The present study was to determine the phytochemical constituents, antioxidant, and anticancer potential of methanolic extract from the seeds of Trachyspermumammi on KB, human oral cancer cell line using in the vitro assay system. The seeds of the T. Ammi were extracted with methanol. Phytochemical screening was done using standard qualitative biochemical tests. The antioxidant activity of the T. Ammi extract was determined by using a phosphomolybdenum assay.MTT assay was used to estimate the antiproliferative activity of the extract against the human oral cancer (KB) cell line. The apoptotic inducing ability of the extract was investigated by intracellular ROS analysis, antioxidant enzyme levels (SOD, CAT, GPx, and GSH), EtBr/AO staining, and TBARS analysis. The qualitative phytochemical analysis revealed the presence of different secondary metabolites. The phytochemical content present in the methanolic extract possessed high antioxidant capacity with transition metal reduction in phosphomolybdate assay. The methanol seed extract was found to be cytotoxic in vitro to (KB) oral cancer cell lines with IC50 value125µg/ml. Significantly, the T. Ammi seed extract showed a broad-spectrum anti-cancer activity through promoting ROS generation via modulating apoptosis induction in KBoral cancer cells. From the results, it can be concluded that T. ammiseed extract was a promising antioxidant and anticancer agent for KB human oral cancer cell line. The results figure the sufficient scientific backgrounds to isolate and purify the bioactive compounds in future for further applications.

Background: Cancer is rapidly evolving life-threatening ailment in the mankind due to changes in daily food intake and lifestyle changes. Oral carcinoma is 6th major cause of cancer death in the world and it is third major reason of cancer mortality in India. Every cell in the human body requires glucose for its metabolic energy. Besides normal cell, cancer cells also require the glucose for its endurance and multiplication. SGLT2 inhibitors which are aimed at diabetes therapy exhibited anticancer properties also in colon and pancreatic cancer lines. Present study aim is to evaluate the anticancer activity of SGLT2 inhibitors against oral cancer cell by MTT Assay.Methods: To evaluate the anticancer activity of SGLT2 inhibitors MTT Cytotoxic assay is performed as per standard protocols. Cancer cells were plated in 24-well plates and incubated at 370C with 5% CO2 condition. After convergence, samples are added to the plates in various concentrations and allowed to incubate then they are detached from the plates and cleansed with the reagents. The wells are coated with the dye and incubated. Later samples are analysed in UV-spectrophotometer.Results: Cytotoxic assay showed decrease in cell viability with increasing dose of SGLT2 inhibitors. IC50 values were determined graphically. The IC50 value of dapagliflozin is 400µg/ml and canagliflozin is 250µg/ml respectively after 24 hours of Assessment.Conclusions: The results of the current study give us an evidence that SGLT2 inhibitors dapagliflozin and canagliflozin exhibits anticancer property in Oral Cancer cell line.

Objective: Berries including strawberry may have beneficial effects against oxidative stress-mediated diseases such as cancer. The purpose of the present study was to investigate the cytotoxic effects of strawberry extract on oral cancer cell line.Methods: Strawberry methanolic extract (SBE) was prepared, and cytotoxic activity of different concentration of SBE on KB cell lines was determined by (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay and neutral red dye incorporation test.Results: Strawberry extract exhibits cytotoxic activity over the oral cancer cell lines. On administration of about 100 μg/ml of strawberry extract, about 50% of cell viability could be observed and assessed from the cell lines.Conclusion: Strawberries have a cytotoxic effect on oral cancer cell line due to the presence of anticancer constituents in the berries. These berries can be used as a natural medicine for cancer sufferers.

ABSTRACTAim: To assess the apoptotic induction potentials of Bacopa monnieri against oral cancer cell.Objective: This research is done to find the apoptotic induction potentials of B. monnieri against oral cancer cell line.Background: The apoptosis is a process of programed cell death that occurs in multicellular organisms. Biochemical events lead to characteristic cellchanges and death. The initiation of apoptosis is tightly regulated by activation mechanisms, because once apoptosis has begun, it inevitably leadsto the death of the cell. The two best-understood activation mechanisms are the intrinsic pathway (also called the mitochondrial pathway) and theextrinsic pathway.Methods: The apoptotic induction potential of B. monnieri was studied on KB cell line by investigating the activation of caspase 3 and caspase 9.Reason: Oral cancer is a growing concern due to the changes in lifestyle. The treatment for this disease has been degraded the patient’s health bcausing various side effects. An effective treatment for the long run is necessary. Usable of herbal plants can serve to be a possible and very effectivremedy.Results: Apoptotic induction potentials of B. monnieri against oral cancer cell line were studied.Conclusion: Caspase 3 and caspase 9 was activated thus proving the apoptotic potential of B. monnieri.Keywords: Bacopa monnieri, oral cancer, apoptotic induction.
