Benthic mapping using local aerial photo interpretation and resident taxa inventories for designing marine protected areas

Given the frequent socioeconomic, political and concomitant ecological failures of science-driven marine protected area (MPA) programmes, it is now important to design MPAs by integrating natural and social science research more comprehensively. This study shows how indigenous peoples assisted in the design of MPAs by identifying marine substrates and related resident taxa on aerial photos, information which was then incorporated into a geographical information system (GIS) database, along with dive survey data. Two questions were asked: (1) Is indigenous ecological knowledge accurate enough for mapping the benthos and associated taxa? (2) Is such an approach an appropriate way for assisting in the biological and social design of MPAs in Oceania? Conventional quadrat field dive surveys were used to measure the accuracy of substrate identification by local informants and a visual survey was used to test hypotheses formulated from local knowledge regarding the spatial distribution and relative abundance of non-cryptic species within certain benthic habitats. Equivalence rates between indigenous aerial photo interpretations of dominant benthic substrates and in situ dive surveys were 75–85% for a moderately detailed classification scheme of the benthos, which included nine locally-defined abiotic and biotic benthic classes for the MPA seabed. Similarly, the taxa inventory showed a strong correspondence between the qualitative predictions of local fisherfolk and the quantitative analysis of non-cryptic species distribution, including their relative abundance and geophysical locations. Indigenous people's predictions about the presence or absence of fish in different benthic habitats corresponded 77% and 92% of the time (depending on scoring schema) with in situ visual measurements. These results demonstrate how incorporating local knowledge of benthic heterogeneity, existing biological communities, and particular spatio-temporal events of biological significance into a GIS database can corroborate the production of scientifically reliable base resource maps for designing MPAs in an environmentally and culturally sound fashion. This participatory approach was used to design and then establish MPAs in the Roviana and Vonavona region of the Western Solomon Islands. Under appropriate conditions, interdisciplinary work can complement the design of scientific fishery management and biodiversity conservation prescriptions for coastal Oceania.

ABSTRACTHigh-throughput sequencing (HTS) is revolutionizing environmental surveys of microbial diversity in the three domains of life by providing detailed information on which taxa are present in microbial assemblages. However, it is still unclear how the relative abundance of specific taxa gathered by HTS correlates with cell abundances. Here, we quantified the relative cell abundance of 6 picoeukaryotic taxa in 13 planktonic samples from 6 European coastal sites using epifluorescence microscopy on tyramide signal amplification-fluorescencein situhybridization preparations. These relative abundance values were then compared with HTS data obtained in three separate molecular surveys: 454 sequencing of the V4 region of the 18S ribosomal DNA (rDNA) using DNA and RNA extracts (DNA-V4 and cDNA-V4) and Illumina sequencing of the V9 region (cDNA-V9). The microscopic and molecular signals were generally correlated, indicating that a relative increase in specific 18S rDNA was the result of a large proportion of cells in the given taxa. Despite these positive correlations, the slopes often deviated from 1, precluding a direct translation of sequences to cells. Our data highlighted clear differences depending on the nucleic acid template or the 18S rDNA region targeted. Thus, the molecular signal obtained using cDNA templates was always closer to relative cell abundances, while the V4 and V9 regions gave better results depending on the taxa. Our data support the quantitative use of HTS data but warn about considering it as a direct proxy of cell abundances.IMPORTANCEDirect studies on marine picoeukaryotes by epifluorescence microscopy are problematic due to the lack of morphological features and due to the limited number and poor resolution of specific phylogenetic probes used in fluorescencein situhybridization (FISH) routines. As a consequence, there is an increasing use of molecular methods, including high-throughput sequencing (HTS), to study marine microbial diversity. HTS can provide a detailed picture of the taxa present in a community and can reveal diversity not evident using other methods, but it is still unclear what the meaning of the sequence abundance in a given taxon is. Our aim is to investigate the correspondence between the relative HTS signal and relative cell abundances in selected picoeukaryotic taxa. Environmental sequencing provides reasonable estimates of the relative abundance of specific taxa. Better results are obtained when using RNA extracts as the templates, while the region of 18S ribosomal DNA had different influences depending on the taxa assayed.

Image-sharpening process integrates lower spatial resolution multispectral bands with higher spatial resolution panchromatic band to produce multispectral bands with finer spatial detail called pan-sharpened image. Although the pan-sharpened image can greatly assist the process of information extraction using visual interpretation, the benefit and setback of using pan-sharpened image on the accuracy of digital classification for mapping remain unclear. This research aimed at 1) highlighting the issue of using pan-sharpened image to perform benthic habitats mapping and 2) comparing the accuracy of benthic habitats mapping using original and pan-sharpened bands. In this study, Quickbird image was used and Kemujan Island was selected as the study area. Two levels of hierarchical classification scheme of benthic habitats were constructed based on the composition of in situ benthic habitats. PC Spectral sharpening method was applied on Quickbird image. Image radiometric corrections, PCA transformation, and image classifications were performed on both original and pan-sharpened image. The results showed that the accuracy of benthic habitats classification of pan-sharpened image (maximum overall accuracy 64.28% and 73.30% for per-pixel and OBIA, respectively) was lower than the original image (73.46% and 73.10%, respectively). The main setback of using pan-sharpened image is the inability to correct the sunglint, hence adversely affects the process of water column correction, PCA transformation and image classification. This is mainly because sunglint do not only affect object’s spectral response but also the texture of the object. Nevertheless, the pan-sharpened image can still be used to map benthic habitats using visual interpretation and digital image processing. Pan-sharpened image will deliver better classification accuracy and visual appearance especially when the sunglint is low.

Seahorse (Hippocampus spp.) has a high economic value in Bintan Island that leads to its exploitation by coastal community intensively. This study was to assess local knowledge on seahorse ecology which was captured across shoreline near the coastal communities. The research was conducted in SebongPereh village Bintan Island. The data were collected by conducting depth interviews and focus group discussion on seahorse fishermen which was selected purposively. The results revealed local fishermen have ecological knowledge related to habitat types, behavior, and monthly variation in abundance, size and reproduction pattern. The result also showed that the existence of the seahorse during the catch season moves around, follow the condition of the seaweed and tidal of sea water. This local knowledge was used by fishermen as a strategy in determining the fishing time and catchment area of a seahorse. Through such local wisdom, local fishing communities established a local institution to manage conversation and sustainability use of seahorse in waters of SebongPereh village.

Abstract A geographical information system (GIS) database was compiled of Permo-Triassic tetrapod fossils from the Karoo Supergoup in South African museum collections. This database is the first of its kind and has great time applicability for understanding tetrapod biodiversity change though time more than 200 million years ago. Because the museum catalogues all differed in recorded information and were not compliant with field capture requirements, this information had to be standardised to a format that could be utilised for archival and research application. Our paper focuses on the processes involved in building the GIS project, capturing metadata on fossil collections and formulating future best practices. The result is a multi-layered GIS database of the tetrapod fossil record of the Beaufort Group of South Africa for use as an accurate research tool in palaeo- and geoscience research with applications for ecology, ecosystems, stratigraphy and basin development.

There is a lot of uncertainty about how we pick the best invasive species management strategies to improve the environment, local economy, and human well-being, as invasive species management involves complex and multidimensional challenges. Invasive species management on inhabited islands is especially challenging, often due to perceived socio-political risks and unexpected technical difficulties. Failing to incorporate local knowledge and local perspectives in the early stages of planning can compromise the ability of decision-makers to achieve long-lasting conservation outcomes. Hence, including local knowledge and accounting for subjective stakeholder perceptions is essential for invasive species management, yet this often remains unaddressed. To address this gap, we present an application of invasive species management based on structured decision-making, and the resource allocation tool INFFER, on Minjerribah-North Stradbroke Island (Australia). We assessed the cost-effectiveness of six management scenarios, co-developed with local land managers and community groups, aimed at preserving the environmental and cultural significance of the island by eradicating European red foxes and feral cats. We further conducted a survey eliciting local stakeholders’ perspectives regarding the significance of the Island, their perception of the benefits of the proposed management scenarios, funding requirements, technical feasibility of implementation, and socio-political risk. We found that the best decisions when the budget is low are less cost-effective than when the budget is high. The best strategy focusses on control of European red fox on Minjerribah. However, our results also highlight the need for more research on feral cat management. This work demonstrates how to use a structured decision support tool, like INFFER, to assess contesting management strategies, this is particularly important when stakeholders’ perceptions regarding management outcomes are heterogeneous and uncertain.

Abstract Numerical modeling efforts in support of restoration and protection activities in coastal Louisiana have traditionally been conducted externally to any stakeholder engagement processes. This separation has resulted in planning- and project-level models built solely on technical observation and analysis of natural processes. Despite its scientific rigor, this process often fails to account for the knowledge, values, and experiences of local stakeholders that often contextualizes a modeled system. To bridge this gap, a team of natural and social scientists worked directly with local residents and resource users to develop a participatory modeling approach to collect and utilize local knowledge about the Breton Sound Estuary in southeast Louisiana, USA. Knowledge capture was facilitated through application of a local knowledge mapping methodology designed to catalog local understanding of current and historical conditions within the estuary and identify desired ecological and hydrologic end states. The results of the mapping endeavor informed modeling activities designed to assess the applicability of the identified restoration solutions. This effort was aimed at increasing stakeholder buy-in surrounding the utility of numerical models for planning and designing coastal protection and restoration projects and included an ancillary outcome aimed at elevating stakeholder empowerment regarding the design of nature-based restoration solutions and modeling scenarios. This intersection of traditional science and modeling activities with the collection and analysis of traditional ecological knowledge proved useful in elevating the confidence that community members had in modeled restoration outcomes.

The relationship (over the normal range) between work, the consumption of oxygen, glucose and lactic acid and the RQ have been studied on the heart in situ in controlled experiments. The following conclusions were reached: a) The correlation found between work and oxygen consumption was imperfect because the behavior of a heart when its work is increased is different as regards oxygen consumption when it is performing pressure-work than when it is performing an equal amount of volume-work. b) The heart rate is an important factor in determining the oxygen consumed by the heart in situ at a given work load. c) The RQ was found to be unrelated to the work level of the heart. It remained relatively constant over the normal range indicating that the nature of the metabolism is probably not altered. d) A low value of ‘efficiency’ was found in this preparation. It does not necessarily indicate an incompetent or insufficient heart. The biological significance of the term, ‘efficiency’ which actually represents merely the ratio of work done to the amount of oxygen consumed, is considered. e) The availability of glucose and lactic acid is not dependent upon the work of the heart; their availability is dependent upon the coronary flow and therefore varies with oxygen consumption. f) The consumption of exogenous glucose by the heart is related to the availability of glucose but not to its arterial blood level. The exogenous glucose consumption of the heart is not related to the cardiac oxygen consumption. g) The consumption of exogenous lactic acid by the heart shows a positive relation to its availability, to its arterial blood level and to the myocardial consumption of oxygen. It appears to be one of the substrates readily used for the work performance of the heart. h) The relative consumption of exogenous glucose and lactic acid by the heart varies with their relative availabilities. i) The significance of these findings is discussed.

ABSTRACTThree types of contaminated soil from three geographically different areas were subjected to a constant supply of benzene or benzene/toluene/ethylbenzene/xylenes (BTEX) for a period of 3 months. Different from the soil from Brazil (BRA) and Switzerland (SUI), the Czech Republic (CZE) soil which was previously subjected to intensivein situbioremediation displayed only negligible changes in community structure. BRA and SUI soil samples showed a clear succession of phylotypes. A rapid response to benzene stress was observed, whereas the response to BTEX pollution was significantly slower. After extended incubation, actinobacterial phylotypes increased in relative abundance, indicating their superior fitness to pollution stress. Commonalities but also differences in the phylotypes were observed. Catabolic gene surveys confirmed the enrichment of actinobacteria by identifying the increase of actinobacterial genes involved in the degradation of pollutants. Proteobacterial phylotypes increased in relative abundance in SUI microcosms after short-term stress with benzene, and catabolic gene surveys indicated enriched metabolic routes. Interestingly, CZE soil, despite staying constant in community structure, showed a change in the catabolic gene structure. This indicates that a highly adapted community, which had to adjust its gene pool to meet novel challenges, has been enriched.
