Relationships between the diet selected by grazing sheep and the herbage on offer

Oesophageal extrusa samples were collected from sheep grazing monospecific swards of four temperate perennial grasses. The in vitro digestibility and the proportion of green plant material in the extrusa samples were related exponentially to the yield of herbage on offer. The digestibility of ingesta was highly correlated with the yield of green herbage. When the yield of green herbage exceeded c. 550 kg/ha the sheep selected a diet more digestible than the mean of the green plant material on offer, and when there were low amounts of green herbage available the ingesta was less digestible than the green plant material on offer because of the low proportion of green plant material eaten. A relationship between the proportion of green plant material in extrusa samples and the green herbage yield was determined for each species, and no significant difference in regressions between species was found. It is suggested that the pooled relationship obtained in this study may be used to predict the proportion of green plant material in the ingesta of sheep over a range of grass species and yields.

Single plots of wheat, oats and barley stubbles were grazed by sheep and cattle in common on a 19 day rotation for 76 days. Initially the stubbles contained 600-1200 kg dry matter ha-1 of green weeds and 3300-4800 kg dry matter ha-1 of cereal residues. Serial estimates were made of diet selection, in vitro digestibility and intake by both species. Sheep consistently selected a diet with a higher proportion of green material than did cattle, but neither species utilized the crop residues to any extent. The overall mean digestibilities of the diet were 68 and 54 per cent fop sheep and cattle respectively (P < 0.01) ; the mean N concentration in the faeces of sheep was 0.64 percentage units higher than for cattle (P < 0.01). Differences in selection by the animals were greatest at the lowest level of green plant material. Estimates of the mean daily intakes of DOM (g kg-1) per unit of metabolic liveweight (W0.9) were 32 and 15, and of dead plant material dry matter (g kg-1) 4.2 and 13.2 for sheep and cattle respectively. There were no differences in selection between stubble species. The results are discussed in relation to maximizing the use of cereal stubbles for animal production.

Over the period 1952-1954 there was no significant difference in the yield of herbage produced annually by the addition to a Phalaris tuberosa L.–Trifolium subterraneum L. pasture mixture, of any one of the following grasses: Agropyron obtusiusculum Lange., Bromus coloratus Steud., Bromus inermis Leyss., Dactylis glomerata L., or Festuca arundinacea Schreb. There were indications of a small change in seasonal production where certain grasses, notably D. glomerata, were included in the mixture. Over the 3-year period the proportion of the sown grass component, in the mixtures where either D. glomerata, B. inermis, or B. coloratus were included, increased by a greater amount than where the simple mixture of phalaris and subterranean clover was used. The addition of each grass also lessened the amount of P. tuberosa in the sown grass component of the yield. In the third year, despite the varying proportions of the phalaris and associated sown grass species, the mean population of the sown perennial grasses in each treatment did not differ significantly from the mean figure of 1.34 plants/sq. lk. The increased production of the sown grass yield component following the association of certain of the above species with P. tuberosa suggests that the latter does not fully exploit the environment. The principle of including another perennial grass when sowing phalaris and subterranean clover might have wide application as a means of combatting "phalaris staggers".

Foxtail millet is considered a ‘smart food’ because of nutrient richness and resilience to environments. A diversity panel of 92 foxtail millet landraces preserved by Taiwan indigenous peoples containing amylose content (AC) in the range of 0.7% to 16.9% exhibited diverse physiochemical properties revealed by a rapid viscosity analyzer (RVA). AC was significantly correlated with 5 RVA parameters, and some RVA parameters were also highly correlated with one another. In comparison to rice, foxtail millet contained less starch (65.9–73.1%) and no significant difference in totals of resistant starch (RS), slowly digestible starch (SDS), hydrolysis index (HI), and expected glycemic index (eGI) according to in vitro digestibility assays of raw flour with similar AC. RS was significantly positively correlated with AC and four RVA parameters, cold paste viscosity (CPV), setback viscosity (SBV), peak time (PeT), and pasting temperature (PaT), implying that suitable food processing to alter physicochemical properties of foxtail millet might mitigate hyperglycemia. This investigation of pasting properties and digestibility of diverse foxtail millet germplasm revealed much variation and showed potential for multi-dimensional utilizations in daily staple food and food industries.

SummaryThe plasma tissue factor (TF) concentration was correlated to factor VII concentration (FVIIag) and factor VII activity (FVIIc) in 498 healthy volunteers ranging in age from 17 to 64 years. Immunoassays using monoclonal antibodies (mAbs) were developed for the determination of TF and FVIIag in plasma. The mAbs and the test systems were characterized. The mean value of the TF concentration was 172 ± 135 pg/ml. TF showed no age- and gender-related differences. For the total population, FVIIc, determined by a clotting test, was 110 ± 15% and the factor VIlag was 0.77 ± 0.19 μg/ml. FVII activity was significantly increased with age, whereas the concentration demonstrated no correlation to age in this population. FVII concentration is highly correlated with the activity as measured by clotting assay using rabbit thromboplastin. The ratio between FVIIc and FVIIag was not age-dependent, but demonstrated a significant difference between men and women. Between TF and FVII we could not detect a correlation.

Abstract Objectives The study aimed to evaluate and compare various commercially available local anesthetic solutions. Materials and Methods A total of 150 commercially available local anesthetic cartridges of similar composition (2% lidocaine with epinephrine 1:100,000) were randomly collected and divided into 3 groups. The designations of groups were selected from their product names such that each group consisted of 60 cartridges. Group S (Septodont, France) Group M (Medicaine, Korea) and Group H (HD-Caine, Pakistan). The samples were divided into five sub-groups, each consisting of 10 cartridges from each group to investigate each parameter. Results The acquired data was statistically analyzed and compared (using SPSS version 12). Compositional analysis revealed a non-significant (P>0.05) difference when the three Groups were compared with standard lidocaine and epinephrine solutions. The mean pH values of samples from group S, M and H respectively fell within the range of pH values of commercially available solutions. Non-significant difference in EPT values of Group S and H was found when efficacy was compared (p = 0.3), however a significant difference (p < 0.01) was observed in contrast to Group M. Anti-bacterial activity was observed in all the group and a non-significant difference in cell viability values of Group S and M was found (p = 0.6), while the difference was significant in comparison to Group H. Conclusion Within the limitations of these investigations, it appears that the properties of different manufacturers fall within the recommended ranges as mentioned in literature and do not appear to be statistically different in the variables we have tested.

Abstract The use of native entomopathogenic nematodes as biocontrol agents is a strategy to decrease the environmental impact of insecticides and achieve sustainable agriculture crops. In this study, the effect of the surface culture of Steinernema sp. JAP1 over two solid media at 23–27°C on infective juvenile (IJ) production and pathogenicity against Galleria mellonella larvae were investigated. First, the bacterial lawn on the surface of the media with egg yolk (P2) or chicken liver (Cl) were incubated in darkness at 30°C for 48 and 72 h, and 100 surface-sterilized IJs were added. Four harvests were conducted within the next 35 days and the mean accumulated production was superior on Cl (210 × 103 IJs) than on P2 (135 × 103 IJs), but the productivity decreased up to 10% when the incubation time of the bacterial lawn was of 72 h. The mean pathogenicity of in vitro- and in vivo-produced IJs were of 47–64% and 31%, respectively. It is worth noting that none of the two solid media had a statistically significant difference in IJ pathogenicity. Considering that the maximum multiplication factor of IJs on solid media was 2108 and that the pathogenicity against G. mellonella was outstanding, Steinernema sp. has a good potential for in vitro mass production.

In response to the need to find better adapted and more persistent perennial pasture plants for the dryland pastures in the cool-temperate low to medium rainfall (500-700 mm) regions, over 1000 accessions representing 24 species of perennial legumes and 64 species of perennial grasses, were introduced, characterised and evaluated for production and persistence under sheep grazing at sites throughout Tasmania. The work has identified four alternative legume species in Talish Clover (Trifolium tumens). Caucasian Clover (T. ambiguum), Stoloniferous Red Clover (T. pratense var. stoloniferum), Lucerne x Yellow Lucerne Hybrid (Medicago sativa x M.sativa subsp. falcata); and two grass species in Coloured Brome (Bromus coloratus) and Hispanic Cocksfoot (Dactylis glomerata var hispanica). The objective of the present study was to evaluate the effect of hyaluronan (HA) during IVM on meiotic maturation, embryonic development, and the quality of oocytes, granulosa cells (GC), and obtained blastocysts. COCs were maturedin vitroin control medium and medium with additional 0.035% or 0.07% of exogenous HA. The meiotic maturity did not differ between the analysed groups. The best rate and the highest quality of obtained blastocysts were observed when 0.07% HA was used. A highly significant difference (P<0.001) was noted in the mean number of apoptotic nuclei per blastocyst and in the DCI between the 0.07% HA and the control blastocysts (P<0.01). Our results suggest that addition of 0.035% HA and 0.07% HA to oocyte maturation media does not affect oocyte nuclear maturation and DNA fragmentation. However, the addition of 0.07% HA during IVM decreases the level of blastocysts DNA fragmentation. Finally, our results suggest that it may be risky to increase the HA concentration during IVM above 0.07% as we found significantly higherBaxmRNA expression levels in GC cultured with 0.07% HA. The final concentration of HA being supplemented to oocyte maturation media is critical for the success of the IVP procedure.

SummaryGroups of lactating cows and heifers were milked four times daily in two diagonally opposed glands for 4 weeks, and the effects on milk yield studied relative to twice-daily milked glands as controls. Mammary enzyme activities, in vitro synthesis rates of milk constituents and histological scoring were determined in mammary biopsy samples obtained at the end of this period. These were used for assessment of mammary function. Frequent milking increased milk yield only in the treated glands, the contralateral control glands continuing to decline in yield at ~ 2%/week. There was no significant difference in response between cows and heifers; the mean increase in yield was 10·4%. The rate of decline in milk yield tended to decrease with frequent milking, to ~ 1%/week. Consequently the yield of the treated glands continued to be elevated above that of the controls for some time after reversion to overall twice daily milking. Milk protein content was increased slightly by frequent milking. Mammary enzyme activities were ~ 18% higher in the treated glands than in the controls. Synthesis rates of lactose, casein and total protein were unaffected by milking frequency, but were all lower in the gland selected for the second biopsy, reflecting the reduction in milk yield caused by the first biopsy. DNA synthesis was increased by milking frequency, as were the size and number of epithelial cells in histological sections.

In a study that included C4 tropical grasses, C3 temperate grasses and C3 pasture legumes, in vitro dry matter digestibility of extrusa, measured as in vitro dry matter loss (IVDML) during incubation, compared with that of the forage consumed, was greater for grass extrusa but not for legume extrusa. The increase in digestibility was not caused by mastication or by the freezing of extrusa samples during storage but by the action of saliva. Comparable increases in IVDML were achieved merely by mixing bovine saliva with ground forage samples. Differences were greater than could be explained by increases due to completely digestible salivary DM. There was no significant difference between animals in relation to the saliva effect on IVDML and, except for some minor differences, similar saliva effects on IVDML were measured using either the pepsin–cellulase or rumen fluid–pepsin in vitro techniques. For both C4 and C3 grasses the magnitude of the differences were inversely related to IVDML of the feed and there was little or no difference between extrusa and feed at high digestibilities (>70%) whereas differences of more than 10 percentage units were measured on low quality grass forages. The data did not suggest that the extrusa or saliva effect on digestibility was different for C3 grasses than for C4 grasses but data on C3 grasses were limited to few species and to high digestibility samples. For legume forages there was no saliva effect when the pepsin–cellulase method was used but there was a small but significant positive effect using the rumen fluid–pepsin method. It was concluded that when samples of extrusa are analysed using in vitro techniques, predicted in vivo digestibility of the feed consumed will often be overestimated, especially for low quality grass diets. The implications of overestimating in vivo digestibility and suggestions for overcoming such errors are discussed.
