Protective effect of the hydro-alcoholic extract of Rubia cordifolia roots against ethylene glycol induced urolithiasis in rats

Abstract Nephrolithiasis appear to be a major health crisis among the population with serious medical related consequences throughout the lifetime of patient. The aim of the study was to evaluate the preventive effect of the hydro-alcoholic extract of A. lanata roots on Urolithiasis rats. Thirty adults male wistar Albino rats weighing 200 – 250 g were divided into five groups comprising 6 rats in each. Group I Served as positive control with water ad libitum. Group II as negative control which is disease treated group receiving 0.75% ethylene glycol mixed with drinking water for 28 days. Group III chosen as standard group receiving ethylene glycol for first 14 days and Cystone 750 mg/Kg from day 15 till day 28. Group IV and V received ethylene glycol for first 14 days and treatment regimen of LD (400 mg/Kg) and HD 800 mg/Kg orally from day 15 till day 28. Invitro studies like Nucleation, Aggregation and Growth assays were performed. Urine volume and pH was collected and observed for change in appearance, pH, odour and turbidity. Extract was given by preparing suspension and stability was observed by measuring its parameters. On Day 29, the kidneys were dissected and histopathology was done to check tubular injury. There was Increase in urine volume, enhanced excretion of urinary constituents like citrate, oxalate etc. and improving clearance rate. Improvement in pH and antioxidant activity was observed in treated groups. The extract showed that it has prominent effect on nephrolithiasis and has better safety profile in the dose given.

The efficacy of a crude hydro-alcoholic extract of Cassia fistula (golden shower tree) fruit to protect the kidney against bromobenzene-induced toxicity was studied. Negative control mice received normal saline; positive control mice were given 460 mg/kg of bromobenzene; Cassia fistula treated mice received 200, 400, 600 and 800 mg/kg of Cassia fistula fruit extract followed by 460 mg/kg bromobenzene (daily by oral gavage for 10 days). On the 11th day, the mice were sacrificed, blood samples were obtained to assess blood urea nitrogen (BUN) and creatinine levels, and kidneys were removed for histological examination. We found that bromobenzene induced significant nephrotoxicity reflected by an increase in levels of BUN and creatinine that was dose dependently prevented by the Cassia fistula fruit extract. The nephroprotective effect of the Cassia fistula fruit extract was confirmed by the histological examination of the kidneys. To the best of our knowledge, this is the first study to demonstrate the protective effect of Cassia fistula in nephrotoxicity.

The present study investigated the protective effect of kolaviron, a biflavonoid from the seed of Garcinia kola, on ethylene glycol monoethyl ether (EGEE)–induced reproductive toxicity in male rats. The protective effect of kolaviron was validated using vitamin E, a standard antioxidant. EGEE was administered at a dose of 200 mg/kg. Other groups of rats were simultaneously treated with kolaviron (100 and 200 mg/kg) and vitamin E (50 mg/kg) for 14 days. EGEE treatment resulted in significant decrease in glutathione (GSH) level, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) activities but markedly increased the glutathione-S-transferase (GST) and lactate dehydrogenase (LDH) activities in the testes. In the spermatozoa, administration of EGEE caused significant decrease in the activities of CAT, GPx, GST and LDH as well as in the level of GSH but significantly increased SOD activity with concomitant increase in hydrogen peroxide and malondialdehyde levels in both testes and spermatozoa. EGEE-exposed rats showed marked testicular degeneration with concomitant decrease in spermatozoa quantity and quality. Overall, EGEE causes reproductive dysfunction in rats by altering antioxidant systems in the testes and spermatozoa. Kolaviron or vitamin E exhibited protective effects against EGEE-induced male reproductive toxicity by enhancement of antioxidant status and improvement in spermatozoa quantity and quality.
