Oxygen uptake and tetrazolium reduction during skin cycle of the mouse

Quantitative measurements of oxygen consumption and triphenyl tetrazolium chloride reduction by intact skin were made during various stages of the skin cycle of the mouse. Values are highest during early anagen, when growth is most rapid: Qo2 is 2.47 ± 0.10 (mean ± se) and TTC is 537 ± 24.6. In late anagen the activities decrease 40–50%: Qo2 is 1.24 ± 0.10 and TTC is 315 ± 20.8. There is a close relationship between tetrazolium reduction and oxygen consumption during all phases of the growth cycle of mouse skin. The coefficient of regression of TTC on Qo2 is 163.2 ( P < .001).

In the present study, we analyzed the main parameters related with the potential fertilization ability of two grapevine varieties, Godello and Mencía, during the years 2017 and 2018. The research was carried out in two vineyards of the Galician winegrowing Designation of Origin areas of Ribeiro and Ribeira Sacra. Ten vines of each variety were selected for bunch and flower counting, pollen calculations, pollen viability studies by means of aceto-carmine (AC) stain and 2, 3, 5-triphenyl tetrazolium chloride (TTC) methods, and the determination of their germination rate. In all vineyards the 50% fruitset was reached, except for Godello in Cenlle during 2017. The mean coulure value was higher for Godello (40.5%) than for Mencía (31%). Analyzing the pollen production per plant and airborne pollen levels, we observed important discordances between them, which can be due to the influence of weather conditions and be related with self-pollination processes. We found important differences on pollen viability depending on the applied method and variety, with higher values for the AC method than the TTC for both varieties in all study plots, and higher values for Mencía variety than Godello. Regarding germination rates, we observed a marked reduction in 2017 with respect to 2018, in all study sites and for both varieties. The analyzed parameters were useful to explain the different productive abilities of Godello and Mencía varieties in the two studied bioclimatic regions of Ribeiro and Ribeira Sacra.

Certain dehydrogenases in the breast muscle of the pigeon and fowl and the leg muscle of the fowl and frog were studied histochemically by the use of 2:3:5: triphenyl tetrazolium chloride. The dehydrogenase activity was found to have a relationship with the colour and the mitochondrial content of the individual muscle fibres. In the pigeon breast muscle, however, the broad white fibres did not show the presence of any of the enzymes studied. It is therefore concluded that these fibres in the pigeon breast muscle are a unique system in which none of the oxidative processes concerned takes place; they cannot be considered as analogous to the white fibres of the other muscles studied.

Eosinophils were isolated from normal human blood by separation over Percoll gradients, which resulted in eosinophil suspensions of a purity higher than 95% and recoveries of about 65%. Normal human eosinophils were found to respond to formyl-methionyl-leucyl-phenylalanine (fMLP) at concentrations greater than 10(-7) mol/L with an increase in the concentration of intracellular free calcium, oxygen consumption, nitroblue tetrazolium reduction, and chemiluminescence. The maximal response of eosinophils to fMLP was lower than that of neutrophils isolated from the same blood samples and required at least ten times as much fMLP as was needed for neutrophils. Low fMLP concentrations (approximately 10(-8) mol/L), which in themselves did not stimulate O2 consumption by either eosinophils or neutrophils, primed these cells to respond to a suboptimal concentration of another stimulus. Purification of eosinophils after treatment of whole blood with fMLP showed that these eosinophils had lost their ability to respond to fMLP. We conclude that normal eosinophils do respond to fMLP and that therefore fMLP should not be used to isolate eosinophils.

Abstract The dynamics of potential oxygen consumption at the sediment surface in a seasonally hypoxic bay were monitored monthly by applying a tetrazolium dye (2-(4-iodophenyl)-3-(4-nitrophenyl)-5-phenyl-2H-tetrazolium chloride [INT]) reduction assay to intact sediment core samples for two consecutive years (2012–2013). Based on the empirically determined correlation between INT reduction (INT-formazan formation) and actual oxygen consumption of sediment samples, we inferred the relative contribution of biological and non-biological (chemical) processes to the potential whole oxygen consumption in the collected sediment samples. It was demonstrated that both potentials consistently increased and reached a maximum during summer hypoxia in each year. For samples collected in 2012, amplicon sequence variants (ASVs) of the bacterial 16S rRNA genes derived from the sediment surface revealed a notable shift in the bacterial community composition before and after the INT assay incubation. Within the bacterial community that was predominated by the ASVs closely related to Woeseia (Woeseiaceae, Gammaproteobacteria), the relative abundance of ASVs affiliated with Arcobacter (Arcobacteraceae, Campylobacteria), a putative sulfur-oxidizing bacterial genus, increased markedly in the summer samples. These findings have implications not only for the group of bacteria that are consistently responsible for the consumption of dissolved oxygen (DO) year-round in the sediment, but also for those that might grow rapidly in response to episodic DO supply on the sediment surface during midst of seasonal hypoxia.

Abstract Tetrazolium chloride was used to visualize the in vitro dehydrogenase activity of splenic tissues from cases with and without hypersplenism. In addition, quantitative measurements of such activity were made in the presence and absence of fluoride, malonate and azide. The data indicate that spleen slices from cases of hypersplenism differed from the controls and were characterized by (a) increased endogenous dehydrogenase activity of the lymphocytes, particularly around the follicles, (b) a decreased azide stimulation in those cases having an initial thrombocytopenia which responded to splenectomy, and (c) a decreased fluoride stimulation in those cases having an initial leukopenia which responded to splenectomy. Routine histological studies confirmed other reports that a constant structural feature of the spleen in hypersplenism is a perifollicular lymphocytic rimming.

Murcha (an amylolytic starter) from different parts of Eastern Nepal were screened for fermentative yeasts. The most potential one was UV-mutated (8W lamp at λ = 254 nm and an intensity of 44.21 Wm-2 for 5-50 s) to study the effect of mutation on growth and fermentation properties. Respiratory-deficient mutants (RDMs) that resulted from the mutation were identified by triphenyl tetrazolium chloride (TTC) overlay technique and replica-plated for further isolation. Cell growth, substrate utilization, and ethanol yield of the mutants were compared with normal cells by carrying out fermentation in high-test cane molasses broth of 30 °Bx. An exhaustive screening of the samples resulted in only two murcha viz., from Laxmimarga (LM) and Udayapur (UD), having the desirable fermentation properties. UV-mutation study of UD and LM yeasts (both identified as strains of Saccharomyces cerevisiae) showed 8-12% survival and ~ 22% RDMs yield of the survived cells. Out of the 8 randomly selected RDMs, only UDm4 (colony No. 4 from UD) showed fermentation properties worth further investigation. Comparison of UD, LM and UDm4 by fermenting molasses (high test) broth of 30°Bx showed the least growth of UDm4 but the highest alcohol yield (9% and 16% more compared to UD and LM, respectively). The present finding indicates that it is possible to improve fermentation properties of feral yeasts from murcha by relatively simple UV-mutation approach. Finding the right mutant (the selective screening part), however, may involve considerable time and effort.

The production of red triphenyl formazan (hereinafter referred to as TPF) from colorless 2,3,5-triphenyl tetrazolium chloride (hereinafter referred to as TTC) by living cells was used as a vitality indicator for examining the effects of 6,7-dihydrodipyrido[1,2-a:2′,1′-c] pyrazidiinium salt (diquat) in elodea (Elodea canadensisMichx) shoots at varying light intensities. Diquat at 10 ppmw completely killed the elodea in 6 hr at 183 ft-c and at almost the same rate at 13,000 ft-c due to light saturation near the lower level. Over 24 hr were required for this effect in the dark. Between 29 and 183 ft-c, TPF production varied inversely with the light intensity. A comparison is made with the destruction of chlorophyll by diquat.

The response of sapwood of swamp white oak (Quercus bicolor Willd.) to mechanical injury was studied over a 28-day period. In cells within 0.5 cm of a wound stimulus, starch grains disappeared and cells were unable to reduce the vital stain, triphenyl tetrazolium chloride (TTC), 8 to 10 days after mechanical injury. Extraneous material and tannins began to accumulate 10 to 12 days after mechanical injury. Nuclei began to disintegrate 12 to 14 days after mechanical injury, but some persisted throughout the period of investigation.With increasing distance from the cambium, starch grains disappeared from ray parenchyma. Cells in the annual rings of sapwood adjacent to the heartwood were unable to reduce TTC. Nuclei were present in all annual rings of sapwood but were lacking in heartwood. Tannins appeared at the heartwood boundary and accumulated in the heartwood.With respect to the criteria used, this investigation indicates that formation of discoloration in sapwood and conversion of sapwood to heartwood appear similar.
