Abstract The sand tampan, Ornithodoros savignyi (Audouin, 1827), is an economically important soft tick of the Afrotropics parasitising a wide range of livestock and humans. These ticks are known to inflict painful bites which may be fatal in susceptible hosts. Historically thought to be a single species, Ornithodoros savignyi is now considered to be a complex of four tick subspecies based on molecular and morphological studies. They include Ornithodoros (Ornithodoros) kalahariensis, O. (O.) pavimentosus, O. (O.) noorsveldensis and O. (O.) savignyi. As such there may be significant implications for previous biological studies conducted on this tick. Therefore, for the purposes of this review, sand tampan toxicosis and potentially useful biological molecules have been discussed for O. (O.) savignyi sensu lato since most reported work was based on ticks collected from the Kalahari and Lake Chad region. An overview of the host range and vector biology for the O. (O.) savignyi species complex will also be examined.

Abstract Background In free-ranging reptile populations, bacterial, fungal, viral and parasitic pathogens may affect hosts through impairment in movements, thermoregulation, reproduction, survival, and population dynamics. The speckled dwarf tortoise (Chersobius [Homopus] signatus) is a threatened species that is mostly restricted to the Succulent Karoo biome in South Africa, and little information on pathogens of this species is available yet. We derived baseline parameters for five males and five females that were captured to genetically enhance a conservation breeding program in Europe. Upon collection of the tortoises, ticks were removed and identified. Immediately upon arrival in Europe, ocular, nasal, oral and cloacal swabs were taken for viral, bacteriological and mycological examinations. Fecal samples were collected before and 1 month after fenbendazole treatment, and analyzed for parasites. A panel of PCR, aiming to detect herpesviruses, adenoviruses and iridoviruses, was carried out. Results Samples were negative for viruses, while bacteriological examination yielded detectable growth in 82.5% of the swabs with a mean load of 16 × 107 ± 61 × 108 colony forming units (CFU) per swab, representing 34 bacterial species. Cloacal and oral swabs yielded higher detectable growth loads than nasal and ocular swabs, but no differences between sexes were observed. Fungi and yeasts (mean load 5 × 103 ± 13 × 103 CFU/swab) were detected in 25% of the swabs. All pre-treatment fecal samples were positive for oxyurid eggs, ranging from 200 to 2400 eggs per gram of feces, whereas after the treatment a significantly reduced egg count (90–100% reduction) was found in seven out of 10 individuals. One remaining individual showed 29% reduction, and two others had increased egg counts. In five tortoises, Nycthocterus spp. and coccidian oocysts were also identified. Soft ticks were identified as Ornithodoros savignyi. Conclusions Our baseline data from clinically healthy individuals will help future studies to interpret prevalences of microorganisms in speckled dwarf tortoise populations. The study population did not appear immediately threatened by current parasite presence.

Background and Aim: The soft tick Ornithodoros savignyi is distributed throughout Africa, including Egypt. It primarily attacks camels, cattle, donkeys, and cows; and rarely affects humans. This study evaluated the acaricidal efficacy of ethanolic Curcuma longa extract (Turmeric) on the second nymphs of O. savignyi and then investigated the safety of this herb in rabbits. Materials and Methods: The nymphs were immersed in 10, 5, 2.5, 1.25, and 0.625 mg/ml ethanolic C. longa extract. An additional group was immersed in ethanol as a control. On the 1st, 7th, and 15th-day post-treatment, the mortality percentages, LC50 and LC95 were calculated. The ticks exposed to 10 mg/ml ethanol C. longa extract were investigated by scanning electron microscopy (SEM). Three male New Zealand White rabbits were orally administered 2 ml (two doses) of 10 mg/ml ethanolic C. longa extract, and another three rabbits were orally given two doses of 2 ml of absolute ethanol as a negative control. Histopathological examination of the kidney and liver hematology and the kidney and liver function was performed. Chemical analysis of the extract was determined by gas chromatography-mass spectrometry (GC-MS). Results: The LC50 and LC95 were 1.31 and 15.07, 1.07 and 8.56, and 0.81 and 6.97 mg/ml on the 1st, 7th, and 15th day, respectively. SEM revealed that mamillae and spots on the surfaces of the treated ticks were not discriminating except for some clefts on the surfaces. The histological examination, blood profile, and biochemical analyses revealed no significant differences between the treated and untreated rabbits (p>0.05). GC/MS analysis revealed 50 compounds, and curcumene and t umerone were found to be the major constituents of this ethanolic extract. Conclusion: The ethanolic C. longa extract produced a strong acaricidal effect on the second nymph of O. savignyi, and it was safe to use in rabbits.

ABSTRACT Endemic tick-borne relapsing fever (TBRF) has not been documented in Nigeria, yet clinically compatible cases have been described, and soft tick species are endemic in surrounding countries. Consequently, our aim was to investigate if TBRF-associated Borrelia is present in Nigeria. To address this, we examined 49 soft tick pools to identify the tick species and to screen for Borrelia. The tick species was revealed by 16S rRNA gene amplification and Sanger sequencing to be Ornithodoros savignyi, an aggressive, multihost, rapidly feeding species with significant veterinary impact. We detected a Borrelia organism in 3 of 49 pooled samples (6%). Molecular analysis of amplified 16S rRNA and flagellin genes and intragenic spacer fragments disclosed that this Borrelia organism was synonymous with the recently described organism “Candidatus Borrelia kalaharica,” identified in a tourist returning to Germany from South Africa. Given the widespread endemic range of this tick vector, TBRF should be considered part of the differential diagnosis for patients with fever returning from arid areas of Africa and further afield.

The distributions of endemic tick vector species as well as the presence of species not endemic to Free State Province, South Africa, were determined during surveys or opportunistic collections from livestock, wildlife and vegetation. Amongst endemic ticks, the presence of Rhipicephalus appendiculatus was confirmed in the north of the province, whilst Rhipicephalus decoloratus was collected at 31 localities mostly in the centre and east, and Ixodes rubicundus at 11 localities in the south, south-west and centre of the province. Amongst the non-endemic species adult Amblyomma hebraeum were collected from white rhinoceroses (Ceratotherium simum) on four privately owned farms, whilst the adults of Rhipicephalus microplus were collected from cattle and a larva from vegetation at four localities in the east of the province. The collection of Rhipicephalus evertsi mimeticus from a sheep in the west of the province is the second record of its presence in the Free State, whereas the presence of Haemaphysalis silacea on helmeted guineafowl (Numida meleagris) and vegetation in the centre of the province represents a first record for this species in the Free State. The first collection of the argasid tick, Ornithodoros savignyi, in the Free State was made from a domestic cow and from soil in the west of the province. The localities at which the ticks were collected have been plotted and the ticks’ role in the transmission or cause of disease in domestic livestock and wildlife is discussed.
