AbstractIn this study, the extraction conditions extracted maximize amounts of phenolic and bioactive compounds from the fruit extract of Ficus auriculata by using optimized response surface methodology. The antioxidant capacity was evaluated through the assay of radical scavenging ability on DPPH and ABTS as well as reducing power assays on total phenolic content (TPC). For the extraction purpose, the ultrasonic assisted extraction technique was employed. A second-order polynomial model satisfactorily fitted to the experimental findings concerning antioxidant activity (R2 = 0.968, P < 0.0001) and total phenolic content (R2 = 0.961, P < 0.0001), indicating a significant correlation between the experimental and expected value. The highest DPPH radical scavenging activity was achieved 85.20 ± 0.96% at the optimum extraction parameters of 52.5% ethanol (v/v), 40.0 °C temperature, and 22 min extraction time. Alternatively, the highest yield of total phenolic content was found 31.65 ± 0.94 mg GAE/g DF at the optimum extraction conditions. From the LC–ESI–MS profiling of the optimized extract, 18 bioactive compounds were tentatively identified, which may regulate the antioxidant activity of fruits of F. auriculata.

Phaleria macrocarpa (ver. name: ‘mahkota dewa’) is a plant which has many medically useful antioxidant activities (Anggraini & Lewandowsky, 2015). The polyphenols responsible for this antioxidant activity has to be extracted before it can be routinely used (Shwter et al., 2016). This study investigates the extraction of polyphenols from P. macrocarpa fruits and its antioxidant activity (DPPH-RSA) under influence of extraction time and temperature. By employing maceration technique, the P. macrocarpa fruits extract showed the maximum total phenolic content (TPC), total flavonoid content (TFC) and DPPH-RSA with value of 69.5 mg QE/g DW, 183.2 mg GA/g DW and 171. 8 mg BHA/g DW, respectively at  optimum extraction conditions of 60 min and 80 ºC (Fig. 1). Excellent and positive Pearson correlation coefficient with R2 > 0.91 between the TPC, TFC and antioxidant activities was observed.

Abstract This work investigated the seasonal variations in biosynthetic path selectivity of nine individual flavonoids, and content and antioxidant activity of three major metabolites in Tetrastigma hemsleyanum. The results revealed that under conditions of precipitation (2.0~6.6 mm), temperature (17.5~24.1°C), humidity (67.3~80.2%) and sunshine duration (3.4~5.8 h) in April and May, the total content of flavonoids reached higher levels between 282 and 394.5 μg/g; in the second half of April with the highest accumulation of flavonoids, the production selectivity (PS) of isoorientin, orientin, rutin, isoquercitin, kaempferol-3-0-rutinoside, astragalin, quercetin, apigenin and kaempferol were 0.30, 0.06, 0.07, 0.07, 0.00, 0.04, 0.38, 0.05 and 0.03, respectively; also according to the reaction network consisting of three main pathways for flavonoids syntheses in T. hemsleyanum, the selectivity of reaction Path II to synthesize quercetin, isoquercitin and rutin in April, May and June fluctuated between 0.52 and 0.66, much higher than that of reaction Path I or Path III.

ABSTRACTGrey mangrove (Avicenia marina) is one of the abundant types of mangrove in Indonesia, which can be used as a natural antioxidant source. This study aimed to determine the effect of extraction duration and fractionation on the antioxidant activity of A. Marina leaves. A. marina leaves powder was macerated by ethyl acetate solvent for 1, 2, 3, and 4 days. Meanwhile, fractionation was carried out using column chromatography with an isocratic method. The antioxidant activity of crude extracts and fractions was analyzed by DPPH (1,1-diphenyl-2-picrylhydrazyl) method. The result showed that one-day treatment was the optimum extraction duration to obtain the extract’s yield (1.65±0.01%) with the highest antioxidant activity (IC50 186.67±2.82 ppm). The best eluent in fractionation was a mixture of ethyl acetate: n-hexane (3:7) eluents, which produced eight fractions. However, fractionation reduced the antioxidant activity of fractions, compared to the crude extract. This might have happened because of the synergistic effect among compounds in the extract. Further research can be carried out to isolate the active compounds in the A. marina leaves extract.

This study aimed to investigate the extract with high antioxidant activity of Tetrastigma hemsleyanum Diels et Gilg and identify the antioxidant components in vitro. α, α-Diphenyl-β-picrylhydrazyl (DPPH) radical assay, Trolox equivalent antioxidant capacity (TEAC) assay, ferric reducing antioxidant power (FRAP), and hydroxyl radical scavenging method were used to screen the extract with high antioxidant activity. The antioxidant capacity of the extracts was evaluated by the free radical scavenging ability of DPPH. The ability of extracts to scavenge 2, 2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radical was evaluated by TEAC assay. The FRAP method was used to evaluate the ability of extracts to reduce Fe3+. The ability to scavenge hydroxyl radicals produced by the interaction of hydrogen peroxide and Fe2+ was measured by monitoring the change in the absorbance of the reaction mixture at 536 nm. Then, high-performance liquid chromatography-DPPH (HPLC-DPPH) and HPLC-hydroxyl radical scavenging methods were used to screen the antioxidant components in the extract. The molecular weight of the above antioxidant components was investigated using the qualitative analytical method of high-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry (HPLC-Q-TOF LC/MS). Based on the concentrations of the samples (0.2–4 mg/mL), the DPPH free radical scavenging ability, ABTS+ free radical scavenging ability, hydroxyl free radical scavenging ability, and Fe3+ reducing ability of the ethyl acetate extract (EAE) were stronger than that of the crude extract (CE), petroleum ether extract (PEE), and n-butanol extract (BE). The EAE has higher antioxidant activity than CE, PEE, and BE. Six antioxidant components, rutin, quercetin, isoquercetin, astragalin, kaempferol, and kaempferol-3-o-rutoside, were identified in the EAE.
