AbstractWith growing demand for food production in Africa, protecting wetlands and combining increased agricultural production with conservation of the ecological integrity of wetlands is urgent. The role of aboveground biomass of papyrus (Cyperus papyrus) in the storage and retention of nitrogen (N) and phosphorus (P) was studied in two wetland sites in East Africa under seasonally and permanently flooded conditions. Nyando wetland (Kenya) was under anthropogenic disturbance from agriculture and vegetation harvesting, whereas Mara wetland (Tanzania) was less disturbed. Maximum papyrus culm growth was described well by a logistic model (regressions for culm length with R2 from 0.70 to 0.99), with culms growing faster but not taller in Nyando than in Mara. Maximum culm length was greater in permanently than in seasonally flooded zones. Total aboveground biomass was higher in Mara than in Nyando. The amounts of N and P stored were higher in Mara than in Nyando. In disturbed sites, papyrus plants show characteristics of r-selected species leading to faster growth but lower biomass and nutrient storage. These findings help to optimize management of nutrient retention in natural and constructed wetlands.

In planta genome editing represents an attractive approach to engineering crops/varieties that are recalcitrant to culture-based transformation methods. Here, we report the direct delivery of CRISPR/Cas9 ribonucleoproteins into the shoot apical meristem using in planta particle bombardment and introduction of a semidwarf1 (sd1)-orthologous mutation into wheat. The triple knockout tasd1 mutant of an elite wheat variety reduced culm length by 10% without a reduction in yield.

AbstractHeterosis helps increase the biomass of many crops; however, while models for its mechanisms have been proposed, it is not yet fully understood. Here, we use a QTL analysis of the progeny of a high-biomass sorghum F1 hybrid to examine heterosis. Five QTLs were identified for culm length and were explained using the dominance model. Five resultant homozygous dominant alleles were used to develop pyramided lines, which produced biomasses like the original F1 line. Cloning of one of the uncharacterised genes (Dw7a) revealed that it encoded a MYB transcription factor, that was not yet proactively used in modern breeding, suggesting that combining classic dw1or dw3, and new (dw7a) genes is an important breeding strategy. In conclusion, heterosis is explained in this situation by the dominance model and a combination of genes that balance the shortness and early flowering of the parents, to produce F1 seed yields.

Miscanthus is a close relative of Saccharum and a potentially valuable genetic resource for improving sugarcane. Differences in flowering time within and between Miscanthus and Saccharum hinders intra- and interspecific hybridizations. A series of greenhouse experiments were conducted over three years to determine how to synchronize flowering time of Saccharum and Miscanthus genotypes. We found that day length was an important factor influencing when Miscanthus and Saccharum flowered. Sugarcane could be induced to flower in a central Illinois greenhouse using supplemental lighting to reduce the rate at which days shortened during the autumn and winter to 1 min d-1, which allowed us to synchronize the flowering of some sugarcane genotypes with Miscanthus genotypes primarily from low latitudes. In a complementary growth chamber experiment, we evaluated 33 Miscanthus genotypes, including 28 M. sinensis, 2 M. floridulus, and 3 M. ×giganteus collected from 20.9° S to 44.9° N for response to three day lengths (10 h, 12.5 h, and 15 h). High latitude-adapted M. sinensis flowered mainly under 15 h days, but unexpectedly, short days resulted in short, stocky plants that did not flower; in some cases, flag leaves developed under short days but heading did not occur. In contrast, for M. sinensis and M. floridulus from low latitudes, shorter day lengths typically resulted in earlier flowering, and for some low latitude genotypes, 15 h days resulted in no flowering. However, the highest ratio of reproductive shoots to total number of culms was typically observed for 12.5 h or 15 h days. Latitude of origin was significantly associated with culm length, and the shorter the days, the stronger the relationship. Nearly all entries achieved maximal culm length under the 15 h treatment, but the nearer to the equator an accession originated, the less of a difference in culm length between the short-day treatments and the 15 h day treatment. Under short days, short culms for high-latitude accessions was achieved by different physiological mechanisms for M. sinensis genetic groups from the mainland in comparison to those from Japan; for mainland accessions, the mechanism was reduced internode length, whereas for Japanese accessions the phyllochron under short days was greater than under long days. Thus, for M. sinensis, short days typically hastened floral induction, consistent with the expectations for a facultative short-day plant. However, for high latitude accessions of M. sinensis, days less than 12.5 h also signaled that plants should prepare for winter by producing many short culms with limited elongation and development; moreover, this response was also epistatic to flowering. Thus, to flower M. sinensis that originates from high latitudes synchronously with sugarcane, the former needs day lengths >12.5 h (perhaps as high as 15 h), whereas that the latter needs day lengths <12.5 h.

The genotype of gal and d60 were investigated in 33 rice varieties chosen from representative semidwarf and dwarf rice varieties. These were crossed with three tester lines, the d60Gal line (genotype d60d60GalGal), the D60gal line (Koshihikari, D60D60galgal), and the D60Gal line (D60D60GalGal). Each F1 plant was measured for culm length, and seed fertility. As a result, all F1 lines with the d60Gal line showed tallness and partial sterility, reduced by 25% in average from those with the D60gal line (Koshihikari) and the D60Gal line. These data indicated that the genotype of the 33 varieties is D60D60galgal and that the d60 locus is not allelic to those of sd1, d1, d2, d6, d18k, d29, d30, d35, d49, d50, and qCL1 involved in the 33 varieties. In addition, the gal gene is not complementarily activated with the semidwarf and dwarf genes described above, other than d60. The Gal gene will be ubiquitously distributed in rice. It is emphasized that Gal is a rare and valuable mutant gene essential to the transmission of d60. The double dwarf genotype of homozygous d30d60 was rarely gained in the F3 of the d30 line × d60 line by breaking their repulsion d60-D30 linkage on chromosome 2.

Gene effects on the yield performance were compared among promising semidwarf genes, namely, novel gene d60, representative gene sd1 with different two source IR8 and Jukkoku, and double dwarf combinations of d60 with each sd1 allele, in a Koshihikari background. Compared with the culm length of variety Koshihikari (mean, 88.8 cm), that of the semidwarf or double dwarf lines carrying Jukkoku_sd1, IR8_sd1, d60, Jukkoku_sd1 plus d60, or IR8_sd1 plus d60 was shortened to 71.8 cm, 68.5 cm, 65.7 cm, 48.6 cm, and 50.3 cm, respectively. Compared with the yield of Koshihikari (mean, 665.3 g/m2), that of the line carrying Jukkoku_sd1 allele showed the highest value (772.6 g/m2, 16.1% higher than Koshihikari), while that of IR8_sd1, d60 and IR8_sd1 plus d60, was slightly decreased by 7.1%, 5.5%, and 9.7% respectively. The line carrying Jukkoku_sd1 also showed the highest value in number of panicles and florets/panicle, 16.2% and 11.1% higher than in Koshihikari, respectively, and these effects were responsible for the increases in yield. The 1000-grain weight was equivalent among all genetic lines. Except for the semidwarf line carrying Jukkoku_sd1, semidwarf line carrying d60 was equivalent to line carrying IR8_sd1in the yield of unpolished rice, and yield components such as panicle length, panicle number, floret number /panicle. Therefore, the semidwarfing gene d60 is one of the best possible choices in practical breeding.

Genetic variability, heritability and selection response were studied among rice genotypes for yield attributing traits. Nine rice genotypes were evaluated in randomized complete block design with two replications at the University of Agriculture Peshawar-Pakistan during 2018. Significant differences were observed among the rice genotypes for all the traits. Pakhal took maximum value for Culm length (113.6 cm), Panicle length (29.05 cm), Secondary branches panicle-1 (31.4), Grain length (10 mm), Spikelets panicle-1 (162.8) and Grains panicle-1 (122.7 g). However genotype NIAB-IR-8 took maximum value for 100 grain weight (2.9 g) and Grain yield plant-1 (30.28 g). High heritability and genetic advance were observed for all the studied traits viz. Culm length (0.95 and 27.77), Panicle length (0.84 and 3.83), Grain length (0.62 and 0.78), Secondary branches panicle-1 (0.66 and 6.19), Spikelets panicle-1 (0.59 and 21.82), Grains panicle-1 (0.75 and 15.82), 100 grain weight (0.76 and 0.31) and Grain yield plant-1 (0.95 and 13.34). On the basis of superiority for yield attributing traits Pakhal and NIAB-IR-8 could be recommended for future use in rice breeding programs. J. Biodivers. Conserv. Bioresour.
