Short-term aerated hydration for the improvement of seed quality in Brassica oleracea L

AbstractSeeds of the Brussels sprouts cultivar Asmer Aries and the cauliflower cultivar Hipop were subjected to ageing at 20% moisture content and 45°C for 24 or 30 h, respectively; all seeds retained high germination after ageing. Aerated hydration of unaged and aged seeds of both cultivars for 4–8 h at a range of temperatures (10–30°C), followed by drying, resulted in improved performance, except that germination percentage and rate of cauliflower were lower at 10°C. Thus, all treated seeds showed greater germination rate and seedling root length than the control, which may have resulted from the advancement of the process of germination. The deleterious effect of aerated hydration at 10°C on cauliflower could be explained by damage due to rapid imbibition; seeds that had imbibed slowly to close to full imbibition (41% moisture content) before aerated hydration showed no decrease in germination. The improvement of aged seeds after aerated hydration was also revealed by higher germination after the controlled-deterioration test, which indicated less deterioration in treated seeds. Furthermore, the optimum improvements for all seeds were observed at 25°C and were greater when the water was aerated than non-aerated. These observations indicate the activation of metabolic repair processes during aerated hydration, leading to a reversal of the deterioration sustained during ageing.

A number of important factors may affect seed priming response, including seed quality. Effects of seed vigor on seed priming response were investigated using seed lots of two muskmelon (Cucumis melo L.) cultivars. Seeds of muskmelon, cvs. Mission and Top Net SR were artificially aged at 43°C for 0, 20 and 40 hours. Seeds were primed for six days in darkness at 25°C in KNO3 (0.35 mol L-1) aerated solution. Aged seeds germinated poorly at 17°C. Priming increased germination rate at 17 and 25°C and germination percentage at 17°C. An interaction effect on germination performance between vigor and priming was observed, especially at low temperature. Priming increased germination performance in seeds of low vigor, and the response was cultivar dependent.

This article describes an idea about improving the aged soybean seed quality through ozone application based on the consideration that the ozone's oxidative and reactive nature could preserve agriculture commodities during storage. Using soybean seeds that were naturally aged in room temperature storage (25±5°C) for two months, gaseous ozone's efficacy in rejuvenating the aged seeds was examined. The aged seeds were divided into three different packages: open container, polypropylene woven sack, and vacuum polyethylene plastic. Gaseous ozone at a capacity of 150 g/h was continuously exposed on packaged seeds during six months of storage under low temperature (18±5°C). The authors found that ozone in specific limit exposure could improve physiological characteristics and inhibit some chemical properties deterioration of aged soybean seeds during storage. Our technique allows for improvement in germination percentage and germination rate of aged seeds in the fourth month of ozone exposure (p<0.05). However, these physiological parameters decreased significantly in the sixth month of ozone exposure, signing that prolonged ozone exposure would lead to adverse effects due to excessive oxidation. The result also showed that ozone storage significantly retard the elevation of moisture and free fatty acid content of aged soybean seeds. The ozone effectiveness evaluation is confirmed in all packaging conditions, but the vacuum packaging offered better preservation on almost entirely seed quality parameters during storage, except for protein. This research result provides a promising technique to restore aged seed quality and can be used for better seed provision in the seed industry.

Coffee seeds are classified into intermediate seeds which require high moisture content during the storage and possess a short shelf life. Seed germination testing aims to determine the potential of a seed lot, evaluate seed quality during the storage and also to estimate the success of seedling in the nursery. This experiment aimed to determine the germination performance of several Arabica coffee varieties after being stored for six months at controlled temperature. Nine varieties of Arabica coffee, namely Abesinia-3, Andungsari-1, BLP, Gayo-2, Goiaba, Kartika-1, Kayumas, MP-3, and S-795 were used in this study. The seeds were harvested in the 2019 harvest season (September) and the germination was tested in April 2020. The germination test was carried out in laboratory by using petridish. The result showed that the tested Arabica coffee seeds still had a high germination percentage which was more than 85%. Only three varieties namely S-795, Abesinia-3, and Goiaba had low germination growth (<80%). Meanwhile, Kayumas, BLP, and Kartika-1 coffee varieties had the best germination growth with an average length of 34.19 mm; 39.79 mm and 39.41 mm, respectively. Kartika-1 variety needs 6–7 days to germinate; Kayumas, BLP, and Goiaba varieties took 7–8 days; S-795 and MP-3 took 10 days; as well as Andungsari-1, Abesinia-3, and Gayo-2 which took 10–12 days. Arabica coffee seeds that had a short germination time will had a high germination rate and otherwise. Each variety displayed a different germination performance. BLP, Kartika-1, Gayo-2 and MP-3 varieties had high potential germination, 86.67%; 89.17%; 86.67%, and 93.33%, respectively. Kayumas, BLP, and Kartika-1 varieties showed significantly longer germination, namely 34.19 mm; 39.79 mm and 39.41 mm, respectively. The fastest of average germination time was possessed by Kartika-1 variety (6.54 days) and the highest seed germination rate was also possessed by Kartika-1 variety (0.153 per day).

Laboratory tests were carried out to study the effect of different fungicides i.e. Bavistin DF, Vitavax and Genuine 50WP on the seed quality of lentil during storage condition. After processing and drying, seeds were preserved separately with three fungicides at the rate of 2.5g kg-1 of seeds and stored in earthen pots till next planting time. The seed quality i.e. moisture content (%), germination rate (%), dry weight (g) of seedlings and vigour index were measured at the end of storage period. The average germination percentage of the initial seed lot was 90% and moisture content was 9%. Among the three fungicides, Bavistin DF showed higher values for germination percentage (91.3% in 2004 and 92.7% in 2005), dry weight (0.70 g in 2004 and 0.67 g in 2005) and vigour (63.8 in 2004 and 62.1 in 2005).Jahangirnagar University J. Stratified, imbibed Douglas fir (Pseudotsugamenziesii (Mirb.) Franco) seeds were exposed to 100% ethylene for times between 0 and 366 h. Germination rate and germination percentage were increased by treatments up to 48 h. The 12-h treatment gave largest stimulation; 30% enhancement of final germination percentage over control. Treatment for 96 h caused increased germination rate for the first 5 days but reduced the germination percentage. Germinants were subject to continuous exposure to atmospheres containing 0.1 – 200 000 ppm ethylene in air, but it did not stimulate growth, and the gas was inhibitory above 100 ppm. Although some effects of high concentrations of ethylene may have been due to the lowering of oxygen supplies, this alone was insufficient to account for the full inhibitory effect. The mechanism of stimulation by short-term exposure to ethylene is discussed.

The speed of the deterioration of oil-seeds depends on conditions of the storage environment and understanding dormancy release time is important to increase the amount and uniformity in germination. Therefore, the present study was conducted to evaluate the effect of different packaging materials and storage environments on viability and dormancy of sesame. The black sesame seed was stored in three packaging materials (woven polypropylene bag, IRRI super bag, metal bin) under two environment conditions (ambient and cold storage) for eight-month (initial storage, 2-month, 4-month, 6-month and 8-month). The effect of storage environments, packaging materials and storage durations on germination percentage, germination index, seedling vigor index I and II were significant. The moisture content decreased in the early parts of storage periods and then increased in later. The moisture content of seeds in the packaging materials increased in long storage due to the effect of seed respiration and heat accumulation in a package. Higher viability was observed in ambient storage, and in a woven polypropylene bag. Because the dormancy release was slow in low- temperature condition and the carbon dioxide concentration in the airtight package inhibited the dormancy break. It also increased during storage and the earliest fully dormancy break has occurred in six-month of woven polypropylene bag storage under ambient condition. Therefore, the germination and dormancy release of sesame were influenced by storage environments packaging materials and storage durations.

Small, 25-kg quantities of moist and dry Brassica napus L. ’Tower’ rapeseed were stored in drums located outdoors for 65 wk. Quality changes occurring in the moist (10.9–15.0% moisture content) rapeseed, which was deliberately allowed to spoil, were compared to those in dry (4.3–4.9% moisture content) seed. Moist seeds in the first 7 wk had high respiration and declined in quality during the 65 wk of storage as indicated by decreased seed germination, increased leakage of seed electrolytes, and increased free fatty acid (FAV) levels. Levels of post-harvest molds, including Aspergillus candidus, A. glaucus group species, A. versicolor, Penicillium verrucosum var. cyclopium and Wallemia sebi, also increased. Mycelia of P. verrucosum var. cyclopium caused seeds of 12.9–17.8% moisture content to become cemented together at the bottom of some drums necessitating removal of samples by chipping at 65 wk. Dry seeds, originally 11.2–12.2% moisture content before drying, had low seed respiration, high germination, low levels of most post-harvest molds but also moderate levels of P. verrucosum var. cyclopium and high FAV and therefore were also of low quality. Drying of the seeds occurred too late to prevent initial rapid loss in seed quality caused by seed enzymatic action and Penicillium activity; to be effective, drying of high moisture seeds should be carried out immediately after combining.

The response of germination and early seedling growth to levels of salinity (0, 50, 100, 150, 200 and 250 mM NaCl) were examined in single seed lots of ten modern rice genotypes. Unaged and deteriorated rice seeds were germinated in rolled paper towels and in Petri dishes. Initial seed quality, final germination, germination rate and early seedling growth were assessed. The samples of the rice genotypes differed in their initial seed quality (measured in terms of Ki). The effect of deterioration varied depending upon the initial seed quality and the severity of the treatment imposed. Ageing (using the technique of controlled deterioration, CD) for up to 24 h had no effect on final germination levels. Although CD for 30 h only reduced final germination slightly, ageing for 36 or 48 h reduced it greatly. Controlled deterioration for 36 h or longer reduced the final length and the rate of extension of both the plumule and radicle. Combining information about germination in salt solution with that about seed quality enabled a distinction to be made between varieties which performed poorly because they were genetically salt-susceptible from those which germinated poorly due to poor seed quality. It is argued that the seed vigour of seed lots used in genotype evaluation should be assessed in order to avoid discarding potentially useful genotypes because of poor physiological seed quality.

Abstract Peanut (Arachis hypogaea L.) germplasm accessions in ICRISAT genebank are conserved as pods under medium-term conditions (4 C and 30% RH). Pod storage requires far greater space than seed storage and is more likely to be expensive, especially in a controlled environment. With the objective to evolve cost effective strategies for conservation, the survival of in-shell and shelled seeds of two peanut cultivars, ICGS 76 (virginia bunch) and JL24 (spanish), was studied under three different storage conditions— ambient (20-40 C and 30-80% RH), short term (23-25 C and 40-50% RH), and medium term (4 C and 30% RH). In-shell seeds had marginally greater longevity than shelled seed in all storage conditions. The differences in time for regeneration of in-shell and shelled seeds stored under medium term conditions were estimated to be less than 4 mo for both the cultivars. Because of the much reduced volume required for storage and the insignificant differences in regeneration interval, conservation of shelled seeds would be highly cost-effective under the controlled environmental conditions, as compared to in-shell seeds. Since storage at very low moisture contents was suggested as a simple and low cost option for conservation of seed lots required for short-term use, the longevity of peanut seeds (cv. ICGS 76) hermetically sealed with 3.6% moisture content was studied in comparison with seeds held at 5.8% moisture. The studies showed that peanut seeds hermetically stored at room temperature (23-25 C) with low moisture content (below 4%) could retain high germination (&gt; 85%) for up to 8 yr.

The experiment was conducted to investigate the seed quality of Indian spinach for three months stored at different containers (Plastic pot, polythene bag and cloth bag) during 2013-2014 at Seed Technology Laboratory of Bangabandhu Sheikh Mujibur Rahman Agricultural University (BSMRAU), Gazipur. The initial moisture content of the seed was 9.89% with 75.00% germination. However, after two months, the seed had a different range of increased moisture content with reduced germination percentage depending upon the packing materials. Among the three containers, seeds stored at cloth bag absorbed more moisture (14.36%) from surrounding atmosphere followed by the moisture content, 12.67% and 11.50% of polythene bag and airtight plastic pot, respectively. Seeds stored at plastic pot, polythene bag and cloth bag exhibited 65.00, 64.00 and 50.00 per cent germination, respectively. Similarly, the fresh weight and dry weight of seedling and seedling vigor were also decreased. The seeds of plastic pot were good in term of germination capacity, fresh and dry weight and vigor index in comparison to those stored at cloth bag and polythene bag.Int. J. Agril. Res.
