Abstract Objectives To induce Neurolathyrism in guinea pig fed with Lathyrus sativus diet. Methods Twenty four guinea pigs were randomly distributed into 3 groups, 8 animals each. Groups I control fed with control diet, Group II low toxin group fed with 90% Lathyrus diet containing 100 mg β-ODAP/100 g Lathyrus and Group III fed with 90% Lathyrus diet with 1060 mg β-ODAP/100 gLathyrus. The experiment was for 2 months. Diet intake, body weight, protein carbonyl, catalase, MDA, specific activity of electron transport complexes I, II, III and IV in cortex and cerebellum were analyzed. Histopathological and morphological changes were studied by electron microscopy Results The diet intake and bodyweight was significantly reduced in both the experimental groups (II & III) as compared group I. Protein carbonyls levels were significantly increased in motor cortex region of group II & III as compared to group I. The catalase activity in cortex region was significantly decreased in group II & III compared to group I. The MDA levels in motar cortex region were significantly higher in the group II & III as compared to group I. Whereas the MDA levels in the cerebellum was significantly higher in group III as compared to group I. The activity of complex I and II of cortex was decreased significantly in group II and III as compared to group I. However, there was no significant change in specific activity of electron transport complex II and complex III. The nitrate levels in the group II & III were significantly higher than group I in the cortex region. However, there were no significant differences in the nitrate levels in the cerebellum region. Electron microscopic study of motor cortex showed that the extensive degeneration of motor neuron was observed in group II and III compared to group I. Conclusions Consumption of ninety % Lathyrus sativus dietfor 2 months induces neurolathyrism in guinea pigs. Abstract Background Our aim was to study the morphologic and functional changes in the retina and sclera induced by form deprivation high myopia (FDHM) in guinea pigs and explore the possible mechanisms FDHM formation. Methods Forty 3-weeks-old guinea pigs were randomized into the blank control (Group I, 20 cases) and model groups (20 cases). In the model group, the right eyes of the guinea pigs were sutured 8 weeks to induce FDHM (Group II) and the left eyes were considered a self-control group (Group III). The diopters were measured with retinoscopy. The anterior chamber depth (AC), lens thickness (L), vitreous depth (V) and axial length (AL) were measured using ultrasonometry A. Retinal and scleral morphology and ultrastructural features were observed with light and electron microscopy. The content of malondialdehyde (MDA) and the activity of superoxide dismutase (SOD) in the retina and sclera were detected with a chemical colorimetric assay. Results After remaining stitched for eight weeks, the diopters of Group II changed from (+3.59±0.33) D to (-7.96±0.55) D, and these values were significantly higher than those of Group I (+0.89±0.32) D and Group III (-0.55±0.49) D. The vitreous chamber depth (4.12±0.13) mm and axial length (8.93±0.22) mm of Group II were significantly longer than that of Group I [(3.71±0.23) mm and (7.95±0.37) mm, respectively] and Group III [(3.93±0.04) mm and (8.01±0.15) mm, respectively] (P < 0.05). With the prolongation of form deprivation, the retina and scleral tissue became thinner, the ganglion cell and inner and outer nuclear layers of the retina became decreased, and the arrangement was disordered. In Group II, the activity of SOD was significantly lower than that in Group I and Group III, and the content of MDA was significantly higher in Group II than in Group I and Group III. The differences were statistically significant (P < 0.05). Conclusions These findings suggested that in the FDHM guinea pigs model, the diopter, the vitreous chamber depth, and axial length increased significantly with prolongation of monocular FD time, and morphological structural changes in the retina and sclera were observed. Oxygen free radicals might participate in the formation of FDHM.

Background: The study evaluates the effect of a high supplemental dose of ascorbic acid (AA) on plasma concentrations of total cholesterol (TC), triglycerides (TG), total lipids (TL), and lipoprotein fractions high-density, very-low-density-, and low-density lipoprotein (HDL, VLDL, LDL) in guinea pigs fed with atherogenic diet. Methods: Group I consisted of 5 normally fed guinea pigs plus a low dose of AA (1 mg/100 g/day), group II consisted of 7 guinea pigs fed with food enriched with 2% cholesterol plus a low dose of AA (1 mg/100 g/day), and group III consisted of 7 guinea pigs fed with food enriched with 2% cholesterol plus a high dose of AA (30 mg/100 g/day). Cholesterolemic factors concentrations were determined after nine weeks. Results: Concentrations of TC, TG, TL, LDL, and VLDL were increased in group II compared to group I (p < 0.01 for all differences). Supplementation with a high dose of AA resulted in decreased concentrations of TC (p < 0.01), TG (p < 0.01), TL (p < 0.01), and LDL (p < 0.01) in group III compared to group II. Additionally, concentration of HDL was increased in group III compared to group II (p < 0.01). Conclusion: High-dose AA supplementation to an atherogenic diet decreases concentrations of TC, TG, TL, and LDL and increases concentration of HDL compared to low-dose AA.

Background: Garlic is naturally occurring sulphur containing dietary agent belong to Allium family. Members of this family i.e. Garlic, Onions are found to have beneficial effect on atherosclerosis and ischemic heart diseases in both experimental animals and in human beings. Objective: To compare the potency of garlic as antihyperlipidemic and antiplatelet and antithrombotic between its watery and alcoholic extract for their effects. Methods: 25 guinea pigs were fed cholesterol (0.5g/kg body weight) for an initial period of 4 weeks. Cholesterol was then discontinued and the animals were divided into 3 groups. Group-I (n=7) was fed with stock diet and was taken as control. Group-II (n=9) was given 1ml of watery extract and Group-III (n=9) was fed with 1ml of alcoholic extract of garlic with normal diet daily for 4 weeks. The garlic content of both the extract was 2 gm/ml by wt/vol. Fasting blood samples were collected at the end 4 weeks and finally at the end of the study i.e.8 weeks for estimation of serum cholesterol, serum triglyceride, LDC-C, HDL-C, VLDL-C, atherogenic index and platelet adhesiveness index. Bleeding time and clotting time were also detected in all the 3 groups. Results: In the present study, the watery and alcoholic extract of garlic shows significant hypolipidemic activity as they reduced significantly serum cholesterol, serum triglyceride, LDC-C, VLDL-C and atheriogenic index in hyperlipidemic guinea pigs (p<0.01) compared to control. The significant rise in HDL-C was observed in group II but not in group III animals. On comparison between two extracts, watery extract of garlic was more potent hypolipidemic agent, compared to alcoholic extract. Both extract significantly reduces platelet adhesiveness and bleeding time. But between two extract the watery extract was found more potent anti-thrombotic agent as it showed significant decrease in platelet adhesiveness (P<0.01). BT was significantly increased in group II compared to group I and III. Conclusion: It can be concluded that watery extract of Allium Sativum is more potent hypolipidemic and antiplatelet, and anti-thrombotic agent. C57BL male mice (Jackson Lab., Bar Harbor, Maine) weighing about 18 gms were randomly divided into three groups: group I was fed sweetened liquid alcohol diet (modified Schenkl) in which 36% of the calories were derived from alcohol; group II was maintained on a similar diet but alcohol was isocalorically substituted by sucrose; group III was fed regular mouse chow ad lib for five months. Liver and heart tissues were fixed in 2.5% cacodylate buffered glutaraldehyde, post-fixed in 2% osmium tetroxide and embedded in Epon-araldite.

SummaryMost anticardiolipin antibodies (ACA) associated with antiphospholipid syndrome (APS) are directed against epitopes expressed on β2-glycoprotein I (β2GPI). Despite a good correlation between standard ACA assays and those using purified human β2GPI as the sole antigen, some sera from APS patients only react in the latter. This is indicative of heterogeneity in anti-β2GPI antibodies. To characterize their reactivity profiles, human and bovine β2GPI were immobilized on γ-irradiated plates (β2GPI-ELISA), plain polystyrene precoated with increasing cardiolipin concentrations (CL/β2GPI-ELISA), and affinity columns. Fluid-phase inhibition experiments were also carried out with both proteins. Of 56 selected sera, restricted recognition of bovine or human β2GPI occurred respectively in 10/29 IgA-positive and 9/22 IgM-positive samples, and most of the latter (8/9) were missed by the standard ACA assay, as expected from a previous study. Based on species specificity and ACA results, IgG-positive samples (53/56) were categorized into three groups: antibodies reactive to bovine β2GPI only (group I) or to bovine and human β2GPI, group II being ACA-negative, and group III being ACA-positive. The most important group, group III (n = 33) was characterized by (i) binding when β2GPI was immobilized on γ-irradiated polystyrene or cardiolipin at sufficient concentration (regardless of β2GPI density, as assessed using 125I-β2GPI); (ii) and low avidity binding to fluid-phase β2GPI (Kd in the range 10–5 M). In contrast, all six group II samples showed (i) ability to bind human and bovine β2GPI immobilized on non-irradiated plates; (ii) concentration-dependent blockade of binding by cardiolipin, suggesting epitope location in the vicinity of the phospholipid binding site on native β2GPI; (iii) and relative avidities approximately 100-fold higher than in group III. Group I patients were heterogeneous with respect to CL/β2GPI-ELISA and ACA results (6/14 scored negative), possibly reflecting antibody differences in terms of avidity and epitope specificity. Affinity fractionation of 23 sera showed the existence, in individual patients, of various combinations of antibody subsets solely reactive to human or bovine β2GPI, together with cross-species reactive subsets present in all samples with dual reactivity namely groups III and II, although the latter antibodies were poorly purified on either column. Therefore, the mode of presentation of β2GPI greatly influences its recognition by anti-β2GPI antibodies with marked inter-individual heterogeneity, in relation to ACA quantitation and, possibly, disease presentation and pathogenesis.

<p class="p1"><span class="s1"><strong>Background:</strong> The inspiratory oxygen fraction (FiO<sub>2</sub>) is usually set between 60% and 100% during conventional extracorporeal circulation (ECC). However, this strategy causes partial oxygen pressure (PaO<sub>2</sub>) to reach hyperoxemic levels (&gt;180 mmHg). During anesthetic management of cardiothoracic surgery it is important to keep PaO<sub>2</sub> levels between 80-180 mmHg. The aim of this study was to assess whether adjusting FiO<sub>2</sub> levels in accordance with body temperature and body surface area (BSA) during ECC is an effective method for maintaining normoxemic PaO<sub>2</sub> during cardiac surgery.</span></p><p class="p1"><span class="s1"><strong>Methods:</strong> After approval from the Ethics Committee of the University of Acıbadem, informed consent was given from 60 patients. FiO<sub>2</sub> adjustment strategies applied to the patients in the groups were as follows: FiO<sub>2</sub> levels were set as 0.21 × BSA during hypothermia and 0.21 × BSA + 10 during rewarming in Group I; 0.18 × BSA during hypothermia and 0.18 × BSA + 15 during rewarming in Group II; and 0.18 × BSA during hypothermia and variable with body temperature during rewarming in Group III. Arterial blood gas values and hemodynamic parameters were recorded before ECC (T1); at the 10th minute of cross clamp (T2); when the esophageal temperature (OT) reached 34°C (T3); when OT reached 36°C (T4); and just before the cessation of ECC (T5).</span></p><p class="p1"><span class="s1"><strong>Results:</strong> Mean PaO<sub>2</sub> was significantly higher in Group I than in Group II at T2 and T3 (<em>P</em> = .0001 and <em>P</em> = .0001, respectively); in Group I than in Group III at T1 (<em>P</em> = .02); and in Group II than in Group III at T2, T3, and T4 <br /> (<em>P</em> = .0001 for all). </span></p><p class="p1"><span class="s1"><strong>Conclusion: </strong>Adjustment of FiO<sub>2</sub> according to BSA rather than keeping it at a constant level is more appropriate for keeping PaO<sub>2</sub> between safe level limits. However, since oxygen consumption of cells vary with body temperature, it would be appropriate to set FiO<sub>2</sub> levels in concordance with the body temperature in the <br /> rewarming period.</span></p>

The article presents the comparative analysis of the state of the cervix in women with endometrial polyps and micropolyps. Patients and methods. The study involved 130 patients aged 18-35 years: 70 patients with endometrial polyps (group I), 30 patients with micropolyps (group II) and 30 patients of the control group (group III). Results. According to the anamnesis of women in the I group were significantly more frequent diseases of the cervix, which corrected physical surgery methods, in particular cryodestruction. In group II, the representatives of these indicators were similar to healthy. Normal colposcopic picture met significantly less frequently in patients and I, and II group. The differences in the incidence of HPV high oncogenic risk in all groups were not statistically significant. Conclusion. Destructive methods used in the detection of any changes in the cervix are often overly aggressive, form scars and contributing to inflamaciones process. In the chain of events leading to the formation of PE, cervical pathology and its correction can take the basic place.
