative matrix factorization (CoGAPS, Coordinated Gene Activity in Pattern Sets) and self-organizing map (FlowSOM) algorithms were used to distinguish changes in functional markers across cellular neighborhoods in the single cell data sets. Brisk immunological infiltration into the tumor microenvironment was observed in non-random, organized cellular neighborhoods. Systemically, combination therapy led to marked promotion of effector cytotoxic T cells and effector memory helper T cells. Natural killer cells also increased with therapy. The patient remains without disease recurrence and with a normal alpha-fetoprotein approximately 2 years from presentation. Our study provides proof-of-concept that borderline resectable or locally advanced HCC warrants consideration of downstaging with effective neoadjuvant systemic therapy for subsequent curative resection.
