Neutral and polar lipid fatty acids in five families of demersal and pelagic fish from the deep Northwest Atlantic

AbstractFatty acids in neutral and polar lipids were used to investigate trophic connections in species from five families of demersal (Rajella fyllae, Malacoraja senta, Alepocephalus bairdii, Borostomias antarcticus) and pelagic fish (Bathytroctes macrolepis, Lampanyctus spp., Chaulidos sloani, Serrivomer beanii) sampled in the deep Atlantic Ocean off Newfoundland, Canada. Lipid extracts were fractionated into neutral, acetone-mobile polar, and phospholipids to separate fatty acids in storage from those in membranes. Multivariate analysis of fatty acids showed that there were greater differences among the three lipid fractions than there were among the species when all fatty acid fractions were considered together. Neutral lipid fatty acids were characterized by monoenes, acetone-mobile polar lipids (AMPL) by C18 polyenes, and phospholipids by 16:0 and DHA. Multivariate analysis of fatty acids in phospholipids showed a strong grouping by taxonomic family (>80% similarity), while the neutral lipid fatty acids showed a weaker grouping by family (72.5% similarity) but groupings that also related to habitat and vertical migration. The neutral lipid data supported the use of 20:1ω9 as a biomarker of calanoid copepods and of 16:1ω7 as a marker of diatoms to determine food web connections in deep-sea fish, but not some other common markers. In addition, correlations with δ15N showed that series of ω6 and ω7 fatty acids were trophically transferred though neutral lipids, especially the essential fatty acid, ARA. Neutral lipids were also reservoirs for essential ω3 fatty acids in demersal species, but the major reservoir for DHA was AMPL and phospholipid in pelagic species.

Chemical parameters involved in technological meat quality for dry cured processing of Gascon pigs were studied in longissimus dorsi (LD) and biceps femoris (BF) muscles. Muscles from Gascon pigs showed 2.60 and 2.84% of intramuscular fat content, 23.64 and 22.14% protein content and 1.34 and 4.63mg of myoglobin per gram of muscle (respectively LD and BF). Intramuscular fat (IMF) and myoglobin levels were higher than those reported for commercial pigs, but lower than those previously found in Iberian pigs. A similar situation was detected in the fatty acid composition of neutral and polar lipids of both muscles. Thus, monounsaturated fatty acids (MUFA) in neutral lipids of Gascon pig muscles (LD and BF respectively) were 58.27 and 51.98%, while polyunsaturated fatty acids (PUFA) levels were 5.61 and 14.13% respectively; values similar to those found in Iberian pigs and quite different to usual values in commercial pig breeds. The same trend was found in polar lipids. Both muscles showed a low susceptibility to induced lipid oxidation, in agreement with their fatty acid composition. These results pointed out that meat from pigs of the Gascon breed showed optimal characteristics for dry cured processing.

1. Rat intestinal microvillus plasma membranes were prepared from previously isolated brush borders and the lipid composition was analysed. 2. The molar ratio of cholesterol to phospholipid was greatest in the membranes and closely resembled that reported for myelin. 3. Unesterified cholesterol was the major neutral lipid. However, 30% of the neutral lipid fraction was accounted for by glycerides and fatty acid. 4. Five phospholipid components were identified and measured, including phosphatidylethanolamine, phosphatidylcholine, phosphatidylserine, sphingomyelin and lysophosphatidylcholine. Though phosphatidylethanolamine was the chief phospholipid, no plasmalogen was detected. 5. In contrast with other plasma membranes in the rat, the polar lipids of the microvillus membrane were rich in glycolipid. The cholesterol:polar lipid (phospholipid+glycolipid) ratio was about 1:3 for the microvillus membrane. Published data suggest that this ratio resembles that of the liver plasma membrane more closely than myelin or the erythrocyte membrane. 6. The fatty acid composition of membrane lipids was altered markedly by a single feeding of safflower oil. Membrane polar lipids did not contain significantly more saturated fatty acids than cellular polar lipids. Differences in the proportion of some fatty acids in membrane and cellular glycerides were noted. These differences may reflect the presence of specific membrane glycerides.

AbstractThe lipid composition of three batches of single generation cysts of Globodera rostochiensis, stored dry at 4°C for 1,7 and 13 years, comprised 81%, 74% and 53% neutral lipids, 14%, 18% and 27% non-acidic phospholipids and 5%, 8% and 20% free fatty acids, respectively. Lipids in eggs from two batches of G. pallida cysts, stored for 3 and 7 years, comprised 80% and 67% neutral lipids, 15% and 23% non-acidic phospholipids and 5% and 10% free fatty acids, respectively. All batches contained the same fatty acids which were dominated by C18:l, C20:l and C20:4. The fatty acid profiles of hatched J2 of G. rostochiensis from two batches, stored for 1 and 9 years, differed only in their free fatty acid fractions. Thus, while it is not possible to determine the age of cysts by their fatty acid profile, it may be possible to use the relative amounts of the main lipid classes as an indicator of age. Four batches of hatched J2 of G. pallida were investigated, with sample A hatched during the second week in potato root diffusate, B during week 3, C during week 4 and D during weeks 5 and 6 and stored for 3.5 days (on average) after hatching. Total lipid content was 27.2%, 31.5%, 18.5% and 6.3% of the dry weight for A, B, C and D, respectively. In the neutral lipid fraction of D an increase in C18:l and to a lesser extent C18:2 was observed. In the free fatty acid fraction of sample D, the percentages of C18:l, C18:2 and C18:3 were greater but the percentages of C20:3 and C20:4 were smaller compared with sample C. Fresh early hatched J2 of G. rostochiensis were compared with later hatched and stored (for 13 days on average) individuals for their lipid content and fatty acid composition. The lipid content was 26.1% and 11.4% in fresh and stored J2, respectively. Total lipid consisted of 77% and 70% neutral lipid, 18% and 26% non-acidic phospholipid and 6% and 4% free fatty acid in fresh and stored J2, respectively. In the neutral lipid fraction of stored J2 C18:l, C16:0 and C18:0 increased, whereas C20:4, C20:l and C20:3 decreased. Therefore, both neutral lipid and free fatty acid fractions showed changes in their fatty acid profiles after long delayed hatching and/or storage in both PCN species.

When perithecia appear (on the 7th day of growth), the mycelial lipid content falls and the degree of fat insaturation increases; fatty acid synthetase activity greatly increases. Lipid turnover is therefore accelerated. Among neutral lipids, triglycerides constitute the largest pool and their proportion increases when perithecia appear. Free fatty acids also increase, confirming the lipid turnover's acceleration. Polar lipids especially disappear; the phosphatidylcholine/phosphatidylethanolamine ratio increases when the fungus ascosporulates.

A thorough analysis of the fatty acid profiles of stipe and blade from the kelp species Laminaria hyperborea is presented. Lipid extracts were fractionated into neutral lipids, free fatty acids, and polar lipids, prior to derivatization and GC-MS analysis. A total of 42 fatty acids were identified and quantified, including the n-3 fatty acids α-linolenic acid, stearidonic acid, and eicosapentaenoic acid. The fatty acid amounts are higher in blade than in stipe (7.42 mg/g dry weight and 2.57 mg/g dry weight, resp.). The highest amounts of n-3 fatty acids are found within the neutral lipid fractions with 590.6 ug/g dry weight and 100.9 ug/g dry weight for blade and stipe, respectively. The amounts of polyunsaturated fatty acids are 3.4 times higher in blade than stipe. The blade had the highest PUFA/SFA ratio compared to stipe (1.02 versus 0.76) and the lowest n-6/n-3 ratio (0.8 versus 3.5). This study highlights the compositional differences between the lipid fractions of stipe and blade from L. hyperborea. The amount of polyunsaturated fatty acids compared to saturated- and monounsaturated fatty acids is known to influence human health. In the pharmaceutical, food, and feed industries, this can be of importance for production of different health products.

The fatty acid composition of neutral lipids (NL), polar lipids (PL) and free fatty acids (FFA) was analysed in dry-cured loins obtained from the native pig breed Chato Murciano (CM) and from a modern crossbreed pig genotype (CG) during the ripening stage (between the 30 and 60 days of processing). Fatty acid concentrations from neutral lipids, polar lipids and free fatty acid fractions were affected by breed. With respect to ripening time, lipolysis was more intense in the CG than in CM product, resulting in a decrease in the concentrations of fatty acids in the NL and PL fractions, accompanied by a corresponding increase in FFA. Results for lipid determination provided evidence that the concentrations of the different groups of fatty acids within the lipid fractions depend on the breed. In order of abundance, the groups of fatty acids in the neutral lipid fraction were monounsaturated fatty acids (MUFA) > saturated fatty acids (SFA) > polyunsaturated fatty acids (PUFA) (59%, 37% and 4% in CM; 58%, 35% and 6% in CG) at 60 days of processing. In the polar lipid fraction, the order was SFA > PUFA > MUFA (44%, 29% and 27% in CM; 42%, 38% and 20% in CG), and in free fatty acid fraction, the order was MUFA > PUFA > SFA (40%, 30%, and 30% in CM; 39%, 32%, and 29% in CG) at 60 days of processing.

The fatty acid compositions of total, neutral, and polar lipids of Beauveria bassiana, Metarrhizium anisopliae, Isaria farinosa, and Hirsutella gigantea were determined. Major fatty acids were palmitic, oleic, and linoleic; smaller amounts of palmitoleic, stearic, and α-linolenic were also present. Neutral lipid contained a higher proportion of oleic and stearic acids than did total and polar lipid, whilst polar lipid had the highest proportion of linoleic acid. The rest of the acids were evenly distributed between the two fractions.

We quantified seasonal changes in the total fatty acid content and the content of major fatty acid classes (i.e., saturates, polyunsaturates, etc.) in the neutral lipids and polar lipids of ovaries, liver, white muscle, and adipopancreatic tissue of female northern pike in central Alberta. Concentrations (percent wet weight) of polar lipid fatty acids (PLFAs) increased during fall and winter in all the organs and tissues examined. As the ovaries were recrudescing between July and March, ovary PLFA content increased from 0.03 to 4.60 g and the combined PLFA content of the three somatic tissues increased from 1.87 to 2.68 g (quantities are expressed per kilogram carcass weight of pike). During this period the quantity of neutral lipid fatty acids (NLFAs) (per kilogram of carcass weight) in the ovary increased from 0.01 to 1.95 g and liver NLFA content decreased from 0.60 to 0.14 g. The NLFA content of adipopancreatic tissue was 3.5–5.5 g/kg carcass weight and that in muscle was 2–4 g/kg carcass weight, but neither showed significant seasonal variation. There were no significant seasonal changes in either the total fatty acid content or the content of any fatty acid class in the combined neutral lipids or combined total lipids (neutral lipids plus polar lipids) of the three somatic tissues. Thus, there do not appear to be large seasonal transfers of fatty acids from somatic to ovarian tissues or from neutral to polar lipids in female pike.

1. Total lipids were extracted from digesta obtained from the rumen, abomasum and upper small intestine (jejunum) of each of four slaughtered sheep. The lipids were fractionated into unesterified fatty acids, neutral lipids and phospholipids and the proportional contribution of each fraction to the total fatty acids was determined.2. The contribution made by phospholipids to the total fatty acids in the digesta showed a marked increase in the samples from the small intestine compared with those from the rumen and abomasum. This increase was apparently due to the presence of biliary phospholipids.3. Total lipids and conjugated bile acids were extracted from sheep bile, the lipids were fractionated and their fatty-acid composition was determined. Phospholipids predominated and these consisted mainly of phosphatidylcholine, together with some lysophosphatidylcholine.4. Both phospholipids contained significant amounts of unsaturated C18 components which could account, at least in part, for the previously reported increament to the proportion of these acids in the digesta when it enters the upper jejunum.5. The overall fatty acid compositions of the two biliary phospholipids were very similar and, in common with other naturally occurring phosphatidylcholines, the fatty acids present in position 2 of the phosphatidylcholine of bile were found to consist almost entirely of unsaturated components.6. Total lipids and conjugated bile acids were extracted from samples of digesta obtained from three sheep with cannulas in different positions in the jejunum. Analysis of the lipids indicated that biliary phospholipids, in particular phosphatidylcholine, underwent progressive hydrolysis in the intestinal lumen.7. The distribution of conjugated bile acids, unesterified fatty acids and phospholipids between the solid (particulate) and liquid (micellar) phases of the intestinal digesta was determined. These chyme constituents were, for the most part, associated with the particulate matter and thus, at any given time, it appears that only a small fraction of the total fatty acids is available for absorption in micellar form. It is suggested that the micellar solubilization of fatty acids may be facilitated by the presence of lysophosphatidylcholine.
