The reticulum plays an important part in the regeneration of lymph nodes autoplastically transplanted into the abdominal wall of the albino rat. The necrosis which follows transplantation involves mainly the lymphocytes. A margin of lymphocytes is preserved only at the periphery. This may be due to early lymphatic connection with the marginal sinus of the node. The reticulum cells are apparently more resistant since for the most part they do not degenerate. There are three possible origins of the lymphocytes of the regenerated node. They may arise by proliferation of lymphocytes retained at the periphery of the plant. The presence of a few mitotic figures among lymphocytes in the marginal region confirms the possibility of this mode of origin. They may also be derived from lymphocytes brought into the transplant by the blood or lymph circulation. Though some lymphocytes are present in the marginal sinus, and an occasional lymphocyte is seen in the capillaries which enter at the hilus, we believe that this source of origin of the lymphocytes is negligible. On the other hand, the hyperplastic reticulum appears to be the important source of lymphocyte production. These may be derived from the reticulum directly as small lymphocytes, or may be formed through the intermediary stage of large lymphoid cells. This capacity is not limited to lymphatic reticulum, as small foci of lymphocyte formation are found in the connective tissue in the vicinity. of the transplants. The reticular origin of lymphoctes is most easily observed in the earlier stages of regeneration before the picture is obscured by the numerous small lymphocytes. Two structures in the regenerating node are directly traceable to the same structures of the transplanrt. These are the marginal sinus and the hilus. The marginal sinus of thef transplant is preserved, and is probably an important means by which lymphatic communication is established with the surrounding tissue. The hilus is the site of entry of the blood vessels, and the hilus of the regenerated node is the same as that of the transplant.

The findings in a fatal case of liver necrosis in a child given diphenylhydantoin sodium are presented. Typical lymph node hyperplasia of the reticulum cells, with mitotic figures and eosinophils, were seen in this instance. These are a well-documented finding in hypersensitivity to this drug.

Four patients who received bone marrow transplants were studied sequentially during the posttransplant period to define the pattern of recovering lymphoid cell types. Three patients received T cell-depleted, HLA-matched marrow, and one received untreated marrow from an identical twin. Blood lymphoid cells were labeled with 25 different pairs of monoclonal antibodies. In each sample, one antibody was conjugated to fluorescein and one to phycoerythrin, thus allowing simultaneous assessment of the expression of the two markers using the fluorescence activated cell sorter. A total of 14 antibodies were used, routinely including HLE, Leu-M3, Leu-4, Leu-1, Leu-5, Leu-9, Leu-6, Leu-2, Leu-3, HLA-DR, Leu-7, Leu-11, Leu-15, and Leu-12. Other antibodies were used to further define some populations. This study has allowed us to define six distinct cell types that have appeared in all four patients by day 90 posttransplantation, and which account for 90-100% of all circulating lymphoid cells. These cell types are (a) T helper cells expressing Leu-1, Leu-4, Leu-9, Leu-5, Leu-3, and variable amounts of HLA-DR; (b) T suppressor cells expressing Leu-1, Leu-4, Leu-9, Leu-5, Leu-2, and variable amounts of HLA-DR; (c) B cells expressing Leu-12, B1, HLA-DR, IgD, and IgM, but none of the T cell antigens; (d) an unusual B cell phenotype (Leu-1 B) expressing all of the B cell markers, and also having low amounts of Leu-1, but none of the other T cell antigens; (e) natural killer (NK) cells expressing Leu-11, Leu-15, Leu-5 but none of the other T cell or B cell markers; (f) NK cells expressing Leu-11, Leu-15, Leu-5, and low levels of Leu-2. Both NK types also express Leu-7 on some, but not all cells. The relative frequencies of these cell types varied among the patients and with time, but the striking findings were the presence of relatively few mature T cells, large numbers of NK cells, and the preponderance of the unusual Leu-1 B cell over conventional B cells in all three patients who developed B cells. Sorting experiments confirmed the NK activity of the major NK cell phenotypes, and DNA analysis confirmed that all of the cells studied were of donor origin. In addition, analysis of Ig genes in one patient showed that the Leu-1 B cells were not clonally rearranged.(ABSTRACT TRUNCATED AT 400 WORDS)

Modulation of lymphocyte flow in the lymphatic compartment of the lymph node may serve, in part, to promote lymphocyte sensitization during an antigenic challenge. This study was undertaken to show where this might occur by examining the structural relationships of the intranodal lymphatic pathways, blood vessels, and connective tissue support with respect to lymphocyte and lymph flow. Differently stained plastic resins were injected into the blood vessels and lymphatics of the submandibular lymph node and visualized with a confocal laser scanning microscope. The specimens were corroded to study the three-dimensional cast structures by scanning electron microscopy. Alkali digestion was also used to prepare the reticular fiber network in the lymph node for scanning electron microscopic examination. At the hilus of the node, two to three arteries gave off arterioles running in medullary cords towards the cortex. The medullary cords, the periphery of the deep cortex, and the perifollicular zones had dense capillary networks. In contrast, the center of the follicle and the center of the deep cortex were less highly vascularized. High-endothelial venules were restricted to the perifollicular zone and the periphery of the deep cortex. At the cortico-medullary boundary, they abruptly transformed into medullary venules with a normal endothelium. The marginal sinus of the lymph node was crossed by thick reticular fibers that arose from the inner sheets of the capsule. The lymph pathway went through the marginal sinus, into the trabecular sinus, to the cortical perifollicular sinus, the dense lymphatic sinus around the deep cortex, and finally into the medullary sinus. At present, the exact functional significance of the complex lymph node lymphatic architecture is not clear. However, the highly organized structural organization may play a significant role in regulating and directing lymphocyte flow to facilitate antigen presentation.

Background: Malignant myoepithelioma is a rare neoplasm in humans and rats, poorly reported in dogs and not previously described in rabbits. This study aimed to report a case of malignant mammary myoepithelioma in a domestic rabbit (Oryctolagus cuniculus). Case: A domestic, hybrid, female, entire, nulliparous, seven-year-old rabbit was attended in the Veterinary Hospital Prof. Ricardo Alexandre Hippler at Universidade Vila Velha (UVV) with a history of apathy, lethargy and anorexia. In the clinical examination a 5 cm tumour was observed in the right abdominal mammary gland (M2), along with floating smaller lesions in M1 and M3. There were no palpable abnormalities in the axillary and inguinal lymph nodes. Complete blood count and serum biochemistry were within the normal range. Fine needle aspiration was performed for cytopathological evaluation of the tumour in M2 and it was suggestive of a malignant glandular neoplasm, with probable epithelial origin. Complete staging included chest and abdominal radiographs, but they were unremarkable. The patient was submitted to a right unilateral mastectomy, involving resection of the inguinal lymph node. The sample was sent to the Laboratory of Animal Pathology of UVV for routine histological evaluation. Histopathological evaluation in M2 was compatible with malignant myoepithelioma based on the literature, while nodules in M1 and M3 were classified as mammary cysts. The inguinal lymph node showed no significant abnormalities. An immunohistochemical panel was performed in the Comparative Pathology Laboratory of the Institute of Biological Sciences of the Universidade Federal de Minas Gerais (UFMG), using immunomarkers (pan-cytokeratin, p63 and vimentin) to characterize the tumour and confirm its diagnosis. Immunohistochemistry revealed expression of p63, and was negative for pan-cytokeratin and vimentin, which confirmed the myoepithelial origin, without involvement of the epithelial compartment. Clinical follow-up was recommended every 2-3 months, including abdominal ultrasonography, chest X-rays, complete blood count and serum biochemistry. Ultrasonography showed thinning of the uterine wall, dilation of 3.7 cm in diameter and presence of endometrial cysts of varied sizes, around 0.7 cm. Neutering was recommended but it was not elected by the pet´s responsible. These findings remained stable throughout the follow-up, which was greater than 405 days.Discussion: This is the first report of a malignant myoepithelioma in rabbit, with a detailed description of the clinical, surgical, pathological and immunophenotypic aspects of this neoplasm in this species. This is a tumour characterized by proliferation of epithelioid to spindle-like myoepithelial cells, as observed in this case, forming an extensive and well delimited solid area with marked central necrosis, compatible with what has already been described in the literature on bitches and women. As observed in this case, the neoplastic cells usually exhibit moderate cellular and nuclear pleomorphism, moderate anisocytosis and anisokaryosis, and several mitotic figures; different from what is observed in women, in which mitotic figures vary from small to moderate amount. Neoplasia and mammary cysts were removed by mastectomy, and the owner choose not to castrate the animal. On ultrasonography, after 12 months, the endometrium presented structures similar to cysts and was hyperplastic, which is commonly reported in association to mammary neoplasia. Nervertheless, the pet´s owner choose not to spay the rabbit and the lesions were stable on imaging follow-up

Abstract Diffuse sclerosing variant (DSV) is a rare subtype of papillary thyroid cancer (PTC). Whether it represents a higher grade subtype than conventional PTC is not quite clear. Furthermore, there are limited data on its long-term outcome and its molecular genetics. In this report, we studied all cases of DSV PTC seen at our center during the last 20 years. Out of more than 6000 patients (pts) with differentiated thyroid cancer, only 37 were DSV. We reviewed the clinical and histopathological features, management and outcome of these cases. In addition, molecular genetics is partially achieved; 17 out of these 37 cases have been genotyped for BRAFV600E, TERT promotor mutations, NRAS, HRAS and KRAS mutations. The molecular profiling of the other 20 cases is being done. A total of 37 pts were studied {(12 Males:25 Females, median age 21 years (8-89)}. One pt had lobectomy and the other 36 pts (97.3%) had a total thyroidectomy. Central only (4 pts) or central/lateral lymph node dissection (29 pts) were performed. The median tumor size was 4.5 cm (1.5-8.1). The tumor was multifocal in 27 cases (73%), with extrathyroidal invasion in 27 (73%) and lymphovascular invasion in 24 pts (64.8%). A background lymphocytic thyroiditis was present in 12 pts (32.4%). Lymph node metastases were present in 34 pts (92%) and distant metastases in 13 pts (35%). The sites of metastasis are lungs in 12 pts (32.4%) and lungs and bone in 1 pt. Twenty pts (54.1%) were in TNM8 stage 1, 10 pts (27%) in stage 2, 1 (2.7%) in stage 4a, 3 (8.1%) in stage 4b and 3 unstageable. The ATA risk classification for these pts was 4 pts (10.8%) in low, 12 (32.4%) in intermediate, 19 (51.4%) in high-risk groups and 2 could not be assessed. I-131 was administered to 33 pts (89.2%). The median administered activity was 136 mCi (46-218). Fifteen pts (40.5%) received additional therapies (3 surgeries, 7 RAI, 5 surgeries, and RAI). In 17 pts (46%) which were genotyped, only 3 tumors (8.1%) had BRAFV600E mutation, 1 (2.7%) had TERT promotor C228T mutation and none had RAS mutations. At the last follow up, 15 pts (40.5%) achieved an excellent response, 9 (24%) an indeterminate response, 6 (16.2%) with a structural disease, and 7 (19%) were lost for follow up. Conclusion: DSV PTC is a rare variant, occurs mostly in adolescent and young pts, characterized by aggressive histopathological features and high rates of lymph node and distant metastases but the commonly reported mutations in PTC are rare in DSV and mortality is absent.

Two mastocytomas of the liver of sheep are described. The specimens were obtained during a survey of hepatic neoplasia that was conducted on abattoir-slaughtered sheep. Although tissues other than the liver were not examined, both tumours had metastasised, one to the portohepatic lymph node, and the other intrahepatically. It is believed that this is the first report of mastocytoma in this species.

Abstract Dissociated thymus cells are capable of initiating graft-versus-host reaction in (C3Hf/Gs x DBA/2)F1 hybrids only when derived from parental donors previously sensitized against the antigens of the other parental strain. The lower immunologic activity of thymus cells as compared with other lymphoid cells is presumably due to quantitative rather than qualitative differences in immunologically competent cells.

Abstract The origin and fate of small lymphoid cells in the dog bone marrow were studied autoradiographically by observing the effect of clamping of the femoral artery during in vivo availability of H3-thymidine. Heavily labeled small lymphoid cells appeared in the bone marrow of the clamped leg 3 hours after injection of the tracer and increased in number up to 6 days. The labeling indices of these cells, however, were significantly lower than those of control marrow. A possible interpretation is that dog bone marrow contains two populations of small lymphoid cells, one migrating into the marrow via the blood stream, the other originating from local precursor cells within the marrow. There was no evidence for a transformation of migrated small lymphoid cells into erythroblasts during the first 48 hours after injection of H3-thymidine.
