Four fistulated adult Murrah buffaloes were fed on a basal diet consisting of wheat straw and concentrate mixture in a 4 × 4 Latin square design to study the effects of feeding plants containing secondary metabolites on rumen metabolites and methanogen diversity. The four groups were Control (no additive), Mix-1 (ajwain oil and lemon grass oil in a 1 : 1 ratio @ 0.05% of dry matter intake), Mix-2 (garlic and soapnut in a 2 : 1 ratio @ 2% of dry matter intake) and Mix-3 (garlic, soapnut, harad and ajwain in a 2 : 1 : 1 : 1 ratio @ 1% of dry matter intake). In each phase of 30 days’ duration, after 19 days of feeding, rumen liquor was sampled for two consecutive days at 0, 2, 4, 6 and 8 h post-feeding, whereas rumen content was sampled at 0 h feeding. The pH of the rumen liquor was recorded at every collection and then the rumen liquor of every collection was pooled day-wise and animal-wise. These pooled samples were used for estimation of rumen metabolites like ammonia, lactic acid and volatile fatty acids. Microscopic counting of protozoa was done in both 0 h and pooled samples of rumen liquor. Rumen contents collected from different locations of rumen were processed for enzyme estimation. The rumen contents were squeezed and the liquid portion was used for DNA isolation, which was further processed to determine methanogen diversity. Daily intake of feed was similar (P > 0.05) in all the four groups. The ammonia-N concentration and ciliate protozoa population were reduced significantly in the treatment groups supplemented with additives. Rumen pH, lactic acid, volatile fatty acids and enzyme activities were not affected (P > 0.05) by feeding of any of these additives. Methanogenic diversity comparison was made between the Control and Mix-1 group. The basic local alignment search tool (BLAST) analysis of the 133 (44 from the Control group and 89 from the Mix-1 group) sequences showed similarity of the sequences of rumen archaea by up to 97% to the known sequences of rumen methanogens. The sequences with minimum length of 750 bp were selected for phylogenetic analysis. Per cent identity of these sequences with that of the available nearest neighbour as calculated by MEGA 5.03 software showed identity of the clones in the range of 88–97%. The clones were similar with Methanobrevibacter smithii ATCC 35061, uncultured Methanobrevibacter sp. clone MEME95 and M. ruminantium M1. Overall, feeding of any of these feed additives to fistulated buffaloes did not affect feed intake, rumen pH, or rumen metabolites except ammonia and enzyme profile. Methanogen diversity showed the possibility of Methanobrevibacter as the major methanogen in buffalo rumen liquor.

The interest in new alternative forages in Europe has increased in recent years. The nutritional composition and fermentation characteristics during different stages of ensiling were studied with Italian ryegrass (Lolium multiflorum Lam.) and winter cereal mixtures. The trial was carried out on a large-scale farm Galgamenti Agricultural Limited Company, Tura, Hungary. Two different forage mixes were studied: Mixture A (three types of Italian ryegrass 40% + two types of triticale 20%+ two types of oats 20%+ wheat 15%+ barley 5%) and Mixture B (three types of Italian ryegrass 55% + two types of winter oats 45%). Experimental field was 30.600 m 2 by the treatments, respectively. The two different forage mixes were sown on 11th September 2017 (Mixture A: 75 kg seed/ha; Mixture B: 75 kg seed/ha) with depth of 2-5 cm. Plant protection treatment was not applied during the growing season. Cutting was carried out in heading stage of triticale by hand at 10 cm stubble height. The fresh Mixture A (dry matter 189 g/kg; crude protein: 161 g/kg DM; NDF: 485 g/kg DM) and the fresh Mixture B (dry matter 195 g/kg; crude protein: 159 g/kg DM; NDF: 519 g/kg DM) were wilted to 28-32% DM (24h) without any movement on the windrow. The wilted forage was picked up by hand and chopped by a forage harvester (John Deere 7300) on concrete surface with theoretical chop length of 9 mm (weight: 800 kg). Wilted and chopped material of 510 g were packed by hand into a glass jars (0.00072 m3 volume, n=5, total no. of minisilos = 15). Five laboratory silos per mixtures were opened on 7, 14 and 90 days after ensiling. Dry matter (DM), crude protein (CP), crude fiber (CF), neutral detergent fiber (NDF), acid detergent fiber (ADF), acid detergent lignin (ADL), ether extract (EE), ash, and total sugar of all treatments were determined. Additionally, pH and the concentration of ammonia-N, volatile fatty acids were measured in the ensiled mixtures. At the end of 90 days of ensiling in both mixture silages, there were significant differences (p<0.05) in all nutrient contents except for ADF in Mixture A and CP, ash and CF in Mixture B which was not affected by fermentation duration. Ensiling caused a significant decrease in pH (p<0.05), due to the production of lactic acid and succeeded to achieve lactic acid type fermentation. Values for ammonia-N, ethanol and acetic acid, butyric acid were all low. These results indicated that the fermentation quality of Italian ryegrass and winter cereal silages underwent rapid fermentation and were well-preserved.

1. Propane-1:2-diol (loog/d) was infused through a cannula into the rumen of sheep receiving a ration of hay and dried grass. The concentration of volatile fatty acids, propanediol, lactic acid and of added polyethylene glycol, and the pH of the rumen contents were measured. The energy metabolism of the sheep was also determined.2. Most of the propanediol disappeared from the rumen within 4 h of its infusion. The infusion of propanediol resulted in a 10% decrease in the concentration of total volatile acids; the concentration of acetic acid decreased by about 30%, that of propionic acid increased by up to 60% and there was no change in the concentration of butyric acid.3. The methane production of the sheep decreased by about 9% after the infusion of propanediol and there were increases in the oxgyen consumption, carbon dioxide production and heat production of the animals; each of these increases was equivalent to about 40% of the theoretical value for the complete metabolism of 100 g propanediol.4. It is concluded that, when propanediol is introduced into the rumen, a proportion is metabolized in the rumen and a large proportion is absorbed directly. Our thanks are due to Dr J. H. Nine lambs fitted with rumen cannulae were used in two 5 × 5 Latin squares to measure the effect of alfalfa hay, C4- and C5-branched- and straight-chain volatile fatty acids (C4/C5 VFAs), cell wall from alfalfa hay (ALFCW) and a soybean protein preparation on intake and rumen digestion of a basal corncob diet with urea included at 30 g kg−1. Relative to the control, alfalfa hay decreased (P < 0.05) intake of the basal diet but increased (P < 0.05) total dry matter intake. ALFCW and soybean protein did not affect intake of the basal diet and total dry matter (P > 0.05). C4/C5 VFAs increased intake of basal diet by sheep in one square (P < 0.05) but had no effect (P > 0.05) on intake with sheep in the other square. Alfalfa hay, ALFCW and soybean protein increased (P < 0.05) total VFA concentration in rumen fluid while only alfalfa hay and C4/C5 VFAs consistently increased isobutyrate, isovalerate and valerate levels in rumen fluid (P < 0.05). In comparison with the control diet, soybean protein elevated (P < 0.05) mean rumen ammonia-N levels whereas C4/C5 VFAs depressed them; however, values exceeded 17 mg dL−1 rumen fluid on all treatments. Most measures of digestion kinetics of fiber from corncobs were unaffected by supplementation (P > 0.05). Alfalfa hay and ALFCW increased (P < 0.05) rate of passage of digesta from the reticulo-rumen. It was concluded that alfalfa hay has a reduced role in improving intake of corncob diets when there is also sufficient NPN in the diet. The pentose sugar l-arabinose is one of the most abundant components released by complete hydrolysis of non-starch polysaccharides of feed ingredients of vegetable origin. Two studies were conducted to investigate the apparent ileal digestibility and urinary excretion of l-arabinose at dietary inclusion levels of 50 and 100 g/kg, and 25, 50, 75 and 100 g/kg respectively, in pigs. As a reference, d-glucose was included in the studies. Water intake, ileal flow of volatile fatty acids and ileal and faecal digestibilities of dietary nutrients in pigs fed on the different diets were also examined. Castrated pigs were prepared with a post-valvular T-caecum cannula to measure ileal digestibility. Faecal digestibility was measured in non-cannulated pigs. Apparent ileal digestibility of l-arabinose was found to be approximately 70%. The presence of l-arabinose in the diet increased ileal flow of volatile fatty acids and lactic acid, suggesting the occurrence of microbial degradation of l-arabinose in the pig small intestine. l-arabinose was partly excreted in the urine. The extent of this urinary excretion as a percentage of intake increased linearly (P < 0.01) as the dietary level increased. In pigs fed on the 25 g l-arabinose/kg diet, 10.9% of the l-arabinose consumed appeared in the urine. This level was increased to 14.7% when pigs were fed on a diet containing 100 g l-arabinose/kg diet. Faecal digestibility and retention of nitrogen decreased significantly in pigs fed on the l-arabinose diets.

ABSTRACTA model has been developed to predict the true metabolizable energy (ME,) concentration in forages given to ruminants.The chemical description of forages is based on the concentrations of ash, crude protein, ether extract, sugars and α-glycans, β-glycans and lignin, volatile fatty acids and lactic acid. The model assumes complete fermentation of sugars, α-glycans and lactic acid. The extent of fermentation of β-glycans and crude protein is determined in part by rumen solid-phase outflow rate, which can itself be predicted from dry-matter intake.The model was tested using 121 graminaceous forages whose chemical composition and concentration of metabolizable energy had been measured in the Feedingstuffs Evaluation Unit at the Rowett Research Institute. The agreement between observed and predicted ME, for all classes of forage was nearly always as good as, or better than, the best prediction from single attributes of food chemistry which could only be determined retrospectively and were not consistent even within classes of forage. The model predicts a decline in ME, with increasing rumen solid-phase outflow rate which is determined mainly by the extent of fermentation of β-glycans.
