The objective of this research was to analyze the chemical composition, antimicrobial and cytotoxic activity of Heracleum pyrenaicum subsp. pollinianum (Bertol.) F. Pedrotti & Pignatti (HPP) and H. orphanidis Boiss. (HO) essential oils. The composition of the oils was analyzed by GC and GC-MS. β-Pinene (35.1%) was the most abundant compound in HPP root oil, while ( Z)-falcarinol (80.0%) dominated in HO root oil. ( E)-Nerolidol (28.5%) was the main constituent in HPP leaf oil. HPP fruit oil, as well as HO leaf and fruit oils mainly contained aliphatic esters, mostly octyl acetate (50.5–84.5%). Antimicrobial screening was performed by microdilution method against eight bacterial and eight fungal strains. The strongest antibacterial activity was shown by both root oils (MICs 0.02–0.60 mg/mL and MBCs 0.04–2.50 mg/mL for HPP, and MICs 0.02–1.25 mg/mL and MBCs 0.04–2.50 mg/mL for HO), while the best antifungal potential was exhibited by HPP fruit oil (MICs 0.30–0.60 mg/mL and MFCs 0.60–1.25 mg/mL) and HO leaf oil (MICs 0.15–0.63 mg/mL and MFCs 0.30–1.25 mg/mL). The tested root and fruit oils exhibited strong cytotoxic effect, which was determined by MTT test against HeLa (IC50 7.53–21.07 μg/mL) and LS174 (IC50 24.16–58.86 μg/mL) cell lines.

AbstractThe chemical composition of the essential oils from the leaves and fruit ofEucalyptus camaldulensisgrown in Mersin, Turkey was analyzed using gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS) techniques. The biological activities (antibacterial and antifungal) were examined using the agar well diffusion method. The main leaf oil constituents werep-cymene (42.1%), eucalyptol (1,8-cineole) (14.1%), α-pinene (12.7%) and α-terpinol (10.7%). The main constituents of the fruit oil were eucalyptol (1,8-cineole) (34.5%),p-cymene (30.0%), α-terpinol (15.1%) and α-pinene (9.0%). Our results showed that both types of oils are rich in terms of monoterpene hydrocarbons and oxygenated monoterpenes. The leaf and fruit essential oils ofE. camaldulensissignificantly inhibited the growth of Gram-positive (Staphylococcus aureusandBacillus subtilis) and Gram-negative (Escherichia coliandStreptococcussp.) bacteria (p<0.05). The oils also showed fungicidal activity againstCandida tropicalisandC. globrata. Leaf essential oils showed more activity than fruit essential oils, probably due to the higher p-cymene concentration in leaves.

In this study, anti-mildew activities of the essential oils of the leaves, wood, twigs, flowers, and their constituents from Michelia compressa var. formosana were evaluated in vitro against 7 mildew fungi. The main compounds responsible for the anti-mildew activities were isolated and identified. The essential oils from the fresh leaves, wood, twigs, and flowers of M. compressa var. formosana were isolated using hydrodistillation in a Clevenger-type apparatus, and characterized by GC-FID and GC-MS. The essential oil yield from the flowers was the highest, and the oils obtained from the 4 parts of the tree were composed mainly of sesquiterpenoids. The leaf oil consisted primarily of α-cadinol (18.9%) and germacrene D (18.5%); the twig oil's main constituent was α-cadinol (19.1%); the wood oil's major components were α-cadinol (25.7%) and β-eudesmol (20.2%); and the flower oil contained α-cadinol (11.4%). Comparing the anti-mildew activities of the oils suggested that wood oil was the most effective. Further fractionation of the wood oil produced α-cadinol, β-eudesmol, τ-cadinol, and elemol. The 4 compounds exhibit very strong anti-mildew activities, and these in descending order are α-cadinol, elemol, β-eudesmol, and τ-cadinol. For the anti-mildew activities of the wood oil, the active source compounds were determined to be α-cadinol, elemol, β-eudesmol, and τ-cadinol.

Essential oils from leaves and fruits of Protiumheptaphyllum collected in Tamandaré beach Pernambuco/Brazil were analysed by GC/MS and tested for toxicity and repellent effect against the two spotted spider mite (Tetranychus urticae). The major constituent identified in the fruits was alpha-terpinene (47.6 %) whereas oil from leaf contained mainly sesquiterpenes such as 9-epi-caryophyllene (21.4 %), trans-isolongifolanone (10.7 %) and 14-hydroxi-9-epi-caryophyllene (16.7 %). The fruit oil was found to be more effective against the mite when compared to the leaf oil. Both showed mortality properties and oviposition deterrence in higher concentration (10 µl.l-1 air), but only the essential oil from fruits induced repellence on T. urticae.

The essential oils from the leaves, pseudostems, rhizomes and fruits of Alpinia rafflesiana were isolated by hydrodistillation. The oils were analysed by capillary GC and GC-MS. The most abundant components in the leaf oil were trans-caryophyllene (32.61%), caryophyllene oxide (8.67%), (2 E,6 Z)-farnesol (4.91%) and α-terpineol (4.25%), while 1,8-cineole (32.25%), myrcene (13.63%), α-terpineol (9.90%) and trans-caryophyllene (9.80%) were the main constituents in the pseudostem oil. The rhizome constituted of tetracosane (42.61%), τ-cadinol (7.46%), α-terpineol (6.71%) were the major components, whereas tetracosane (13.39%), (2 E,6 E)-farnesol (7.31%), α-terpineol (8.51%) and caryophyllene oxide (8.05%) were the main components in the fruit oil. Antimicrobial assay revealed that all the essential oils showed moderate to weak inhibition against the tested microorganisms. The leaf oil was the most active and inhibited both S. aureus and E. coli with MIC values of 7.81 μg/mL and 15.6 μg/mL, respectively.

The major aromatic components of the essential leaf oil of the New Zealand lemonwood tree Pittosporum eugenioides are octyl acetate (33%), terpinen-4-ol (13%), decanol (6%) and (Z)-hex-3-enol (5%). These products are responsible for the characteristic Citrus-like aroma which is detected when the leaves are crushed, a phenomenon which provided the species with its common name. The major component of the oil, octyl acetate is also an abundant component of the essential oils of Heracleum and Boswellia species.

The chemical composition of a leaf oil sample from Ivoirian Xylopia staudtii Engler & Diels (Annonaceae) has been investigated by a combination of chromatographic [GC(RI)] and spectroscopic (GC-MS, 13C NMR) techniques. Thirty-five components that accounted for 91.8% of the whole composition have been identified. The oil composition was dominated by the furanoguaiadienes furanoguaia-1,4-diene (39.0%) and furanoguaia-1,3-diene (7.5%), and by germacrene D (17.5%). The composition of twelve other leaf oil samples demonstrated qualitative homogeneity, but quantitative variability. Indeed, the contents of the major components varied substantially: furanoguaia-1,4-diene (24.7–51.7%) and germacrene D (5.9–24.8%). The composition of X. staudtii leaf oil is close to that of X. rubescens leaf oil but varied drastically from those of the essential oils isolated from other Xylopia species. 13C NMR spectroscopy appeared as a powerful and complementary tool for analysis of sesquiterpene-rich essential oils.

The compositions of hydrodistillated essential oils of Laser trilobum (L.) Borkh. rhizomes and fruits from Serbia, were investigated using GC and GC/MS. In the dark-blue rhizome oil forty-six compounds (93.1% of the total oil) were identified, with α-pinene (31.5%), γ-terpinene (9.0%), p-cymene (7.9%), β-pinene (6.1%) and 1,4-dimethylazulene (6.0%) as the major components. In the colorless fruits oil, twenty components (96.8% of the total oil) were identified, and the main constituents were limonene (51.6%) and perillaldehyde (26.8%). The antimicrobial activity of the oils was tested using the broth microdilution method against nine bacterial and two fungal strains. The oils revealed significant antimicrobial effect, mainly better than that of thymol, used as a reference compound. The strongest activity was recorded for the rhizome oil against Escherichia coli, Klebsiella pneumoniae and Candida albicans (MICs=25 μg/mL), and the fruit oil against C. albicans ATCC 10259 (MIC=12.5 μg/mL).

The chemical composition of the essential oils obtained by hydrodistillation from air dried leaves, bark and roots of Glossocalyx staudtii Engl. grown in Gabon was analyzed by GC and GC-MS. The essential oil content was 0.84% (w/w), 0.28% (w/w), and 0.74% (w/w), respectively. The leaf oil was characterized by a high content of monoterpene hydrocarbons (64.8%), with β-pinene (30.6%) and α-pinene (22.6%) as the major constituents. The oil obtained from the bark contained 30.5% of oxygenated monoterpenes with cryptone as the main constituent (11.5%) and 28.9% of aliphatic compounds, with 9.3% of 2-tridecanone; in the roots oil, the main component was 2-tridecanone (55.2%). The antimicrobial activity of the oils was studied by means of the agar disc diffusion and broth microdilution methods. The Gram positive bacteria were the most sensitive to the essential oils. A significant anticandidal effect of the bark essential oil was observed.

Eugenia dysenterica DC is a native species from the Cerrado biome and can be found in several states of Brazil. This study evaluated the chemical composition, antimicrobial and antioxidant activities from the essential oils of a population collected in São Paulo state. Essential oils were obtained by conventional means, and their compositions were analyzed by GC-MS. Screening assays for antimicrobial activity were carried out by the microdilution method and the antioxidant potential was assessed by the DPPH scavenging method. The GC-MS analysis indicated that 52.63% of the essential oil is composed by oxygenated sesquiterpenes and the major compound is (-)-elema-1,3,11(13)-trien-12-ol (24.86%). The antimicrobial assay indicated MIC 42.1 µg/mL for S. aureus and MIC > 10000 µg/mL for the other tested microorganisms, Gram negative bacteria and fungi. The oil showed an IC50 of 5.4±0.632 mg/mL for in the DPPH assay. The essential oil had a different chemical composition from previous studies. The essential oil did not present a potent antioxidant activity. However, it can be considered a promising antimicrobial agent against S. aureus.

Nigella sativa L. (NS) and its volatile compounds are well known for their broad spectrum of effects. This study aimed to investigate the variability of the chemical composition and the in vitro antibacterial activity of five essential oils (Eos) originated from Morocco, Saudi Arabia, Syria, India, and France. These five samples were grown under different edaphic and climatic conditions. The agar diffusion method and microdilution method in 96-well plates were used to test the sensitivity of multidrug-resistant strains clinically isolated from patients (methicillin-resistant Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and Acinetobacter baumannii), for the determination of the minimum inhibitory concentration and bactericidal concentration. Among all the investigated Eos, the monoterpenes were highly present in the chemical composition. Moroccan, Saudi Arabian, and Syrian seeds were characterized by the presence α-phellandrene (20.03–30.54%), β-cymene (12.31–23.82 %), and 4−caranol (9.77–14.27%). The Indian seeds were rich with 4-caranol (18.81%), β-cymene (14.22%), α-phellandrene (10.58%), and β-chamigrene (9.54%), while France NS was rich with estragole (20.22%) and D-limonene (14.63%). The minimum inhibitory (MIC) and bactericidal concentration (MBC) obtained for the four Eos (with the exception of France because of the low yield) tested were ranging from 3 to 40 μl/ml. Gram-positive (+) bacteria were slightly sensitive to the Eos tested than the Gram-negative (−) bacteria. The results of this study showed that the Eos of NS seeds show interesting antibacterial activity which could be associated to the existence of different bioactive compounds. Indeed, these compounds can be used for preventive or curative purposes in the face of the noncontrolled emergence of resistance to antibiotics.
