Melastoma malabathricum Linn. commonly known as ‘senduduk’ in Malaysia, belongs to Melastomataceae family. The study was carried out to screen the phytochemicals present in methanol extract of leaf, flower, fruit, and stem and to evaluate the antioxidant activity, total flavonoid and total phenolic contents of these different parts. Phytochemical screening showed that all parts of this plant contained tannins, steroids, phenols and flavonoids. The flower extract was found to have highest total phenolic whereas the leaf demonstrated the highest flavonoid content followed by flower. The results of antioxidant activity from the DPPH assay showed that the flower has the highest radical scavenging activity comparable to quercetin standard while stem showed the lowest activity. The higher content of total phenolics, flavonoids and antioxidant capacity of flower suggests the possibility of its incorporation and exclusion of stem in M. malabathricum preparations for development of newer effective drugs.

The flowers of Rhododendron arboreum Smith is a source of polyphenolic compounds. A flavonol, quercetin, was isolated from ethyl acetate fraction of the methanol extract of flower petals of R. arboreum by repeated Sephadex LH-20 column chromatography. Isolated quercetin was characterized by comparing melting point, Rf values, UV and IR spectra with authentic quercetin. The isolated quercetin was used as a standard for the estimation of total flavonoids.  Total phenolic and total flavonoid content in different parts of R. arboreum was carried out spectrophotometrically using Folin-Ciocalteu reagent and Aluminium chloride reagent respectively. Gallic acid and quercetin were used as standard for the construction of calibration curve of phenolic and flavonoid respectively. The results showed that the highest total phenolic content was detected in the 70% acetone extract of the flowers (600 mg GAE/g extract) and petals (600 mg GAE/g extract) and the lowest amount was detected in methanol extract of stem (188 mg GAE/g extract). Similarly, the highest total flavonoid was detected in the 70% acetone extract of the twigs (170 mg QE/g extract) and the lowest amount was detected in the methanol extract of stem (45 mg QE/g extract).  The antioxidant activity of the methanol extracts obtained from different parts of R. arboreum was determined by 2, 2- diphenyl-1-picryl hydrazyl (DPPH) assay and the radical scavenging activity (IC50) was calculated. The highest free radical scavenging effect was observed in leaves with IC50=8.34 ?g/ml and lowest was observed in stem with IC50=67.83 ?g/ml. The IC50 values, total phenolic and total flavonoid content (correlation coefficient R2= 0.923 for phenolic, R2= 0.965 for flavonoid) were correlated which showed strong correlation indicating that the major components responsible for antioxidant activity is phenolics. The highest the phenolic content, the lowest the IC50 value observed. The result indicated that R. arboreum is a rich source of high value polyphenols as natural antioxidant to use in preventive medicine as well as in food and pharmaceutical industry.Scientific World, Vol. 12, No. 12, September 2014, page 34-40

Abstract Background: Rhus vulgaris is rich in various classes of polyphenols and flavonoids that act as free radical scavengers and reduce oxidative stress and cure various harmful human diseases. The plant is a traditionally known medicinal plant which is used against a number of diseases including cancer. Methods: The current investigation points towards the investigating quantitative phenolic contents, flavonoid contents and the free radical scavenging activity & antioxidant activity of Rhus vulgaris extract in n-hexane, acetone, and 80% of aqueous methanol. The phenolic contents were estimated by Folin–Ciocalteu procedure and gallic acid as a reference molecule whereas the total flavonoid contents were determined by using aluminum chloride and catechin. In addition, the dilution serial method was used to evaluate the leaves extract of rhus vulgaris, and the DPPH (2,2-diphenyl-1-picrylhydrazyl) method was utilized to assess the above-mentioned extracts against oxidative stress. Results: The result revealed that the ranges of total phenolic content from 5.82 ± 4.6 to 83.15 ± 7.6 mg GAE/g of the dry weight of extract, expressed as gallic acid equivalents. The total flavonoid concentrations were varied from 2.21 ± 7.34to 23.47 ± 4.87 mg CE/g, expressed as catechin equivalents. Antioxidant activity of extracts was expressed as the concentration of DPPH radical’s inhibition ranges from 1.2 ± 0.32 to 22.86 ± 3.71 mg AAE/ g. Conclusion: The 80% aqueous methanolic extract of Rhus vulgaris showed the highest phenolic and flavonoid contents and strong antioxidant potential and it could be used as antibiotics for different curable and incurable diseases.

The current investigation is intended to evaluate the content of phytochemical constituents and antioxidant potential of hydroalcoholic extracts of stem and root of Grewia serrulata DC (HAESGS & HAERGS) and leaf and bark of Grewia Nervosa (Lour.) panigrahi (HAELGN & HAEBGN). Initially, all the extracts at different concentrations were estimated for their total phenolic content and total flavonoid content. The study was further extended for their antioxidant potential evaluation using various in vitro methods such as 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical, hydroxyl radical and superoxide radical scavenging assays. The total phenolic content (mg gallic acid equivalent per gram of extract) was high in HAELGN (170.82±0.19) and HAERGS (123.00±0.48) than HAESGS (111.2±0.26) and HAEBGN (119.60±0.23). The total flavonoid content (mg quercetin equivalent per gram) is greater in HAERGS (71.24±0.50) and HAESGS (65.68±0.27) than HAELGN (55.82±0.35) and HAEBGN (62.38±0.45). The IC50 values (µg/ml) of different plant extracts inferred that DPPH radical scavenging activity is greater in HAELGN (42.91±0.88) and HAEBGN (53.87±0.35) than HAESGS (126.73±1.20) and HAERGS (88.87±1.25). However, hydroxyl and superoxide radical scavenging activity is more in HAERGS (135.41±1.19 & 88.00±1.42) and HAELGN (172.28±1.91 & 108.163±1.09) than HAESGS (237.3±1.65 & 110.074±0.87) and HAEBGN (204.7±1.04 & 125.54±1.07). The results of present comprehensive analysis demonstrated that both the plants Grewia serrulata DC and Grewia Nervosa (Lour.) panigrahi possess high phenolic, flavonoid contents and potential antioxidant activity, and could be used as a valid source of natural antioxidants and might be utilized for pharmacological screening of various therapeutic activities. While extensive research has been performed on the composition and cooking quality of commodity beans, relatively little is known about pigmented heirloom varieties and the effects of processing on their antioxidant capacity. The aim of this study was to evaluate the effect of soaking and cooking on antioxidants in four heirloom bean varieties compared to Pinto. Water absorption kinetics, soaking and cooking time, DPPH radical scavenging activity, and total phenolic and total flavonoid content were determined in raw, soaked, and cooked samples. Heirlooms required less time to hydrate compared to Pinto, whereas cooking times were similar. The effect of soaking on antioxidant capacity and flavonoids was minimal compared to cooking, which led to losses of up to 57%. Each pigmented heirloom bean had specific characteristics, and three of them had equal or higher amounts of antioxidants or antioxidant activity than Pinto at every processing step. Among heirlooms, Koronis Purple and Jacob’s Cattle had the highest antioxidant activity and Jacob’s Cattle and Tiger’s Eye the highest amount of flavonoids, even after cooking.

In this work, we reported a comparative study on the phytochemical screening and antioxidant activity of aqueous extract from different parts of Moringa oleifera plant, i.e. leaves, twigs, stem barks, and woods. The phytochemical content of each extract was screened through qualitative analysis and followed by quantitative analysis for the determination of total phenolic content and total flavonoid content. The results showed that different parts of Moringa oleifera plant would give different extracted natural products. Either leaf or twig part of Moringa oleifera contained alkaloids, flavonoids, saponins, carbohydrates, polyphenols, proteins, amino acids, and phenolics. Meanwhile, only flavonoids, carbohydrates, and phenolics were found in the aqueous extract of the wood part of Moringa oleifera. In agreement to the phytochemical assay, the aqueous extract of leaf part of Moringa oleifera gave the highest total phenolic content (684 ± 16.7 µg GAE mL-1) and the highest flavonoid content (514 ± 26.1 µg CE mL-1) than other parts. Consequently, the leaf extract exhibited the highest antioxidant activity (64.6 ± 0.69%) which is close to the antioxidant activity of ascorbic acid (71.7 ± 0.56%) as the positive control. This finding is critical for the utilization of Moringa oleifera leaf part as the most potent source of antioxidant agents in the future.

Subcritical water extraction (SWE) has been employed for the extraction of bioactive compounds from plant materials with cost-effectiveness, less consuming time, and environmental sustainability. To explore the effects of thermal processing during SWE, total organic content (TOC), total sugar, polysaccharides, total phenolic content (TPC), total flavonoid content (TFC), and antioxidant activity (ABTS and DPPH assays) of eight aqueous extracts have been quantitatively investigated. The results indicated that elevated temperatures indeed resulted in significant changes in the constituents and antioxidant activities of okara extracts. Among them, the extract obtained at 220°C exhibited the highest total phenolic, flavonoid content, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical-scavenging activity, and ABTS [2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonate)] radical-scavenging activity. However, phenolic compounds were destroyed after the treatment above 230°C, suggesting that any polymer processing is improper to undertake at higher than this value to achieve the high antioxidant activity. Moreover, a significant positive correlation between TPC or TFC and antioxidant capacity (DPPH and ABTS) values was detected.

In the present study the extracts ofin vivoandin vitrogrown plants as well as callus tissue of red clover were tested for their antioxidant activities, using different extraction solvent and different antioxidant assays. The total flavonoid and phenolic contents as well as extraction yield of the extracts were also investigated to determine their correlation with the antioxidant activity of the extracts. Among all the tested extracts the highest amounts of total phenolic and total flavonoids content were found in methanol extract ofin vivogrown plants. The antioxidant activity of tested samples followed the orderin vivoplant extract > callus extract >in vitroextract. The highest reducing power, 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging, and chelating power were found in methanol extracts ofin vivogrown red clover, while the chloroform fraction ofin vivogrown plants showed the highest 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, superoxide anion radical scavenging and hydrogen peroxide scavenging compared to the other tested extracts. A significant correlation was found between the antioxidant activity of extracts and their total phenolic and total flavonoid content. According to the findings, the extract ofin vitroculture of red clover especially the callus tissue possesses a comparable antioxidant activity to thein vivocultured plants’ extract.

Citrus fruits exhibit a high level of different phytoconstituents, of which the changes in the different parts of the fruit during ripening have not been thoroughly studied yet. Thus, in this study, we have investigated how different parts of pomelo fruit (Citrus grandis L.) are modified throughout the development of two consecutive growing seasons. In detail, the main phytochemical compounds, such as total phenolic content, total flavonoid content, antioxidant capacity, DPPH free radical scavenging activity, Ferric reducing antioxidant power (FRAP), and naringin and tannin content, were analyzed. A systematic metabolism of these compounds was found during the development of the fruit, but some pomelo tissues showed a fluctuating trend, suggesting a dependence on the different growing season. Focusing on the tissue distribution of these compounds, the fruit membrane contained the highest level of total phenolic and flavonoid content; fruit flavedo displayed the highest antioxidant capacities and FRAP activities, whereas maximum accumulation of naringin was noticed in fruit albedo. Instead, the highest DPPH free radical scavenging activity and tannin contents were found in the pomelo juice. Regarding the distribution of compounds, a possible bias pattern for the accumulation of those compounds has been noticed throughout the fruit development. From the GC-MS analysis, a total of 111 compounds were identified, where 91 compounds were common in both seasons. Overall, these results could be useful for the food processing industry as guidelines for excellent quality foods and for introducing health-beneficial products and components into our daily diets.

This experiment aims to determine the correlation of total phenolic and flavonoid content of jati putih leaves fraction (Gmelina arborea Roxb.) towards Antioxidant activity .  Sample was extracted by maceration method using ethanol 70% to obtain the ethanol extract (EE), followed by liquid-liquid extraction method to obtain fraction of ethyl acetate (EA) and n-Hexane (EH). The phytochemical screening  and determination of total phenolic and flavonoid content were done by colorimetric method. Antioxidant activity were done by DPPH, FRAP and ABTS methods. Phytochemical screening showed positive results for flavonoids, phenolic and saponins.  The largest total phenolic content was found on EA (11,59 µg/ml ± 0,3 %b/b EAG) and the largest total flavonoid content was on EA (3,88 µg/ml ± 0,02 %b/b EK). The total phenolic and flavonoid content of Jati putih leaves has a correlation with antioxidant activity. The coefficient correlation of activity on reducingDPPH radical was 56,7% (total of phenolic content) and 57,8% (total of flavonoid content) and on iron reduction power in FRAP method  was 99,9% (total of phenolics and flavonoids content). The relationship with the activity in reducing radical ABTS obtained coefficient correlation of 57,0% and 58,1% for total phenolic and flavonoids contents, respectively.
