Currently, seed priming is reported as an efficient and low-cost approach to increase crop yield, which could not only promote seed germination and improve plant growth state but also increase abiotic stress tolerance. Salinity represents one of the most significant abiotic stresses that alters multiple processes in plants. The accumulation of polyamines (PAs) in response to salt stress is one of the most remarkable plant metabolic responses. This paper examined the effect of osmopriming on endogenous polyamine metabolism at the germination and early seedling development of Brassica napus in relation to salinity tolerance. Free, conjugated and bound polyamines were analyzed, and changes in their accumulation were discussed with literature data. The most remarkable differences between the corresponding osmoprimed and unprimed seeds were visible in the free (spermine) and conjugated (putrescine, spermidine) fractions. The arginine decarboxylase pathway seems to be responsible for the accumulation of PAs in primed seeds. The obvious impact of seed priming on tyramine accumulation was also demonstrated. Moreover, the level of ethylene increased considerably in seedlings issued from primed seeds exposed to salt stress. It can be concluded that the polyamines are involved in creating the beneficial effect of osmopriming on germination and early growth of Brassica napus seedlings under saline conditions through moderate changes in their biosynthesis and accumulation.

Abstract Transcriptionally active chromosome (TAC) is a component of protein-DNA complexes with RNA polymerase activity found in chloroplasts. Although TAC in Arabidopsis thaliana has been extensively investigated, how the rice (Oryza sativa L.) TAC complex functions remains largely unknown. We report the characterization of the mutant thermosensitive chlorophyll-deficient7 (tcd7) and the cloning of TCD7. tcd7 mutant seedlings displayed an albino phenotype specifically at low temperatures and before the four-leaf stage. We identified TCD7 by map-based cloning followed by transgenic rescue and genome editing tests, showing that TCD7 encodes the putative TAC component FRUCTOKINASE-LIKE 2 (OsFLN2). TCD7 transcripts were highly abundant in green tissues, and the protein localized to chloroplasts. In agreement with the albino phenotype, transcript levels of genes controlling chloroplast development and the establishment of photosynthetic capacity were severely reduced in tcd7 seedlings at low temperatures, but were expressed as in the wild type at high temperatures, implying that TCD7 regulates the PEP pathway and chloroplast development. Moreover, TCD7 interacted with the thioredoxin OsTRXz to form an OsTRXz-TCD7 regulatory module, which might regulate plastid transcription under cold stress. Our results demonstrate that the nucleus-encoded TAC protein TCD7 protects chloroplast development from cold stress via a TRXz-FLN regulatory module.

Abstract Iron (Fe) is an essential plant micronutrient since photosynthesis, respiration, the scavenging of reactive oxygen species and many other cellular processes depend on adequate Fe levels. Nonetheless, non-complexed Fe ions can be dangerous for cells, as they can act as a pro-oxidant. Therefore, plants possess a complex homeostatic control system for safely taking up Fe from the soil, transporting it to the various cellular destinations and for its subcellular compartmentalization. At the end of the plant’s life cycle, maturing seeds are loaded with the required amount of Fe for germination and early seedling establishment. In this review, we discuss recent findings on how the microbiota in the rhizosphere influence and interact with the strategies adopted by plants to take up iron from the soil. We also focus on the process of seed loading with Fe and take into account the Fe metabolism in wild crops’ relatives. These aspects of plant Fe nutrition can represent promising avenues for a better comprehension of the long road of Fe from soil to seeds.

AbstractWith increasing human global population, increased yield under saline conditions is a desirable trait for major food crops. Use of endophytes, isolated from halophytic hosts, seems to be an exciting approach for conferring salt tolerance to a salt sensitive crop. Therefore, in the current study, fungal endophytes were isolated from halophytic plants’ roots and their ability to withstand in vitro salt stress was evaluated. They could withstand upto 1M NaCl concentrations and this tolerance was independent of their host or tissue source. When inoculated on salt sensitive wheat seeds/seedlings several of the endophytes showed a positive impact on germination and biomass related parameters upon salt stress, both in vitro and under glasshouse conditions. One of the isolate from dicot plants (identified as Microsphaeropsis arundinis) could successfully colonize wheat and promote its growth under salt and no salt conditions. Amongst the fungal isolates that are known to be natural endophytes of wheat, Chaetomium globosum was the best performing isolate which has been reported as an effective biocontrol agent earlier. Based on the results of our preliminary study, we suggest that these fungal endophytes could prove beneficial for salt stress tolerance enhancement of wheat crop.

(1) Background: Seed treatment with potassium nitrate (KNO3) has been associated with dormancy breaking, improved germination and enhanced seedling growth and uniformity in a variety of plant species. However, the KNO3 effect seems to be dependent on plant species and treatment conditions. (2) Methods: We describe the effect of incubation of dry pea seeds with different KNO3 concentration on water uptake kinetic, early seedling growth, antioxidant metabolism and hormone profile in pea seedlings. (3) Results: Low (0.25 mM) KNO3 levels increased seedling water uptake and growth, whereas high (40 mM) levels decreased seedling growth. KNO3 treatment differentially affected the antioxidant defences. Low KNO3 levels maintained the activity of antioxidant enzymes, while high levels reduced the activity of H2O2-scavenging enzymes. KNO3 induced a progressive decline in ascorbate levels and reduced (GSH) and oxidised (GSSG) glutathione. Low KNO3 levels strongly increased GA1 and decreased ABA in both seedlings and cotyledons, resulting in a decline in the ABA/GAs ratio. (4) Conclusions: Pea seed treatment with a low KNO3 level promoted early seedling growth. In this process, an interaction among KNO3, antioxidant defences and ABA/GAs ratio is proposed.

CTP synthases (CTPS) comprise a protein family of the five members CTPS1-CTPS5 in Arabidopsis, all located in the cytosol. Specifically, downregulation of CTPS2 by amiRNA technology results in plants with defects in chlorophyll accumulation and photosynthetic performance early in development. CTP and its deoxy form dCTP are present at low levels in developing seedlings. Thus, under conditions of fast proliferation, the synthesis of CTP (dCTP) can become a limiting factor for RNA and DNA synthesis. The higher sensitivity of ami-CTPS2 lines toward the DNA-Gyrase inhibitor ciprofloxacin, together with reduced plastid DNA copy number and 16S and 23S chloroplast ribosomal RNA support this view. High expression and proposed beneficial biochemical features render CTPS2 the most important isoform for early seedling development. In addition, CTPS2 was identified as an essential enzyme in embryo development before, as knock-out mutants were embryo lethal. In line with this, ami-CTPS2 lines also exhibited reduced seed numbers per plant.

This study investigated the effects of potting media on seed germination and early seedling growth of Pterocarpus erinaceus Poir at the Forestry Nursery in Jos, Nigeria. Using Completely Randomized Design (CRD) with three replicates, laboratory-tested soil samples, top soil, sharp sand, sharp sand + top soil, sharp sand + top soil + cow dung and sharp sand + top soil + poultry droppings were used in various combinations to assess the growth parameters of P. erinaceus (germination percentage, emergence, plant height, number of leaves, length of leaves and stem diameter) for 12 weeks. Descriptive and inferential statistics were employed to analyze collected data. Result indicated that sharp sand + top soil + poultry droppings had the highest nitrogen concentration (2.19%), sharp sand + top soil + cow dung (2.07%), sharp sand + top soil (1.50%), top soil (0.72%) and Sharp sand (0.38%). Potting media with poultry droppings recorded an overall higher percentage germination of 42.9% by the end of the germination period. Analysis of variance (ANOVA) of collected data on combined soil aggregate on growth parameters indicated a significant (p<0.05) difference in plant height, number of leaves, leaf length and stem diameter. Potting with poultry dropping gave the best potting media growth results when compared to other treatments that enhanced seed germination and seedling growth of P. erinaceus. This superior observation of the poultry droppings incorporated potting mixtures over the cow dung provides an outstanding potentials to enhance P. erinaceus plantation establishment. Consequently, recommended for raising seedlings in the nursery as well as ensuring sustainable management.
