In Finland, all laboratories carrying out diagnostics of infectious diseases in humans are approved by the Regional State Administrative Agencies and are obligated to participate in External Quality Assurance rounds. Performance in these rounds is thought to reflect the quality of laboratory work. In the 6-year study period, 17 Finnish laboratories received 48 simulated faecal specimens for the culturing of diarrhoeal pathogens, yielding altogether 586 faecal culture External Quality Control specimens and 581 reports. The results were correct in 92% of all reports and in 67% of all specimens. False-negative Salmonella results were given for 2 of the 18 specimens, one with biochemically atypical Salmonella strain and the other with a low count of Salmonella cells. False-negative Shigella report was given for 6 of the 7 specimens in some participating laboratory. Detection of all common faecal pathogens is especially relevant to patient safety, public health, and epidemiological surveillance.

Die casting technology is applied widely for mass production of non-ferrous metal parts. How to guarantee the quality of die castings is always a concerned topic. A quality control system for die casting machine was developed based on statistical analysis of technology parameters. Firstly the original data is acquired from die casting machine and the technology curves are displayed. Secondly, the technology parameters are identified automatically and a database of technology parameters is established. Thirdly, a statistical analysis based on the database is performed. The statistics parameters of expecta -tion and deviation are figured out and the probability distribution diagrams for technology parameters are displayed. Finally, Tolerances for technology parameters are figured out and then the system will monitor the die casting process according to the tolerances. A practical system realization in a die casting machine is demonstrated by its monitoring interfaces. Statistical information and on-line monitoring provided by this system are helpful to guarantee the quality of die castings.

The Southern Ocean Carbon and Climate Observations and Modeling (SOCCOM) project has deployed 194 profiling floats equipped with biogeochemical (BGC) sensors, making it one of the largest contributors to global BGC-Argo. Post-deployment quality control (QC) of float-based oxygen, nitrate, and pH data is a crucial step in the processing and dissemination of such data, as in situ chemical sensors remain in early stages of development. In situ calibration of chemical sensors on profiling floats using atmospheric reanalysis and empirical algorithms can bring accuracy to within 3 μmol O2 kg–1, 0.5 μmol NO3– kg–1, and 0.007 pH units. Routine QC efforts utilizing these methods can be conducted manually through visual inspection of data to assess sensor drifts and offsets, but more automated processes are preferred to support the growing number of BGC floats and reduce subjectivity among delayed-mode operators. Here we present a methodology and accompanying software designed to easily visualize float data against select reference datasets and assess QC adjustments within a quantitative framework. The software is intended for global use and has been used successfully in the post-deployment calibration and QC of over 250 BGC floats, including all floats within the SOCCOM array. Results from validation of the proposed methodology are also presented which help to verify the quality of the data adjustments through time.

We used an evolutionary test to ask whether the congression of chromosomes to the spindle equator is important in itself or just a mitotic happenstance. If congression matters, then it might evolve if absent initially. Previous workers established that newly made trivalents, meiotic units of three chromosomes, generally do not congress to the spindle equator. Instead, these young trivalents lie close to the pole to which two of the three chromosomes are oriented. We studied ancient sex-chromosome trivalents that arose hundreds of thousands to several million years ago in several species of praying mantids and one grasshopper. All these old trivalents lie near the spindle equator at metaphase; some of them congress as precisely to the equator as the ordinary chromosomes in the same cells. We conclude that congression evolved independently two or three times in the materials studied. Therefore, the metaphase position of chromosomes midway between the poles appears to matter, but why? In the praying mantids, the evident answer is that metaphase is a quality-control checkpoint. Sometimes the three chromosomes are not associated in a trivalent but rather are present as a bivalent plus an unpaired chromosome, which lies near one pole. Earlier workers showed that such cells are blocked in metaphase and eventually degenerate; this prevents the formation of sperm with abnormal combinations of sex chromosomes. We suggest that the quality-control system would have trouble distinguishing an unpaired chromosome from an uncongressed, newly arisen trivalent, both of which would lie near a spindle pole. If so, the confused quality-control system would block anaphase imprudently, causing a loss of cells that would have produced normal sperm. Hence, we conclude that the congression of the trivalent to the equator probably evolved along with the metaphase quality-control checkpoint. The mechanism of congression in old trivalents is uncertain, but probably involves an interesting force-sensitive regulation of the motors associated with particular chromosomes. We also examined the congression of two newly made quadrivalents when they orient with three kinetochores to one pole and one to the other. As others have described, one of these quadrivalents does not congress, while the other quadrivalent comes closer than expected to the spindle equator. Such variation in the extent of congression may provide materials on which natural selection can act, leading to the evolution of congression. The trivalents of praying mantids are attractive materials for further studies of the mechanism of congression and of the idea that metaphase is a checkpoint for progression through the cell cycle.
