Japanese carotenoid database with α- and β-carotene, β-cryptoxanthin, lutein, zeaxanthin, lycopene, and fucoxanthin and intake in adult women

Abstract. Carotenoid intake is associated with low mortality and cancer risks; data on non-provitamin carotenoid intake is limited especially in Asians. We aimed to estimate carotenoid intake in Japanese adult women. Carotenoid content database comprises 196 food items, including 39 fruits, 87 vegetables and mushrooms, and 11 seaweeds, and was established using data from the literature and analyses of foods available in Japan. We surveyed the intake of these foods in Japanese women aged 21–56 years (n=109). Total intake of 7 carotenoids (mean±SD [range]) was 7,450±3,840 (1,160–21,300) μg/day; α-carotene, β-carotene, β-cryptoxanthin, lutein, zeaxanthin, lycopene, and fucoxanthin represented 4.3%, 23%, 3.4%, 15%, 2.0%, 39%, and 13% of total intake, respectively. Lutein intake was 1,132±686 (294–3,490) μg/day; its best sources were spinach, cucumber, chicken egg, green onion, and Chinese chives, representing 51% of total intake. Lutein can be obtained from a variety of sources. Thus, lutein intake levels did not vary widely among individuals and very few individuals consumed insufficient levels of lutein. Intake of zeaxanthin, lycopene, and fucoxanthin was 149±93 (2–479), 2,890±2,970 (0–17,100), and 980±1,230 (0–5,660) μg/day, respectively. Their intake required rich sources including chicken egg for zeaxanthin (52%); tomato products for lycopene (98%), and wakame seaweed for fucoxanthin (76%). The carotenoid content database including all food items consumed in Japan will be helpful for further investigations on carotenoid intake and its health benefits.

AbstractObjectiveThere is an increasing concern of anaemia in Japanese women, but no national trend data has existed to date. We analysed long-term national trends of anaemia in adult women.DesignSecondary analyses of 15 consecutive cross-sectional nationwide surveys conducted during the period 1989–2003. Analyses were based on 5-year intervals (1989–1993, 1994–1998, 1999–2003).SettingJapan.SubjectsThe subjects included the National Health and Nutrition Survey of Japan (NHNS-J) population. Analyses were based on 50 967 non-pregnant/non-lactating women aged 20 years and over, with complete data sets. We stratified subjects into six age groups (20–29 years, 30–39 years, 40–49 years, 50–59 years, 60–69 years and 70+ years), and three age groups (20–49 years, 50–69 years, 70+ years) for analyses on residential areas (metropolitan, cities and towns).ResultsDecreases in trends of mean haemoglobin values (intravenous) were significant in all age groups. Changes in the prevalence of anaemia were significant only in women aged 30–39 and 40–49 years. Anaemia (haemoglobin < 12.0 g dl−1) increased from 16.8% to 20.6% in women aged 30–39 years, and from 20.2% to 26.9% in women aged 40–49 years, respectively. Prevalence was highest in women aged 70 years and older in all times, but no significant change was observed.ConclusionsHaemoglobin values have declined significantly in all ages in the last 15 years. Anaemia was especially significant in women aged 30–39 years and 40–49 years. Our findings suggest that a large number of young women in Japan are at risk of anaemia, so continuous monitoring and controlling efforts of this trend are needed.

Narcissus pseudonarcissus is an important bulbous plant with white or yellow perianths and light yellow to orange-red coronas, but little is known regarding the biochemical and molecular basis related to flower color polymorphisms. To investigate the mechanism of color formation, RNA-Seq of flower of two widely cultured cultivars (‘Slim Whitman’ and ‘Pinza’) with different flower color was performed. A total of 84,463 unigenes were generated from the perianths and coronas. By parallel metabolomic and transcriptomic analyses, we provide an overview of carotenoid biosynthesis, degradation, and accumulation in N. pseudonarcissus. The results showed that the content of carotenoids in the corona was higher than that in the perianth in both cultivars. Accordingly, phytoene synthase (PSY) transcripts have a higher abundance in the coronas than that in perianths. While the expression levels of carotenoid biosynthetic genes, like GGPPS, PSY, and LCY-e, were not significantly different between two cultivars. In contrast, the carotenoid degradation gene NpCCD4 was highly expressed in white-perianth cultivars, but was hardly detected in yellow-perianth cultivars. Silencing of NpCCD4 resulted in a significant increase in carotenoid accumulation, especially in all-trans-β-carotene. Therefore, we presume that NpCCD4 is a crucial factor that causes the low carotenoid content and color fading phenomenon of ‘Slim Whitman’ by mediating carotenoid turnover. Our findings provide mass RNA-seq data and new insights into carotenoid metabolism in N. pseudonarcissus.

Refining of crude red fruit oil (CRFO) through the degumming and neutralization steps intended to produce oil free of impurities (non triglycerides) such as phospholipids, proteins, residues and carbohydrates, and also reducing the amount of free fatty acids (FFA). This study aims to determine the effect of red fruit oil purification through degumming and neutralization stages on chemical properties, fatty acid composition, carotenoid content and tocopherol of red fruit oil (RFO). The results showed that degumming of CRFO did not affect the decrease in water content, FFA levels, peroxide numbers, iodine values, carotenoids and tocopherols content; but decrease in levels of phosphorus, β-carotene and α-tocopherol. Neutralization of degummed-RFO (DRFO) did not affect the decrease in water content, iodine value, carotenoid, tocopherol and α-tocopherol; but the FFA levels, peroxide number, phosphorus and β-carotene levels decreased significantly. The fatty acid composition of RFO was dominated by unsaturated fatty acids (± 75%), which increases through degumming and neutralization stages. β-carotene is more sensitive than α-tocopherol during refining process of crude oil, but in general, this process can improve the RFO quality.

ABSTRACT Sweetpotato (Ipomoea batatas) is a root crop grown in many countries. This tuberous root is a source of energy, nutrients, and phytochemicals. In this study, bioactive compounds and physical and physicochemical qualities of sweetpotato genotypes were evaluated. Eight new genotypes of sweetpotato produced by Embrapa Hortaliças (orange-fleshed: MD09026-OF and MD09024-OF; cream-fleshed: MD09011-CF, MD09004-CF, MD10039-CF, and MD10004-CF; yellow-fleshed: MD09017-YF and MD12002-YF) and two cultivars used as controls (Beauregard and Brazlândia Roxa) were evaluated for color, soluble solids, dry matter, phenolic compounds, total carotenoids and β-carotene. Hue angles differed even between those sweetpotatoes with the same flesh color. The orange-fleshed genotypes MD09024-OF, MD09026-OF, and Beauregard, had the lowest L*, showing to be darker than the others. These sweetpotatoes also had the brightest flesh colors with higher C*. The orange-fleshed genotypes MD09026-OF and MD09024-OF were sweeter (10.55oBrix and 9.23oBrix) than Beauregard (5.12oBrix). Brazlândia Roxa had the highest dry matter content (38.05%), followed by the genotypes MD10004-CF, MD09017-YF, MD09026-OF MD10039-CF, and MD09011-CF, which showed similarity, ranging from 32.33% to 29.12%. The highest contents of total carotenoids were found for the orange-fleshed genotypes MD09026-OF (80.06 mg g-1) and MD09024-OF (70.56 mg g-1) and Beauregard (73.12 mg g-1). These same genotypes showed the highest total phenolic compounds (0.815 mg g-1 and 0.686 mg g-1, respectively). MD09026-OF showed the highest content of β-carotene (46.47 mg g-1). MD09026-OF was the most prominent genotype among those evaluated, as it showed the highest total carotenoid, β-carotene, phenolic compounds, and soluble solids content, in addition to a high dry matter content.

Efforts to increase β-carotene in cassava have been successful, but the ability of high-β-carotene cassava to prevent vitamin A deficiency has not been determined. Two studies investigated the bioefficacy of provitamin A in cassava and compared the effects of carotenoid content and variety on vitamin A status in vitamin A-depleted Mongolian gerbils (Meriones unguiculatus). Gerbils were fed a vitamin A-free diet 4 weeks prior to treatment. In Expt 1, treatments (ten gerbils per group) included 45 % high-β-carotene cassava, β-carotene and vitamin A supplements (intake matched to high-β-carotene cassava group), and oil control. In Expt 2, gerbils were fed cassava feeds with 1·8 or 4·3 nmol provitamin A/g prepared with two varieties. Gerbils were killed after 4 weeks. For Expt 1, liver vitamin A was higher (P < 0·05) in the vitamin A (1·45 (sd 0·23) μmol/liver), lower in the control (0·43 (sd 0·10) μmol/liver), but did not differ from the β-carotene group (0·77 (sd 0·12) μmol/liver) when compared with the high-β-carotene cassava group (0·69 (sd 0·20) μmol/liver). The bioconversion factor was 3·7 μg β-carotene to 1 μg retinol (2 mol:1 mol), despite 48 % cis-β-carotene [(Z)-β-carotene] composition in cassava. In Expt 2, cassava feed with 4·3 nmol provitamin A/g maintained vitamin A status. No effect of cassava variety was observed. Serum retinol concentrations did not differ. β-Carotene was detected in livers of gerbils receiving cassava and supplements, but the cis-to-trans ratio in liver differed from intake. Biofortified cassava adequately maintained vitamin A status and was as efficacious as β-carotene supplementation in the gerbil model.

Apricots are well known for the high content of bioactive compounds such as carotenoids, polyphenols, vitamins and minerals. Several studies have pointed out the chemical composition or the biological effects of apricots, but limited information are available regarding the stability of active compounds during storage or processing. The aim of this study was to determine the stability of major carotenoids in commercial dried apricots during storage.Carotenoids were extracted monthly from dried apricots kept in a dark environment, at room temperature, for twelve months. Total carotenoids were determined using the spectrophotometric method while the most relevant carotenoids were analyzed by high-performance liquid chromatography-photodiode array detection (HPLC-PDA) on a C30 column and using a gradient elution system.Initial carotenoid content of dried fruits was 6.72 mg/100g, while after six months of storage it decreased to 2.46 mg/100g. After twelve months of storage the total carotenoid content was 0.82 mg/100g, representing 20.35 % of the initial concentration. The major carotenoids identified in apricots were: all trans β-carotene, its geometrical isomers (9-cis-β-carotene; 13-cis-β-carotene; 9,13-di-cis β-carotene); β-carotene-5,8-epoxide; β-cryptoxanthin and β-cryptoxanthin palmitate. Significant decreases were observed for all pigments but all trans β-carotene appears to be the most sensitive pigment, with 15.7 % residual concentration. Although the concentrations of β-cryptoxanthin palmitate is small, it has shown significant increased stability compared to carotenes.

Vitamin A deficiency (VAD) is a serious problem in sub-Saharan Africa (SSA) and other parts of the world. Understanding the effect of marker-based improvement (MARS) of two maize synthetics (HGA and HGB) representing different heterotic groups on their agronomic performance, carotenoid content, and combining abilities could help identify suitable sources to develop divergent inbred lines for optimizing heterosis. This study involved three selection cycles each of the two synthetics and their nine varietal-cross hybrids together with a released check variety was conducted across four diverse locations in Nigeria in 2018 and 2019. Environment and hybrid effects were significant on grain yield and other agronomic traits as well as provitamin A content and other carotenoids. Genetic improvement per cycle of MARS in the parental synthetics was 15% for provitamin A, 25% for β-carotene and 26% for lutein in HGA and 4% for grain yield, 3% for zeaxanthin and 5% for α-carotene in HGB. Grain yield and agronomic traits of the two maize synthetics were controlled by additive and non-additive gene effects, while provitamin A content and other carotenoids were mainly controlled by additive gene effects. Some selection cycles which were high in grain yield and provitamin A content were identified as potential sources of new and divergent maize inbred lines in maize breeding programs. Some varietal-cross hybrids expressed significant mid-parent heterosis for grain yield and moderate mid-parent heterosis for provitamin A, β-carotene and xanthophylls. These hybrids could be commercialized at reasonable prices to small-scale farmers in rural areas that are most affected by vitamin A deficiency.

Carotenoids are important secondary metabolites in wine grapes and play a key role as potential precursors of aroma compounds (i.e., C13-norisoprenoids), which have a high sensorial impact in wines. There is scarce information about the influence of pre-harvest inactivated yeast treatment on the norisoprenoid aroma potential of grapes. Thus, this work aimed to study the effect of the foliar application of yeast extracts (YE) to Negro Amaro and Primitivo grapevines on the carotenoid content during grape ripening and the difference between the resulting véraison and maturity (ΔC). The results showed that β-carotene and (allE)-lutein were the most abundant carotenoids in all samples, ranging from 60% to 70% of total compounds. Their levels, as well as those of violaxanthin, (9′Z)-neoxanthin, and 5,6-epoxylutein, decreased during ripening. This was especially observed in treated grapes, with ΔC values from 2.6 to 4.2-fold higher than in untreated grapes. Besides this, a principal components analysis (PCA) demonstrated that lutein, β-carotene, and violaxanthin and (9′Z)-neoxanthin derivatives principally characterized Negro Amaro and Primitivo, respectively. Thereby, the YE treatment has proved to be effective in improving the C13-norisoprenoid aroma potentiality of Negro Amaro and Primitivo, which are fundamental cultivars in the context of Italian wine production.
