Abstract. In the development of new psychiatric drugs and the exploration of their efficacy, behavioral testing in mice has always shown to be an inevitable procedure. By studying the behavior of mice, diverse pathophysiological processes leading to depression, anxiety, and sickness behavior have been revealed. Moreover, laboratory research in animals increased at least the knowledge about the involvement of a multitude of genes in anxiety and depression. However, multiple new possibilities to study human behavior have been developed recently and improved and enable a direct acquisition of human epigenetic, imaging, and neurotransmission data on psychiatric pathologies. In human beings, the high influence of environmental and resilience factors gained scientific importance during the last years as the search for key genes in the development of affective and anxiety disorders has not been successful. However, environmental influences in human beings themselves might be better understood and controllable than in mice, where environmental influences might be as complex and subtle. The increasing possibilities in clinical research and the knowledge about the complexity of environmental influences and interferences in animal trials, which had been underestimated yet, question more and more to what extent findings from laboratory animal research translate to human conditions. However, new developments in behavioral testing of mice involve the animals’ welfare and show that housing conditions of laboratory mice can be markedly improved without affecting the standardization of results.

ZusammenfassungEine Immunaktivierung geht häufig mit Verhaltensänderungen (sickness behavior) einher, die Teil der unspezifischen Immunantwort sind, über inflammatorische Zytokine gesteuert werden und durch veränderte Priorisierung von Aktivitäten zur erfolgreichen Immunabwehr beitragen. Bei chronischer Immunaktivität kann sich aus diesen Verhaltensänderungen ein depressives Syndrom entwickeln. Dieser Zusammenhang wurde anhand humaner Modelle (immunaktivierende Behandlung mit Interferon-alpha) bestätigt. Umgekehrt finden sich erhöhte Immunparameter auch bei primär depressiven Patienten und Studien zeigten antidepressive Effekte einer anti-inflammatorischen Add-on-Therapie. Psychischer Stress, der eine Aktivierung des peripheren Immunsystems bewirkt und Hauptrisikofaktor für die Entwicklung einer depressiven Episode ist, könnte hier als Bindeglied fungieren. Mikroglia sind an vorderster Front in inflammatorische Prozesse im ZNS involviert und parallel an der Aufrechterhaltung synaptischer Verbindungen beteiligt. Sie bilden eine wichtige Schnittstelle zwischen Inflammation und Neurotransmission. Sowohl inflammatorische Prozesse in der Peripherie als auch psychischer Stress führen zur Aktivierung von Mikroglia und triggern die Induktion eines pro-inflammatorischen, neurotoxischen Phänotyps. Freigesetzte Zytokine und neurotoxische Metabolite induzieren über vielfältige Mechanismen depressive Symptome und triggern degenerative Prozesse (Neuropilreduktion). Eine Beteiligung der Mikroglia in der Pathophysiologie der Depression wird vermutet.

SummaryWe have demonstrated previously that fibrin enhanced plasmin formation by the vascular plasminogen activator was significantly impaired, when components isolated from the plasma of three uncontrolled diabetic patients (type I) were used to study plasminogen activation in vitro. In the present study it can be demonstrated that functional properties of the vascular plasminogen activators as well as of the plasminogens from the same three diabetic patients are significantly improved after normalization of blood sugar levels and improvement of HbAlc values. Most pronounced the Km of diabetic vascular plasminogen activator in the presence of fibrin returned to normal values, and for diabetic plasminogen the prolonged lag period until maximal plasmin formation occurred was shortened to almost control values. From these data we conclude that the observed abnormalities of in vitro fibrinolysis are not primarily associated with the diabetic disease, but might be secondary to metabolic disorders caused by diabetes.

Mycotoxicological monitoring of forages shows that the problem of mycotoxicosis has been relevant for several decades. Minimal doses of mycotoxins in feed lead to a decrease in milk productivity, increased sensitivity to infectious and non-infectious diseases. When several mycotoxins enter the body simultaneously, a synergistic effect develops, causing a significant increase in toxicity. Feed contaminated with several types of fungi and their toxins is dangerous for dairy cattle. It was found that the feed mixture contained the types of associations of Aspergillus spp. fungi + Fusarium; Aspergillus spp. + Penicillium spp. + Mucor spp; Fusarium + Penicillium; Mucor spp. + Fusarium + Ustilaginales. Pathogenetic features of metabolic and morphological changes in highly productive cows with polymycotoxicosis were studied. Feeding food contaminated with various metabolites of mold fungi leads to the development of signs of chronic toxemia in animals. Clinical manifestations are the development of diarrhea and dehydration, with a decrease in milk productivity. Metabolic disorders feature the development of an inflammatory process, metabolic acidosis, hyperfermentonemia, with an increase in the amount of creatinine and urea in the blood serum. Metabolic signs indicate the development of hepatorenal syndrome due to structural disorders of the liver and kidneys. Histological signs of polymicotoxicosis are intracapillary and hemorrhagic glomerulonephritis, hepatocyte micronecrosis, and proliferation of connective tissue stroma cells, which leads to the development of atrophic cirrhosis in the interstitial and circular phases.
