Effects of fasting and restricted refeeding on utilization of ingested energy in rats

Ingested metabolizable energy was partitioned into 3 components--storage, cost of storage, and maintenance--to study energy utilization during 20 days of refeeding following a 3-day fast in adult male Wistar rats. During the refeeding period, one experimental group was refed ad libitum, whereas two others were restricted to approximately 75 and 50%, respectively, of prefast food intake. Energy utilization in the experimental groups was compared with that in nonfasted controls. Energy conservation occurred in all experimental groups during refeeding, and the primary form of that conservation was a decrease in the energy required for maintenance. The decreased requirement for maintenance allowed a greater proportion of ingested energy to be used for restoration of carcass energy. The degree of energy conservation was, in general, proportional to the degree of food restriction during refeeding.

Relevance. Purulent complications occur in 35-45% of patients with surgical diseases, lethality rate from these conditions achieves 25%. The purpose of the study. To investigate the wound process while treating the purulent wounds with immobilized forms of miramistin and benzalkonium chloride in gel of sodium salt of carboxymetylcellulose. Materials and methods. There were analysed results of experimental study of the wound process on 108 male Wistar rats. Experimental animals were divided into 3 groups, each group included 36 rats. In a comparison group ‘Levomekol’ ointment was used for treatment. In the first experimental group treatment was done by the ointment with the following composition: 0,01% solution of miramistin — 100 g, metronidazol –1,0 g, sodium salt of carboxymetylcellulose — 4,0 g, while in the second experimental group ointment composition included benzalkonium chloride 0,02 g, metronidazol — 1,0 g, sodium salt of carboxymetylcellulose — 4,0 g, purified water — up to 100,0 g. The wound process was evaluated by planimetric, microbiological and histological methods of investigation. Results and its discussion. The reduction of wound area in the first experimental group was higher than in a comparison group on the 3rd day of the experiment by 9,7%, on the 5th day — by 7,6%, on the 10th and 15th days — by 10,5%, while in the second experimental group the change was 37,2%, 27,3%, 13,8% and 6,9% respectively. In the first experimental group microbial contamination of the wound was less than in a comparison group by 5,8х106, on the 5th day — by 3,1х105, on the 10th day — by 6,1х104, while in the second experimental group on the 3rd day there were no differences, on the 5th day — by 6,3х105, on the 10th day — 1,1х104. Histological investigations of the wound biopsies on the 10th day of experiment showed that in a comparison group the bottom of the wound was covered by granulation tissue without signs of epithelization, there was full coverage of the wound with a newly formed epithelium in experimental groups. Conclusions. The use of prolonged quaternary ammonium antiseptics in treatment of purulent wounds is pathogenetically justified and efficient.

Ad libitum (AL) supply of standard chow is the feeding method most often used for rodents in animal experiments. However, AL feeding is known to result in a shorter lifespan and decreased health as compared with restricted feeding. Restricted feeding and thus limiting calorie intake prevents many health problems, increases lifespan and can also increase group uniformity. All this leads to a reduced number of animals needed. So-called standard chows are known to be prone to variation in composition. Synthetic foods have a more standard composition, contributing to group uniformity which, like diet reduction, may decrease the number of animals necessary to obtain statistical significance. In this study, we compared the effects of AL versus restricted feeding (25% reduction in food intake) on standard chow versus synthetic food of three different suppliers on body weight (BW), growth, several blood parameters and organ weights in growing female Wistar rats over a period of 61 days. Diet restriction led to a decreased growth and significantly reduced variation in BW and growth as compared with AL feeding. AL feeding on synthetic diets caused a significantly higher BW gain than on chow diets. Due to experimental design, this same effect occurred on food restriction. Blood parameters and organ weights were affected neither by diet type nor by amount. Incidentally, variations were significantly reduced on food restriction versus AL, and on synthetic diets versus chow diets. This study demonstrates that food restriction versus AL feeding leads to a significantly reduced variation in BW and growth, thereby indicating the potential for reduction when applying this feeding schedule.

The effects of differences in meal frequency on body weight, body composition, and energy expenditure were studied in mildly food-restricted male rats. Two groups were fed approximately 80% of usual food intake (as periodically determined in a group of ad libitum fed controls) for 131 days. One group received all of its food in 2 meals/day and the other received all of its food in 10-12 meals/day. The two groups did not differ in food intake, body weight, body composition, food efficiency (carcass energy gain per amount of food eaten), or energy expenditure at any time during the study. Both food-restricted groups had a lower food intake, body weight gain, and energy expenditure than a group of ad libitum-fed controls. In conclusion, these results suggest that amount of food eaten, but not the pattern with which it is ingested, has a major influence on energy balance during mild food restriction.

A group of 16 cows, each suckling one calf, were fed on rations composed of maize meal, cottonseed meal, urea and rice straw in one of four different ratios and at two levels, (a) ad libitum and (b) in restricted amounts. The composition of the feed affected intake. Both the gross intake and the estimated intake of metabolizable energy were closely correlated with liveweight change. The intake of metabolizable energy was correlated with blood glucose in the cows fed ad libitum, but not in the cows fed on restricted amounts of the rations. It was concluded that feed quality significantly affected the blood glucose concentration of the lactating cows, but that within the ranges used in this experiment, feed intake and the intake of metabolizable energy had little effect.

To determine the effects of brief food restriction on fatty acid (FA) metabolism, hindlimbs of F344/BN rats fed either ad libitum (AL) or food restricted (FR) to 60% of baseline food intake for 28 days were perfused under hyperglycemic-hyperinsulinemic conditions (20 mM glucose, 1 mM palmitate, 1,000 μU/ml insulin, [3-3H]glucose, and [1-14C]palmitate). Basal glucose and insulin levels were significantly lower ( P < 0.05) in FR vs. AL rats. Palmitate uptake (34.3 ± 2.7 vs. 24.5 ± 3.1 nmol/g/min) and oxidation (3.8 ± 0.2 vs. 2.7 ± 0.3 nmol · g−1 · min−1) were significantly higher ( P < 0.05) in FR vs. AL rats, respectively. Glucose uptake was increased in FR rats and was accompanied by significant increases in red and white gastrocnemius glycogen synthesis, indicating an improvement in insulin sensitivity. Although muscle triglyceride (TG) levels were not significantly different between groups, glucose uptake and total preperfusion TG concentration were negatively correlated ( r 2 = 0.27, P < 0.05). In conclusion, our results show that under hyperglycemic-hyperinsulinemic conditions, brief FR resulted in an increase in FA oxidative disposal that may contribute to the improvement in insulin sensitivity.

Abstract Background and Aims Vascular calcification (VC) is an important contributor to the high rate of cardiovascular mortality associated to chronic kidney disease. The inability to eliminate phosphorus (P) and the subsequent P retention promotes CV. P metabolism and uremic VC are influenced by obesity and by the caloric content of the diet. Caloric restriction (CR) has been shown to have multiple beneficial effects on health, for example, CR has been reported to improve vascular health and retard vascular ageing. However, to our knowledge the effect of CR on the development of uremic VC has not been explored. We hypothesize that CR may be beneficial to prevent the development of uremic VCs. Thus, the objective of the present study was to determine if rats subjected to CR were protected against VC. Method 48 Wistar rats were divided in four groups. The control diet provided Metabolizable Energy = 3.528 kcal/g and contained 0.6% Calcium (Ca) and 0.6% P. Additional diets of identical composition to the control diet but containing varying levels of Ca and P: 0.9% Ca, 0.9% P; 0.6% Ca, 1.2% P; and 0.9% Ca, 1.8% P, were also used in the experiments. Rats in Group 1 and 3 were fed 15 g/day of the control diet. Rats in Group 2 and 4 were calorie restricted and fed 10 g/day of diet with Ca/P = 0.9%/0.9%. Thus the daily P intake should be identical in the four groups. Uremia was induced by 5/6 nephrectomy (Nx). After Nx rats in Group 1 and 2 were fed ad libitum a diet with 0.6% Ca and 1.2% P. While rats in Group 3 and 4 were fed ad libitum a diet with 0.9% Ca and 1.8% P. Rats were supplemented with calcitriol. At the end of the experiment, rats were sacrificed to obtain blood samples and tissue samples (thoracic and abdominal aortas). After blood collection, plasma was separated by centrifugation and stored at –20° C until assayed. Plasma creatinine, Ca and P were measured by spectrophotometry. Energy intake was calculated based on food intake. VC was studied by histology and by measuring the tissue Ca content. Values are expressed as mean ± standard error (SE), the difference between groups was assessed by ANOVA. Fisher LSD test was used as a post-hoc procedure. p&lt;0.05 was considered significant. Results Before Nx, caloric intake was significantly lower in calorie restricted rats (35.4 ± 0.1 and 35.8 ± 0.1 kcal/day) than in rats eating normal calories (52.7 ± 0.1 and 52.8 ± 0.2 kcal/day); however, P intake was almost identical in the four groups and ranged between 89.8 and 91.6 mg/day. After Nx, rats in all groups reduced food intake and, consequently, caloric intake. Thus, although the P content of the diet was increased after Nx, daily P intake was not increased in Groups 1 and 2; however, P intake was significantly increased in Groups 3 and 4 (120.9 ± 4.6 and 122.2 ± 6.2 mg/day, respectively). In all groups, rats had high plasma concentrations of creatinine and P, and low plasma concentrations of Ca. Also, all rats had elevated Ca content in the aorta. No significant differences between the study groups were found in any of these parameters (Table 1). Von Kossa staining of the aortas showed abundant mineral deposition in the four groups. Conclusion This study shows that, contrary to what was expected, CR did not prevent or ameliorate uremic calcifications.

ABSTRACTPost-weaning metabolizable energy intake, growth of empty-body chemical components and efficiencies of energy utilization were evaluated for Hereford intact males (17) and females (16) and Simmental intact males (15) and females (16) during a 212-day feeding period. Within each breed × sex subclass, animals were assigned to one of three levels of metabolizable energy (ME) intake: (1) 544 kJ/kg M0·75 per day, (2) 795 kJ/kg M0·75 per day, and (3) ad libitum. Body composition of each animal was estimated at the beginning and end of the feeding period by deuterium oxide dilution.Protein and water gain of Hereford and Simmental cattle were similar at restricted levels of intake but were greater for Simmental than for Hereford cattle at ad libitum intakes. Similarly, rates of protein and water gain tended to increase more rapidly in response to increased energy intake by males than by females. Hereford males gained fat and energy slightly more rapidly than Hereford females, but Simmental males gained fat and energy at slower rates than Simmental females.Males had higher maintenance requirements and tended to use ME with less efficiency for maintenance and gain than females. Hereford cattle had lower maintenance requirements and used ME with greater efficiency for both maintenance and gain than Simmental cattle.
