The aim of this study was to investigate the phytochemicals using reverse-phase high pressure liquid chromatography (RP-HPLC), antioxidant, antifungal and antibacterial activities of Seriphidium oliverianum stem extracts. The extraction was carried out by conventional shaking process (CSP) and ultrasonic assisted process (UAP). The highest total phenolic contents (97.85 ± 0.735 mg gallic acid equivalent (GAE)/g sample) and flavonoid contents (188.15 ± 0.53 mg catechin equivalent (CE)/g sample) were found in methanol extract obtained by CSP. Antioxidant activity was investigated using DPPH° scavenging assay and reducing power assay. Methanol extract using UAP showed the highest DPPH° scavenging activity (79.95% ± 1.80%) followed by methanol and butanol extracts obtained through CSP. Moreover, methanol extracts using CSP showed highest reducing activity (1.032 ± 0.0205 absorbance). In-vitro antimicrobial activity was studied using most common infection causing fungal and bacterial strains. Anti-fungal activity of methanol extract using CSP showed the highest zone of inhibition (10.5 mm) against F. avenaceum fungal strain, while aqueous extracts obtained through showed the highest antibacterial activity (22 ± 1.32 mm zone of inhibition) against S. aureus. The results showed that the methanol stem extract of S. oliverianum is a valued candidate for further screening and could be processed for in-vivo infection induced animal trials.

Objectives: The work aims to screen the antioxidative potentials of different crude extracts of the fronds of two medicinally important pteridophytes of Southern Assam, India, namely, Cyclosorus interruptus (Willd.) H. Itô and Pronephrium nudatum (Roxb. ex Griff.) Holttum. Methods: Frond extracts of the pteridophytes were prepared by Soxhlet hot extraction method. Total phenolic content (TPC) and total flavonoid content (TFC) of the hexane, ethyl acetate, acetone, and methanol extracts of the fronds of the plants were done by following standard protocol. In vitro assessment of the antioxidative behavior of the extracts was performed using standard 2,2-diphenyl-1-picrylhydrazyl scavenging assay, reducing power assay, metal chelating assay, hydroxyl, superoxide, and 2,2’- azino-bis (3-ethylbenzothiazoline - 6 – sulfonic acid) radical scavenging methods in different in vitro systems. Results: Preliminary phytochemical analysis implicated the presence of phenolic and flavonoid compounds in all the frond extracts. The methanol extract of the fronds of both the plants showed maximum phenolic and flavonoid contents in comparison to the other extracts, however, that of C. interruptus was found to be higher than P. nudatum. Antioxidative potentials of the said extracts were also found to be impressive and noteworthy. The decreasing order of the antioxidative efficacies of the extracts was found to be same as that of TPC and TFC of the extracts. Conclusion: It is pertinent to comment that the methanol extract of the fronds of both the plants may be treated as a potential source of natural antioxidants.

The present study aims at comparative study between plant and callus extract, with respect to analysis of phytochemical constituents, antioxidant, antibacterial activity and cytotoxicity properties of Centratherum punctatum using aqueous medium and different solvents such as, methanol and ethyl acetate. In vitro studies in MS media supplemented with BAP 4.5 mg/L + Kn 4.0 mg/L has shown the high callus induction percentage of 92.33% with a maximum callus weight of 1.08 g. The phytochemical analysis of aqueous, methanol and ethyl acetate extract of C. punctatum in vivo plant and in vitro callus showed the presence of alkaloids, flavonoids, phenols and carbohydrates. The aqueous extract of both plant and callus showed the presence of tannins, proteins and steroids whereas the methanol extract showed the presence of tannins, amino acids and terpenoids. The ethyl acetate extract showed terpenoids and protein. FTIR analysis of plant and callus aqueous extract had a maximum characteristic band at 3399.87 cm-1 and 3412.73 cm-1 respectively indicating the presence of N-H stretching. GC-MS analysis revealed the presence of 11 different compounds in ethyl acetate extracts of plant and the callus extract revealed the presence of 15 different compounds which was absent in the plant extract. Plant extract exhibited maximum total phenol content than callus extract. The in vitro callus extract showed higher DPPH radical scavenging activity with lower inhibition percentage than in vivo plant extract. A maximum zone of inhibition was observed in methanol extract of in vivo plant and in vitro callus (15 mm and 14 mm respectively) against Bacillus subtilis. The ethyl acetate extract of in vivo plant and in vitro callus had a zone of 14 mm and 12 mm against E. coli. A maximum zone of inhibition (12 mm and 11 mm respectively) was observed in both methanol and ethyl acetate of in vivo plant and in vitro callus against Staphylococcus aureus. Antiproliferative analysis revealed that in vivo plant has inhibitory percentage of 23.6 whereas callus exhibited 28.5% against HeLa cells.

  The Himalayan region is rich in flowering plants diversity, including medicinal and wild edible plants. These plants possess variety of therapeutically important compounds, like alkaloids, flavonoids, phenolics, saponins, steroids, tannins and terpenoids in different parts. Presence of significant amount of these compounds makes a species valuable to be used as super foods or medicines. Furthermore, plant extracts especially those of fruits are basically attributed with natural antioxidants, either in form of vitamins or color-inducing pigments, which have become the target to replace the synthetic antioxidants. In this regard, fruits of Berberis species, one of the popular wild edible fruits of Nepal, are noted for their antioxidant property since time immemorial. The present study was done with an attempt to quantify the antioxidant potential and antibacterial activities of the fruit extracts of four taxa of Berberis (B. angulosa var. angulosa, B. angulosa var. fasciculata, B. aristata and B. asiatica) from Nepal and to correlate the antioxidant potential with various phytochemicals present in the extracts. Methanolic fruit extracts were used to spectrophotometrically quantify total phenolic and flavonoid contents. DPPH free radical scavenging assay and antibacterial assay were carried out in in-vitro condition. Preliminary phytochemical analysis revealed high polyphenol content (52.60 ± 3.73 and 58.07 ± 1.44 mg GAE/g) and better antioxidant property (35.29 ± 3.01 and 29.15 ± 2.01 μg/ml) respectively in B. angulosa var. angulosa and B. angulosa var. fasciculata than in the fruit extracts of B. aristata and B. asiatica. In contrast, fruit extracts of B. asiatica showed the highest total flavonoid content (27.52 ± 0.56 mg QE/g) than did by the extracts of other taxa studied. The fruit extract of B. aristata and B. angulosa var. fasciculata at very high concentration (200 mg/ml) showed maximum zone of inhibition (ZOI) against tested bacterial strains, Staphylococcus aureus and Escherichia coli. In contrast, B. asiatica did not show any ZOI for both of the tested bacterial strains. As fruits are better sources of antioxidant with greater accumulation of flavonoids and phenolics, wild fruits should be equally spaced for their better efficiency as cultivated ones.Botanica Orientalis – Journal of Plant Science (2016) 10: 6–11

Abstract Background The objective of the study is to evaluate the hepatoprotective activity of methanolic extract fractions of Lindernia ciliata (LC) and development of qualitative analytical profile of the bioactive fraction using HPLC fingerprinting analysis. All the fractions of methanolic extract of Lindernia ciliata (LCME) are assessed for their total phenolic, flavonoid contents and in vitro antioxidant properties by using DPPH, superoxide, nitric oxide, hydroxyl radical scavenging activities and reducing power assay. Acute toxicity study was conducted for all the fractions and the two test doses 50 and 100 mg/kg were selected for the hepatoprotective study. Liver damage was induced in different groups of rats by administering 3 g/kg.b.w.p.o. paracetamol and the effect of fractions were tested for hepatoprotective potential by evaluating serum biochemical parameters and histology of liver of rats. The effective fraction was evaluated for its antihepatotoxic activity against D-Galactosamine (400 mg/kg b.w. i.p.) and in vivo antioxidant parameters viz., Glutathione (GSH), Melondialdehyde (MDA) and Catalase (CAT) levels are estimated using liver homogenate. Results Among all the fractions, butanone fraction of LCME, (BNF-LCME) has shown better hepatoprotective activity and hence it is selected to evaluate the antihepatotoxicity against D-GaIN. The activity of BNF-LCME is well supported in in vitro and in vivo antioxidant studies and may be attributed to flavonoidal, phenolic compounds present in the fraction. Hence, BNF-LCME was subjected to the development of qualitative analytical profile using HPLC finger printing analysis. Conclusions All the fractions of LCME exhibited significant hepatoprotective activity and BNF-LCME (50 mg/kg) was identified as the most effective fraction.

Objective: phytochemical screening and antioxidant activity analyses of selected indoor plants and to evaluate commercial applications.Methods: Qualitative and quantitative phytochemical analyses of alcoholic and aquatic crude extracts of leaves of selected non-flowering indoor plants were assessed using standard protocols and later compared with FTIR analyses. Antioxidant and antibacterial activities of the extracts were studiedResults: Phytochemical analysis of polar solvent extractions of the four selected plants Pedilanthus tithymaloides, Cordyline terminalis, Tradescantia zebrine and Rhoeo discolou. Indicated the presence of tannins in all four varieties terpenoids in 3, flavonols, phytosterols and phenols in two plants, followed by alkaloids. The phytochemical analyses were supported by FTIR reports. Quantitative studies indicated variations in flavonol, tannin and phenols concentrations among the four species. High concentrations of Total flavonols (P. tithymaloides) and Tannins (C. terminalis) were observed. C. terminalis extract showed comparatively highest reducing power followed by R. discolour and P. tithymaloides extracts. Antibiotic Sensitivity Testing indicated P. tithymaloides showed a maximum zone of inhibition compared to R. discolor. C. terminalis plant leaf extract showed a faint zone of inhibition against E. fecalis while others couldnot. Intense colors of C. terminalis and T. zebrine plants could be used as a natural dye as well as pH indicator. Conclusion: The rich concentrations of the tannins from non-flowering indoor plants could be the future option of dyes and dyeing industry as natural colorants as well as pH indicators. These plants were rich sources of phytochemicals (phenols, flavonols, tannins, and phytosterols), with antioxidant and antibacterial activity.

The antioxidant activities of crude extract ofPhaulopsis fascisepalaleaf were evaluated and compared with α-tocopherol and BHT as synthetic antioxidants and ascorbic acid as natural-based antioxidant.In vitro, we studied its antioxidative activities, radical-scavenging effects, Fe2+-chelating ability and reducing power. The total phenolic content was determined and expressed in gallic acid equivalent. The extract showed variable activities in all of thesein vitrotests. The antioxidant effect ofP. fascisepalawas strongly dose dependent, increased with increasing leaf extract dose and then leveled off with further increase in extract dose. Compared to other antioxidants used in the study, α-Tocopherol, ascorbic acid and BHT,P. fascisepalaleaf extract showed less scavenging effect on α,α,-diphenyl-β-picrylhydrazyl (DPPH) radical and less reducing power on Fe3+/ferricyanide complex but better Fe2+-chelating ability. These results revealed thein vitroantioxidant activity ofP.fascisepala.Further investigations are necessary to verify these activitiesin vivo.

Objective: In this study, the leaf powder of Aegle marmelos (L.) Correa. was subjected to preliminary phytochemical and in vitro antioxidant analysis. Methods: The freshly prepared plant leaf extract was subjected to preliminary phytochemical screening, which revealed the presence of alkaloids, tannins, saponins, flavonoids, glycosides, phenolic compounds, terpenoids, and steroids. Reducing power, superoxide (SO) anion radical, nitric oxide (NO) radical, and hydroxyl radical scavenging assays were carried out to evaluate the antioxidant potential of the methanolic leaf extract of this plant. The amounts of total phenolic and flavonoid compounds were also determined. Results: This study has revealed that the A. marmelos (L.) Correa. leaf extract showed considerably high amounts of most of the phytochemicals, total antioxidant capacity, total phenolic, and flavonoid content. The study also indicated that the A. marmelos (L.) Correa. showed comparatively good scavenging activity, i.e., inhibition of hydroxyl radical, NO and SO anion scavenging and reducing power activities when compared with the respective standards. Conclusion: The leaf powder of A. marmelos (L.) Correa. can be used as easily accessible source of natural antioxidant and as a possible food supplement or in pharmaceutical industry.

Objective: In the present study is investigated of phytochemicals and antioxidant activities of the leaf extracts from Meliosma simplicifolia (L.). Methods: The seaweed sample was subjected to extraction with solvents of different polarities (hot water, methanol, acetone, chloroform, and petroleum ether) and screened for phytochemicals according to standard methods. The ability of the plant extract to act as hydrogen/electrons donor or scavenger of radicals was determined by in vitro antioxidant assays using 2,2-diphenyl-2-picryl-hydrazyl free radical (DPPH) scavenging, reducing power assay, and superoxide radical (O2•) scavenging activity. The ICPMS and GC-MS analysis of the methanol leaf extract of M. simplicifolia was revealed the presence of antibacterial. Results: Phytochemical analysis revealed the presence of Preliminary metabolites Quantitative studies of estimated phenol, flavonoid and tannin, as for the methanol extract of stem showed the highest content of phenolic compounds (40.71±0.94 GAE mg/100). Antioxidant activities were concluded the estimation M. simplicifolia leaf for as followed the studies. In leaf the highest DPPH scavenging activity (132.3 μg/ml), ICP-MS analysis of the leaf extract showed the presence minerals such as Mg, Fe, Cu, Na, and Zn in excess. The leaf extract of the plant was also tested for its antibacterial activity and results confirmed that it has potential activity. Conclusion: The preliminary studies in the methanol extract of the leaf of M. simplicifolia are suggestive of the antibacterial potentials of the methanol extract of leaves of M. simplicifolia.

Objective: To evaluate the preliminary phytochemical content and antioxidant potential of the hydroalcoholic leaf extracts of Hemionitis arifolia. Methods: Total phenolic, flavonoid and alkaloid contents were evaluated using spectrophotometric methods. The free radical scavenging activity of the leaf hydroalcoholic extract were evaluated against DPPH+, ABTS+, Reducing power assay and nitric oxide assay were determined. Results: The hydroalcoholic concentrate of H. arifolia uncovered the most elevated polyphenol content when contrasted and the other phytoconstituents. Absolute phenol content of the hydroalcoholic separate was observed to be 31.78%, flavonoid content is 1.02% and Alkaloid content is 30.40% individually. The Solvent concentrates showed huge cell reinforcement movement, with hydroalcoholic extract. ABTS Assay, DPPH assay, Reducing power assay and Nitric oxide assay where the Inhibition concentration were 667.75µg/ml, 734.25 µg/ml, 791.58 µg/ml and 899.67 µg/ml. Conclusion: This study suggests that hydroalcoholic leaf extracts of H. arifolia could be a potential source of natural antioxidant and justifies its use in ethno-medicine.

Animal African trypanosomiasis (AAT) also known as Nagana is a resurgent disease in Africa. Medicinal plants are being used in less developed countries for the treatment of various diseases including trypanosomiasis, due to the high cost of currently available drugs. Most of these plants have been useful sources of treatment of various diseases based on information obtained from folk medicine but have not been scientifically certified. Here, we investigated the in vitro and in vivo anti-trypanosomal potentials of the methanol extract of Aformorsia laxiflora and Khaya senegalensis against T. b. brucei. Phytochemical screening as well as LD50 of the plant extracts was carried out following standard procedures. Parasitemia was monitored daily while Packed Cell Volume was determined at three time points (days 1, 4 and 7) during the course of the infection. The phytochemical analysis showed the presence of saponins, alkaloids, flavonoids, antraquinones, resins and tanins. However, steriods/terpenoids were absent in K. senegalensis but present in A. laxiflora. The toxicity of methanol extract of both A. laxiflora and K. senegalensis was above 5000mg/kg body weight. Methanol extracts of A. laxiflora (leaves) and K. senegalensis (stem bark) showed promising trypanocidal potential in vitro against T. b. brucei at concentrations of 10, 15, 25mg/ml and 40 and 20mg/ml respectively. At these concentrations, both extracts immobilized the parasites within 55mins post-incubation. In general, A. laxiflora leaf extract demonstrated prophylactic activity against T. b. brucei in vivo at a dose of 500mg/Kg body weight particularly in group C animals where a delayed pre-patent period (6 days post-infection), extended survival (14 days post-infection) and significant (P<0.05) reduction in the parasite burden confirmed by an absence of anemia (PCV 47.00±0.8 %) was observed when compared to the infected untreated control group. K. senegalensis extract on the other hand did not show anti-trypanosomal activity in the treated groups (1, 2, and 3). Based on these observations, it was therefore deduced that the methanol extract of leaves of A. laxiflora possessed the ability to ameliorate the burden of the disease and could be a plausible candidate for drug development against the disease.Keywords: Trypanosoma brucei brucei, Afromosia laxiflora, Khaya senegalensis, anti-trypanosomal, in vitro, in vivo
