Selected bacterial isolates from the surface water of Buriganga river were characterized by morphological, biochemical characteristics and sequence-based PCR-amplified fragments of 16S rRNA. All isolates were rod shaped and Gram-positive. The isolates were confirmed as Chryseobacterium arthrosphaerae strain FDAARGOS 519, Bacillus cabrialesii strain TE 3, Bacillus tequilensis KCTC strain 13622(T), Bacillus amyloliquefaciens DSM 7 strain ATCC 23350, Bacillus subtilis strain E20 and Bacillus subtilis subsp. subtilis strain 168 based on sequence analysis. A phylogenetic tree was constructed that showed only one major cluster comprising of two sub-clusters grouping Chryseobacterium arthrosphaerae in one and Bacillus tequilensis in another. Chryseobacterium arthrosphaerae strain FDAARGOS 519 was susceptible to all antibiotics at different ranges, while Bacillus tequilensis KCTC strain 13622(T) and Bacillus amyloliquefaciens DSM 7 strain ATCC 23350 were found to be resistant to polymyxin only but sensitive to other antibiotics. However, Bacillus cabrialesii strain TE 3 was resistant to polymyxin and neomycin, while Bacillus subtilis subsp. subtilis strain 168 was resistant to polymyxin, vancomycin and rifampicin.

AbstractRescue of the ribosomes from dead-end translation complexes, such as those on truncated (non-stop) mRNA, is essential for the cell. Whereas bacteria use trans-translation for ribosome rescue, some Gram-negative species possess alternative and release factor (RF)-dependent rescue factors, which enable an RF to catalyze stop-codon-independent polypeptide release. We now discover that the Gram-positive Bacillus subtilis has an evolutionarily distinct ribosome rescue factor named BrfA. Genetic analysis shows that B. subtilis requires the function of either trans-translation or BrfA for growth, even in the absence of proteotoxic stresses. Biochemical and cryo-electron microscopy (cryo-EM) characterization demonstrates that BrfA binds to non-stop stalled ribosomes, recruits homologous RF2, but not RF1, and induces its transition into an open active conformation. Although BrfA is distinct from E. coli ArfA, they use convergent strategies in terms of mode of action and expression regulation, indicating that many bacteria may have evolved as yet unidentified ribosome rescue systems.

Bacillus subtilis ∆6 is a genome-reduced strain that was cured from six prophages and AT-rich islands. This strain is of great interest for biotechnological applications. Here, we announce the full-genome sequence of this strain. Interestingly, the conjugative element ICE Bs 1 has most likely undergone self-excision in B. subtilis ∆6.

Intracellular serine proteinase was isolated from sporulating cells of Bacillus subtilis Marburg 168 by gramicidin S-Sepharose 4B affinity chromatography. The enzymological characteristics, the amino acid composition and the 19 residues of the N-terminal sequence of the enzyme are reported. The isolated proteinase was closely related to, but not completely identical with, the intracellular serine proteinase of B. subtilis A-50. The divergence between these two intracellular enzymes was less than that between the corresponding extracellular serine proteinases (subtilisins) of types Carlsberg and BPN′!, produced by these bacterial strains. This may be connected with the more strict selection constraints imposed in intracellular enzymes during evolution.
