The Effect of Hemodialysis and Acetate-Free Biofiltration on Anemia

The authors monitored, for a period of 12 months, anemia-, nutrition-, and free radical-related parameters and the rHuEPO dose required to maintain target hemoglobin (Hb) in 20 patients with chronic renal failure. Ten patients each were randomized for treatment by either acetate-free biofiltration (AFB) or low-flux hemodialysis (HD). At baseline, Hb levels were 102±2 (AFB) vs. 98±2 g/L (HD) (not significant difference, NS), the rHuEPO dose was 4050±976 vs. 5100±1538 IU/week (NS). Compared with baseline and with HD, lower rHuEPO doses were required during AFB at months 8, 9, 10 and 11, and 12 when they were 2100±510 (AFB) vs. 6000±1153 (HD), p=0.008. Prealbumin, transferrin and cholinesterase levels rose in the AFB group. Kt/V, albumin, transferrin saturation, aluminium, bicarbonate in serum, superoxide dismutase and glutathione peroxidase in erythrocytes, and malondialdehyde and antioxidant capacity in plasma did not differ between the AFB and HD groups. In terms of anemia control, AFB using an AN69 membrane was found to be more advantageous than low-flux HD, AFB improves some nutritional parameters. Aim: To assess the role of antioxidants and lipid peroxidation in the pathogenesis of pre-eclampsia. Study Design: This is a case-control study designed to evaluate the levels of malondialdehyde and total antioxidant capacity with glutathione peroxidase and superoxide dismutase activities in pre-eclamptic. One hundred (100) participants were randomly selected to include fifty pre-eclamptic (test), twenty-five (25) healthy pregnant normotensive women (control A) and twenty-five healthy non-pregnant normotensive women (control B). Place and Duration of Study: This study was carried out at the Holy Rosary Hospital waterside, Onitsha, Anambra state. Nigeria. The study lasted for 13 months. Methodology: One hundred women (50 pre-eclamptic, 25 pregnant normotensives and 25 non-pregnant normotensives) aged 25-40 years were recruited for this study. Blood samples were collected from the participants for the estimation of total antioxidant capacity (TAC), malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities using spectrophotometric methods. Blood pressures were measured using accosson sphygmomanometer. Data were analysed using analysis of variance and Pearson’s correlation coefficient at p < 0.05. Results: The TAC was significantly higher in the pregnant normotensives (p< 0.05) compared with the pre-eclamptic and non-pregnant normotensives. There was no significant difference in the MDA in pre-eclamptic compared with pregnant normotensives and non-pregnant normotensives (p>0.05). A significant difference was observed in the SOD and GPx activities in the pregnant normotensive and non-pregnant normotensives compared with pre-eclamptic. A positive correlation was observed between the blood pressures (systolic and diastolic) and the activities of TAC, SOD and GPx in pre-eclamptic. Furthermore, a positive correlation was observed between MDA levels and SBP (r = 0.019) while a negative correlation (r = -0.225) existed between MDA and DBP. Conclusion: Diminished ability of antioxidants to scavenge free radicals may affect the onset of pre-eclampsia and therefore possible prognostic tool in its management.

Ре-зультати досліджень вказують на те, що хронічнийкадмієвий токсикоз призводить до посиленої акти-вації процесів ліпопероксидації. The features of the antioxidant system of rats with chronic cadmium toxicosiare disclosed. It wasresearched that cadmium chloride in toxic doses reduces enzyme activity of antioxidant system, asindicated by the decrease in enzyme glutathione peroxidase, hlutationreduktazy, superoxide dismutase,catalase and restored glutathione in the liver and blood of rats. The results indicate that chroniccadmium toxicosis leads to enhanced activation of lipid peroxidation.

The objective of research was to study the state of prooxidant and antioxidant systems in the tissues of the lungs, kidneys, pancreas and salivary glands at burn disease. The intensity of the free radical processes was evaluated on the basis of the content of malondialdehyde (MDA) and antioxidant system – based on the indexes of its enzymatic chain: superoxide dismutase and catalase in homogenates of the studied organs. It has been found that changes in experimental burn disease depend on the studied organs and the stage of burn disease. The activation of free radical processes observed in all investigated organs (lungs, kidneys, pancreas and salivary glands). Reactive oxygen species induce lipid peroxidation, which is a universal marker of tissue damage. MDA appears in the body during degradation of polyunsaturated fatty acids and it’s a marker of lipid peroxidation and oxidative stress. It was found the increasing of MDA in all organs, especially in the lungs and kidneys at stage of burn shock. Under these conditions it was observed the decrease of superoxide dismutase and catalase in all investigated organs. At burn disease there is development of disbalance between the action of prooxidant and antioxidant systems due to the activation of free radical processes.

The aim of this study was to evaluate the association between estrogen removal, antioxidant enzymes, and oxidative stress generated by obesity in a MS female rat model. Thirty two female Wistar rats were divided into 4 groups: Control (C), MS, MS ovariectomized (Ovx), and MS Ovx plus estradiol (E2). MS was induced by administering 30% sucrose to drinking water for 24 weeks. After sacrifice, intra-abdominal fat was dissected; adipocytes were isolated and lipid peroxidation, non-enzymatic antioxidant capacity, and the activities of Cu-Zn and Mn superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) were determined. There were no significant differences in the activities of Cu-Zn, Mn SOD, CAT, and GPx between the C and MS groups, but in the MS Ovx group there was a statistically significant decrease in the activities of these enzymes when compared to MS and MS Ovx+E2. The increased lipid peroxidation and nonenzymatic antioxidant capacity found in MS Ovx was significantly decreased when compared to MS and MS Ovx+E2. In conclusion, the removal of E2by ovariectomy decreases the activity of the antioxidant enzymes in the intra-abdominal tissue of MS female rats; this is reflected by increased lipid peroxidation and decreased nonenzymatic antioxidant capacity.

During aging, oxidative stress affects the normal function of satellite cells, with consequent regeneration defects that lead to sarcopenia. This study aimed to evaluate tocotrienol-rich fraction (TRF) modulation in reestablishing the oxidative status of myoblasts during replicative senescence and to compare the effects of TRF with other antioxidants (α-tocopherol (ATF) andN-acetyl-cysteine (NAC)). Primary human myoblasts were cultured to young, presenescent, and senescent phases. The cells were treated with antioxidants for 24 h, followed by the assessment of free radical generation, lipid peroxidation, antioxidant enzyme mRNA expression and activities, and the ratio of reduced to oxidized glutathione. Our data showed that replicative senescence increased reactive oxygen species (ROS) generation and lipid peroxidation in myoblasts. Treatment with TRF significantly diminished ROS production and decreased lipid peroxidation in senescent myoblasts. Moreover, the gene expression of superoxide dismutase(SOD2), catalase(CAT),and glutathione peroxidase(GPX1)was modulated by TRF treatment, with increased activity of superoxide dismutase and catalase and reduced glutathione peroxidase in senescent myoblasts. In comparison to ATF and NAC, TRF was more efficient in heightening the antioxidant capacity and reducing free radical insults. These results suggested that TRF is able to ameliorate antioxidant defense mechanisms and improves replicative senescence-associated oxidative stress in myoblasts.

AbstractData on the values of selected blood antioxidant parameters, i.e. total antioxidant capacity, glutathione peroxidase, and superoxide dismutase in healthy dogs, are lacking. There are no published accepted standard reference methods for their determination. The aim of this study was to determine the values of plasma total antioxidant capacity and the activities of whole blood glutathione peroxidase and erythrocyte superoxide dismutase in 30 healthy client-owned dogs (19 females, 11 males). The effect of age and sex on the measured antioxidant parameters was also investigated. Antioxidant parameters were determined with an automated biochemical analyser, using the commercially available Randox kits. No significant difference in age, weight, and antioxidant parameters was determined between females and males. A significant positive effect of age (p = 0.002, r2= 0.284) on superoxide dismutase activity was confirmed. There was no effect of sex on any of the antioxidant parameters measured. However, we observed a tendency of the effect of sex (p = 0.063, r2= 0.118), as well as age (p = 0.073, r2= 0.111), on the activity of glutathione peroxidase. Our results are in part comparable with the results of other studies in which the same types of methods and samples were used to determine antioxidant parameters. In conclusion, the sex and age of dogs should be taken into consideration when planning a study on antioxidant status parameters.

The purpose of this study was to examine the effects of polysaccharides fromLycium barbarumfruits (LBP) on free radical metabolism of mice after exhaustive swimming exercise. The results showed that LBP supplementation could alleviate fatigue and enhanced the exercise tolerance. Furthermore, LBP supplementation effectively attenuates the exhaustive exercise-induced increased lipid peroxidation and elevates antioxidant enzymes (superoxide dismutase, glutathione peroxidase, and catalase) activities. LBP may have therapeutic role in preventing liver tissue damage during exhaustive exercise.

This article presents the results of research on the influence of cadmium loading on the state level of enzymatic and non-enzymatic antioxidant links of the antioxidant defense system of the organisms of young cattle, such as the activity of catalase, superoxide dismutase, glutathione peroxidase, glutathione levels, selenium, vitamins A and E. It was found that feeding bull calves with cadmium chloride at doses of 0.03 and 0.05 mg/kg of body weight helped to reduce both the enzymatic and non-enzymatic link of antioxidant protection (superoxide dismutase 31%, catalase 13%, glutathione peroxidase 23%, reduced glutathione 10%, vitamin A 28%, vitamin E 31%, selenium 20%). Toxic effects of cadmium promotes change in steady-state concentrations of radical metabolites О2–,ОН˙, НО2˙, which, in turn, trigger the process of lipid peroxidation. The lowest level of indicators of antioxidant defense system in the blood of young cattle was registered on the sixteenth and twenty-fourth days of the experiment, which is associated with increased activation of lipid peroxidation and the disturbaance of the balance between the antioxidant system and lipid peroxidation intensity. The activity of the antioxidant defense system in the blood was different for calves fed with cadmium chloride at doses of 0.03 and 0.05 mg/kg of animal mass. The more cadmium chloride in the feed, the lower the activity of the antioxidant defense system of the calves’ organisms was registered. Thus cadmium chloride depresses the antioxidant defense system, which specifically involves lowering the activity of enzymatic links (catalase, superoxide dismutase, glutathione peroxidase) and non-enzymatic links (reduced glutathione, selenium, vitamins A and E).
