The aromatic species Piper gaudichaudianum Kunth (Piperaceae) is widely used in Brazil for medicinal and ritualistic applications. In the current study, chemophenetic patterns were realized across season and circadian rhythm based on the chemical profile of essential oils (EOs) from leaves. Hydrodistilled essential oils were analyzed by GC-MS and GC-FID, and a new calculation of metabolite oxidation level, averaged for each individual molecule component of the EO, was used to explore the patterns of metabolism/biosynthesis. This new index used an intermediate calculation, the ‘weighted average redox standard’ (SRO), to enable a value for mixtures of metabolites to be generated, the ‘general mixture redox index’ (GMOR). The indices were subjected to a proof-of-concept approach by making comparison to outcomes from multivariate analyses, i.e., PCA and HCA. Chemical analysis demonstrated that the essential oils were dominated by sesquiterpenes, constructed of 15 classes of compound (C-skeletons), and 4 C-skeletons were recognized in the monoterpene group, giving a total of 19. The variation of chemical profiles was distinct at different phenological stages, but stronger chemical variation was evident between day and night as compared to season. Furthermore, due to comprehensive sampling across different regions, nine chemotypes were recognized, including those previously reported. The SRO and GMRO indices demonstrate that phenological variation of chemistry is mainly an outcome of redox fluctuations in terpene biosynthesis, changing from day to night. These indices also corroborate that chemical diversity is increased with oxidative metabolism. Lastly, the current study demonstrates pronounced phenotypic plasticity in P. gaudichaudianum, which makes it a suitable candidate to help further our understanding of chemophenetics and chemical ecology.

Abstract Piper gaudichaudianum Kunth (Piperaceae) is widely used in Brazil as medicinal and ritualistic. In this study, chemophenetic patterns were evaluated based on leaves’ essential oils (EOs) chemical composition. Several collections were performed to accomplish circadian rhythm and seasonal studies. Besides, a predictive methodology was developed and submitted to Proof of Concept (PoC) to determine the metabolism pattern and evaluate the reduction-oxidation of complex mixtures: Weighted Average Redox Standard (SRO) and General Mixture Redox Index (GMOR). Fresh leaves EOs obtained by hydrodistillation were analyzed by GC-MS and GC-FID. The main identified compounds were sesquiterpenes. Nineteen terpene skeletons were registered. There was chemical composition variation at different phenological stages. EOs varied more between day and night than seasonally. Nine chemotypes are proposed based on our results and those from literature. SRO and GMRO analyzes highlighted a possible redox balance throughout day and night. Compounds per carbon skeleton diversification in EOs are matched by an increase in compounds SRO. We also report for the first time high chemical phenotype plasticity based on EOs analysis and its implications for P. gaudichaudianum chemophenetics, chemosystematics and ecology.

The chemical composition of seedlings and adult plants of several Piper species were analyzed by 1H NMR spectroscopy combined with principal component analysis (PCA) and HPLC-DAD, HPLC-HRESIMS and GC-MS data. The chromatographic profile of crude extracts from leaves of Piper species showed remarkable differences between seedlings and adult plants. Adult leaves of P. regnellii accumulate dihydrobenzofuran neolignans, P. solmsianum contain tetrahydrofuran lignans, and prenylated benzoic acids are found in adult leaves of P. hemmendorffii and P. caldense. Seedlings produced an entirely different collection of compounds. Piper gaudichaudianum and P. solmsianum seedlings contain the phenylpropanoid dillapiole. Piper regnellii and P. hemmendorffii produce another phenylpropanoid, apiol, while isoasarone is found in P. caldense. Piper richadiaefolium and P. permucronatum contain dibenzylbutyrolactones lignans or flavonoids in adult leaves. Seedlings of P. richardiaefolium produce multiple amides, while P. permucronatum seedlings contain a new long chain ester. Piper tuberculatum, P. reticulatum and P. amalago produce amides, and their chemistry changes less during ontogeny. The chemical variation we documented opens questions about changes in herbivore pressure across ontogeny.

AbstractThe objective of this study was to determine the chemical profile and to evaluate the antibacterial activity of the essential oils of Piper species and modulation of the antibiotic activity, using the microdilution method to determine the minimum inhibitory concentration. The chemical components were characterized by gas chromatography coupled to mass spectrometry, which revealed β-copaen-4-α-ol (31.38%), spathulenol (25.92%), and germacrene B (21.53%) as major constituents of the essential oils of Piper arboreum, Piper aduncum, and Piper gaudichaudianum, respectively. The essential oils analyzed in this study did not present a clinically relevant activity against standard and multiresistant Escherichia coli. However, in the case of multiresistant Staphylococcus aureus, there was a significant activity, corroborating with reports in the literature, where Gram-positive bacteria are more susceptible to antimicrobial activity. The essential oils modulated the effect of the antibiotics norfloxacin and gentamicin, having on the latter greater modulating effect; however, for erythromycin, no statistically significant effect was observed. In conclusion, the results obtained in this study demonstrated that the essential oils of the analyzed Piper species present an inhibitory effect against S. aureus and modulate antibiotic activity, most of which presents synergistic activity.

Piper gaudichaudianum Kunth (Piperaceae) é uma planta com ampla distribuição no território brasileiro. Além de sua importância ecológica, é utilizada na medicina popular e muitas de suas atividades biológicas já foram comprovadas. O objetivo deste trabalho foi verificar o efeito da sazonalidade sobre o teor de óleo essencial (OE) em folhas de Piper gaudichaudianum e averiguar a possível influência do processo de secagem do material vegetal no rendimento deste extrativo. Foram coletadas aleatoriamente folhas de indivíduos de uma população nativa de Santa Maria - RS, Brasil, ao longo de um ano. O OE foi extraído de folhas frescas e secas à temperatura ambiente pelo método de hidrodestilação. Quando verificada a presença de órgãos reprodutivos, os mesmos foram extraídos, a fim de examinar a sua influência no rendimento do OE de folhas. Para determinação do teor de umidade (TU%), 20 g de folhas e 10 g de órgãos reprodutivos foram secos separadamente em estufa a 50°C até peso constante. O rendimento de OE de folhas (% m/m) foi calculado para o material fresco com base na massa seca, considerando o TU%, bem como para o material seco à temperatura ambiente. Apenas pequenas variações sazonais nos teores de OE foram detectadas para as amostras analisadas, não havendo diferença estatística entre eles. O maior rendimento foi observado na primavera, tanto para o OE de folhas (1,61±0,09%), quanto de órgãos reprodutivos (5,17%), enquanto o menor teor de OE em folhas ocorreu no verão (1,32±0,14%), a única estação em que não houve a presença de órgãos reprodutivos. A ausência de diferença significativa entre os rendimentos indica que não houve variabilidade sazonal quantitativa na produção de OE em folhas desta população. Portanto, a coleta deste órgão vegetal pode ser realizada em qualquer época do ano visando à obtenção deste extrativo. Uma vez que não foi verificada diminuição do teor de OE durante o processo de secagem à temperatura ambiente, esse processamento pós-colheita poderá ser adotado.

ABSTRACT: This study evaluated the potential relaxing and/or molluscicidal effects of eugenol and essential oils of Origanum majorana, Ocimum americanum, Hesperozygis ringens, and Piper gaudichaudianum in the gastropod Pomacea canaliculata. Compounds were tested at concentrations of 100, 250, 500, and 750µL L-1 to evaluate the relaxing effects. In the second experiment, animals were exposed to 10, 25, and 50µL L-1 of essential oils of H. ringens and P. gaudichaudianum for a period of 24h for the evaluation of molluscicidal effects. Eugenol and essential oils of O. majorana and O. americanum showed relaxing effects at ≥250µL L-1, but the essential oils of H. ringens and P. gaudichaudianum did not promote relaxing or molluscicidal effects within the times and concentrations studied. Therefore, only eugenol and the essential oils of O. majorana and O. americanum can be used for relaxation purposes in P. canaliculata.
