Renodulation and nitrogen fixing potential of Acacia nilotica inoculated with Rhizobium isolates

Renodulation and nitrogen fixation potential of indigenous and exotic isolates of Rhizobium were studied in a field experiment with Acacia nilotica from July 1990 to June 1991. The inoculum isolates belonged to different serotypes and did not show cross-reaction with the native population of Rhizobium. Nitrogenase activity of root nodules observed at 4-month intervals covering three seasons snowed a decline during winter months, which corresponded with the senescence of the nodules. Maximal renodulation as checked by serotyping and antibiotic resistance pattern was observed with isolate AB 3 followed by AD 4 and USDA 3325. The highest nitrogenase activity was exhibited in plants inoculated with AD 4 at 12 months. Plants inoculated with Rhizobium isolate USDA 3325 showed the highest increase in dry-matter yield at 12 months. Except for AB 3, dual inoculation with Rhizobium isolates and Glomus fasciculatum did not enhance dry matter production as compared with uninoculated controls or single inoculation of either Rhizobium isolate or G. fasciculatum.Key words: Rhizobium, Acacia nilotica, nitrogenase activity, renodulation.

ABSTRACT The elephant-grass undergoes successive cutting and periodical evaluations that it possible to identify clones with high stability for dry matter production, which can be used for energy production. The present study was carried out to evaluate stability dry matter yield for different parametric and non-parametric methods in elephant grass genotypes for biomass production undergoes successive cutting in the agroclimatic conditions of the Norte Fluminense (RJ, Brazil). The variable measured in the 40 genotypes was dry matter yield (DMY) at 2009, 2010 and 2011 in a field study designed as randomized block design with two replicates. Each sample was grown in different environment condition. The stability methods tested were the Yates and Cochran’s, Plaisted and Peterson’s, Wricke’s ecovalence, Annicchiarico’s, Lin and Binns’ and Kang and Phan’s. Results indicated that cutting (E) and genotypes (G) influenced significantly on the performance of dry matter yield. The non-parametric stability methods were effective for the evaluation of stability in dry matter yield. Genotypes Mercker, Pinda-México, Mercker 86-México, Guaçu/IZ, Mercker Pinda, P-241-Piracicaba and Cubano Pinda were stable stability dry matter yield. Hence, there are genotypes may be exploited in future breeding programmes in order to improve productivity of upland elephant grass over environment.

ABSTRACTSilage cut twice annually (June and August) from a tetraploid red clover/grass sward and three times annually (May, July and September) from a low nitrogen (N) and high N perennial ryegrass/white clover sward was fed in proportion to dry-matter yield from each cut, over a 10-week period, each winter for 3 years to castrated male cattle of initial live weight 401 kg in year 1 and 425 kg in years 2 and 3. The silages were supplemented with 0, 1, 2 and 3 kg concentrate per head daily.Total dry-matter yield from the red clover/grass sward was similar to that from the perennial ryegrass/white clover sward (high N grass) receiving 360 kg N per ha but the digestibility, particularly of first cut material was much lower. Dry-matter production of the low N grass/white clover sward was 0·73 of high N grass sward and produced silages of similar digestibility and fermentation.Dry-matter intakes by the cattle were higher on the legume-based silages in years when clover made a worthwhile contribution to total yield, but this did not significantly improve utilization or animal performance compared with high N grass silage. Mean daily carcass gain per head on red clover/grass silage was 0·41 kg which was significantly less than the 0·61 kg on white clover/grass silage and 0·59 on high N grass (P < 0·001). Carcass output from red clover/grass silage was 618 kg/ha and 629 kg/ha from white clover/grass, both of which were significantly less than the 863 kg/ha from the high N grass silage (P < 0·001). Dressing proportion was also significantly poorer in animals fed red clover/grass silage compared with the other silage types.

Variation in water-use efficiency (W, g of total dry matter produced/kg water used), and its correlation with cultivar isotope discrimination in leaves (Δ) was assessed in peanut plants grown in small canopies in the field. Plants were grown in separate minilysimeters that were both embedded in the ground and positioned above the crop. Differences among cultivars were found in W and � and the relationship between W and Δ was compared for plants grown in open and closed canopies. Genetic variability in W in plants grown in the field under non-limiting water conditions was demonstrated, with Tifton-8, of Virginia habit, having the highest W (3.71 g/kg) and Rangkasbitung, an Indonesian cultivar of Spanish habit, the lowest (2.46 g/ kg). Variability in W was due to variation in total dry matter production more than that of water use. A strong negative correlation was found between Δ and W, and also between Δ and total dry matter. The relationship between whole plant W, including roots, and Δ was stronger than that between shoot W, without roots and Δ. The improvement occurred because of variation among cultivars in the root to shoot ratio. This highlights the importance of taking account of root dry matter in studies concerning W. There were significant differences in W and Δ between plants in pots above-ground compared to pots in the ground, with above-ground plants having significantly lower values of both W and Δ. The ranking of W and Δ among cultivars was not affected by the contrast in environment, which suggests these parameters are under strong genetic control. Total above-ground dry matter yield at maturity was negatively correlated with Δ, while pod yield was not. It appears a negative association between harvest index and Δ may exist; however not all cultivars used in this and other studies follow this response. Both water-use efficiency, Wand total dry matter production are negatively correlated with Δ in leaves of peanut plants grown in small canopies in the field. Measurement of Δ may prove a useful trait for selecting cultivars with improved W and total dry matter yield under field conditions.

In two experiments, one with alfalfa and the other with birdsfoot trefoil, we have been able to show that a late cut in October does not adversely affect dry matter yield (DAI) of the legumes. Three cuts during a harvesting season ending on September 5 were detrimental to alfalfa, since lower yields were obtained the following year of harvest. Birdsfoot trefoil varieties were affected differentially. Three cuts did not affect the yield of Viking the following year, but did so of Empire. Significant differences in dry matter yields were obtained between DuPuits and Vernal and between Viking and Empire. Protein in the forage followed inversely the same pattern as that of DM yields, that is, where the intervals were shortest, the protein content was highest, and conversely. The total available carbohydrate and the nitrogen content of the roots were lowest following the treatments which had the shortest intervals between them.

Under clearly defined conditions one-step acquisition of viomycin resistance by a normally effective strain of Rhizobium meliloti resulted in one-step acquisition of ineffectiveness in nitrogen fixation, which probably occurred with a one-gene change in the R. meliloti genome. Two-step mutants retained their ability to produce root nodules but such nodules also were ineffective. Increased sensitivity of the viomycin-resistant mutants to glycine and D-alanine was not noted. Bacteroids were not seen in nodules formed by the viomycin-resistant mutants on their homologous host plant. Nitrogenase activity was not detected, by acetylene reduction, in detached ineffective nodules, whereas effective nodules formed 10.6 μmoles of ethylene per hour per gram of nodules. Growth of the effective parent strain in a low concentration of viomycin resulted in elongation and swelling of the cells so that they appeared as artificially produced bacteroids. Viomycin-resistant mutants did not undergo this transformation. Antigens could be readily extracted by hot- and cold-saline extraction of wet packed cells of both resistant and sensitive cultures but antigenic differences, which may have indicated cell wall differences, were not noted.

Two experiments were established 1 May through 1 December 1991 and 1992 to determine the monthly nutrient uptake and dry matter production of the 'Gunung' yam (Dioscorea alata) grown on an Ultisol. During the first year the plants were fertilized with 0; 667; 1,333; 2,000 and 2,667 kg/ha of a 15-5- 15-5 (N, P2O5, K2O and MgO) fertilizer supplemented with a minor element mixture. No fertilizer was applied the second year. Biomass harvests were conducted at 2, 3, 4, 5, 6 and 7 months after planting. At each harvest, the plants were dug-up and separated into leaf-laminas, vine and petioles, roots and tubers. Fresh and oven-dry weights of the plant components were determined and samples from each were analyzed for N, P, K, Ca and Mg. Regardless of the year, tuber dry matter yield was not significantly affected by the fertilizer treatment. Maximum nutrient uptakes were 214 kg/ha of N, 19 kg/ha of P, 223 kg/ha of K, 95 kg/ha of Ca and 9 kg/ha of Mg. Nitrogen, K and Ca uptake peaks occurred about five months after planting. Maximum dry matter production was 11,303 kg/ha, 8,672 kg/ha of which was tuber dry weight. The dry matter production peak occurred at the completion of the 7-month cropping cycle. The plants utilized 24.7 kg/ha of N, 2.2 kg/ha of P, 25.7 kg/ha of K, 11.0 kg/ha of Ca and 1.0 kg/ha of Mg, for every 1,000 kg/ha of edible dry matter produced.

Abstract This study evaluated dry matter yield and nutritional characteristics of different oat genotypes (Avena spp.) for ensiling. Treatments consisted of genotypes of white oat IPR 126 (Avena sativa), black oat Cabocla IPR and Agrocoxilha (Avena strigosa), and BRS Madrugada and BRS Centauro (Avena vertis). Oats were harvested at the phenological stage of milk/dough grain. The design was a completely randomized block scheme, with five treatments and three replications per treatment. The IPR Cabocla genotype showed the highest dry matter content (before and after silo opening) and in vitro dry matter digestibility of fresh forage. No differences were observed for in vitro dry matter digestibility and dry matter losses among silages. The highest silage digestible dry matter yield (kg ha-1) was observed for the BRS Centauro genotype. Thus, despite the better nutritional quality presented by the IPR Cabocla genotype before ensiling, BRS Centauro genotype presented a higher yield of digestible dry matter per hectare.

In Brazil, elephant grass has been researched for energy generation, as it represents an alternative energy source by virtue of its biomass production. The present study was developed to examine the adaptability and energy-biomass production stability of 73 elephant-grass genotypes under a biannual-harvest regime, using the methodologies proposed by Eberhart and Russell and Cruz. The experiment was carried out at the northern region of Rio de Janeiro State, Brazil. Nine harvests and subsequent evaluations were performed at six-month intervals. Each harvest was considered an environment of genotype evaluation. After the plants were harvested, their dry matter yield (DMY) was estimated in t ha-1 harvest. Combined analysis of variance revealed highly significant effects of genotypes, harvests, and genotype &times; harvest interaction, by the F test. In five of the nine evaluated harvests, the genotypes had an average dry matter yield greater than the overall mean. The method of Eberhart and Russel was effective in identifying highly adaptable elephant-grass genotypes with high dry matter production stability throughout the nine harvests. When the method of Cruz was used, no genotypes were found comprising high yielding ability, adaptability to unfavorable environments, responsiveness to environmental improvement, and high stability altogether.

Neste estudo, objetivou-se estudar a utilização do o capim tifton 85 (Cynodon sp.) em wetlands. Para isso, avaliou-se a produção de matéria seca quando submetido a diferentes tempos de detenção hidráulico. Os resultados permitiram concluir que o capim tifton 85 mostrou-se adequado à utilização em sistemas de tratamento do tipo wetland, sendo o tempo de detenção hidráulico de dois dias (referente a uma taxa de aplicação superficial de 110 kg ha-1 d-1 de DBO) proporcionou a maior produção de matéria seca e, por consequência, maior de remoção de nutrientes.   Palavras-chave: Cynodon spp., massa seca, sistema alagado construído, água residuária.     GUIMARÃES, J. The objective of this study was to study the use of tifton 85 grass (Cynodon sp.) in wetland systems. For this, the dry matter production was evaluated when subjected to hydraulic retention times. The results allowed to conclude that tifton 85 grass was suitable for use in SAC treatment systems, with a two-day hydraulic retention time (referring to a surface application rate of 110 kg ha-1 d-1 of BOD) provided the highest dry matter yield and, consequently, higher nutrient removal.   Keywords: Cynodon spp., dry mass, wetlands, wastewater.
