The 150 independently isolatedthy−mutants ofE. coliK12 Hfr 3.OSO were studied genetically and phenotypically. Variants were found among the mutants in respect to the lag period of thymineless death, and temperature sensitivity. The latter correlates with mutations located at a specific site on the genetical map.Thethylocus is located between thecysandser/glygenes, and is a linear structure where 134thymutants are distributed over more than 17 sites. The site distribution of the mutants is not regular: about a half of them (62) are localized within one site and all these are temperature-sensitive.Two further genes involving utilization of thymine—tlrandtd—were found. Mutations oftlrlead to a reduced thymine requirement (0·5 μg./ml.instead of 20 μg./ml.). A mutation oftdresults in thymidine sensitivity.This latter character is expressed when thetd-sallele is transferred intoE. coliK12, prototrophic for thymine, by conjugation. Thymidine inhibition can be reversed by the addition of any riboside to the growth medium. Both genes map at the proximal end of the Hfr 3.OSO chromosome and are linked with thethrgene. The most probable gene order is:tlr-td-thr.The following results have been obtained from14C-thymine incorporation experiments with wild-type cells, as well as withthy−tlr+andthy−tlr−cells: (1) Wild-type cells incorporate exogenous thymine extremely poorly, but incorporate thymidine better. (2) Thethy−tlr+mutants are able to incorporate thymine only when high concentration are used, but can utilize a low concentration of thymidine. (3) Thethy−mutants are able to incorporate exogenous thymine as well as thymidine at low concentration. (4) Thetlrmutation is a thymine-specific one.

ABSTRACT Mutants resistant to selenomethionine were isolated, and their properties studied. Mapping studies indicate that the mutation sites are located near the eth-1r locus in linkage group I, about ten map units away from the mating type locus. The sites of new mutation are either allelic to or very close to eth-1r. They are resistant not only to selenomethionine but also to ethionine, while the ethionine-resistant mutant, eth-1r, is sensitive to selenomethionine. The selenomethionine-resistant mutants are also temperature-sensitive mutants. However, they can grow at higher temperatures in medium containing 1 M glycerol.—It is very unlikely that the resistance is due to a change in the permeability of the membrane. Aryl sulfatase of se-metr mutants is not repressed by a high concentration of methionine (5 mM), although inorganic sulfate (2 mM) still can cause total repression. The γ-cystathionase levels of the mutants are normal, but the S-adenosylmethionine synthetase levels are only one-tenth of that observed in the wild-type strain. The heat-stability of this enzyme in the mutant is also different from that of the wild-type enzyme suggesting that the mutation might affect the structural gene of S-adenosylmethionine synthetase.

<b><i>Background/Aims:</i></b> We previously identified the Triclo1 fosmid in a functional metagenomic selection for clones that increased triclosan tolerance in <i>Escherichia coli</i>. The active enzyme encoded by Triclo1 is ucFabV. Although ucFabV is homologous to FabV from other organisms, ucFabV contains substitutions at key positions that would predict differences in substrate binding. Therefore, a detailed characterization of ucFabV was conducted to link its biochemical activity to its ability to confer reduced triclosan sensitivity. <b><i>Methods:</i></b> ucFabV and a catalytic mutant were purified and used to reduce crotonoyl-CoA in vitro. The mutant and wild-type enzymes were introduced into <i>E. coli</i>, and their ability to confer triclosan tolerance as well as suppress a temperature-sensitive mutant of FabI were measured. <b><i>Results:</i></b> Purified ucFabV, but not the mutant, reduced crotonoyl-CoA in vitro. The wild-type enzyme confers increased triclosan tolerance when introduced into <i>E. coli</i>, whereas the mutant remained susceptible to triclosan<i>. </i>Additionally, wild-type ucFabV, but not the mutant, functionally replaced FabI within living cells. <b><i>Conclusion:</i></b> ucFabV confers increased tolerance through its function as an enoyl-ACP reductase. Furthermore, ucFabV is capable of restoring viability in the presence of compromised FabI, suggesting ucFabV is likely facilitating an alternate step within fatty acid synthesis, bypassing FabI inhibition.

This study was based on determining concentration of essential and toxic heavy metal in coconut water available at a local Hazaribagh area in Dhaka, Bangladesh. All essential minerals, if present in the drinking water at high concentration or very low concentration, it has negative actions. In this study, fifteen samples and eight heavy metals were analyzed by Atomic Absorption Spectroscopy (AAS) method which was followed by wet ashing digestion method. The concentration obtained in mg/l were in the range of 0.3 to 1.5, 7.77 to 21.2, 0 to 0.71, 0 to 0.9, 0 to 0.2, 0.9 to 17.3, 0.1 to 0.9, 0 to 0.9 and 0 to 0.7 for Fe, Ni, Cu, Cd, Cr, Zn, Pb and Se respectively. From this data it was concluded that any toxic heavy metals like Cd, Cr, Pb and Ni exceed their toxicity level and some essential nutrients were in low concentration in those samples.

This study was based on to determine the concentration of macro and micro nutrients as well as toxic and nontoxic heavy metals present in the chicken feed available in Dhaka city of Bangladesh. All macro nutrients, if present in the feed at high concentration have some adverse effect, at the same time if this nutrient present in the feed at low concentration this have some adverse effect too. So that this nutrient level should be maintained at a marginal level. On the other side toxic heavy metals if present in the feed at very low concentration those can contaminate the total environment of the ecosystem. In this study six brand samples (starter, grower, finisher and layer) which was collected from different renowned chicken feed formulation industry in Bangladesh. Those samples were prepared for analysis by wet ashing and then metals were determined by Atomic Absorption Spectroscopy. It was found that 27.7 to 68.4, 57.3 to 121.9, 0.21 to 4.1, 0.32 to 2.1, 0.11 to 1.58, 0.28 to 2.11 and 0.28 to 1.78 for zinc, iron, copper, mercury, cadmium, nickel and cobalt respectively. It was found that essential macro and micro nutrients were present in the feed in low concentration on the other side mercury was present in high concentration in the feed samples.

A total of 64 beach water samples with various bacteriological quality (Grades 1 to 4) were analysed for their bacteriological and parasitological contents (E coli and Giardia cysts respectively). Results indicated that Giardia cysts were detected in less than 10% of the Grade 1 beach water samples with E coli concentrations of &lt;24/100mL. For Grades 2, 3 & 4 beach water samples, Giardia cysts were found, respectively, in 85, 50 and 64% of the samples. Except for one beach water sample which had an unusually high concentration of Giardia cysts (23 cysts/L), they were generally present at moderate concentrations (&lt;10 cysts/L) in all other beach water samples. Despite moderate levels of Giardia cysts present in beach water of different grades, the potential health risk faced by swimmers bathing in local beach water needs to be carefully assessed as Giardia is known to have a low infectious dose.

Background: The antidepressant mianserin has been shown to extend the lifespan of Caenorhabditis elegans (C. elegans), a well-established model organism used in aging research. The extension of lifespan in C. elegans was shown to be dependent on increased expression of the scaffolding protein (ANK3/unc-44). In contrast, antidepressant use in humans is associated with an increased risk of death. The C. elegans in the laboratory are fed Escherichia coli (E. coli), a diet high in protein and low in carbohydrate, whereas a typical human diet is high in carbohydrates. We hypothesized that dietary carbohydrates might mitigate the lifespan-extension effect of mianserin. Objective: To investigate the effect of glucose added to the diet of C. elegans on the lifespan-extension effect of mianserin. Methods: Wild-type Bristol N2 and ANK3/unc-44 inactivating mutants were cultured on agar plates containing nematode growth medium and fed E. coli. Treatment groups included (C) control, (M50) 50 μM mianserin, (G) 73 mM glucose, and (M50G) 50 μM mianserin and 73 mM glucose. Lifespan was determined by monitoring the worms until they died. Statistical analysis was performed using the Kaplan-Meier version of the log-rank test. Results: Mianserin treatment resulted in a 12% increase in lifespan (P<0.05) of wild-type Bristol N2 worms but reduced lifespan by 6% in ANK3/unc-44 mutants, consistent with previous research. The addition of glucose to the diet reduced the lifespan of both strains of worms and abolished the lifespan-extension by mianserin. Conclusion: The addition of glucose to the diet of C. elegans abolishes the lifespan-extension effects of mianserin.

Background Intestinal and hepatic manifestations of lupus seem to be underestimated in comparison to other major organ lesions. Although recent data point to gut-liver axis involvement in lupus, gut permeability dysfunction and liver inflammation need to be more investigated. Objective This study aims to assess fecal calprotectin, intestinal tight junction proteins and liver inflammation pathway in wild-type murine imiquimod- induced lupus. Methods C57BL/6 mice were topically treated on their right ears with 1.25 mg of 5% imiquimod cream, three times per week for six weeks. Fecal calprotectin was collected at day 0, 22 and 45. Renal, liver and intestinal pathology, as well as inflammatory markers, intestinal tight junction proteins, and E. coli protein in liver were assessed at sacrifice. Results At six weeks, lupus nephritis was confirmed on histopathology and NGAL and KIM-1 expression. Calprotectin rise started at day 22 and persists at day 45. Protein expression of Claudine, ZO-1 and occludin was significantly decreased. E. coli protein was significantly increased in liver with necro-inflammation and increased TLR4, TLR7, and pNFκB/NFκB liver expression. Conclusion This study is the first to demonstrate early fecal calprotectin increase and liver activation of TLR4- NFκB pathway in wild-type murine imiquimod-induced lupus.

Abstract We have applied a genetic system for analyzing mutations in Escherichia coli to Deinococcus radiodurans, an extremeophile with an astonishingly high resistance to UV- and ionizing-radiation-induced mutagenesis. Taking advantage of the conservation of the β-subunit of RNA polymerase among most prokaryotes, we derived again in D. radiodurans the rpoB/Rif r system that we developed in E. coli to monitor base substitutions, defining 33 base change substitutions at 22 different base pairs. We sequenced &gt;250 mutations leading to Rif r in D. radiodurans derived spontaneously in wild-type and uvrD (mismatch-repair-deficient) backgrounds and after treatment with N-methyl-N′-nitro-N-nitrosoguanidine (NTG) and 5-azacytidine (5AZ). The specificities of NTG and 5AZ in D. radiodurans are the same as those found for E. coli and other organisms. There are prominent base substitution hotspots in rpoB in both D. radiodurans and E. coli. In several cases these are at different points in each organism, even though the DNA sequences surrounding the hotspots and their corresponding sites are very similar in both D. radiodurans and E. coli. In one case the hotspots occur at the same site in both organisms.
