The cytokine transforming growth factor (TGF)-β1 is highly induced after encephalopathic brain injury, with data showing that it can both contribute to the pathophysiology and aid in disease resolution. In the immature brain, sustained TGFβ-signaling after injury may prolong inflammation to both exacerbate acute stage damage and perturb the normal course of development. Yet in adult encephalopathy, elevated TGFβ1 may promote a reparative state. In this review, we highlight the context-dependent actions of TGFβ-signaling in the brain during resolution of encephalopathy and focus on neuronal survival mechanisms that are affected by TGFβ1. We discuss the mechanisms that contribute to the disparate actions of TGFβ1 toward elucidating the long-term neurological and neuropsychiatric consequences that follow encephalopathic injury.

ABSTRACT Two different Cd2+ uptake systems were identified inLactobacillus plantarum. One is a high-affinity, high-velocity Mn2+ uptake system which also takes up Cd2+ and is induced by Mn2+ starvation. The calculated Km and V maxare 0.26 μM and 3.6 μmol g of dry cell−1min−1, respectively. Unlike Mn2+ uptake, which is facilitated by citrate and related tricarboxylic acids, Cd2+ uptake is weakly inhibited by citrate. Cd2+ and Mn2+ are competitive inhibitors of each other, and the affinity of the system for Cd2+ is higher than that for Mn2+. The other Cd2+uptake system is expressed in Mn2+-sufficient cells, and noKm can be calculated for it because uptake is nonsaturable. Mn2+ does not compete for transport through this system, nor does any other tested cation, i.e., Zn2+, Cu2+, Co2+, Mg2+, Ca2+, Fe2+, or Ni2+. Both systems require energy, since uncouplers completely inhibit their activities. Two Mn2+-dependent L. plantarum mutants were isolated by chemical mutagenesis and ampicillin enrichment. They required more than 5,000 times as much Mn2+ for growth as the parental strain. Mn2+ starvation-induced Cd2+ uptake in both mutants was less than 5% the wild-type rate. The low level of long-term Mn2+ or Cd2+accumulation by the mutant strains also shows that the mutations eliminate the high-affinity Mn2+ and Cd2+uptake system.

The main aim of this research was to investigate the survival mechanisms employed by tourist guides in the context of the Covid 19 Pandemic. A mixed method research design was adopted, using in-depth interviews with key tourism informants and surveys administered to tourist guides in Gauteng. A total of five key informant interviews and two hundred surveys were collected at the end of the data collection period. Key findings indicate that many tourist guides were unable to access the aforementioned fund and had to implement their own short and long-term survival mechanisms. In many cases, various cost cutting measures were implemented and the utilization of personal savings and loans. Other guides temporarily engaged in different employment to secure an income. This study presents policy and systemic recommendations, which, if accepted and implemented, could assist the reignition of the tourist guide profession post-pandemic and ensure they form part of the tourism sector recovery trajectory. Additionally, further research on tourist guides in South Africa needs to be conducted, to fully understand the various aspects of this profession in the country.

Abstract Background Silages, as a nutritious and long-term preserved fodder, is achieved through fermentation. However, the constant low temperatures and freeze–thaw event are key factors that affect the quality of silages due to low levels of lactic acid production and undesirable microbial community revitalization. In this study, we attempt to investigate the effects of Lactobacillus plantarum QZW5 on the fermentation characteristics of wheat silage under ensiling conditions at the order of the following temperatures: constant low temperatures (5 °C for 30 days), multigelation (10 ℃ at day and – 10 ℃ at night for 30 days) and room temperature (aerobic/anaerobic, 10 days). Results Under multigelation condition, Lactobacillus plantarum QZW5 inoculation significantly increased the contents of lactic acid and acetic (P < 0.05) in the silage. The pH, bacterial richness index (Chao) and diversity index (Shannon) of the silage were significantly decreased by Lactobacillus plantarum QZW5 (P < 0.05). In Lactobacillus plantarum QZW5 inoculated silage, the dominant genera were L. plantarum subsp. plantarum. However, those in the control silage were Leuconostoc mesenteroides and Leuconostoc fallax. LAB (lactic acid bacteria) inoculation increased the abundance of desirable Lactobacillus spp. and inhibited the growth of undesirable Escherichia coli, Saccharomyces cerevisiae, and Filamentous fungi, among others in the silage. Conclusions Therefore, inoculation of L. plantarum QZW5 during ensiling could stimulate favorable fermentation and reconstruct bacterial community for better silage preservation. Graphic abstract

Bacteriocins is a natural preservative (bio-preservatives) that can safely be used for food. Bacteriocinscan be produced by a group of Lactic Acid Bacteria (LAB). Several strains of lactic acid bacteria (LAB)is bactericidal against gram positive and gram negative bacteria. The purpose of this study was toanalyze the bacteriocins produced by Lactobacillus plantarum in chicken meat and determine the levelof the last and long-term. Factors that are seen in this study is the levels of bacteriocins and also theduration of storage of chicken meat. The levels of bacteriocins is 0% and 10% by way of spraying inchicken meat and duration of storage are at 0, 5, 10, 15, 20, and 25 hours at room temperature. Thisstudy used a factorial completely randomized design with three replications. Plantaricin concentrationagainst pH and aw values were significantly different (P<0.05). The test result of storage duration againstwater content were significantly different (P<0.05). Treatment of plantarisin IIA-1A5 10% to maintainthe condition of chicken meat was better than treatment without the adding plantarisin.

ABSTRACT This study aimed to determine the effects of Bifidobacterium bifidum TMC3115, Lactobacillus plantarum 45 (LP45) and their combined use on cognitive performance and gut microbiota in APP/PS1 mice. The APP/PS1 mice were randomly divided into four groups: Alzheimer's disease (AD) model group, TMC3115 group [1 × 109 colony forming unit (CFU)], LP45 group (1 × 109 CFU) and a mixture group of TMC3115 (5 × 108 CFU) and LP45 (5 × 108 CFU). The wild-type littermates were chosen as normal control. The mice were sacrificed at the end of 22 weeks after behavioral evaluation. Collected cecum content was analyzed using 16S rRNA sequencing. Combined use of TMC3115 and LP45 significantly increased the times across the platform, time spent in the target quadrant compared with the AD, TMC3115 and LP45 groups in Morris water maze test. Microbiota analysis showed that combined TMC3115 and LP45 supplementation significantly increased observed species and beta diversity, and reversed gut dysbiosis by decreasing the abundance of Bacteroides and increasing the abundance of Acetatifactor and Millionella. These results indicate the long-term combined administration of TMC3115 and LP45 can improve spatial memory impairment in APP/PS1 mice and suggest that modifying the gut microbiome may provide potential benefits for AD patients.

Abstract Objectives A triathlon, which consists of swimming, bicycling, and running, is a high-intensity and long-term form of exercise that can cause injuries such as muscular damage, inflammation, oxidative stress, and energy imbalance. Previous studies indicated that Lactobacillus supplementation could improve oxidative stress and inflammatory responses. We investigate the effects of Lactobacillus plantarum PS128 supplementation on triathletes for shorten the acute inflammatory phase. Methods The triathletes were assigned to one of two groups with different exercise intensity stimulations with different time-points to investigate the effects of body compositions, inflammation, oxidative stress, performance, and injury-related biochemical indices. Results L.plantarum PS128 supplementation, combined with training, can significantly alleviate oxidative stress (such as creatine kinase, Thioredoxin, and Myeloperoxidase indices) after a triathlon (P &lt; 0.05).This effect is possibly regulated by a 6–13% decrease of indicated pro-inflammation (TNF-α, IL-6, and IL-8) cytokines (P &lt; 0.05) and 55% increase of anti-inflammation (IL-10) cytokines (P &lt; 0.05) after intensive exercise stimulation. Conclusions In conclusion, L. plantarum PS128 may be a potential ergogenic aid for better training management, physiological adaptations to exercise, and health promotion.This study was supported by the Ministry of Science and Technology in Taiwan (grant no.
