An analysis of the role of the ShSUT1 sucrose transporter in sugarcane using RNAi suppression

The role of ShSUT1 in sucrose mobilisation and storage in sugarcane was investigated by employing RNAi technology to reduce the expression of this gene. Transcript profiling in non-transformed plants showed an alignment between expression and sucrose concentration, with strongest expression in source leaves and increasing expression through the daylight period of a diurnal cycle. Five transgenic plant lines were produced with reduced ShSUT1 expression ranging from 52 to 92% lower than control plants. Differential suppression of ShSUT1 sequence variants in the highly polyploid sugarcane genome were also investigated. Amplicon sequencing of the ShSUT1 variants within the transgenic lines and controls showed no preferential suppression with only minor differences in the proportional expression of the variants. A range of altered sugar, fibre and moisture contents were measured in mature leaf and internode samples, but no phenotype was consistently exhibited by all five transgenic lines. Phenotypes observed indicate that ShSUT1 does not play a direct role in phloem loading. ShSUT1 is likely involved with retrieving sucrose from intercellular spaces for transport and storage.

The article explores the role of antropoetonyms in the reader’s “horizon of expectation” formation. As a kind of “text in the text”, antropoetonyms are concentrating a large amount of information on a minor part of the text, reflecting the main theme of the work. As a “text” this class of poetonyms performs a number of functions: transmission and storage of information, generation of new meanings, the function of “cultural memory”, which explains the readers’ “horizon of expectations”. In analyzing the context of the literary work we should consider the function of antropoetonyms in vertical context (the link between artistic and other texts, and the groundwork system of culture), as well as in the context of the horizontal one (times’ connection realized in the communication chain from the word to the text; the author’s intention). In this aspect, the role of antropoetonyms in the structure of the literary text is extremely significant because antropoetonyms convey an associative nature, generating a complex mechanism of allusions. It’s an open fact that they always transmit information about the preceding text and suggest a double decoding. On the one hand, the recipient decodes this information, on the other – accepts this as a sort of hidden, “secret” sense.

Alternative isoforms of exon II HTR2A are associated with the development of mental pathologies and is applicable to predict antipsychotic therapy outcome.

Regardless of advances in detection and treatment, breast cancer affects about 1.5 million women all over the world. Since the last decade, genome-wide association studies (GWAS) have been extensively conducted for breast cancer to define the role of miRNA as a tool for diagnosis, prognosis and therapeutics. MicroRNAs are small, non-coding RNAs that are associated with the regulation of key cellular processes such as cell multiplication, differentiation, and death. They cause a disturbance in the cell physiology by interfering directly with the translation and stability of a targeted gene transcript. MicroRNAs (miRNAs) constitute a large family of non-coding RNAs, which regulate target gene expression and protein levels that affect several human diseases and are suggested as the novel markers or therapeutic targets, including breast cancer. MicroRNA (miRNA) alterations are not only associated with metastasis, tumor genesis but also used as biomarkers for breast cancer diagnosis or prognosis. These are explained in detail in the following review. This review will also provide an impetus to study the role of microRNAs in breast cancer.

The role of certain viruses in malignant brain tumor development remains controversial. Experimental data demonstrate that human herpesviruses (HHVs), particularly cytomegalovirus (CMV), Epstein–Barr virus (EBV) and human herpes virus 6 (HHV-6), are implicated in brain tumor pathology, although their direct role has not yet been proven. CMV is present in most gliomas and medulloblastomas and is known to facilitate oncomodulation and/or immunomodulation, thus promoting cancer cell proliferation, invasion, apoptosis, angiogenesis, and immunosuppression. EBV and HHV-6 have also been detected in brain tumors and high-grade gliomas, showing high rates of expression and an inflammatory potential. On the other hand, due to the neurotropic nature of HHVs, novel studies have highlighted the engagement of such viruses in the development of new immunotherapeutic approaches in the context of oncolytic viral treatment and vaccine-based strategies against brain tumors. This review provides a comprehensive evaluation of recent scientific data concerning the emerging dual role of HHVs in malignant brain pathology, either as potential causative agents or as immunotherapeutic tools in the fight against these devastating diseases.

AbstractAside from their canonical role in hemostasis, it is increasingly recognized that platelets have inflammatory functions and can regulate both adaptive and innate immune responses. The main topic this review aims to cover is the proinflammatory effects and side effects of platelet transfusion. Platelets prepared for transfusion are subject to stress injury upon collection, preparation, and storage. With these types of stress, they undergo morphologic, metabolic, and functional modulations which are likely to induce platelet activation and the release of biological response modifiers (BRMs). As a consequence, platelet concentrates (PCs) accumulate BRMs during processing and storage, and these BRMs are ultimately transfused alongside platelets. It has been shown that BRMs present in PCs can induce immune responses and posttransfusion reactions in the transfusion recipient. Several recent reports within the transfusion literature have investigated the concept of platelets as immune cells. Nevertheless, current and future investigations will face the challenge of encompassing the immunological role of platelets in the scope of transfusion.

AbstractWith global climate change, plants are frequently being exposed to various stresses, such as pathogen attack, drought, and extreme temperatures. Transcription factors (TFs) play crucial roles in numerous plant biological processes; however, the functions of many tomato (Solanum lycopersicum L.) TFs that regulate plant responses to multiple stresses are largely unknown. Here, using an RNA-seq approach, we identified SlNAP1, a NAC TF-encoding gene, which was strongly induced by various stresses. By generating SlNAP1 transgenic lines and evaluating their responses to biotic and abiotic stresses in tomato, we found that SlNAP1-overexpressing plants showed significantly enhanced defense against two widespread bacterial diseases, leaf speck disease, caused by Pseudomonas syringae pv. tomato (Pst) DC3000, and root-borne bacterial wilt disease, caused by Ralstonia solanacearum. In addition, SlNAP1 overexpression dramatically improved drought tolerance in tomato. Although the SlNAP1-overexpressing plants were shorter than the wild-type plants during the early vegetative stage, eventually, their fruit yield increased by 10.7%. Analysis of different hormone contents revealed a reduced level of physiologically active gibberellins (GAs) and an increased level of salicylic acid (SA) and abscisic acid (ABA) in the SlNAP1-overexpressing plants. Moreover, EMSAs and ChIP-qPCR assays showed that SlNAP1 directly activated the transcription of multiple genes involved in GA deactivation and both SA and ABA biosynthesis. Our findings reveal that SlNAP1 is a positive regulator of the tomato defense response against multiple stresses and thus may be a potential breeding target for improving crop yield and stress resistance.

This study illuminates an understudied pathway through which disadvantage is reproduced in the rental housing market: the housing search, application, and tenant screening process. Using in-depth interviews with 25 housing-seekers with criminal conviction records, past evictions, and damaged credit histories, this article examines the direct role of the rental housing search and application process in reproducing economic precarity and social disadvantage among renters with discrediting background records, beyond delimiting their housing options. Its findings suggest that navigating the housing search from a position of acute market disadvantage comes with significant costs for this population, including the financial burden of repeated application fees and the psychological strains associated with the specter of indefinite housing insecurity. The findings also demonstrate how the housing search process may undermine the willingness of stigmatized renters to contest exploitative or unlawful rental practices by reinforcing awareness of their degraded status in the rental market.
