A haloalkaliphilic strain XQ-INN 246T was isolated from the sediment of a salt pond in Inner Mongolia Autonomous Region, China. Cells of the strain were rods, motile and strictly aerobic. The strain was able to grow in the presence of 2.6–5.3 M NaCl (optimum concentration is 4.4 M) at 30–50 °C (optimum temperature is 42 °C) and pH 7.0–10.0 (optimum pH is 8.0–8.5). The whole genome sequencing of strain XQ-INN 246T revealed a genome size of 4.52 Mbp and a DNA G+C content of 62.06 mol%. Phylogenetic tree based on 16S rRNA gene sequences and concatenated amino acid sequences of 122 single-copy conserved proteins revealed a robust lineage of the strain XQ-INN 246T with members of related genera of the family Natrialbaceae . The strain possessed the polar lipids of phosphatidylglycerol and phosphatidylglycerol phosphate methyl ester. No glycolipids were detected. Based on phylogenetic analysis, phenotypic characteristics, chemotaxonomic properties and genome relatedness, the isolate was proposed as the type strain of a novel species of a new genus within the family Natrialbaceae, for which the name Salinadaptatus halalkaliphilus gen. nov., sp. nov. is proposed. The type strain is XQ-INN 246T (=CGMCC 1.16692T=JCM 33751T).

Three species of the family Perlodidae are newly reported or confirmed for China, Isoperla asiatica Raušer from Arxan, Inner Mongolia Autonomous Region, Isoperla eximia Zapekina-Dulkeit from Changbai Mountain, Jilin Province, and Mesoperlina capnoptera (McLachlan, 1886) from Xinjiang Uygur Autonomous Region. Based on new material, additional taxonomic notes and images of another perlodid stonefly, Rauserodes epiproctalis (Zwick, 1997) is provided.

A Gram-positive, rod-shaped, non-spore-forming and strictly aerobic bacterium (Gsoil 161T) was isolated from soil of a ginseng field in Pocheon Province, South Korea. The novel isolate was characterized using a polyphasic approach in order to determine its taxonomic position. On the basis of 16S rRNA gene sequence similarity, strain Gsoil 161T was shown to belong to the family Nocardioidaceae and was related to Aeromicrobium marinum (98.0 % similarity to the type strain), Aeromicrobium alkaliterrae (97.6 %), Aeromicrobium fastidiosum (97.0 %) and Aeromicrobium erythreum (96.7 %); the sequence similarity with other species within the family was less than 94.4 %. It was characterized chemotaxonomically as having ll-2,6-diaminopimelic acid in the cell-wall peptidoglycan, MK-9(H4) as the predominant menaquinone and C16 : 0, 10-methyl C18 : 0 (tuberculostearic acid), C16 : 0 2-OH, 10-methyl C17 : 0 and 10-methyl-C16 : 0 as the major fatty acids. The G+C content of the genomic DNA was 65.5 mol%. These chemotaxonomic properties and phenotypic characteristics support the affiliation of strain Gsoil 161T to the genus Aeromicrobium. Results of physiological and biochemical tests enabled strain Gsoil 161T to be differentiated genotypically and phenotypically from currently known Aeromicrobium species. Therefore, strain Gsoil 161T represents a novel species, for which the name Aeromicrobium panaciterrae sp. nov. is proposed. The type strain is strain Gsoil 161T (=KCTC 19131T=DSM 17939T=CCUG 52476T).

Two yellow, rod-shaped, Gram-reaction-negative, strictly aerobic bacterial strains, designated JC2682T and JC2681T, were isolated from a seawater sample of Jeju Island, Korea. The isolates required sea salts for growth and showed gliding motility. Phylogenetic analyses based on 16S rRNA gene sequences revealed that strains JC2682T and JC2681T shared 96.9 % similarity and formed a robust monophyletic group within the radiation encompassing the family Flavobacteriaceae . This distinct clade, which had varying branch positions depending on the tree-making algorithm used, represents a new genus within the family Flavobacteriaceae . The predominant isoprenoid quinone (menaquinone 6, MK-6) and DNA G+C content (27–33 mol%) were consistent with its assignment to the family Flavobacteriaceae , but the polar lipid profile, fatty acid composition and overall phenotypic traits demonstrated that the test strains were not closely affiliated to any recognized genera. Based on data from the present polyphasic taxonomic study, strains JC2682T and JC2681T are considered to represent two novel species of a new genus in the family Flavobacteriaceae , for which the names Flavivirga jejuensis gen. nov., sp. nov. and Flavivirga amylovorans sp. nov. are proposed, respectively. The type strain of Flavivirga jejuensis is JC2682T ( = KACC 14158T  = JCM 17113T). The type strain of Flavivirga amylovorans is JC2681T ( = KACC 14157T  = JCM 17112T). Amended descriptions of the genera Psychroserpens and Lacinutrix are provided.

A strictly aerobic, Gram-staining-negative, oxidase- and catalase-positive, non-motile, rod-shaped bacterium, designated strain 5416T-29T, was isolated from air and was characterized by using a polyphasic approach. Colonies were reddish pink and circular with entire margins. Flexirubin-type pigments were absent. The strain formed a distinct phylogenetic lineage within the family Cytophagaceae of the phylum Bacteroidetes. Strain 5416T-29T did not show more than 88 % 16S rRNA gene sequence similarity to the type strain of any recognized species. The major cellular fatty acids were C16 : 1 ω5c, iso-C17 : 0 3-OH and iso-C15 : 0. The polar lipids were phosphatidylethanolamine, one unknown amino lipid and several unknown polar lipids. Menaquinone-7 (MK-7) was the major respiratory quinone. The G+C content of the DNA of strain 5416T-29T was 45.5 mol%. Results of phenotypic and phylogenetic analyses clearly indicate that strain 5416T-29T represents a novel species of a new genus in the family Cytophagaceae, for which the name Rhodocytophaga aerolata gen. nov., sp. nov. is proposed. The type strain of Rhodocytophaga aerolata is 5416T-29T (=KACC 12507T =DSM 22190T).

A sponge-associated strain, KMM 7019T, was investigated in a polyphasic taxonomic study. The bacterium was strictly aerobic, heterotrophic, Gram-negative, yellow-pigmented, motile by gliding and oxidase-, catalase-, β-galactosidase- and alkaline phosphatase-positive. A phylogenetic analysis based on 16S rRNA gene sequences revealed that strain KMM 7019T is closely related to members of the genus Salegentibacter, namely Salegentibacter holothuriorum, Salegentibacter mishustinae and Salegentibacter salegens (97·7–98 % sequence similarities). The DNA–DNA relatedness between the strain studied and Salegentibacter species ranged from 27 to 31 %, clearly demonstrating that KMM 7019T belongs to a novel species of the genus Salegentibacter, for which the name Salegentibacter agarivorans sp. nov. is proposed. The type strain is KMM 7019T (=KCTC 12560T=LMG 23205T).

Sampling of agricultural and natural environments in two US states (Colorado and Florida) yielded 18 Listeria-like isolates that could not be assigned to previously described species using traditional methods. Using whole-genome sequencing and traditional phenotypic methods, we identified five novel species, each with a genome-wide average blast nucleotide identity (ANIb) of less than 85 % to currently described species. Phylogenetic analysis based on 16S rRNA gene sequences and amino acid sequences of 31 conserved loci showed the existence of four well-supported clades within the genus Listeria ; (i) a clade representing Listeria monocytogenes , L. marthii , L. innocua , L. welshimeri , L. seeligeri and L. ivanovii , which we refer to as Listeria sensu stricto, (ii) a clade consisting of Listeria fleischmannii and two newly described species, Listeria aquatica sp. nov. (type strain FSL S10-1188T = DSM 26686T = LMG 28120T = BEI NR-42633T) and Listeria floridensis sp. nov. (type strain FSL S10-1187T = DSM 26687T = LMG 28121T = BEI NR-42632T), (iii) a clade consisting of Listeria rocourtiae , L. weihenstephanensis and three novel species, Listeria cornellensis sp. nov. (type strain TTU A1-0210T = FSL F6-0969T = DSM 26689T = LMG 28123T = BEI NR-42630T), Listeria grandensis sp. nov. (type strain TTU A1-0212T = FSL F6-0971T = DSM 26688T = LMG 28122T = BEI NR-42631T) and Listeria riparia sp. nov. (type strain FSL S10-1204T = DSM 26685T = LMG 28119T = BEI NR- 42634T) and (iv) a clade containing Listeria grayi . Genomic and phenotypic data suggest that the novel species are non-pathogenic.

A Gram-negative, pleomorphic, extremely halophilic archaeon, designated strain XH-70T, was isolated from the saline Lake Xilinhot, in Inner Mongolia, China. It formed small (0.9–1.5 mm), red-pigmented, elevated colonies on agar medium. The strain required at least 2.5 M NaCl and 5 mM Mg2+ for growth. The 16S rRNA gene sequence analysis indicated that strain XH-70T belongs to the family Halobacteriaceae, showing 99.5 % similarity to the type strain of Halovivax asiaticus and 94.7 and 94.6 % similarity, respectively, to the type strains of Natronococcus amylolyticus and Natronococcus occultus. Polar lipid analysis supported the placement of strain XH-70T in the genus Halovivax. DNA–DNA hybridization studies (32 % with Halovivax asiaticus CGMCC 1.4248T), as well as biochemical and physiological characterization, allowed strain XH-70T to be differentiated from Halovivax asiaticus. A novel species, Halovivax ruber sp. nov., is therefore proposed to accommodate this strain. The type strain is XH-70T (=CGMCC 1.6204T=DSM 18193T=JCM 13892T).

Abstract Background Ticks (Arthropoda, Ixodida), after mosquitoes, are the second most prevalent vector of infectious diseases. They are responsible for spreading a multitude of pathogens and threatening the health and welfare of animals and human beings. However, given the history of tick-borne pathogen infections in the Inner Mongolia Autonomous Region of China, surprisingly, neither the genetic diversity nor the spatial distribution of haplotypes within ticks has been studied. Methods We characterized the haplotype distribution of Dermacentor nuttalli in four main pastoral areas of the Inner Mongolia Autonomous Region, by sampling 109 individuals (recovered from sheep) in April–August 2019. The 16S rRNA gene, cytochrome c oxidase subunit I (COI), and the internal transcribed spacer 2 region (ITS2) were amplified and sequenced from extracted DNA. Results Twenty-six haplotypes were identified using 16S rRNA sequences, 57 haplotypes were identified with COI sequences, and 75 haplotypes were identified with ITS2 sequences. Among the three genes, total haplotype diversity was greater than 0.7, while total nucleotide diversity was greater than 0.06. Neutrality tests revealed a significantly negative Tajima’s D result, while Fu's Fs was not significantly positive. Fixation index values (FST) indicated that the degree of genetic differentiation among some sampled populations was small, while for others it was moderate. Analysis of molecular variance (AMOVA) revealed that the variation within populations was greater than that among populations. The mismatch analysis of D. nuttalli exhibited double peaks. Conclusion The genetic diversity of D. nuttalli populations in our region can likely adapt to different geographical environments, thereby leading to genetic diversity, and creating genetic differentiation among different populations. However, genetic differentiation is cryptic and does not form a pedigree geographical structure.

The Gram-stain-negative, orange-pigmented, non-spore-forming, non-motile, strictly aerobic, rod-shaped bacterial strain, designated TP-CH-4T, was isolated from a seamount near the Yap Trench in the tropical western Pacific. The optimal growth conditions were determined to be at pH 7–8, 25–30 °C and in the presence of 2 % (w/v) NaCl. The major respiratory quinone was MK-6. The polar lipid profile contained phosphatidylethanolamine, two unidentified aminolipids, two unidentified phospholipids and three unidentified polar lipids. The predominant cellular fatty acids were iso-C15 : 0 and summed feature 1 (composed of C13 : 03-OH and/or iso-C15 : 1H). Phylogenetic analysis of 16S rRNA gene sequences revealed that strain TP-CH-4T was a member of the family Flavobacteriaceae and formed a distinct lineage. Strain TP-CH-4T displayed highest sequence similarities to Pseudozobellia thermophila KMM 3531T (95.1 %) and Flagellimonas flava A11T (93.9 %). Genome sequencing revealed the strain TP-CH-4T has a genome size of 4.5 Mbp and a G+C content of 44.5 mol%. Collectively, based on phenotypic, chemotaxonomic, phylogenetic and genomic evidence, strain TP-CH-4T represents a novel species of a novel genus of the family Flavobacteriaceae , for which the name Pelagihabitans pacificus gen. nov., sp. nov. is proposed. The type strain of Pelagihabitans pacificus is TP-CH-4T (=CGMCC 1.17120T=KCTC 72434T).

A Gram-stain-negative, yellow-pigmented, non-spore-forming, non-motile, rod-shaped, catalase-positive, strictly aerobic bacterial strain, designated CAU 1491T, was isolated from seawater and its taxonomic position was examined using a polyphasic approach. Cells of strain CAU 1491T grew optimally at 30 °C, pH 7.5 and in 2.0 % (w/v) NaCl. Phylogenetic analysis based on the 16S rRNA gene sequence of CAU 1491T showed that it formed a distinct lineage within the family Flavobacteriaceae as a separate deep branch, with 97.0 % or lower sequence similarity to representatives of the genera Lacinutrix , Gaetbulibacter and Aquibacter . The major cellular fatty acids of strain CAU 1491T were iso-C15 : 0, iso-C15 : 1 G, iso-C17 : 0 3-OH and summed feature 3. The polar lipid pattern consisted of diphosphatidylglycerol, phosphatidylserine, phosphatidylethanolamine and an unidentified phospholipid. The strain contained MK-6 as the sole respiratory quinone. Genome sequencing revealed that strain CAU 1491T has a genome size of 3.13 Mbp and a G+C content of 32.4 mol%. On the basis of the phenotypic, chemotaxonomic and genomic data, strain CAU 1491T represents a new genus and species in the family Flavobacteriaceae for which the name Pontimicrobium aquaticum gen. nov., sp. nov. is proposed. The type strain of Pontimicrobium aquaticum is CAU 1491T (=KCTC 72003T=NBRC 113695T).
