The aim of the present study was to investigate and compare the impact of the breeding (BS) and non-breeding (NBS) seasons on scrotal circumference, and native and post-thaw semen characteristics in Karayaka rams. Six (6) Karayaka rams were used as semen donors, and ejaculates were collected with the aid of an artificial vagina during the BS (October-November) and NBS (April-May). Interestingly, sperm freezability was found to be higher in the NBS, when compared to the BS, and the sperm motility rate (36.00±2.74%, 55.0±3.24%) and percentage of abnormal sperm (44.00±1.90%, 57.00±2.09%) were found to significantly differ with season (P<0.05). In conclusion, the post-freezing sperm motility was better during the non-breeding season. Post-thaw sperm abnormality was higher during non-breading season. However, if ram semen is used fresh for AI, it can be collected and used during both during the breeding and non-breeding season.

AbstrakIndonesia memiliki banyak tumbuhan berpotensi obat salah satunya bisa dijadikan sebagai obatkontrasepsi tradisonal yang biasa digunakan untuk menjarangkan anak atau sterilisasi sepertirebusan akar Caesalpinia bonduc (Aru). Tujuan penelitian untuk mengetahui pengaruh obatkontrasepsi tradisional air rebusan akar aru terhadap kualitas pematangan sperma epididimis mencit(Mus musculus). Mencit dipilih secara acak untuk mewakili 4 kelompok dosis yaitu : 0,0 (K0), 16,0(K1) 24,0 (K2) dan 32,0 (K3) μl/gram berat badan/hari. Setiap kelompok perlakuan dilakukandengan 5 kali replikasi. Perlakuan diberikan melalui oral dengan menggunakan jarum gavage selama10 hari berturut-turut. Pembedahan dilakukan pada hari pertama setelah perlakuan selesai.Pengamatan kualitas sperma epididimis dengan perhitungan produksi sperma, % sperm motil, %sperma hidup, % sperma abnormal,dan rangking keaktifan sperma. Untuk mengetahui ada atautidaknya perbedaan yang bermakna antar perlakuan dalam kelompok dilakukan uji Anava satu arah,bila terdapat perbedaan bermakna dilanjutkan uji BNJ untuk membandingkan angka rata-rata antarkelompok perlakuan. Pemberian air rebusan akar Caesalpinea bonduc pada mencit menunjukanpengaruh perbedaan secara signifikan pada jumlah (%) sperma abnormal antara dosis 0,0 μl dengan24,0 dan 32,0 μl serta antara 16,0 μl dengan 24,0 dan 32,0 μl; Jumlah (%) sperma hidup terdapatperbedaan antara dosis 0,0 μl dengan 16,0 , 24,0 dan 32,0 μl; Jumlah (%) sperma motil terdapatperbedaan antara dosis 0,0 μl dengan 24,0 dan 32,0 μl; Rangking keaktifan sperma terdapatperbedaan antara dosis 0,0 μl dengan 24,0 dan 32,0 μl. The aims of this study is to know the effect of root boiled water ofCaesalpinia bonduc as traditional contraception medicine on quality of sperm maturation ofepididymis of mice (Mus musculus). Mice were choosed radomly and doses gouped: 0,0 (K0 ), 16,0(K1) 24,0 (K2) and 32,0 (K3) μl/gram body weight/day. Each of group was replicated 5 times.Treatment were given by oral with using gavage needle for ten days. Surgery was carried out on firstday after completing treatments. Examination of epidymis sperm by counting number of spermproduction, percentage of motil, live, abnormal sperm, and rank of sperm motility. In order to knowthe deferences between control and treated group, was used one way anava analysis and analysedvalue for comparing between teratment group. The treatments of root boiled water of Caesalpiniabonduc have effect significantly on percentage of abnormal sperm between dose of 0,0 μl with 24,0and 32,0 μl, also between 16,0 μl with 24,0 and 32,0 μl. Percentage of live sperem is different betweendose of 0,0 μl with 16,0 , 24,0 and 32,0 μl. Percentage of motile sperm is defferent significantlybetween dose of 0,0 μl with 24,0 and 32,0 μl. Meanwhile percentage of motile rank has differencebetween dose of 0,0 μl with 24,0 and 32,0 μl. The treatment of root boiled water of Caesalpiniabonduc has effect signifiantly on increasing percentange of sperm abnormality, decreasingpercentage of life sperm, motile and rank of motile of mice sperm.

SUMMARYTwo groups of 4 Suffolk rams were each subjected to mating pressures of 90 ejaculates (High) and 45 ejaculates (Low) over a 3-day period during summer and autumn. Every sixth (High) or every third (Low) ejaculate was collected into an artificial vagina and assessed for volume, sperm density, total sperm per ejaculate, motility and percentage of live and abnormal sperm.There was a general lowering of semen quantity and quality with successive ejaculations. Rate of change of the various measures was greater on the High than on the Low mating pressure. Volume and percentage of live sperm showed a greater decline, and percentage of abnormal sperm a greater increase, during the autumn than during the summer. Density showed a greater decline in summer than in autumn.The significance of the changes in semen characteristics observed is considered in relation to requirements for high fertility after pro-gestagen treatment.

Indonesia has agreat variety of roosters, either indigenous type as well as exotic and cross breed. The purpose of this experiment was to study the characteristics of semen from three types of Indonesian local roosters such as Merawang, Kampung and crosses Sentul Kampung with Kedu (SK Kedu). A total of 15 roosters consist of  Merawang roosters,  Kampung, and SK Kedu roosters were 5 each. The semen was collected 3 times a week by dorso-abdominal and cloaca massage method. The parameters evaluation was macroscopic characteristics consist of volume, color, consistency, and pH. Microscopic evaluation of semen such as a mass movement, sperm motility, live sperm, sperm abnormality and sperm concentration. Results of this experiment showed that semen volume of  Merawang  (0.40±0.26 mL) was higher (p<0.05) compare to Kampung (0.24±0.12 mL) or  SK Kedu (0.16±0.10 mL) but no difference on semen color, consistency and semen pH. There were no difference in the mass movement, sperm motility and live sperm as well as on sperm abnormality among three types of roosters.  Sperm  concentration of Merawang (4490 million mL-1) was significantly higher than Kampung (3245 million mL-1) and the SK Kedu roosters (3751 million mL-1). Its was conclude that Merawang roosters had good semen quality better than Kampung and SK Kedu roosters

The present study was designed to determine the effect of different levels of egg-yolk on freezability of Jakhrana buck semen. Six healthy Jakhrana bucks (BW=30 ± 2kg, age=12 ± 0.5 month) were used for semen collection. These bucks were maintained under semi-intensive system at Jakhrana Unit of C.I.R.G. Makhdoom, Mathura. A total of 48 ejaculates (6 bucks × 8 replicates) were collected twice a week using artificial vagina. Each ejaculate was divided into 4 groups (G1, G2, G3 and G4). The G1, G2, G3 and G4 were extended with Tris-egg yolk-citric acid- fructose-glycerol (TEYCFG) extenders containing 5, 10, 15 and 20% egg yolk level, respectively. Each ejaculate was evaluated for sperm motility, viability, abnormality, and hypo-osmotic swelling (HOS) response and acrosome integrity before and after freezing. At pre-freeze stage no significant (P>0.05) difference in sperm motility and viability was found among all groups. Sperm abnormality was significantly (P<0.05) higher in G4 as compared to other groups (G1, G2, G3). The HOS response and acrosomal integrity was significantly (P<0.05) higher in G1, G2 and G3 as compared to G4. However, no significant (P>0.05) difference was observed in HOS response and acrosomal integrity among G1, G2 and G3. At post thaw stage, sperm motility, viability and HOS response was significantly (P<0.05) higher in G1 and G2 as compared to G3 and G4. Sperm abnormality was significantly (P<0.05) lower in G2 as compared to other groups. The acrosomal integrity was significantly (P<0.01) higher in G1 and G2 as compared to G3 and G4. It is concluded that 10% egg yolk in Tris based extender may be the best for successful cryopreservation of Jakhrana buck semen.

A study was carried out to study the effect of freezing on cryosurvival of yak semen. Artificial insemination in yak is still in infancy. Semen cryopreservation and use of artificial insemination can be applied in yak husbandry for conservation and rapid multiplication of superior germplasm. Semen was collected from four adult yak bulls using artificial vagina method managed under uniform conditions. A total of 40 ejaculates comprising of 10 ejaculates each bull were collected following twice a week schedule and evaluated for fresh semen characteristics. The fresh yak semen characteristics viz. ejaculate volume (ml), mass activity (0-4), initial sperm motility (%), sperm concentration (x 106/ml), live sperm (%), sperm abnormality (%) and intact acrosome (%) were 3.10 ± 0.18, 3.53 ± 0.96, 83.89 ± 2.87, 1180.22 ± 42.32, 77.63 ± 4.23, 8.45 ± 3.33 and 93.61 ± 3.78 respectively. The ejaculates were diluted (1:10) with Tris extender consisting of 6.4 ml glycerol and 20 ml of fresh egg yolk. Straws were equilibrated at 5°C for 4 hours followed by exposure to liquid nitrogen vapour for 10 minutes and finally transferred to liquid nitrogen container for storage. The cryosurvival rate was studied after 7 days of storage in liquid nitrogen. The frozen semen was thawed in warm water (37°C) for 30 seconds for evaluation. Mean values of postthaw sperm motility (%), live sperm (%) and intact acrosome (%) in yaks were 55.67 ± 4.67, 65.62 ± 3.23 and 89.26 ± 3.67 respectively. In conclusion, yak semen has a better cryosurvival while freezing in tris extender with 6.4 per cent glycerol and 20 per cent egg yolk following an equilibration period of 4h.SAARC J. A total of sixteen rabbits sourced from two locations in Southeastern Nigeria were used for this study. Semen was collected from all the rabbits twice weekly for eight weeks using an artificial vagina while observation on the effect of time of collection on semen quality were also made. The results obtained indicated that time of collection had no significant effect (P > 0.05) on all the seminal characteristics measured. However, source of the animals showed significant effect (P < 0.05) on semen volume, sperm concentration, total sperm per ejaculate and proportion of abnormal sperm. Average semen volume, sperm concentration and total sperm per ejaculate were: 0.69±0.007 ml, 104.0±1.44 x 106/m1 and 71.40±0.90x106 respectively for the rabbits sourced from Port Harcourt. The corresponding values for rabbits sourced from Owerri were: 0.66±0.008m1, 96.02 ±1.63x106iml and 63.49±1.50 x 106 respectively. Abnormal sperm proportions were 11.05±0.22% and 1216 ± 032% for the rabbits sourced from Port Harcourt and Owerri respectively. There were no significant (P> 0.05) differences observed between locations in live sperm concentration and sperm motility. Live sperm concentration and sperm motility were 87.75±1.09% and 71.62±0.74% for the Port Harcourt group, while the values obtained for the Owerri group were 88.06±034% and 71.19±0.89% respectively.

It could be stated that the diluted semen of Awassi rams taken in the breeding season preserved its fertilizing abilityat different temperatures for different periods of time. The motility of spermatozoa kept at 23 vs. 8oC was checked daily. The largest spread of data was observed 24 hours after taking the semen, then the motility rate of cells showed a linear decrease. Motility results of fresh and frozen-thawed samples were compared also after heat resistance test and significant differences were found between these groups. Significant individual differences were observedin the sperm motility after heat resistance test.

An experiment was undertaken to determine the conception rate of native sheep by using Artificial Insemination with liquid ram semen. The semen was collected from ram using artificial vagina and the was stored in a refrigerator (4°C) for three days. The volume of semen was extended with egg yolk citrate diluter. A total of 63 ewes were inseminated with stored liquid semen collected from 15 rams by AV method. The total number of spermatozoa, live-dead, normal-abnormal, sperm motility and the pH was observed regularly. The average semen volume per ejaculate was 0.76-1.00ml and the sperm concentration was 2.37x109 - 4.30x109 per ejaculate. The number of normal spermatozoa and the pH was almost similar irrespective of days of storage. Number of live spermatozoa and the sperm motility were reduced with the increasing age of semen. The average conception rate (%) was obtained as 63.61, 61.90, 52.38 and 47.61 with sperm in zero, 1st, 2nd and 3rd day storage respectively. The present study aims to research the effect of age on the quality of ejaculates of rams from Synthetic population of Bulgarian dairy breed. Rams divided into two age groups were used for the experiment: G1 (2.5 years) versus G2 (6 years). Ejaculates were obtained by the method of artificial vagina and diluted with medium 6A-G. The tested ejaculates were obtained outside the breeding season. A Sperm Class Analyzer (SCA Microptic, Barcelona, Spain) was used to study sperm motility. The biochemical activity of the enzymes lactate dehydrogenase (LDH) and gamma-glutamyl transferase (GGT) was determined using an automatic Mindrai BA88 spectrophotometer. The activity of the enzymes was studied in sperm plasma, in aqueous and tritonic extract. Significant differences between the groups were found in total sperm motility (P?0.05) and LDH enzyme activity in sperm plasma (P?0.01). Regarding the efficiency of the LDH enzyme in aqueous and tritone extract, no significant differences between the groups was found. No significant differences were found between groups for GGT activity, while age had significant impact on LDH enzyme in sperm plasma.

This study was conducted to examine testosterone concentrations its relationship with the scrotal circumference and physical characteristics of semen in aceh bulls. Semen samples were collected weekly from jugular vein of three aceh bulls aged 4-5 years old for 10 weeks. Testosterone concentration was measured by enzyme-linked immunosorbent assay (ELISA) method. Semens were collected by using artificial vagina and evaluated for physical characteristics namely ejaculatory volume, pH, and sperm motility, concentration, and abnormalities. Data were analyzed using correlation-regresion test. Testosterone concentrations showed a positive correlation with scrotal circumference (r = 0.799), number of sperm (r = 0.703), sperm motility (r = 0.857) and sperm abnormalities (r = -0,877). No correlation, however, was found between testosterone concentrations with semen volume (r = 0.038) and pH (r = 0.418). It can be concluded that testosterone concentrations correlated positively with scrotal circumference, numbers of sperm, sperm of motility and sperm of abnormality.
