Bamboos seldom bloom and almost no seeds could be harvested, and, hence, few works are focused on germination physiology. Systematic research on the physiological effects of colchicine on germination and seedling growth of bamboo seeds is lacking. In this study, we finely recorded seed germination and seedling growth of Dendrocalamus brandisii in media supplemented with different colchicine concentrations. Physiological effects and mechanisms of colchicine were analyzed. The results showed that D. brandisii seeds were non-dormant, and seed lots achieved their highest germination rates on the 4th day and finished the whole germination period after 21 days. Colchicine inhibited seed germination and seedling growth but did not change its germination pattern. Seed germination and seedling growth decreased constantly with colchicine concentration. Colchicine showed more negative effects on seedling growth than on seed germination and root growth. High concentrations of colchicine retarded the development of plumules and even caused their aberrant development. Under tissue culture conditions, seed germination, and seedling growth relied mainly on the endogenous starch and soluble sugar degradation, in which α-amylase, STP, and SUSY played the key role. Colchicine inhibited seed germination and seedling growth by suppressing the α-amylase, STP, and SUSY activities. Colchicine showed more negative effects on sucrose degradation than on starch degradation during seed germination and seedling growth. This study provides new basic information on the seedling physiology for the genetic breeding of bamboo plants.

Anti-insect photoselective nets present a new technology that combines light manipulation and pest protection in orchards. In this study, the effect of the anti-insect photoselective red net on the generative and vegetative traits was studied in an apple orchard near the city of Zadar, Croatia. 'Granny Smith' apples were grown on M9 rootstock and raised as slender spindles. The experiment consisted of two treatments: the trees covered with the red photoselective anti-insect nets (AGRITECH S. r. l., Eboli, Italy; mesh size of 2.4 × 4.8 mm) and uncovered trees as control. Yield, percentage of fruit with diameter >70 mm and fruit mass were not significantly different between treatments. L* color value was higher on fruit skin grown under the red net, and there was no significant difference in other color values (a*, b*, C* and h°). Soluble solids concentration (SSC) and total flavonoid content were significantly lower in fruit grown under red net than in control, and there was no significant difference in titratable acidity (TA), SSC/TA ratio, starch degradation and Streif maturity index. Red net proved to be effective against fruit red blush development (undesirable trait for 'Granny Smith' apple) and sunburn damage occurrence.

Abstract Opisthopappus is a major wild source of Asteraceae with good cold and drought resistance. Two species of this genus (Opisthopappus taihangensis and Opisthopappus longilobus) have been employed as model systems to address the evolutionary history of perennial herb biomes in the Taihang Mountains of China. However, further studies on the adaptive divergence processes of these two species are currently impeded by the lack of genomic resources. To elucidate the molecular mechanisms involved, a comparative analysis of these two species was conducted. Among the identified transcription factors, the bHLH members were most prevalent, which exhibited significantly different expression levels in the terpenoid metabolic pathway. O. longilobus revealed a higher expression than did O. taihangensis in terms of terpenes biosynthesis and metabolism, particularly regarding monoterpenoids and diterpenoids. Analyses of the positive selection genes (PSGs) identified from O. taihangensis and O. longilobus, 1203 genes were found that related to adaptative divergence, which were under rapid evolution and/or have signs of positive selection. Different PSG expressions occurred primarily in the mitochondrial electron transport, starch degradation, secondary metabolism, as well as nucleotide synthesis and S-metabolism pathway processes. Two PSGs were obviously differentially expressed in terpenes biosynthesis that might result in the fragrances divergence between O. longilobus and O. taihangensis, which would provide insights as to how the two species adapted to different environments, characterized by sub-humid warm temperate and temperate continental monsoon climates. The comparative analysis for these two species of Opisthopappus not only revealed how the divergence occurred from molecular perspective, but also provided novel insights into how differential adaptations occurred in Taihang Mountains.

Abstract A laboratory-scale anaerobic moving bed biofilm reactor (AnMBBR) was installed and operated at various hydraulic retention times (HRTs) of 20 to 1.5 d with surface area loading rate (SALR) of 0.86 to 11.43 gCOD/m2/d. Synthetic starch containing desizing wastewater with chemical oxygen demand (COD) of 12.75 g/L was prepared and fed into the reactor. Monod, modified Stover-Kincannon, Grau second-order and First-order substrate removal models were used to evaluate the results of AnMBBR. COD removal efficiency of bioreactor was dwindled by increasing the SALR or reducing the HRT. Decay coefficient (Kd) and yield coefficient (Y) for Monod model were 0.027 1/d and 1.01 mgVSS/mgCOD, respectively. Maximum substrate utilization rate (Umax) and kinetic constant (Kb) for Modified Stover-Kincannon model were estimated as 12.57 and 15.22 g/L/d, respectively. The constants (a and b) for Grau second-order model were found to be 1.09 and 1.31 whilst kinetic coefficient for Second-order model and First-order substrate removal model were 1.62 and 1.55 1/d, respectively. Modified Stover-Kincannon model and Grau second-order model were found to be the best fit for experimental data with R2 value of 0.99. The findings suggest that these models can be applied to predict the behaviour of AnMBBR on various scales.

The weaning period is a stressful period for the gastrointestinal tract (GIT) of piglets. This work aims to evaluate the effects of the commercial polyphenol-based product GreenFIS® on: (1) GIT health and performance of 60 weaned piglets; (2) digestibility in 18 growing pigs. Three diets were tested: a control diet (C), C plus 2.5 g of GreenFIS®/kg C (T1), and C plus 5 g of GreenFIS®/kg C (T2). After the post-weaning trial three piglets per treatment were sacrificed for the GIT histological analysis. No differences between diets were recorded in terms of growing performance or clinical and biochemical blood parameters. The GIT histological analysis did not show any indicators of inflammation for any of the groups. The feces of the two extreme treatments (C and T2) were analyzed for microbiota, revealing a greater presence of the Ruminococcus bromii group, positively associated with starch degradation, in T2. In the second experiment six pigs per treatment were randomly chosen for the digestibility study. The inclusion of GreenFIS® at both levels led to a higher fecal digestibility of gross energy (86.2%, 89.1%, and 89.5%, for C, T1, and T2, respectively) and crude protein (87.0%, 90.2%, and 90.0%). In conclusion, the additive did not improve, in the excellent experimental hygienic conditions, the gut health, but it did increase nutrient digestibility.
