Influence of mating pressure and season on the semen characteristics of rams

SUMMARYTwo groups of 4 Suffolk rams were each subjected to mating pressures of 90 ejaculates (High) and 45 ejaculates (Low) over a 3-day period during summer and autumn. Every sixth (High) or every third (Low) ejaculate was collected into an artificial vagina and assessed for volume, sperm density, total sperm per ejaculate, motility and percentage of live and abnormal sperm.There was a general lowering of semen quantity and quality with successive ejaculations. Rate of change of the various measures was greater on the High than on the Low mating pressure. Volume and percentage of live sperm showed a greater decline, and percentage of abnormal sperm a greater increase, during the autumn than during the summer. Density showed a greater decline in summer than in autumn.The significance of the changes in semen characteristics observed is considered in relation to requirements for high fertility after pro-gestagen treatment.

Semen characteristics were studied prior to and during the paddock joining of rams to varying numbers of ewes in a series of three experiments. A marked fall in ejaculate volume, sperm density, and their product, total number of sperm per ejaculate to levels of c. 0.3 ml, 1.5 x l09 sperm/ml, and 0.45 x l09 total sperm respectively occurred during the first 4 days of joining. Although there were large differences in the number of ewes joined per ram (25 to 100) and consequently the number of ewes served per ram per day, the depression in these semen characteristics occurred to a similar extent in all rams during the first 2 weeks of mating. By examining the data for all collections throughout the joining period, however, highly significant negative correlations were obtained for ejaculate volume, sperm density, and total number of sperm per ejaculate in each case with the number of ewes served during the 4 days preceding semen collection. Increases in the proportion of abnormal sperm were found sporadically in the ejaculates of rams during joining. Rams 2½ years old produced significantly more sperm per ejaculate before, during, and after the 6 week joining period than rams 1½ years old.

A total of sixteen rabbits sourced from two locations in Southeastern Nigeria were used for this study. Semen was collected from all the rabbits twice weekly for eight weeks using an artificial vagina while observation on the effect of time of collection on semen quality were also made. The results obtained indicated that time of collection had no significant effect (P > 0.05) on all the seminal characteristics measured. However, source of the animals showed significant effect (P < 0.05) on semen volume, sperm concentration, total sperm per ejaculate and proportion of abnormal sperm. Average semen volume, sperm concentration and total sperm per ejaculate were: 0.69±0.007 ml, 104.0±1.44 x 106/m1 and 71.40±0.90x106 respectively for the rabbits sourced from Port Harcourt. The corresponding values for rabbits sourced from Owerri were: 0.66±0.008m1, 96.02 ±1.63x106iml and 63.49±1.50 x 106 respectively. Abnormal sperm proportions were 11.05±0.22% and 1216 ± 032% for the rabbits sourced from Port Harcourt and Owerri respectively. There were no significant (P> 0.05) differences observed between locations in live sperm concentration and sperm motility. Live sperm concentration and sperm motility were 87.75±1.09% and 71.62±0.74% for the Port Harcourt group, while the values obtained for the Owerri group were 88.06±034% and 71.19±0.89% respectively.

This study was aimed to explore the effect of adding pentoxifylline (PTX) and nitric oxide (NO) to Tris extender on some post-cryopreserved semen attributes of Holstein bulls for different preservation periods (cooling at 5◦C, 48 hrs., 1, 2 and 3 months post cryopreservation, PC). Seven Holstein bulls of 2.5-3 years old were used in the current study during the period from 20th November, 2017 to 20th August, 2018. Pooled semen was equally divided into three groups within one experiment. PTX (0.192 g / 100 ml extender) and NO (0.018 g / 100 ml extender) were added to Tris extender and comparisons in response were made with the control group (Tris extender, C). The PTX group exhibited greater (P≤ 0.01) sperm's cell individual motility percentage as compared with the C group at cooling (5ºc) and 48 hr PC periods, while, PTX and NO groups were superior in these percentages at the remaining PC periods than C group. Excluding data of 2 months PC, greater (P≤ 0.01) live sperm percentage was observed in PTX and NO groups in comparison with the C group at all preservation periods. Lesser (P≤ 0.01) abnormal sperm percentage were noticed for PTX and NO groups as compared with the C group at all preservation time periods. The PTX and NO groups exhibited greater   (P≤ 0.01) acrosome and plasma membrane integrity percentages in comparison with the C group at all preservation time periods. In conclusion, adding PTX and NO to Tris extender enhanced some of  PC semen characteristics of Holstein bulls at different preservation periods.

The aim of the present study was to investigate and compare the impact of the breeding (BS) and non-breeding (NBS) seasons on scrotal circumference, and native and post-thaw semen characteristics in Karayaka rams. Six (6) Karayaka rams were used as semen donors, and ejaculates were collected with the aid of an artificial vagina during the BS (October-November) and NBS (April-May). Interestingly, sperm freezability was found to be higher in the NBS, when compared to the BS, and the sperm motility rate (36.00±2.74%, 55.0±3.24%) and percentage of abnormal sperm (44.00±1.90%, 57.00±2.09%) were found to significantly differ with season (P<0.05). In conclusion, the post-freezing sperm motility was better during the non-breeding season. Post-thaw sperm abnormality was higher during non-breading season. However, if ram semen is used fresh for AI, it can be collected and used during both during the breeding and non-breeding season.

SUMMARYFour Finnish Landrace and four Scottish Blackface rams were exposed at 21-day intervals over a period of 19 months to two ovariectomized ewes, which had been induced to show oestrus. The behaviour of each ram was observed for 10 minutes with each ewe. Over the period of the experiment the Finnish Landrace rams consistently mounted the ewes more often than did the Blackfaces and in this respect in particular the reproductive behaviour of the males of these breeds was found to be positively correlated with that of the females of their breed.Initially, however, the Finnish Landrace rams mated more ewes than did the Blackfaces, but from the time that all rams became sexually active it was impossible to differentiate between the ability of the two groups of rams to mate the ewes presented to them. Semen collections from each ram were attempted by the use of an artificial vagina, again at three-weekly intervals. The motility, density and proportion of live sperm did not differ between the two groups of rams.The average number of mounts accredited to each ram during the test increased from around 5 in the summer to around 35 in the autumn.

The aim of this study is to investigate the sexual desire and semen characteristics of Friesian bulls born in Iraq by adding different concentration of Glycyrrhizin glabra to nutrient of these bulls and study the interaction between months and these treatments. Nine bulls divided randomly to three equal groups ,first group control T1 ,second and third group T2 ,T3 adding 200,400 mg/ kg body weight of Glycyrrhizin glabra respect .Sexual desire was measured according to reaction time and ejaculates collection by artificial vagina and examined using the following parameters: volume, mass, individual motility ,dead and abnormal sperm , concentration of sperms`/ml and total number of sperms , during December, January and February. Results revealed that the treatment T3 showed significant (P < 0.05) superior sexual desire and some semen characteristics in most month of this study comparative with treatment T2 and control , but treatment T2 it is didn’t appear to causes deleterious changes in reproductive performance of this bulls . Results also indicated that best (P < 0.05) sexual desire was in February and best (P < 0.05) some semen parameters were in January and February

The experiment was carried out to compare the semen characteristics of indigenous and Amo strains of cockerel at poultry unit of teaching and research farm of Federal University of Kashere, Gombe State, Nigeria. Semen samples were collected from nine indigenous and nine Amo breeds of cockerel at three days interval for two weeks using abdominal massage technique. Semen samples were examined macroscopically for semen colour, pH and ejaculation volume. Then, microscopic observation was carried for sperm concentration, mass motility, progressive motility, live and dead sperms percentage, normal and abnormal sperm, all for semen characteristics. The results showed a significant difference (P ≤ 0.05) between mass motility, progressive motility, sperm concentration and head defects of 4.85 ± 0.27 to 4.37 ± 0.19, 95.13 ± 0.43 to 81.63 ± 1.15%, 4.93 ± 1.84 to 3.40 ± 1.07×109/ml and 2.96 ± 0.17 to 3.44 ± 0.12% for indigenous and Amo breeds of cockerel, respectively. There were no significant differences observed as semen colour, ejaculate volume, semen pH, live / dead normal sperm neck (mid-piece), tail defects and sperm total abnormalities were found to be 2.85 ± 0.07 to 2.00 ± 0.090.21 ± 0.17 to 0.20 ± 0.02 /ml, 88.85 ± 0.58 to 72.70 ± 0.54% /ml, 11.14 ± 0.58 to 27.29 ± 0.54%, 81.00 ± 0.78 to 66.22 ± 0.61%,9.03 ± 0.42 to 13.96 ± 0.47%, 9.70 ±  to 13.00 ± 0.30 and 21.70 ± 0.59 to 30.40 ± 0.53% for the indigenous and Amo breed groups of cockerel, respectively. It was concluded that semen quality characteristics could be differed between genetically improved (Amo strain) and indigenous breed of cockerels.

Objective: To determine the effect of age and frequency of collection on quality parameters of Jersey bull semen at National Livestock Breeding Center (NLBC), Pokhara. Materials and Methodology: Nine Jersey bulls were selected randomly from NLBC. To obtain the effect of age, six bulls of three different age interval (3-4, 5-7 and 8-9 years) were selected randomly. Remaining three bulls were used to determine the effect of frequency of collection on semen quality. Studies were conducted for three months (Nov/Dec to Dec/Jan, 2016). Semen quality was analyzed using CASA system and SMILE software. ANOVA and paired t- tests were applied to find out level of significance. All the statistical analyses were done using SPSS 17.0 Results: Sperm concentration, post- thaw motility, live sperm, and abnormal sperm were significantly affected by the age of the bulls. Semen quality was found superior in the bulls belonging to 5-7 years of age intervals. Significantly higher value for semen volume, sperm concentration and post- thaw motility was obtained on second collection (P<0.05). However, volume initial motility, pre-filling motility, live sperm and abnormal sperm percentage did not differ significantly among different age intervals and between the frequencies of collection. Conclusion: Different quality parameters of semen like volume, concentration, initial motility, pre-filling motility, post thaw motility, live and normal spermatozoa percentages were higher in 5-7 years old bulls and in second collection of semen. Sperm concentration (0.551 ±0.004 to 1.084 ± 0.014x109), post- thaw motility (42.44±0.11 to 51.72±0.03 %), live sperm (45.62 ± 0.13 to 51.23±0.15 %), and abnormal sperm (11.57±0.12 to 19.59±0.24%) were significantly affected by the age of the bulls. Semen quality was found superior in the bulls belonging to 5-7 years of age intervals. On the other hand, volume (3.98±0.09 to 6.72±0.06 ml), initial motility (72.68±0.02 to 78.64±0.15 %) and pre-filling motility (71.34±0.17 to 77.62±0.02 %) did not differ significantly among different age intervals. Significantly, higher value for semen volume (3.98±0.12 to 6.02±0.14 ml), sperm concentration (0.572±0.119 to 1.095±0.015) and post- thaw motility (40.34±0.21 to 48.54±0.06% was obtained on second collection. However, initial motility (70.58±0.06 to 75.64±0.13 %), prefilling motility (68.11±0.12 to 71.62±0.02 %), live sperm (53.27±0.54 to 56.17±0.02 %) and abnormal sperm (12.31±0.12 to 14.3±0.47 %) did not differ significantly between the frequencies of collection. Different quality parameters of semen like volume, concentration, initial motility, pre-filling motility, post thaw motility, live and normal spermatozoa percentage were higher in 5-7 years old bulls and in second collection of semen.

The goal of this study was to evaluate and compare the semen characteristics, and testosterone concentration for three breeds raised under hot, desert conditions in Saudi Arabia. A total of 150 rams (Najdi, Naemi, and Harri) were used, aged 12, 18, 24, 30 and 36 months (ten rams from each breed in each age group). Body weight and testicular parameters were measured. In addition, testosterone concentration (TC), ejaculate volume (EV), sperm concentration (SC), total sperm output per ejaculate (TSO), live sperm percentage (LSP), sperm progressive motility (SPM) and sperm mass motility (SMM). The results show that TC and EV differed significantly (P less than 0.01) between breeds. At 36 months, semen characteristics (TSO, SMM and SPM) differed significantly among breeds. Naemi rams were superior to Najdi and Harri in terms of the mean live sperm percentage (LSP) for all age groups. These results may assist in ram selection for breeding programs based on high reproductive performance.
