The myeloid differentiation factor 2 (MD-2)-related lipid-recognition protein is involved in immune responses through recognizing bacteria lipopolysaccharide in mammals, arthropods and plants. However, the physiological roles of MD-2 in other biological processes are largely unknown. Here, we identified three homologue MD-2 genes ( NlML1 , NlML2 and NlML3 ) by searching the genome and transcriptome databases of the brown planthopper Nilaparvata lugens , a hemipteran insect species. Temporospatial analysis showed that the NlML1 gene was highly expressed in the fat body but much less so in the other tissues, while the NlML2 and NlML3 genes were highly expressed in the testis or digestive tract. RNA interference-mediated depletion of the NlML1 gene significantly downregulated the transcription of numerous integument protein genes. The NlML1 knockdown led to moulting failure and mortality at the nymph–adult transition phase, impaired egg laying and hatching, and reduced 20-hydroxyecdysone (20E) production in the nymphs. 20E could rescue the deficient moulting phenotypes derived from ds NlML1 RNAi. These novel findings indicate that NlML1 is required for nymphal moulting and female reproductive success as it plays an important role in regulating 20E synthesis, lipid and chitin metabolisms in N. lugens , thus contributing to our understanding of developmental and reproductive mechanisms in insects.

AbstractThe sex determination mechanism for hemipteran species remains poorly understood. During the sex determination of the brown planthopper (BPH), Nilaparvata lugens, one species of Hemiptera, the functions of doublesex (Nldsx) and NlTra-2 (NlTra-2) genes were identified in our previous studies. Here, we identify an upstream gene for Nldsx in the sex determination cascade, NlFmd, which acts as female determinant gene for N. lugens. The sex-specific transcript of NlFmd (NlFmd-F) encodes an arginine/serine-, and proline-rich protein that is essential for female development. The knockdown of NlFmd resulted in the development of pseudomales, with sex-specific alternative Nldsx processing, and maternal RNA interference (RNAi) against NlFmd generates male-only progeny. Moreover, homologous genes for NlFmd have also been identified in two rice planthopper species, the white-backed planthopper (WBPH, Sogotalla furcifera) and the small brown planthopper (SBPH, Laodelphax striatellus), and these genes appear to be involved in the sex determination cascades for these species. Our data suggest that the sex determination cascade in Delphacidae is conserved.

Ceramides are bioactive sphingolipids that have been implicated in insect development; however, their role in insect reproduction remains poorly understood. Here, we report the pivotal role of neutral ceramidase (NCER) in the female reproduction of the brown planthopper (BPH), Nilaparvata lugens (Stål), a significant pest in rice cultivation in Asia. LC-MS/MS demonstrated that, among different developmental stages of BPH, the levels of ceramides were highest in 1st instar nymphs and lowest in adults. The transcription of NCER was negatively correlated with the levels of ceramides at different developmental stages of BPH, in that the transcript levels of NCER were the highest, whereas ceramides levels were the lowest in BPH adults. Knocking down NCER through RNA interference (RNAi) increased the levels of ceramides in BPH females and ovaries, which resulted in a delay in oocyte maturation, a reduction in oviposition and egg hatching rate, as well as the production of vulnerable offspring. Transmission electron microscopy (TEM) analysis and TdT-mediated dUTP Nick-End Labeling (TUNEL) assays showed mitochondrial deficiency and apoptosis in NCER-deficient oocytes. Taken together, these results suggest that NCER plays a crucial role in female reproduction in BPH, likely by regulating the levels of ceramides.

Growth and reproduction are the two most basic life processes of organisms and the distribution of energy in these processes is a core issue of the life history of organisms. Nilaparvata lugens (Stål), the brown planthopper (BPH), is a single-feeding rice pest. In the present study, this species was used as a model for testing the effects of nutritional conditions on various growth and reproduction indicators. First, the third-instar nymphs were fed with three different concentrations (100, 50, and 25%) of artificial diet until the second day of adulthood. The results showed that as the nutrient concentration decreased, the body development and oviposition of BPH were hindered. The total lipid content in the fat bodies was also significantly reduced. RT-PCR analysis showed compared to the 100% concentration group, the expression levels of vitellogenin (Vg) genes in the fifth-instar nymphs, adults, and in different tissues (ovary, fat body, and other tissues) were significantly decreased in the 50 and 25% treatment groups. Western blot analysis showed that Vg protein expression was highest in the 100% group, followed by the 50% group, with no expression in the 25% group. These results indicate that growth and reproduction in the BPH are regulated by, or correlated with, nutrient concentration. This study is of great significance as it reveals the adaptive strategies of the BPH to nutritional deficiencies and it also provides valuable information for the comprehensive control of this pest.

Non-ATPase regulatory subunits (Rpns) are components of the 26S proteasome involved in polyubiquitinated substrate recognition and deubiquitination in eukaryotes. Here, we identified 15 homologues sequences of Rpn and associated genes by searching the genome and transcriptome databases of the brown planthopper, Nilaparvata lugens , a hemipteran rice pest. Temporospatial analysis showed that NlRpn genes were significantly highly expressed in eggs and ovaries but were less-highly expressed in males. RNA interference-mediated depletion of NlRpn genes decreased the proteolytic activity of proteasome and impeded the transcription of lipase and vitellogenin genes in the fat bodies and ovaries in adult females, and reduced the triglyceride content in the ovaries. Decrease of the proteolytic activity of the proteasome via knockdown of NlRpn s also inhibited the transcription of halloween genes, including NlCYP307A2 , NlCYP306A2 and NlCYP314A1 , in the 20-hydroxyecdysone (20E) biosynthetic pathway in the ovaries, reduced 20E production in adult females, and impaired ovarian development and oocyte maturation, resulting in reduced fecundity. These novel findings indicate that the proteolytic activity of the proteasome is required for female reproductive processes in N. lugens , thus furthering our understanding of the reproductive and developmental strategies in insects.

The brown planthopper, Nilaparvata lugens (Stål)[BPH], is a damaging pest of rice in Asia. Insecticides and rice varietal resistance are widely implemented BPH management practices. However, outbreaks of BPH have been linked to excessive insecticide use—challenging the compatibility of these two management practices. IR62 is a variety with resistance against BPH, the whitebacked planthopper, Sogatella furcifera Horváth [WBPH], and the green leafhopper, Nephotettix virescens (Distant)[GLH]. We compared BPH responses to IR62 and to the susceptible variety IR64 treated with buprofezin, carbofuran, cartap hydrochloride, cypermethrin, deltamethrin, fipronil, or thiamethoxam + chlorantraniliprole. In greenhouse bioassays, cypermethrin, fipronil and thiamethoxam + chlorantraniliprole reduced egg laying on both varieties, and, together with buprofezin, reduced nymph survival to zero. Buprofezin, carbofuran, and cartap hydrochloride stimulated egg laying, and carbofuran increased nymph biomass, but these effects were reduced on IR62. Planthopper populations were ten times higher on deltamethrin-treated rice than untreated rice in a screenhouse experiment. Host resistance failed to buffer against this insecticide-induced resurgence in BPH and WBPH. However, IR62 reduced the effect in GLH. Rice treated with cypermethrin and fipronil had reduced yields compared to untreated controls, suggesting possible phytotoxic effects. We found little evidence of synergies between the two management practices; but host resistance did buffer against the undesirable effects of some insecticides.

The standard seedling seedbox test (SSST) is the most prevalent phenotyping test in research on the genetics and breeding of planthopper-resistant rice. Using 16 rice lines that included plants susceptible, resistant and tolerant to the brown planthopper (Nilaparvata lugens), we compared the SSST to modified seedling seedbox tests (MSSTs) and the days-to-wilt (DTW) test. We also conducted a series of performance tests to assess nymph survival and development; adult longevity and egg-laying; egg survival; honeydew production; and plant weight loss. We also assessed the relative costs of the different phenotyping tests to better recommend test protocols that are suitable for high-throughput phenotyping. The SSST was found to be highly robust but fails to identify late-stage resistance; tolerance; or ovicidal responses. MSSTs improved phenotyping by identifying plants with low damage from planthoppers at later growth stages. Herbivore performance tests such as population or biomass build-up tests reduce space requirements and reduce setup and evaluation costs compared with bulk tests. They can also facilitate the assessment of plant tolerance; albeit with added costs. The DTW test most clearly segregates resistant and susceptible plants, thereby facilitating gene discovery and marker-assisted selection. We recommend that bulk testing be improved by switching from the SSST to a suitable MSST and that donor variety and pre-release lines be assessed for the nature of rice–planthopper interactions using biomass build-up tests—including the DTW test.

Symbionts in the abdomen fat body of brown planthopper (BPH) play an important role in the growth and reproduction of their host, Nilaparvata lugens Stål (Hemiptera: Delphacidae). Thus, controlling BPH infection on rice by inhibiting symbionts with antimicrobials is feasible. However, the effect of antimicrobials on the microbiome in the fat body and the relationship between microbial community and mortality have not been fully elucidated. A decrease in the total number of yeast-like symbiotes in the fat body and elevated mortality were observed after exposure to toyocamycin, tebuconazole, and zhongshengmycin. Additionally, we found that the antimicrobials reduced bacterial diversity and increased fungal diversity in the fat body and altered the bacterial and fungal community structure. Although the total absolute abundance of bacteria and fungi decreased after antimicrobial exposure, the absolute abundance of Serratia increased, indicating that Serratia, which was the most dominant in the fat body, is an important symbiont involved in resistance to antimicrobials. After antimicrobial exposure, seven genera, which probably participated in the nutrition and development function of the host, were totally eliminated from the fat body. Overall, our study enriches the knowledge of microbiomes in the fat body of BPH under antimicrobial treatment and the disturbance of symbionts would be further used to help other pesticides to control pests.
