AbstractMembraneless organelles are cellular compartments that form by liquid-liquid phase separation of one or more components. Other molecules, such as other proteins and nucleic acids, will distribute between the cytoplasm and the liquid compartment in accordance with the thermodynamic drive to lower the free energy of the system. The resulting distribution colocalizes molecular species, to carry out a diversity of functions. Two factors could drive this partitioning: the difference in solvation between the dilute versus dense phase, and intermolecular interactions between the client and scaffold proteins. Here, we develop a set of knowledge-based potentials that allow for the direct comparison between desolvation energy and pairwise interaction energy terms, and use these to examine experimental data from two systems: protein cargo dissolving within phase-separated droplets made from FG repeat proteins of the nuclear pore complex, and client proteins dissolving within phase-separated FUS droplets. We find close agreement between desolvation energies of the client proteins and the experimentally determined values of the partition coefficients, while pairwise interaction energies between client and scaffold show weaker correlations. These results show that client stickiness is sufficient to explain differential partitioning of clients within these two phase-separated systems without taking into account the composition of the condensate. This suggests that selective trafficking of client proteins to distinct membraneless organelles requires recognition elements beyond the client sequence composition.

The present paper describes an investigation of diffusion in the solid state. Previous experimental work has been confined to the case in which the free energy of a mixture is a minimum for the single-phase state, and diffusion decreases local differences of concentration. This may be called ‘diffusion downhill’. However, it is possible for the free energy to be a minimum for the two-phase state; diffusion may then increase differences of concentration; and so may be called ‘diffusion uphill’. Becker (1937) has proposed a simple theoretical treatment of these two types of diffusion in a binary alloy. The present paper describes an experimental test of this theory, using the unusual properties of the alloy Cu 4 FeNi 3 . This alloy is single phase above 800° C and two-phase at lower temperatures, both the phases being face-centred cubic; the essential difference between the two phases is their content of copper. On dissociating from one phase into two the alloy develops a series of intermediate structures showing striking X-ray patterns which are very sensitive to changes of structure. It was found possible to utilize these results for a quantitative study of diffusion ‘uphill’ and ‘downhill’ in the alloy. The experimental results, which can be expressed very simply, are in fair agreement with conclusions drawn from Becker’s theory. It was found that Fick’s equation, dc / dt = D d2c / dx2 , can, within the limits of error, be applied in all cases, with the modification that c denotes the difference of the measured copper concentration from its equilibrium value. The theory postulates that D is the product of two factors, of which one is D 0f the coefficient of diffusion that would be measured if the alloy were an ideal solid solution. The theory is able to calculate D/D 0 , if only in first approximation, and the experiments confirm this calculation. The study reported here compared how younger and older adults processed the same set of media messages which were selected to vary on two factors, arousing content and valence. Results showed that older and younger adults had similar arousal responses but different patterns of attention and memory. Older adults paid more attention to all messages than did younger adults. However, this attention did not translate into greater memory. Older and younger adults had similar levels of memory for slow-paced messages, but younger adults outperformed older adults significantly as pacing increased, and the difference was larger for arousing compared with calm messages. The differences found are in line with predictions made based on the cognitive-aging literature.

This paper discuss the development of marketing knowledge based on the marketing practice to bridge the gap between academicians and practicians. Issues being discussed in this paper are the meaning of marketing knowledge from practician perspective, the theory of marketing in practician’s world, actionable research as the bridge of knowledge development, and the difference perspective between academician and parctician.

In the digital microfluidic experiments, the droplet characteristics and flow patterns are generally identified and predicted by the empirical methods, which are difficult to process a large amount of data mining. In addition, due to the existence of inevitable human invention, the inconsistent judgment standards make the comparison between different experiments cumbersome and almost impossible. In this paper, we tried to use machine learning to build algorithms that could automatically identify, judge, and predict flow patterns and droplet characteristics, so that the empirical judgment was transferred to be an intelligent process. The difference on the usual machine learning algorithms, a generalized variable system was introduced to describe the different geometry configurations of the digital microfluidics. Specifically, Buckingham’s theorem had been adopted to obtain multiple groups of dimensionless numbers as the input variables of machine learning algorithms. Through the verification of the algorithms, the SVM and BPNN algorithms had classified and predicted the different flow patterns and droplet characteristics (the length and frequency) successfully. By comparing with the primitive parameters system, the dimensionless numbers system was superior in the predictive capability. The traditional dimensionless numbers selected for the machine learning algorithms should have physical meanings strongly rather than mathematical meanings. The machine learning algorithms applying the dimensionless numbers had declined the dimensionality of the system and the amount of computation and not lose the information of primitive parameters.

The authors have been investigating the various characteristics of screw-type centrifugal pumps, such as pressure fluctuations in impellers, flow patterns in volute casings, and pump performance in air-water two-phase flow conditions. During these investigations, numerical results of our investigations made it clear that three back flow regions existed in this type of pump. Among these, the back flow from the volute casing toward the impeller outlet was the most influential on the pump performance. Thus the most important factor to achieve higher pump performance was to reduce the influence of this back flow. One simple method was proposed to obtain the restraint of back flow and so as to improve the pump performance. This method was to set up a ringlike wall at the suction cover casing between the impeller outlet and the volute casing. Its effects on the flow pattern and the pump performance have been discussed and clarified to compare the calculated results with experimental results done under two conditions, namely, one with and one without this ring-type wall. The influence of wall’s height on the pump head was investigated by numerical simulations. In addition, the difference due to the wall’s effect was clarified to compare its effects on two kinds of volute casing. From the results obtained it can be said that restraining the back flow of such pumps was very important to achieve higher pump performance. Furthermore, another method was suggested to restrain back flow effectively. This method was to attach a wall at the trailing edge of impeller. This method was very useful for avoiding the congestion of solids because this wall was smaller than that used in the first method. The influence of these factors on the pump performance was also discussed by comparing simulated calculations with actual experiments.

The Brn-3a, Brn-3b, and Brn-3c POU family transcription factors are closely related to one another and are members of the group IV subfamily of POU factors. Here we show that despite this close relationship, the factors have different effects on the activity of a target promoter: Brn-3a and Brn-3c stimulate the promoter whereas Brn-3b represses it. Moreover, Brn-3b can antagonize the stimulatory effect of Brn-3a on promoter activity and can also inhibit promoter activation by the Oct-2.1 POU factor. The difference in the transactivation activities of Brn-3a and Brn-3b is dependent upon the C-terminal region containing the POU domain of the two proteins, since exchange of this domain between the two factors converts Brn-3a into a repressor and Brn-3b into an activator.

Dietary restriction of phenylalanine is the main treatment for phenylketonuria (PKU), and current estimates of requirements are based on plasma phenylalanine concentration and growth. The present study aimed to determine more precisely the phenylalanine requirements in patients with the disease by use of indicator amino acid oxidation, withl-[1-13C]lysine as the indicator. Breath13CO2 production (F13 co 2) was used as the end point. Finger-prick blood samples were also collected for measurement of phenylalanine to relate phenylalanine intake to blood phenylalanine levels. The mean phenylalanine requirement, estimated using a two-phase linear regression crossover analysis, was 14 mg · kg−1 · day−1, and the safe population intake (upper 95% confidence interval of the mean) was found to be 19.5 mg · kg−1 · day−1. A balance between phenylalanine intake and the difference between fed and fasted blood phenylalanine concentration was observed at an intake of 20 mg · kg−1 · day−1. The similarity between these two values (19.5 and 20 mg · kg−1 · day−1) suggests that the maximal phenylalanine intake for children with PKU should be no higher than 20 mg · kg−1 · day−1.

Generalized Procrustes Analysis (GPA) is a multivariate statistic method that is used at the evaluation of sensory analyses in the food industry. GPA provides benefit in terms of decreasing the difference between the panellists and bringing the data obtained from different panellists together. In this study, the aim was to determine the effect of a pre-slaughter fasting period on sensory characteristics of lambs fed with different rations using GPA. Semi-trained panellists formed from twenty-six persons were requested for evaluation of the meat samples such as tenderness, juiciness, flavour and overall liking rated on a scale of 1(extremely dislike) to 9 (extremely good). The first two factors obtained by GPA explained 66.74% of total variability. As a result of the analysis, it was determined that 12 h and 24 h fasting of lambs fed barley supplemented with alfalfa hay were less preferable when compared to lambs fed alfalfa hay only. In addition, lambs in both groups with 48 h fasting were preferred less by the panellists. In conclusion, GPA analysis provides useful data concerning the sensitivity of each panellist in a sensory panel test.

In Ni-13.0at%Si-3.1at%Fe alloy, when γ/γ’ two-phase microstructure formed at 1123 K is isothermally heated at 923 K which is lower than the temperature where the initial γ/γ’ microstructure forms, the phase-separation of γ/γ’ precipitate phase occurs and γ particles newly appear in each cuboidal γ’ precipitate. While in Ni-10.2at%Al-10.8at%Fe alloy, when γ/γ’ two-phase microstructure formed at 1023 K is isothermally heated at 1123 K which is higher than the temperature where the initial γ/γ’ microstructure forms, the phase-separation of γ’ precipitate phase takes place and γ particles newly appear in each cuboidal γ’ precipitate. Such appearance of new γ particles in γ’ precipitates can be explained by the difference in the volume fraction of γ phase that should exist in the γ/γ’ two-phase system depending on the heating temperature.
