Insight into the practical models for prediciting the essential role of the cytochrome P450-mediated biotransformation in emodin-associated hepatotoxicity

Bisphenol A (BPA) is a widespread pollutant threatening the ecosystem and human health. An effective BPA degrader YC-JY1 was isolated and identified as Sphingobium sp. The optimal temperature and pH for the degradation of BPA by strain YC-JY1 were 30 °C and 6.5, respectively. The biodegradation pathway was proposed based on the identification of the metabolites. The addition of cytochrome P450 (CYP) inhibitor 1-aminobenzotriazole significantly decreased the degradation of BPA by Sphingobium sp. YC-JY1. Escherichia coli BL21 (DE3) cells harboring pET28a-bisdAB achieved the ability to degrade BPA. The bisdB gene knockout strain YC-JY1ΔbisdB was unable to degrade BPA indicating that P450bisdB was an essential initiator of BPA metabolism in strain YC-JY1. For BPA polluted soil remediation, strain YC-JY1 considerably stimulated biodegradation of BPA associated with the soil microbial community. These results point out that strain YC-JY1 is a promising microbe for BPA removal and possesses great application potential.

Biosensor experiments on investigation of interaction between prostacyclin synthase (PGIS) and different proteins of the cytochrome P450 monooxygenase systems were perfomed. Interaction of PGIS with microsomal (CYP21A2, CYP2E1) and mitochondrial (CYP27A1, CYP11B1, CYP11B2, CYP11A1) cytochrome P450s was detected. Kinetic and equilibrium parameters of protein complexes formation were determined. Data obtained suggest an essential role of these hemoproteins interaction in regulation of prostacyclin and thromboxane A2 biosynthesis.

Research suggests that debriefing is the most important component of simulation. Debriefing is the time when participants think over and reflect on what happened during the simulated experience. The aim of this research is to gain insight into the experience of facilitating debriefing following a simulated experience in a tertiary health care setting. This qualitative study was guided by phenomenology, as developed by Heidegger and Gadamer, and included 10 health professionals (nurses, doctors, and a midwife) debriefing simulation experiences. Data were collected through individual interviews and analysed through iterative re-writing and the generation of three stories as getting started; supporting the debrief to unfold, and knowing how to end. This study affirmed the role of the facilitator in debriefing following simulated experiences. The facilitator’s role includes leading the dialogue beyond “talk” and into “learning”. The art of asking a question, working with silence, and enabling learning all require skill. Through experience, facilitators learn to respond to whatever unfolds in the session. This study contributed detail of what facilitators do to draw out thinking, which confirmed their essential role. Facilitators cannot prepare a script for debriefing beforehand but must rely on their debriefing know-how to respond in each debrief. The facilitator’s know-how is therefore central to debriefing following simulated experiences.

AbstractThe organization of the genome into transcriptionally active and inactive chromatin domains requires well-delineated chromatin boundaries and insulator functions in order to maintain the identity of adjacent genomic loci with antagonistic chromatin marks and functionality. In plants that lack known chromatin insulators, the mechanisms that prevent heterochromatin spreading into euchromatin remain to be identified. Here, we show that DNA Topoisomerase VI participates in a chromatin boundary function that safeguards the expression of genes in euchromatin islands within silenced heterochromatin regions. While some transposable elements are reactivated in mutants of the Topoisomerase VI complex, genes insulated in euchromatin islands within heterochromatic regions of the Arabidopsis thaliana genome are specifically downregulated. H3K9me2 levels consistently increase at euchromatin island loci and decrease at some TE loci. We further show that Topoisomerase VI physically interacts with S-adenosylmethionine (SAM) synthase MAT3, which is required for H3K9me2 deposition. Topoisomerase VI promotes MAT3 occupancy on heterochromatic elements and its exclusion from euchromatic islands, thereby providing a mechanistic insight into the essential role of Topoisomerase VI in the delimitation of chromatin domains.

Abstract This article revisits Julia Alvarez’s critically acclaimed historical novel In the Time of the Butterflies (1994). While much scholarship has paid attention to the novel as historiographic metafiction, its depiction of Rafael Leónidas Trujillo’s regime (1930-61), and its feminist perspective on the Dominican Republic, its racial politics are under-studied. In particular, scholars have overlooked Fela, the Afra-Dominican servant, spirit medium, and storyteller. I argue that studying Fela’s presence in the text as an unauthorized and unauthored voice not only adds complexity to the production of historiography and storytelling but also provides new insight into postcolonial feminist critiques of voice/lessness, narrative, and marginalized identities in the novel and criticism on it. Closely analyzing Fela’s voice—as it intersects with storytelling, historical slave narratives, Vodou, the maternal, and Haiti’s contribution to the Dominican Republic’s history—makes visible the unacknowledged yet essential role of the Afra-Dominican not only in this novel specifically but also to the Dominican Republic more generally.
