Organic Zn and Cu supplementation imprints on seminal plasma mineral, biochemical/ antioxidant activities and its relationship to spermatozoal characteristics in bucks

The aim of this study was to compare markers of semen quality, sperm membrane damage, and the seminal plasma antioxidant activity in warmblood stallions with and without sport workload stress. Four stallions were used for breeding only (control) and four both for breeding and competition in jumping. Semen samples were collected at 14-day intervals (from June to August) from each stallion (5 ejaculates per stallion). Immediately after sperm collection, a conventional examination of the ejaculate was processed. Catalytic activities of enzymes aspartate aminotransferase, alanin aminotransferase, glutathione peroxidase, superoxide dismutase and indicator of lipoperoxidation - F2α isoprostanes were measured in samples of seminal plasma. Contrary to basic semen quality indicators, the values of seminal plasma pH, aspartate aminotransferase and alanin aminotransferase were significantly (P < 0.05) impaired in the physically stressed stallions. Also, the level of F2α isoprostanes and the activity of superoxide dismutase were significantly (P < 0.05) increased by stress. The antioxidant activities of superoxide dismutase and glutathion peroxidase increased during the monitored period and reflected changes in F2α isoprostane concentration. We can conclude that even the conventional basic sperm indicators stay within the reference ranges of the biochemical indicators of seminal plasma such as pH or AST/ALT activity may be negatively influenced by sport workload stress. Increased concentrations of F2α isoprostanes indicate that lipoperoxidation can be a mechanism of cell membrane destabilization, which is counteracted by an increase of antioxidant enzyme activities. This is the first report of oxidative stress symptoms in normospermic equine semen in relation to stallion sport workload.

Aim: The study aimed to investigate the relationship between some heavy metals and total antioxidant capacities, glutathione levels, fructose, and testosterone in seminal plasma of infertile azoospermic and oligospermic males in Akwa Ibom State of Nigeria. Study Design:  A cross-sectional design with a total of 124 males included in the study of which 32 were azoospermic, 38, oligospermic, and 54, normospermic. Place and Duration of Study: Semen samples were collected from the urology or fertility clinic of UUTH, St. Luke’s Hospital, Anua, and Ibom specialist hospital of Akwa Ibom State. However, laboratory assays were performed at the Department of Medical Laboratory Science, Rivers State University, Port Harcourt between May, 2018 and January, 2021. Methodology: Semen specimens were collected after 3-5 days abstinence according to WHO criteria while seminal plasma were obtained from semen by spinning at 4500 rpm for 10 minutes and stored at -700C prior to laboratory analysis. Atomic absorption spectrometer (AAS) was used to determine the levels of heavy metals while ELISA methods were used to determine testosterone and GSH concentrations. TAC and fructose assays were carried out using spectrophotometric methods. Results: Results showed that non-essential heavy metals such as lead, mercury, and arsenic correlated negatively with testosterone, fructose, and antioxidant activities of the seminal plasma in azoospermic subjects. In addition, lead and mercury correlated positively in the azoospermic subjects. Meanwhile, selenium, an essential heavy metal, correlated positively with testosterone and antioxidant activities in oligospermic subjects with 11-19 x106cells/ml. Conclusion: The levels of non-essential heavy metals in azoospermic subjects precipitated poor anti-oxidant and testosterone activities inducing oxidative stress while in oligospermic subjects, selenium and antioxidant parameters and testosterone were in good association indicating improved antioxidant activities and testicular function.

Elevated levels of reactive oxygen species (ROS) are a major cause of male infertility. However, some men with high seminal ROS levels are still fertile. The main objective of this study was to understand the molecular mechanism(s) responsible for the preservation of fertility in those men. Semen samples from fertile men were divided into two groups: control (n = 10, ROS < 102.2 RLU/s/106 sperm) and ROS+ (n = 10, ROS > 102.2 RLU/s/106 sperm). Proteomic analysis of seminal plasma and spermatozoa was used to identify the differentially expressed proteins (DEPs) between the experimental groups, from which some proteins were validated by Western blot (WB). A total of 44 and 371 DEPs were identified between the study groups in the seminal plasma and spermatozoa, respectively. The identified DEPs were primarily involved in oxidoreductase, endopeptidase inhibitor, and antioxidant activities. We validated by WB the underexpression of NADH:ubiquinone oxidoreductase core subunit S1 (p = 0.01), as well as the overexpression of superoxide dismutase 1 (p = 0.03) and peroxiredoxin 4 (p = 0.04) in spermatozoa of ROS+ group. Our data suggest that fertile men with high ROS levels possess an effective antioxidant defense system that protects sperm proteins, as well as an active proteasomal system for degradation of defective proteins.

Abstract. In this study, cytoprotective and antioxidant activities of Rosa canina (RC) and Salix alba (SA), medicinal plants, were studied on mouse primary splenocytes by comparing Controlled Differential Sieving process (CDSp), which is a novel green solvent-free process, versus a conventional technique, employing hydroethanolic extraction (HEE). Thus, preventive antioxidant activity of three plant powders of homogeneous particle sizes, 50–100 µm, 100–180 µm and 180–315 µm, dissolved directly in the cellular buffer, were compared to those of hydroethanolic (HE) extract, at 2 concentrations (250 and 500 µg/mL) in H2O2-treated spleen cells. Overall, compared to HE extract, the superfine powders, i. e., fractions < 180 µm, at the lowest concentration, resulted in greater reactive oxygen species (ROS) elimination, increased glutathione peroxidase (GPx) activity and lower malondialdehyde (MDA) production. Better antioxidant and preventive effects in pre-treated cells were found with the superfine powders for SA (i. e., 50–100 µm and 100–180 µm, both p < 0.001), and with the intermediate powder for RC (i. e., 100–180 µm, p < 0.05) versus HE extract. The activity levels of catalase (CAT) and superoxide dismutase (SOD) in pretreated splenocytes exposed to H2O2, albeit reduced, were near to those in unexposed cells, suggesting that pretreatment with the fine powders has relatively restored the normal levels of antioxidant-related enzymes. These findings supported that CDSp improved the biological activities of plants, avoiding the use of organic solvents and thus it could be a good alternative to conventional extraction techniques.

In the present study, the nutritional quality of four grains including adlay (AD), buckwheat (BW), glutinous barley (GB), and white rice (WR) were evaluated in terms of plasma lipid parameters, gut transit time, and thickness of the aortic wall in rats. The rats were then raised for 4 weeks on the high-fat diet based on the American Institute of Nutrition-93 (AIN-93 G) diets containing 1 % cholesterol and 20 % dietary lipids. Forty male rats were divided into 4 groups and raised for 4 weeks with a diet containing one of the following grains: WR, AD, BW, or WB. The level of thiobarbituric acid-reactive substances (TBARS) in liver was shown to be higher in rats by the order of those fed WR, AD, GB, and BW. This indicates that other grains decreased oxidative stress in vivo more than WR. The superoxide dismutase, glutathione, glutathione peroxidase, and glutathione reductase levels in the AD, BW, and GB groups were significantly higher than those in the WR group (p < 0.05). Plasma lipid profiles differed significantly according to grain combination, and decreased aortic wall thickness was consistent with the finding of decreased plasma low-density lipoprotein cholesterol (LDL-C) (p < 0.05) and increased high-density lipoprotein (HDL-C) in rats fed AD, BW, and GB (p < 0.001). The antioxidant and hypolipidemic capacities of grains are quite high, especially those of adlay, buckwheat, and glutinous barley. In conclusion, this study has demonstrated that the whole grains had a cardioprotective effect. This effect was related to several mechanisms that corresponded to lowering plasma lipids, decreasing TBARS, and increasing antioxidant activities.
