Abstract The water quality of the Sunter River in Jakarta was classified as heavily polluted due to activities around the river, both domestic and non-domestic. As one of the environmental parameters for water quality, the presence of Escherichia coli (E. coli) is normally found any natural environment, and under certain conditions it can become resistant to antimicrobials due to genetic mutations. The mutated E. coli produces Extended Spectrum Beta-Lactamase (ESBL) enzymes and has a higher survival ability in antibiotic-contaminated river water, thus potentially endangering public health. This study was aimed to evaluate the effect of environmental factors on the abundance of ESBL producing E. coli and their resistance to antibiotic cefotaxime. Sampling was conducted in six locations representing the upstreams and the downstreams of Sunter River, following the Global Surveillance guidelines. E. coli strains were isolated using Tryptone Bile X-glucuronide (TBX) agar medium (with and without the addition of cefotaxime 4μg/ml) and the antibiotic sensitivity test of ESBL E. coli was conducted by performing a double-disk test. The results showed that the highest average abundance of ESBL E. coli was found in the sample taken from Sindang Station (904.24 x 104 colony per unit (CFU) / 100 mL) and the lowest was from Sunter Station (1,58 x 104 CFU / 100 mL). The results of the Bivariate Pearson correlation analysis showed that temperature, pH, and salinity were negatively correlated with the abundance of ESBL-producing E. coli bacteria.

ABSTRACT The aim of this work was to evaluate the microbiological quality of striped venus clams (Chamelea gallina L.) harvested in the north Adriatic Sea during an 8-year monitoring period. A total of 387 samples were analyzed to assess the presence of Escherichia coli. Environmental parameters (salinity, pH, dissolved oxygen, seawater temperature, and freshwater outflow) were collected to find out a possible relationship between the E. coli counts and environmental factors. The results evidenced that the microbiological quality of the clams was good, with only about 5% of the samples not complying with European and Italian regulations for this product, that is, with E. coli counts higher than 230 most probable number (MPN) per 100 g of flesh and intravalvar liquid. Statistical analyses revealed a relationship between microbial contamination and the season and water temperature, probably due to the difference in the filtering activity of the mollusks. However, the main factor affecting the E. coli concentration in the clams turned out to be the flow rate of Marecchia, the major river that reaches the sea in the area of harvesting. In fact, a model fitted to evaluate the probability of finding a higher E. coli count in relation to the environmental parameters evidenced that it was an increase of the level of the Marecchia led to a higher probability of elevated E. coli contamination. This result could be explained by the higher supply of both nutrients and coliforms (including E. coli) when the river is higher and by the anthropogenic characteristics of the lands crossed by the river.

Abstract Faecal contamination of surface water sources is an important water quality issue worldwide. Although quite a few studies exist on surface water faecal contamination and variability of indicator bacteria, most of the studies have been based on larger river basins and in temperate region. The variability is relatively unknown in local scale and in tropical developing countries. In this study we assess how anthropogenic and environmental factors affect faecal contamination and physicochemical parameters in Rupsha and Bhairab rivers around Khulna city, Bangladesh. Water samples were collected from six locations of the rivers during a wet and dry period in 2018 to measure Escherichia coli (E. coli) concentrations. Water physicochemical parameters—temperature, turbidity, pH, dissolved oxygen, biochemical oxygen demand and chemical oxygen demand were also measured. Higher concentrations of E. coli were found in the sampling sites located near the densely populated urban area compared to the downstream site, which receives fewer amounts of discharges from sewer drains. All the E. coli samples violated bathing water quality standards. E. coli concentrations were found to be correlated positively with precipitation and turbidity. A linear regression model was applied, that explains large part of the variation in E. coli concentration (R2 = 0.42). Water quality index assessment was also ranked the water quality as ‘poor’ category; indicate that the water is unsuitable for uses in domestic and recreational purposes and high health risks involved with the water use. The study findings highlight the problem of untreated sewage discharge into the rivers. Implementation of sewage treatment plant with adequate capacity is highly recommended.

Introduction. The increasing bacterial contamination of water bodies requires an increase in water quality control’s reliability to ensure epidemic safety against waterborne infections. Therefore, researchers in both Russia and Europe came to the conclusion that it is necessary to search for indicator microorganisms that can more accurately suggest the presence of pathogens. microorganisms in water than traditional indicators. The aim of the study was to justify the introduction of indicator indices of fecal contamination “generalized coliform bacteria” and Escherichia coli to assess the safety of drinking water Material and methods. The article provides an analysis of domestic and international regulatory documents and literary materials regulating the quality of drinking water in terms of sanitary and microbiological indicators and assessment criteria. The results of many years of experimental and field research carried out by research organizations and practical organizations of the Federal Service for Supervision of Consumer Rights Protection and Human Welfare are presented. Results. On the territory of Russia, “general coliform bacteria,” is one of the indicator indices chosen according to the fermentation of lactose, determine the safety of drinking water. The water does not take into account pathogenic bacteria (Salmonella, Shigella) and a number of lactose-negative opportunistic bacteria, causative agents of intestinal infections. The study of microorganisms isolated from the feces of patients confirms the frequency of occurrence of lactose-negative microorganisms to varying from 20 to 100% of strains. With an annual trend towards a decrease in the percentage of non-standard drinking water samples in terms of microbiological indices, general intestinal infections (GII) of unknown etiology increase, i.e. risk of GII. If the quality of drinking water does not correspond to thermotolerant coliform bacteria (TCB), 95% of samples contain E. coli. Therefore, the determination of E. coli more reliably indicates the intake of fresh fecal contamination and provides efficiency in taking measures to eliminate an unfavorable situation than TCB. Conclusion. Reasons are given for the introduction of more reliable microbiological indicators of water safety control, such as - “generalized coliform bacteria” with the preservation of the abbreviation GCB, combining both lactose-positive and lactose-negative bacteria, determined by the sign of glucose fermentation, negative oxidase test and negative stain according to Gram and E. coli as an indicator of recent faecal contamination, which will allow the assessment of water quality for a wide range of bacteria of the order Enterobacterials, corresponding to the modern taxonomy of Enterobacteriaceae NCBI, will ensure harmonization with international requirements and the safety of drinking water for the population.

Indicator microorganisms – Escherichia coli – in a stormwater wetland controlling non-point source pollution in the Keum River basin was studied and the relationship between water quality parameters and the survival of E. coli was investigated by Pearson correlation analysis based on 16 sets of data collected on dry days. A significant seasonal variation of the density of E. coli was observed in the influent and effluent. A dominant decay of E. coli was found in almost all the wetland components. In the settling pond, the density was observed to vary negatively with pH and the removal of total suspended solids, and the decay was promoted by solar radiation. In the aeration pond, the survival of E. coli was encouraged negatively by the dilution effect, unfavorable increase of dissolved oxygen (DO) and sedimentation with coarse particles. In the marsh wetland, the reduction of E. coli was positively influenced by UV radiation, obvious increase of pH, DO and sedimentation with algae and particles. In the polishing pond, the introduction of E. coli with avian feces and growth of the bacteria were more prevalent than decay. Although this introduction or growth was almost negligible, further study will be conducted in the future.

Results from statutory testing of private water supplies in nine Public Health Laboratories in England were compiled, and the effects of supply class, source, treatment and location on water quality were examined. A total of 6551 samples from 2911 supplies was examined, over a 2-year period, of which 1342 (21%) samples, and 949 (33%) supplies on at least one occasion, failed current regulations for Escherichia coli. Total coliforms, including E. coli, were detected in 1751 (27%) samples from 1215 (42%) supplies. The percentage of samples positive for E. coli was highest in summer and autumn, and lowest in winter. Samples taken from larger supplies and from boreholes were less frequently contaminated than those from other sources. Chlorination, filtration or UV light treatment improved the bacteriological quality of supplies, but still resulted in a low level of compliance with the regulations. The public health implications of the study are discussed.

A água é um recurso fundamental para a vida na Terra. Analisar e monitorar esse bem é uma das formas de auxiliar na conservação da sua qualidade. Este trabalho teve como objetivo analisar a qualidade da água da Cachoeira do Rio São Tomás por meio de variáveis de qualidade da água, Índice de Estado Trófico (IET) e balneabilidade. Foram realizadas sete campanhas, quatro no período chuvoso (novembro e dezembro de 2017 e janeiro e fevereiro de 2018) e três no período de estiagem (junho, julho e agosto de 2018). As variáveis físico-químicas analisadas foram temperatura (T), condutividade elétrica (CE), sólidos totais dissolvidos (STD), potencial hidrogeniônico (pH), turbidez, fósforo total (PT) e nitrato, sendo os valores de referência os estabelecidos pela Resolução CONAMA 357/05 e observados na literatura. O estudo do IET foi realizado considerando os resultados obtidos para PT. Para balneabilidade, a variável obtida foi Escherichia coli (E. coli) e os resultados comparados com os limites descritos na Resolução CONAMA 274/00. A precipitação foi analisada para melhor entender os resultados. A Turbidez e o PT estiveram em inconformidade com os limites determinados na Resolução CONAMA 357/05. Para IET a água foi classificada nos mais altos graus de trofia (supereutrófico e hipereutrófico). Em relação a balneabilidade, a água foi classificada como impropria para banho em três campanhas e para a média geral. Os resultados negativos podem ter relação com o uso e ocupação do solo inadequados associados à precipitação, o que favorece o carreamento de resíduos para o corpo d’água. Water Quality of Cachoeira do Rio São Tomás, in the Municipality of Rio Verde, Goiás, BrazilA B S T R A C TThe water is a primordial resource for life on Earth. Analyzing and monitoring this asset is one of the ways to assist on the preservation of its quality. This paper had as objective to analyze the quality of the water of Cachoeira do Rio São Tomás through variables of water quality, Trophic State Index (TSI) and bathing. Seven campaigns were accomplished, four during the rainy period (November and December of 2017 and January and February of 2018) and three during the drought period (June, July and August of 2018). The physical-chemical variables analyzed were temperature (T), electric conductivity (EC), total dissolved solids (TDS), hydrogen potential (pH), turbidity, total phosphorus (TP) and nitrate, considering the reference values the ones stablished by CONAMA Resolution 357/05 and observed in literature. The study of TSI was accomplished considering the obtained results for TP. For bathing, the obtained variable was Escherichia coli (E. coli) and the results compared to the described limit of CONAMA Resolution 274/00.  The rainfall was analyzed for better understanding of the results. The Turbidity and TP were in nonconformity with the limits determined by the CONAMA Resolution 357/05. For TSI the water was classified in the highest degrees of trophic (supereutrophic and hypereutrophic). In relation to bathing, the water was classified as inappropriate for bathing in three campaigns and for the general average. The negative results can be related to the inappropriate use and coverage of the soil associated to rainfall, which favors the solids transport to the water body.Keywords: anthropic action, bathing, eutrophication, water resources, subsidy for water management.

In the present study, antibiogram of Escherichia coli isolated from farmed Asian clam, Corbiculafluminea was characterised. Asian clam or locally known as ‘etak’ is processed to became smokedclam and consumed as snack by Kelantanese. However, there are many diarrhoea cases afterconsuming smoked clam. Furthermore, there are also insufficient information about the E.coli levelin Asian clam farm and effective antibiotic in controlling the bacteria in the litereature. Hence, thisstudy was carried out to provide information of antibiogram of E.coli to be reference in the future.Eosin Methylene Blue agar (EMB) was used to isolate E. coli. A total of 100 isolated bacteria weresubjected to antibiotic sensitivity test using disk diffusion method. A total of 18 types of antibioticsnamely novobiocin (30µg/disk), fosfomycin (50 µg/disk), tetracycline (30µg/disk), lincomycin(15µg/disk), flumequine (30µg/disk), sulphamethoxazole (25 µg/disk), amoxycillin (25 µg/disk),chloramphenicol (30 µg/disk), oleandomycin (15 µg/disk), spiramycin (100 µg/disk), ampicillin(10 µg/disk), oxytetracycline (30 µg/disk), doxycycline (30 µg/disk), nalidixic acid (30 µg/disk),florfenicol (30 µg/disk), erythromycin (15 µg/disk), kanamycin (30 µg/disk) and oxolinic acid (2µg/disk). The findings of the present study showed total plate count of E. coli was 6.45 x 103 colonyforming unit (CFU/100g) of sampled Asian clam. Hence, the clam is needed to be under cleansingtreatment before can consider safe for human consumption. Antibiotic results showed 51 % wasrecorded as antibiotic resistance case, 44 % antibiotic sensitive case and 5 % as antibioticintermediary sensitive case. None of the tested antibiotics was successfully inhibited the growth ofthe present bacterial isolates indicating more antibiotics are needed to be screen in the future studyto find out the most effective antibiotic in controlling isolated E. coli.

The present study aimed to evaluate the characteristics that most influenced the water quality variability of the Atibaia river in the city of Paulínia/SP, the coordinates of the water collection point are given by 22º44'23”(S) and 47º07 ' 40 ”(W), in the dry and rainy seasons, from 2006 to 2016. The data used in this study come from the monitoring of parameters carried out in the spring by a research team, accompanied by periodic collections of surface water samples and analyzes laboratory tests. The parameters addressed included: precipitation, temperature, Hydrogenionic potential (pH), turbidity, thermotolerant coliforms (Escherichia coli (E. coli)), dissolved oxygen (DO), biochemical oxygen demand (BOD), chemical oxygen demand (COD), total nitrogen (Ntotal), total phosphorus (Ptotal), total dissolved solids (TDS), electrical conductivity (EC) and chlorides (Cl-). Water quality was assessed using linear correlation analysis, using the Pearson Correlation coefficient (r), and Multivariate Analysis, using Principal Component Analysis (PCA). The presence of EC, Cl- and COD in both periods indicates quality characteristics related to the mineralization of organic compounds present in the water and the eutrophication process.

The WSP system serving Mèze and Poussan (French Mediterranean coast) was constructed in 1980 and enlarged and upgraded from 1994 to 1998. Water quality along the waste stabilisation pond to (WSP) system has been monitored over the years, thus allowing us to assess the influence of enlargement and upgrading works. A significant enhancement of the average microbiological quality of the effluent was observed, with respective E. coli and streptococci average abatements of 4.1 and 3.4 log. units. Former seasonal variations of microbiological removal have vanished. The contribution of the different ponds to the disinfection performance of the WSP system was analysed. A microbiological quality model was proposed to evaluate the die-off kinetics related to the different ponds and as a tool for the design and management of WSP systems. Though the relationships between die-off coefficients and environmental factors appeared somewhat frail, this modelling is considered a promising approach for the prediction of WSP microbiological performance.

To identify emerging Escherichia coli that have the potential to cause diarrhea in pigs, the prevalence of E. coli pathotypes was determined among 170 and 120 isolates from diarrheic and nondiarrheic piglets, respectively. The isolates were tested for F4, F5, F6, F18, and F41 fimbriae, for E. coli attaching and effacing (EAE), porcine attaching and effacing–associated (Paa), and adhesin involved in diffuse adherence (AIDA-I) factors, for LT, STa, STb, and enteroaggregative heat-stable (EAST1) enterotoxins, and for Shiga toxins (Stx1, Stx2, and Stx2e), using DNA hybridization and polymerase chain reaction. All isolates were O-serotyped and tested for antibiotic resistance against 10 drugs. Seventeen different pathotypes, accounting for 40.0% of the isolates, were recovered from diarrheic piglets. The main pathotypes included EAST1 (13.5%), F4/LT/STb/EAST1 (6.5%), AIDA-I/STb/EAST1 (4.1%), F5/STa (2.9%), EAE/EAST1 (2.9%), and AIDA-I/F18 (2.3%). Only 3 pathotypes, EAE (11.7%), EAST1 (10.8%), and EAE/EAST1 (3.3%), were recovered from nondiarrheic piglets. Paa factor was detected in 8.8% and 7.5% of isolates from diarrheic and nondiarrheic piglets, respectively, and always was associated with other virulence determinants. Overall, 22.9% of isolates from diarrheic piglets appeared to be enteropathogens: enterotoxigenic E. coli (11.7%), enteropathogenic E. coli (3.5%), and E. coli isolates (3.0%) for which none of the above adherence factors was detected. Pathotypes AIDA-I/STb/EAST1 and AIDA-I/STb were isolated only from diarrheic piglets and accounted for 4.7% of isolates. Strains of these pathotypes induced diarrhea when inoculated into newborn colostrum-deprived pigs, in contrast to an isolate positive only for EAST1, which did not induce diarrhea. Antibiotic sensitivity test showed that isolates of the AIDA-I/STb/EAST1 and AIDA-I/STb pathotypes were the only strains sensitive to enrofloxacin, gentamicin, neomycin, and trimethoprim–sulfamethoxazole. This study showed that at least 20.5% of isolates from diarrheic piglets appeared to be associated with AIDA-I/STb pathotype and that EAST1 pathotype is probably not an important marker for diarrhea in piglets.
