This study investigated the impact of immune-related pathway inhibition, among them indolamine 2,3-dioxygenase (IDO), alone and together with immune cells on growth and viability of colorectal cancer (CRC) cells. A panel of patient-derived CRC cell lines with different molecular characteristics (CpG island methylator phenotype, chromosomal, and microsatellite instability) was included. Initial phenotyping of CRC cell lines (n=17) revealed high abundance of immunosuppressive checkpoint-molecules in general, but an individual profile for IDO. Presence of immune-related molecules was independent of the molecular subtype. Selective treatment of CRC cell lines showing high or low IDO expression (n=2 cell lines each) was performed with single agents and combinations of Indoximod, Curcumin, and Gemcitabine with and without the addition of peripheral blood lymphocytes (PBL) in an allogeneic setting. All substances affected CRC cell growth in a cell line specific manner. The combination of Curcumin and Gemcitabine proved to be most effective in tumor cell elimination. Functional read-out analyses identified cellular senescence, after both single and combined treatment. Curcumin alone exerted strong cytotoxic effects by inducing early and late apoptosis. Necrosis was not detectable at all. Addition of lymphocytes generally boosted antitumoral effects of all IDO-inhibitors, with up to 80 % cytotoxicity for the Curcumin treatment. Here, no obvious differences became apparent between individual cell lines. Combined application of Curcumin and low-dose chemotherapy is a promising strategy to kill tumor target cells and to stimulate antitumoral immune responses.

Abstract OBJECTIVE Diffuse astrocytomas are classified as WHO grade II and its median overall survival (mOS) is 10 to 11 years. The efficacy of chemoradiation in the high-risk feature has been reported. The prognosis is associated with IDH and TERT promoter (TERTp) mutations. Here, we retrospectively analyzed the patients with diffuse astrocytoma treated with vincristine, ACNU, carboplatin and interferon-β plus radiotherapy (VAC-feron-R)in our institute. PATIENTS AND METHODS Between December 2003 to January 2016, 44 patients were diagnosed as diffuse astrocytoma with integrated diagnosis of histological and molecular analysis. The average age was 43.1 years (22–71 years). They received VAC-feron-R as initial treatment in our institute. We analyzed the IDH1/2 and the TERTp mutation using Sangar sequencing and determined the 1p/19q codeletion by the fluorescence in situ hybridization or the multiplex ligation-dependent probe amplification. RESULTS Median follow-up period was 76.5 months, mPFS was 126 months, mOS did not reach, and 10-year survival rate was 60%.IDH status was determined in 29 patients, 9 mutant and 20 wild types. There was no significant difference in PFS and OS between the two groups. TERTp status was determined in 18 patients with IDH wild type, 6 mutant and 12 wild types. mPFS of patients with TERTp wild type did not reach, but that with TERTp mutant type was 34.5 months (p = 0.0356). CONCLUSION Compared with previous clinical studies, VAC-feron-R showed a favorable clinical outcome in diffuse astrocytoma. The impact of TERTp status on prognosis was identified but not IDH status in this cohort.

<p>Antarctic krill (<em>Euphausia superba</em>), hereafter krill, are pivotal to the Antarctic marine ecosystem, forming the base of a highly productive system and contributing significantly to the biogeochemical cycle. The negative effects of anthropogenic climate stressors amplified in the Southern Ocean such as rapid warming and ocean acidification (OA) have been acknowledged for krill. Less explored is the impact of increasing plastic pollution, particularly in conditions that reflect the likely future Southern Ocean environment. We hypothesise that krill have heightened vulnerability to multi-stressor scenarios due to their physiological and behavioural traits coupled with rapid environmental changes of their Antarctic habitats. Here, we investigate the single and combined effects of nanoplastic (NP; spherical, aminated (NP-NH<sub>2</sub>), yellow-green, fluorescent polystyrene nanoparticles) and OA (pCO<sub>2</sub>-manipulated seawater, pH 7.7) on the embryonic development of krill eggs. Krill were collected in the Scotia Sea within the Atlantic sector of the Southern Ocean in austral summer 2019. Eggs from a single female were incubated in seawater at 0.5 °C for 6 days with three treatments: (i) with 0.16 μm NP, (ii) in acidified conditions, and (iii) with a combined treatment of NP (0.16μm) and acidification. All NP treatments were at a concentration of 2.5μg/ml. We found that exposure to the NP-OA multi-stress treatment negatively impacted the development of embryos, decreasing the probability of reaching the limb bud stage by 9% compared with the control, whilst no significant difference was observed for the singular NP or OA treatments. This preliminary study supports our hypothesis regarding the potential impacts of multiple stressors on vulnerable embryonic stages of this ecologically critical Antarctic species.</p>

BackgroundThe monoclonal antibody (mAb) trastuzumab is part of the standard of care for patients with human epidermal growth factor receptor 2 (HER2)-overexpressing breast cancer. Antibody-dependent cell-mediated phagocytosis (ADCP) and cytotoxicity (ADCC) are major mechanisms of action of the mAb trastuzumab. Histone deacetylase inhibitors (HDACi), such as valproic acid (VPA) or vorinostat (SAHA), exert several immunostimulatory properties, which contribute at least in part to their anticancer effect. However, the impact of HDACi-induced immunostimulatory effects on trastuzumab-mediated anti-tumor immune response is not well characterized.MethodsWe analyzed the ADCP and ADCC activity of peripheral blood mononuclear cells (PBMCs) from age and gender-matched healthy volunteers (n=5) against HDACi-treated HER2-overexpressing breast cancer cells (SKBR3), using a well-established in vitro three-color imaging flow cytometry and flow cytometry approach.ResultsVPA and SAHA enhanced trastuzumab-mediated ADCP and trastuzumab-independent cytotoxicity. Mechanistically, VPA upregulated the activating antibody-binding receptor Fc-gamma receptor (FcγR) IIA (CD32A) on monocytes (CD14+). Moreover, VPA and SAHA downregulated the anti-apoptotic protein myeloid leukemia cell differentiation 1 (MCL1) in breast cancer cells. Additionally, VPA and SAHA induced an immunogenic cell death, characterized by the exposure of calreticulin (CALR), as well as decreased the “do not eat me” signal CD47 on tumor cells.ConclusionsHDACi VPA and SAHA increase trastuzumab-mediated phagocytosis and trastuzumab-independent cytotoxicity. The immunomodulatory activities of those HDACi support a rationale combined treatment approach with mAb for cancer treatment.

This article intends to promote the usage of potato peels as efficient substrate for the anaerobic digestion process for energy recovery and waste abatement. This study examined the performance of anaerobic digestion of potato peels in different inoculum-to-substrate ratios. In addition, the impact of combined treatment with cow manure and pretreatment of potato peels was examined. It was found that co-digestion of potato peel waste and cow manure yielded up to 237.4 mL CH4/g VSadded, whereas the maximum methane yield from the mono-digestion of potato peels was 217.8 mL CH4/g VSadded. Comparing the co-digestion to mono-digestion of potato peels, co-digestion in PPW/CM ratio of 60:40 increased the methane yield by 10%. In addition, grinding and acid hydrolysis applied to potato peels were positively effective in increasing the methane amount reaching 260.3 and 283.4 mL CH4/g VSadded respectively. Likewise, compared to untreated potato peels, pretreatment led to an elevation of the methane amount by 9% and 17% respectively and alleviated the kinetics of biogas production.

Abstract The impact of plant growth regulators (PGRs) “Stimpo” and “Regoplant” on Miscanthus x giganteus (Mxg) biomass parameters was investigated when the plant was grown in military soils with different properties from Dolyna, Ukraine and Hradcany, Czech Republic. The results showed that PGRs positively influenced the biomass parameters when the plant was grown in soil in Dolyna with good agricultural characteristics, the influence of “Regoplant” was higher and the best results were obtained with combined treatment: application to rhizomes before planting and spraying on the biomass during vegetation. Using of PGRs did not improve the biomass parameters when the plant was grown in poor soil in Hradcany. In parallel the peculiarities of the metals uptake process were studied for the following metals: chromium (Cr), manganese (Mn), nickel (Ni), copper (Cu), zinc (Zn), strontium (Sr) and lead (Pb). The uptake behavior of the monitored elements differed based on the soil quality. According to the bioconcentration factor uptake of the abiogenic elements, Cr and Pb, was dominant in the plant roots in both soils, whereas Ni was not detected in any plant tissues. The behavior of biogenic elements (Mn, Cu, Zn) and their analogs (Sr) was different. Those elements were more intensively taken up in shoot tissues in low-nutrient sandy Hradcany soils, while they were mainly taken up in plant roots in fertile Dolyna soils. The unusual behavior of biogenic elements in the low-nutrient soils may be explained by the effect of stress. However, more research is needed focused mainly on soil properties and nutrient availability in order to confirm or disprove this hypothesis and to explore the cause of the stress. The summarized results here show that soil properties influenced Mxg biomass parameters, affected the uptake behavior of metals significantly and tested PGRs cannot be utilized universally in the production of Mxg in the poor military soils.

Abstract Introduction A reduction of the hospitalization and reperfusion treatments was reported during COVID-19 pandemic. However, high variability in results emerged, potentially due to logistic paradigms adopted. Here, we analyze stroke code admissions, hospitalizations, and stroke belt performance for ischemic stroke patients in the metropolitan Bologna region, comparing temporal trends between 2019 and 2020 to define the impact of COVID-19 on the stroke network. Methods This retrospective observational study included all people admitted at the Bologna Metropolitan Stroke Center in timeframes 1 March 2019–30 April 2019 (cohort-2019) and 1 March 2020–30 April 2020 (cohort-2020). Diagnosis, treatment strategy, and timing were compared between the two cohorts to define temporal trends. Results Overall, 283 patients were admitted to the Stroke Center, with no differences in demographic factors between cohort-2019 and cohort-2020. In cohort-2020, transient ischemic attack (TIA) was significantly less prevalent than 2019 (6.9% vs 14.4%, p = .04). Among 216 ischemic stroke patients, moderate-to-severe stroke was more represented in cohort-2020 (17.8% vs 6.2%, p = .027). Similar proportions of patients underwent reperfusion (45.9% in 2019 vs 53.4% in 2020), although a slight increase in combined treatment was detected (14.4% vs 25.4%, p = .05). Door-to-scan timing was significantly prolonged in 2020 compared with 2019 (28.4 ± 12.6 vs 36.7 ± 14.6, p = .03), although overall timing from stroke to treatment was preserved. Conclusion During COVID-19 pandemic, TIA and minor stroke consistently reduced compared to the same timeframe in 2019. Longer stroke-to-call and door-to-scan times, attributable to change in citizen behavior and screening at hospital arrival, did not impact on stroke-to-treatment time. Mothership model might have minimized the effects of the pandemic on the stroke care organization.

Objective: The current longitudinal impact of treatment of ADHD on intimate partner violence (ITAP) study aims to investigate whether decrease of ADHD symptoms is associated with reduction of intimate partner violence (IPV) frequency in IPV offenders with ADHD in a forensic psychiatry setting. Method: Of n = 209 offenders of IPV with ADHD, frequency of IPV and ADHD symptoms were assessed at the 8th, 16th, 24th, and 52nd weeks of their combined treatment for ADHD and IPV. Results: We observed a significant decrease of self-reported ADHD symptoms (large effect size, d ≥ 0.80) and all IPV outcomes (small, d > 0.20, to large, d > 0.80, effect sizes). The decrease in IPV was mainly associated with the decrease in ADHD symptoms. Conclusion: As IPV treatment alone is not effective in the reduction of IPV in forensic psychiatry, we now have strong indications that the combined treatment of adult ADHD and IPV is more effective in offenders with ADHD.
