Rapid and effective evaluation of the antioxidant capacity of propolis extracts using DPPH bleaching kinetic profiles, FT-IR and UV–vis spectroscopic data

Cannabis sativa L. (hemp) is a plant used in the textile industry and green building material industry, as well as for the phytoremediation of soil, medical treatments, and supplementary food products. The synergistic effect of terpenes, flavonoids, and cannabinoids in hemp extracts may mediate the biogenic synthesis of metal nanoparticles. In this study, the chemical composition of aqueous leaf extracts of three varieties of Romanian hemp (two monoecious, and one dioecious) have been determined by Fourier-Transformed Infrared spectroscopy (FT-IR), high-performance liquid chromatography, and mass spectrometry (UHPLC-DAD-MS). Then, their capability to mediate the green synthesis of silver nanoparticles (AgNPs) and their pottential antibacterial applications were evaluated. The average antioxidant capacity of the extracts had 18.4 ± 3.9% inhibition determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH•) and 78.2 ± 4.1% determined by 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS™) assays. The total polyphenolic content of the extracts was 1642 ± 32 mg gallic acid equivalent (GAE) L−1. After this, these extracts were reacted with an aqueous solution of AgNO3 resulting in AgNPs, which were characterized by UV−VIS spectroscopy, FT-IR, scanning electron microscopy (SEM-EDX), and dynamic light scattering (DLS). The results demonstrated obtaining spherical, stable AgNPs with a diameter of less than 69 nm and an absorbance peak at 435 nm. The mixture of extracts and AgNPs showed a superior antioxidant capacity of 2.3 ± 0.4% inhibition determined by the DPPH• assay, 88.5 ± 0.9% inhibition as determined by the ABTS•+ assay, and a good antibacterial activity against several human pathogens: Escherichia coli, Klebsiella pneumoniae, Pseudomonas fluorescens, and Staphylococcus aureus.

The 4-(?-bromo acetyl)-4?-toluene sulfonanilide (2) was used as key intermediate to synthesize new heterocyclic compounds. This bromo compound was synthesized via sulfonation of amino group of p-amino acetophenone using Hinsburg method with 4-toluene sulfonyl chloride to form 4-acetyl-4?-toluene sulfonanilide (1) which is used as a starting material in this work. This compound was brominated to yield compound (2) which is used as a precursor to synthesize new five and seven membered heterocyclic compounds such as substituted 1,3-oxazoles (3,4), 1,3-thiazole derivatives (5-7), thiourea compounds (8a,b), 1,3-Thiazoline-2-thione compounds (9a-f) and 1,2,5-triazepine compounds (11a-d). The synthesized compounds were identified depending upon physical, FT-IR and UV spectroscopic data.

Fourier transform infrared (FT-IR) spectroscopy is a physicochemical technique based on the vibrations of a molecule energized by infrared radiation at a specific wavelength range. Abiotic stresses can induce the production of secondary metabolites, increasing bioactivity. The objectives of the study were to evaluate the impact of heat treatments on the bioactivity of pineapple by-products, and whether FT-IR analysis allows understanding of the changes imparted by abiotic stress. The by-products were treated at 30, 40, and 50 °C for 15 min, followed by storage at 5 ± 1 °C for 8 and 24 h. Lyophilized samples were characterized for total phenolic content and antioxidant capacity and analyzed by FT-IR. Thermal treatments at 50 °C reduced the content of phenolic compounds (21–24%) and antioxidant capacity (20–55%). Longer storage time (24 h) was advantageous for the shell samples, although this effect was not demonstrated for the core samples. The principal components analysis (PCA) model developed with the spectra of the pineapple shell samples showed that the samples were grouped according to their total phenolic compounds content. These results allow the conclusion to be drawn that FT-IR spectroscopy is a promising alternative to the conventional chemical analytical methodologies for phenolic and antioxidant contents if there are significant differences among samples.

The comparison of the antioxidant activity of the studied forest fruits emphasized a hierarchy of the antioxidant capacity in rosehip, blackthorn, lingonberry and cornelian cherry. The purpose of the study was to investigate the antioxidant capacity and nutritional value of rosehip, blackthorn, lingonberry and cornelian cherry. In the current study, the FT-IR spectroscopy technique was applied to detect molecular components in forest fruits samples. Antioxidant capacity was evaluated with photochemical assay as well as humidity, protein, fibre, lipid and carbohydrate content. The FT-IR results revealed the presence of different bio-active compounds in berries such as flavonoids, tannins, sugars, acids, proanthocyanidins, carotenoids, citric metabolites and others. The highest antioxidant capacity was observed in rosehip 105.67±1.38 and blackthorn 49.89±1.92 (μg/mg equivalent ascorbic acid). Regarding nutritional parameters, rosehip showed the most increased content of protein displaying average values of 1.60, carbohydrates 38.20 and fibre 24.10 (g/100 g/sample). These results can provide useful information providing a research interest for the identification of new molecular compounds from Romanian flora samples.   ********* In press - Online First. Deoxyphomalone (1), dimethyl 4-methyl-2,6-pyridinedicarboxylate (2), stemphyperylenol (3), and N-methyl-2-pyrrolidone (4) were isolated from the fermentation broth of a strain of the fungus, Alternaria tenuissima. This fungus was isolated from the soil underlying the lichen, Peltigera didactyla, which had been collected from Wapusk National Park in Northern Manitoba. The structures of the compounds were determined by comprehensive analysis of their spectroscopic data including FT-IR, 1D and 2D NMR spectroscopy and mass spectrometry; and their bioactivities were tested against E.coli cells. The taxonomic identity of the fungus was confirmed by ITS sequencing of its ribosomal DNA.

Saliva provides a valuable tool for assessing oral and systemic diseases, but concentrations of salivary components are very small, calling the need for precise analysis methods. In this work, Fourier transform infrared (FT-IR) spectroscopy using transmission and photoacoustic (PA) modes were compared for quantitative analysis of saliva. The performance of these techniques was compared with a calibration series. The linearity of spectrum output was verified by using albumin–thiocyanate (SCN–) solution at different SCN– concentrations. Saliva samples used as a comparison were obtained from healthy subjects. Saliva droplets of 15 µL were applied on the silicon sample substrate, 6 drops for each specimen, and dried at 37 ℃ overnight. The measurements were carried out using an FT-IR spectrometer in conjunction with an accessory unit for PA measurements. The findings with both transmission and PA modes mirror each other. The major bands presented were 1500–1750 cm–1 for proteins and 1050–1200 cm–1 for carbohydrates. In addition, the distinct spectral band at 2050 cm–1 derives from SCN– anions, which is converted by salivary peroxidases to hypothiocyanate (OSCN–). The correlation between the spectroscopic data with SCN– concentration (r > 0.990 for transmission and r = 0.967 for PA mode) was found to be significant ( P < 0.01), thus promising to be utilized in future applications.
