Abstract The effects of pure oxygen aeration on compositional characteristics of soluble microbial products (SMP) and extracellular polymeric substances (EPS) of the activated sludge acclimated in a sequential batch reactor (SBR) were explored in batch mode. The structure of the extracellular products would change with different aeration methods or aeration rates. The proportion of SMP of most oxygen aerated sludge was less than 10%, while that in air aerated sludge was as high as 30%–40%. The proportion of TB-EPS decreased from 56.95% to 30.63%, and the proportion of LB-EPS increased obviously with the increase of oxygen aeration rate. The contents of the protein (PN) and the polysaccharide (PS) of extracellular products with oxygen aeration were significantly different, and the PN was much higher than PS (p < 0.05). The zeta potential of each component in activated sludge was negative, gradually decreasing with the progress of biological treatment. The fluorescence peaks in SMP, LB-EPS and TB-EPS with pure oxygen aeration were attributed to tryptophan PN-like and humic acid-like fractions. The results showed that the proportion of the components in the extracellular products could be regulated by adjusting the aeration rate and aeration mode, so as to optimize the treatment process of activated sludge.

Dendritic cells (DC) are professional antigen-presenting cells that activate T cells to kill cancer cells. The extracellular products of DCs have also been reported to perform the same function. In this study, we examined the in vitro differentiation of umbilical cord blood monocytes into DCs in the presence of GM-CSF, and interferon (IFN)-α. The resulting DC population (called IFN-DCs) were then matured in the presence of TNF-α, and pulsed with total protein extracted from A549 cancer cell line. The pulsed DCs and their conditioned medium were then used to stimulate allogeneic lymphocytes (alloLym). The proliferation and cytotoxicity of alloLym were then determined. The results showed that after 5 days of differentiation, the stimulated monocytes had the typical morphology and characteristic surface markers of DCs. Both unpulsed and pulsed IFN-DCs can induce the proliferation of alloLym, especially Vγ9γδ T cells. The conditioned medium from pulsed and unpulsed IFN-DCs culture also prompted the growth of Vγ9γδ T cells. Moreover, alloLym stimulated with pulsed DCs and their conditioned medium had a greater cytotoxic effect on A549 cells than the ones that were not stimulated. Our results indicated that IFN-DCs and their conditioned medium could induce the anti-tumor immunity in vitro, providing evidence for application of cord blood monocytes-derived, interferon-α- stimulated dendritic cells and their extracellular products in anti-cancer therapy.

Some strains of Pseudomonas corrugata (Pco) and P. mediterranea (Pme) efficiently synthesize medium-chain-length polyhydroxyalkanoates elastomers (mcl-PHA) and extracellular products on related and unrelated carbon sources. Yield and composition are dependent on the strain, carbon source, fermentation process, and any additives. Selected Pco strains produce amorphous and sticky mcl-PHA, whereas strains of Pme produce, on high grade and partially refined biodiesel glycerol, a distinctive filmable PHA, very different from the conventional microbial mcl-PHA, suitable for making blends with polylactide acid. However, the yields still need to be improved and production costs lowered. An integrated process has been developed to recover intracellular mcl-PHA and extracellular bioactive molecules. Transcriptional regulation studies during PHA production contribute to understanding the metabolic potential of Pco and Pme strains. Data available suggest that pha biosynthesis genes and their regulations will be helpful to develop new, integrated strategies for cost-effective production.
