Improvement of Ketoacidosis in the Diabetic Rat after the Administration of the Oral Antilipolytic Agent Gr 79236

1. We assessed the effect of a novel oral antilipolytic agent, N-[(1S, trans) − 2 – hydroxycyclopentyl] adenosine (GR 79236), in experimental diabetic ketoacidosis. Ketotic rats were gavaged with GR 79236 (1 mg/kg) or water (vehicle) and their blood/plasma/serum biochemistry and haematological profile was determined. 2. We found that GR 79236 reduced the plasma non-esterified fatty acid concentration. This effect was associated with the correction of blood/plasma/serum biochemical variables (β-hydroxybutyrate, acetoacetate, triacylglycerol) directly related to diabetic ketoacidosis, and of others (cholesterol, creatinine, creatine kinase and aspartate transaminase) which are not directly related to this metabolic abnormality. 3. There was, however, no evidence of GR 79236 lowering blood glucose in this model. One possible explanation for this observation is that GR 79236 stimulated gastric emptying leading to enhanced absorption of stomach contents when compared with untreated animals.

The influence of age on serum biochemical components in the domestic rabbit was investigated. The serum total protein (g/dL), albumin (g/dL), globulin (g/dL), creatinine (mg/dL), urea mg/dL, cholesterol (mg/dL), Alkaline phosphatase (ALP; iu/l), Aspartate aminotranferase (AST; iu/l) and Alanine amimotransferase (ALT; iu/l) were assessed in 24 male rabbits at 7, 14 and 21 weeks of age to represent weaners, growers and pubertal stages. Data were analysed using descriptive statistics and analysis of variance at á . Results of 0.05 serum biochemistry revealed a steady significant increase in total protein (4.2±0.3; 5.3±0.5; 6.9±0.2), albumin (2.2±0.7; 2.8±0.4; 4.0±0.2), globulin (1.9±0.3; 2.5±0.4, 2.9±0.2), cholesterol (49.5±1.1; 56.4±4.2; 62.4±5.4) and creatinine (0.8±0.1; 1.2±0.1; 1.9±0.3) at 7, 14 and 21 weeks respectively. Serum urea levels were similar across the ages. The values recorded for ALP and AST at 7 weeks (39.6±7.5; 101.1±6.7) and 14 weeks (38.7±6.4; 104.5±8.7) were significantly higher than the 21 weeks (33.5±2.7; 95.8±5.9) while ALT at 7 weeks (27.33±5.6) was significantly lower than 14 (38.2±3.9) and 21 weeks (37.4±3.0). The age of the rabbits had a significant effect on their serum biochemical parameters. They should therefore be grouped according to age when setting serum reference values for them.

The effects of dietary replacements of fish meal with soybean meal on growth performance, heamatology and serum biochemistry in juvenile Labeo cubie were evaluated. Four hundred and fifty (450) apparently healthy juvenile Laboe cubie were distributed into one of fifteen 1000 L tanks divided into five groups and fed any of diets A (0% soybean meal); B (30% soybean meal); C (40% soybean meal); D (45% soybean meal) and diet E (75% soybean meal) for 56 days. At set time points day 7, 14, 28 and day 56, five fish were randomly selected per tank for growth assessments, heamatology and serum biochemical evaluations. Growth performance indices decreased significantly (p < 0.05) along the gradient of soybean meal inclusion and the duration of feeding. The erythrocytes count increased significantly (p < 0.05) at day 14 in fish fed soybean meal diets and decreased significantly (p < 0.05) beginning at day 28 in fish fed soybean meal-based diets. The haematocrit of fish fed diets D and E were significantly (p < 0.05) lower at day 28 compared to day 56. There were no significant variations in haemoglobin concentration and the leucocytes counts (p > 0.05). The proteins, blood glucose and the transaminases increased significantly (p < 0.05) along gradient of dietary inclusion and duration of feeding in the first 28 days while creatinine kinase activity, blood urea nitrogen and uric acid concentration remained significantly elevated (p < 0.05) in fish fed the soybean meal-based diets. The results of the study show juvenile L. cubie may tolerate up to 40% dietary inclusion of soybean meal as replacement for fish meal.

Background. Mantidis Ootheca (MO), described as the ootheca of Hierodula patellifera Serville, 1839, Tenodera angustipennis (Saussure, 1869), or Statilia maculate (Thunberg, 1784) in Korean Herbal Pharmacopoeia, is an important herbal material that has been traditionally used for treating several medical conditions including renal failure, spermatorrhea, and pediatric enuresis in Korea. Objective. The present study investigated the potential subacute toxicity of MO water extract during a 2-week repeated oral administration of doses of 0, 50, 150, or 450 mg/kg/day to C57BL/6 male mice by gavage. Methods. The following parameters were examined during the study period: mortality, clinical signs, body weight, hematology, serum biochemistry, gross findings, organ weight, and histopathology. All the mice were euthanized at the end of the treatment period. Results. No treatment-related changes in mortalities, clinical signs, body weight, gross finding, and organ weight change were detected after 14 days of oral MO extract administration. In addition, no meaningful MO extract treatment-related changes were observed in the hematological, serum biochemical, and histopathological parameters compared with the normal control group following treatment with doses of up to 450 mg/kg/day. Conclusion. Based on these findings, we concluded that treatment of mice with the water extract of MO did not result in significant toxicity and, therefore, it could be considered safe for further pharmacological studies.

The effect of Emblica officianalis and Spirulina platensis was studied in New Zealand White rabbits. Weaned rabbits of both sex were allocated to three different feeding treatments via T1 (Control), T2 (supplemented with 2% Emblica officianalis) and T3 (supplemented with 5% Sprulina platensis) in addition to ad lib. berseem fodder. The concentrate diets formulated were iso-nitrogenous and iso-caloric. The final body weight, weight gain and average daily weight gain of rabbits in all the groups followed the same trend. While, T1 and T2 were found to have better FCR than T3 supplemented feeding treatments, there were no significant changes in the serum biochemical indices, only the serum globulin value was significantly (P less than 0.05) lower (2.41 vs. 3.55, 3.10 g/dl) and albumin value was found significantly higher (4.52 vs. 3.27, 3.56 g/dl) in the groups fed spirulina supplemented diets as compared to groups fed with non supplemented diets. Amla supplementation had no effect on the serum biochemistry indices.

Serum creatine kinase, aspartate transaminase, and hydroxybutyrate dehydrogenase activities were abnormal in 76, 50, and 28% respectively of 50 patients studied within 26 hours of surgery. No patient showed clinical evidence of myocardial infarction. Creatine kinase MB isoenzyme elevation, and lactate dehydrogenase LD1 activity greater than LD2 (LD1 > LD2) were infrequent (6 and 10% respectively). No patient showed the combination of transient MB isoenzyme elevation and LD1 > LD2, although their rare association without infarction after surgery is to be anticipated.

This study was designed to investigate the effect of oral curcumin pretreatment (200 mg/kg) on isoproterenol-induced myocardial injury in rats. Isoproterenol (85 mg/kg, s.c., in two divided doses at 24 h intervals) administration induced a statistically significant increase ( P < 0.01) in serum lactate dehydrogenase, creatine kinase, aspartate transaminase, and alanine transaminase activities and significant increase ( P < 0.01) in myocardial lipid peroxides levels as compared to vehicle control rats. Furthermore, significant depletion ( P < 0.01) of myocardial endogenous antioxidants viz. superoxide dismutase, catalase, and tissue glutathione levels were also found in the pathogenic control group, that is, isoproterenol only treated animals. Curcumin (200 mg/kg) pretreatment for 20 days in isoproterenol treated rats significantly lowered ( P < 0.01) the serum lactate dehydrogenase, creatine kinase, aspartate transaminase, alanine transaminase, and myocardial lipid peroxides levels and increased the levels of myocardial endogenous antioxidants (superoxide dismutase, catalase, and tissue glutathione) as compared to pathogenic control rats. Furthermore, histological examination of rat’s heart section confirmed myocardial injury with isoproterenol administration and near normal pattern with curcumin pretreatment. The results of our study provide clear evidence that the curcumin pretreatment enhances the antioxidant defense against isoproterenol-induced oxidative myocardial injury in rats and exhibit cardioprotective property.

Food-feed competition between human and animals has necessitated farmers to turn to alternative feed resources such as Acha Offal. Forty eight rabbits of mixed breeds and similar live weight were utilized in an experiment to evaluate the effect of feeding acha offal meal- based diets supplemented with Maxigrain enzyme on their haematology and serum biochemistry. The experiment had two phases (weaner and grower) each lasting for 42 days. Six treatments diets namely T1, T2, T3, T4, T5 and T6 were compounded to be iso-caloric (2700kcal/kg) and iso-nitrogenous (18% crude protein) for the weaner phase while (15% crude protein) and isocaloric (2500kcal/kg) for the grower phase. The Acha Offal (AO) was included at 0, 15 and 30% while the enzyme was supplemented at 0 and 200ppm. The experiment was arranged in a 2x3 factorial fitted into a Completely Randomized Design. Each treatment was replicated 4 times having two rabbits per replicate. Feed and water were provided to the animals daily and all standard routine management practices were strictly observed throughout the experimental period. At the end of the feeding trial, blood samples were collected from each animal for blood analyses. The results show that there was no significant (P>0.05) variation on the haematological and biochemical parameters due to enzyme or AO effect. In the growing phase, main effect of AO was not significant (P>0.05) on the haematological parameters except on white blood cell (WBC) and mean cell haemoglobin concentration (MCHC). Effect of enzymes on growing rabbit was not significantly different (P>0.05) except on WBC at 0ppm (5.75 x109/L) and 200ppm (6.60 x109/L) and on MCHC at 0ppm (33.27 x109/L) and 200ppm (33.20 x109/L). The Interactive effect of AO and enzyme showed that T2 had the highest WBC (8.80 x109/L) while T4 had the lowest MCHC (33.10 x109/L). Serum biochemical parameters were not affected by Acha offal except phosphorus where there was a significant (P<0.05) reduction in 30% AO (1.35g/dL). Enzymes effect showed that animals on 0ppm had significantly (P<0.05) higher phosphorus (1.83g/dl) than 200ppm (1.38g/dL). Acha offal supplemented with Maxigrain enzyme can be included in the diets of grower rabbits without adverse effect on haematological and serum biochemical indices.     La concurrence entre les humains et les animaux en matière d'aliments pour animaux a obligé les agriculteurs à se tourner vers d'autres ressources alimentaires comme 'acha'. Quarante-huit lapins de races mixtes et de poids réel similaire ont été utilisés dans une expérience pour évaluer l'effet de l'alimentation d'acha complétés par l'enzyme 'Maxigrain' sur leur hématologie et biochimie sérique. L'expérience a eu deux phases (sevreur et producteur) chacune durant 42 jours. Six régimes de traitements, à savoir T1, T2, T3, T4, T5 et T6, ont été composés pour être iso-caloriques (2700kcal/kg) et iso-azotés (18% de protéines brutes) pour la phase de sevrage tandis que (15% de protéines brutes) et isocaloriques (2500 kcal/kg) pour la phase de croissance. L'alimentation d'acha (AO) a été inclus à 0, 15 et 30% tandis que l'enzyme a été complétée à 0 et 200ppm. L'expérience a été organisée dans un factorial 2x3 intégré dans une conception complètement randomisée. Chaque traitement a été reproduit 4 fois ayant deux lapins par réplique. Des aliments et de l'eau ont été fournis quotidiennement aux animaux et toutes les pratiques de gestion de routine standard ont été strictement observées tout au long de la période expérimentale. À la fin de l'essai d'alimentation, des échantillons de sang ont été prélevés sur chaque animal pour des analyses sanguines. Les résultats montrent qu'il n'y avait aucune variation significative (P>0.05) sur les paramètres hématologiques et biochimiques dus à l'effet d'enzyme ou d' 'AO'. Dans la phase de croissance, l'effet principal de l' 'AO' n'était pas significatif (P>0,05) sur les paramètres hématologiques, sauf sur les globules blancs (le 'WBC') et la concentration moyenne d'hémoglobine cellulaire (le 'MCHC'). L'effet des enzymes sur la croissance du lapin n'était pas significativement différent (P>0,05) excepté sur WBC à 0ppm (5.75 x109/L) et 200ppm (6.0ppm 60 x109/L) et sur le 'MCHC' à 0ppm (33.27 x109/L) et 200ppm (33.20 x109/L). L'effet interactif de l'AO et de l'enzyme a montré que T2 avait le 'WBC' le plus élevé (8,80 x109/L) tandis que T4 avait le 'MCHC' le plus bas (33,10 x109/L). Les paramètres biochimiques du sérum n'ont pas été affectés par le phosphore d'alimentation d'acha sauf où il y avait une réduction significative (P<0.05) de 30%AO (1.35g/dL). L'effet d'enzymes a prouvé que les animaux sur 0ppm ont eu sensiblement (P<0.05) le phosphore plus élevé (1.83g/dl) que 200ppm (1.38g/dL). L'alimentation d'Acha complété avec l'enzyme 'Maxigrain' peut être inclus dans les régimes des lapins 'grower' sans effet défavorable sur les indices biochimiques hématologiques et sériques.

Exercise has been suggested as a powerful intervention for health care and fitness management in humans; however, few studies have demonstrated the benefits of exercise training in dogs. The purpose of this study was to examine the effects of exercise training on heart rate (HR), bone mineral density (BMD), muscle volume (MV), and hematological and serum biomarkers in dogs. Six healthy beagles completed the interval treadmill exercise, developed on the basis of the FITT principle, two times a week for 12 weeks. To evaluate the physiological parameters, the HR values were analyzed using the Polar H10 system during the entire exercise period. At pre-and post-exercise, quantitative computed tomography and hematological and serum biochemical parameters were analyzed. The interval exercise resulted in a normal HR response and no adverse behavioral or physiological effects on the dogs. We showed that exercise improved BMD in the femur (541.6 ± 16.7 vs. 610.2 ± 27.8 HA, p < 0.01) and increased serum total alkaline phosphatase (TALP; 68.6 ± 9.2 vs. 81.3 ± 17.2, p < 0.01), aspartate aminotransferase (23.5 ± 1.0 vs. 33.5 ± 1.6, p < 0.01), and creatine kinase (114.8 ± 5.3 vs. 214.0 ± 20.8, p < 0.01) levels. There was a positive relationship between BMD and TALP (femur: r = 0.760, p = 0.004; vertebrae: r = 0.637; p = 0.025). Our findings suggest that interval exercise training is beneficial to increase BMD in the femur, and an increased TALP level would be a concomitant mechanism for enhancing BMD with exercise in dogs.
