Nigella sativa L. (NS) and its volatile compounds are well known for their broad spectrum of effects. This study aimed to investigate the variability of the chemical composition and the in vitro antibacterial activity of five essential oils (Eos) originated from Morocco, Saudi Arabia, Syria, India, and France. These five samples were grown under different edaphic and climatic conditions. The agar diffusion method and microdilution method in 96-well plates were used to test the sensitivity of multidrug-resistant strains clinically isolated from patients (methicillin-resistant Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and Acinetobacter baumannii), for the determination of the minimum inhibitory concentration and bactericidal concentration. Among all the investigated Eos, the monoterpenes were highly present in the chemical composition. Moroccan, Saudi Arabian, and Syrian seeds were characterized by the presence α-phellandrene (20.03–30.54%), β-cymene (12.31–23.82 %), and 4−caranol (9.77–14.27%). The Indian seeds were rich with 4-caranol (18.81%), β-cymene (14.22%), α-phellandrene (10.58%), and β-chamigrene (9.54%), while France NS was rich with estragole (20.22%) and D-limonene (14.63%). The minimum inhibitory (MIC) and bactericidal concentration (MBC) obtained for the four Eos (with the exception of France because of the low yield) tested were ranging from 3 to 40 μl/ml. Gram-positive (+) bacteria were slightly sensitive to the Eos tested than the Gram-negative (−) bacteria. The results of this study showed that the Eos of NS seeds show interesting antibacterial activity which could be associated to the existence of different bioactive compounds. Indeed, these compounds can be used for preventive or curative purposes in the face of the noncontrolled emergence of resistance to antibiotics.

Antibiotics are becoming ineffective against resistant bacteria. The use of essential oils (EOs) may constitute an alternative solution to fight against multidrug-resistant bacteria. This study aims to determine the chemical composition of EOs from five populations of the endemic Algerian Origanum glandulosum Desf. and to investigate their potential antibacterial activity against multidrug-resistant uropathogenic E. coli strains. The EOs were obtained by hydrodistillation and their composition was investigated by gas chromatography/mass spectrometry (GC/MS). The antibacterial activity was evaluated by the disc diffusion method against eight E. coli strains (six uropathogenic resistant and two referenced susceptible strains). Minimum inhibitory and bactericidal concentrations (MIC/MBC) were obtained by the broth microdilution method. The main EO components were thymol (15.2–56.4%), carvacrol (2.8–59.6%), γ-terpinene (9.9–21.8%) and p-cymene (8.5–13.9%). The antibacterial tests showed that all the EOs were active against all the strains, including the multidrug-resistant strains. The EO from the Bordj location, which contained the highest amount of carvacrol (59.6%), showed the highest antibacterial activity (inhibition diameters from 12 to 24.5 mm at a dilution of 1/10). To our knowledge, this is the first description of the activity of O. glandulosum EOs against resistant uropathogenic strains. Our study suggests that O. glandulosum EO could be used in some clinical situations to treat or prevent infections (e.g., urinary tract infections) with multidrug-resistant strains.

Objective: The aim of the current study is to determine the chemical composition and evaluate antibacterial activity of Vitex agnus-castus L. (VAC) essential oils against some bacteria causing nosocomial infections in the neonatal and intensive care rooms at the university hospital center of Fez Morocco. Methods: The phytochemical screening of essential oils was determined using gas chromatography (GC) and GC-mass spectrometry analysis. The antibacterial test was evaluated against Gram-positive bacteria Staphylococcus aureus and Gram-negative bacteria species (Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, and Proteus mirabilis) using disc diffusion method. Results: Twenty-nine components were identified in the fruits’ oil representing 93.1% of total oil. The major components in the fruits oil are 1,8-cineole (11.6%), α-thujene (9.3%), phyllocladene (8.2%), α-pinene (7.9%), caryophyllene (5.9%), and cubenol (5%). Furthermore, 28 components were identified in the leaf essential oil. The main component was caryophyllene (9.5%), followed by 1,8-cineole (8.7%), manoyl oxide (7.3%), eugenyl acetate (7.1%), phyllocladene (6.8%), and α-pinene (5.2%). Antibacterial activity of both oils showed a strong activity against nosocomial bacteria tested. Conclusion: Essential oils of Moroccan VAC could be exploited as natural drugs for bacteria, especially those who have acquired resistance to conventional antibiotics.

Background. Emerging of multidrug-resistant bacteria can compromise the effectiveness of antibiotics used to treat skin infections. Those bacteria imposed public health problems and questioning medical care in the 21st century. In this circumstance, essential oils of medicinal plants origin are supreme sources of structural and functionally divergent compounds, which inhibited the growth of common wound colonizing MRSA and ESBL producing P. aeruginosa. The aim of this study was to evaluate the combined antibacterial activity of essential oils extracted from Rumex abyssinicus, Cucumis pustulatus, and Discopodium penninervium against multidrug-resistant (MDR) isolates of skin ulcers. Methods. Essential oils (EOs) were extracted from aerial parts of R. abyssinicus, C. pustulatus, and D. penninervium with steam distillation. A mixture of each oil (1 : 1) was adsorbed to a disc and placed on Mueller Hinton Agar. Then, minimum zone of inhibition and bactericidal concentration of EOs was measured after incubeted for 18–24 hours at 37 °C. Their combined antibacterial effect was determined by the fractional inhibitory concentration index. Results. The antibacterial activity of mixed oil varied in their doses and bacteria species, of which a mixture of essential oil of R. abyssinicus and D. penninervium had inhibition zone (32 mm); its MIC and MBC values range from 1-2 μl/ml against MRSA. It had an inhibition zone (36 mm), MIC value 4 μl/ml, and MBC (8 μl/ml) against ESBL producing P. aeruginosa, whereas combined effects of R. abyssinicus and C. pustulatus had MIC values ranging from 2–8 μl/ml for E. coli and K. pneumoniae and 2 μl/ml for MRSA. There was a strong synergistic effect between R. abyssinicus and D. penninervium and promising antibacterial effect more specifically on MRSA and P. aeruginosa. Conclusion. This in vitro study of the combined effect of EOs has significant antibacterial activity on wound colonizing bacteria and reduces delaying wound healing as that of modern drugs tested in parallel. Hence, further structural elucidation of active compounds helps us to properly design or synthesis of topical antibiotics for wound care.

The antimicrobial activity of the n-hexane, chloroform, ethyl acetate and ethanol extracts of the aerial parts of C. drabifolia S.M. subsp. cappadocica (DC.) Wagenitz (Asteraceae) was evaluated against microorganisms including multi-antibiotic resistant bacteria using the paper disc diffusion method. The chemical composition of the chloroform extract of this plant was determined by gas chromatography and gas chromatography-mass spectrometry. The chloroform extract exhibited significant antibacterial activity against all the bacteria tested, except Stenotrophomonas maltophilia MU63. The major compounds of the chloroform extract were spathulenol (14.1%), caryophyllene oxide (12.5%), octadecanol (10.2%), ethyl palmitate (7.7%), [Z,Z]-10,12-hexadecadienal (6.0%), 3-hydroxy p-anisaldehyde (5.9%) and pentacosane (5.8%).

A rapid emergence of resistant bacteria is occurring worldwide, endangering the efficacy of antibiotics and reducing the therapeutic arsenal available for treatment of infectious diseases. In the present study, we developed a new class of compounds with antibacterial activity obtained by a simple, two step synthesis and screened the products for in vitro antibacterial activity against ATCC® strains using the broth microdilution method. The compounds exhibited minimum inhibitory concentrations (MIC) of 1–32 μg/mL against Gram-positive ATCC® strains. The structure–activity relationship indicated that the thiophenol ring is essential for antibacterial activity and the substituents on the thiophenol ring module, for antibacterial activity. The most promising compounds detected by screening were tested against methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant Enterococcus faecium (VREF) clinical isolates. We found remarkable activity against VREF for compounds 7 and 16, were the MIC50/90 were 2/4 µg/mL and 4/4 µg/mL, respectively, while for vancomycin the MIC50/90 was 256/512 µg/mL. Neither compound affected cell viability in any of the mammalian cell lines at any of the concentrations tested. These in vitro data show that compounds 7 and 16 have an interesting potential to be developed as new antibacterial drugs against infections caused by VREF.

<p>The purpose of this study was to assess <em>in vitro</em> the activity of four essential oils (<em>Cymbopogon citratus, Eucalyptus citriodora, Lippia multiflora, Melaleuca quinquenervia</em>) and the biobactericide Neco® on Gram-positive bacteria.</p><p><strong>T</strong>he aromatogram and antibiogram were assessed by the agar well diffusion method and the Muller Hinton disk-agar diffusion method, respectively. Also, the minimum inhibitory concentration and the minimum bactericidal concentration were determined by the microdilution method in liquid medium.</p><p>The aromatogram showed that the biobactericide Neco® induced the largest inhibition diameters (34.53 ± 11.82 - 43.92 ± 5.38 mm) of all strains combined, followed by the essential oils of <em>Eucalyptus Citriodora</em> (30.01 ± 3.02 - 41.89 ± 1.77 mm) and <em>Lippia multiflora</em> (20.72 ± 4.72 - 37.61 ± 2.80 mm). However, the essential oils of <em>Melaleuca quinquenervia</em> (19.99 ± 3.93 - 26.20 ± 13.27 mm) and <em>Cymbopogon citratus</em> (13.52 ± 3.59 - 29.08 ± 2.35 mm) had the smallest inhibition diameters. Moreover, the comparison of the activities of the aromatogram and antibiogram revealed generally that activities were higher with essential oils than with antibiotics.</p><p>At the end of this study, the essential oils of <em>Cymbopogon citratus</em>, <em>Eucalyptus citriodora, Lippia multiflora, Melaleuca quinquenervia</em> and the biobactericide Neco® had an antibacterial activity on Gram+ bacteria.</p><p><strong><br /></strong></p>

Nigella sativa L. is a plant with high medicinal profile in the treatment of diseases. This study aimed to assess the chemical composition and in vitro antibacterial and cytotoxic effect of Nigella sativa L. seed extract. Seeds were purchased from supermarket and blended into powder using electric blender. The powder was macerated with 96% methanol. The extracts were then qualitatively screened to test the presence of secondary metabolites and agar well diffusion method was employed to study the antibacterial activity of extracts. In this evaluation, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay was used to examine the cytotoxic effect of extract on peripheral blood mononuclear cells (PBMCs). MTT assay was done on different extract concentrations for 3 days and the results were recorded daily. The findings revealed the presence of tannins, terpenoids, steroids, saponins, alkaloids, phenolic compounds and flavonoids. The antibacterial activity was observed with the inhibition zones ranging from 11.3 ±1.1 to 16.3 ±1.5mm while the minimum inhibitory concentration (MIC) values of extract ranged between 0.5 and 1.25 μg/mL. The black seed extracts contain bioactive substances with germicidal activity and in vitro administration of black seed extract up to the dose of 100 μg/mL resulted no cytotoxic effect on PBMCs.

AbstractStaphylococcus aureus and coagulase-negative staphylococci are among the major causes of mastitis in sheep. The main goal of this research was to determine the in vitro antibacterial activity of several essential oils (EOs, n 30), then five of them were chosen and tested alone and in blends against staphylococci isolates. Five bacteria were isolated from episodes of ovine mastitis (two S. aureus and three S. xylosus). Biochemical and molecular methods were employed to identify the isolates and disk diffusion method was performed to determine their antimicrobial-resistance profile. The relative percentage of the main constituents in the tested essential oils and their blends was detected by GC-EIMS analysis. Antibacterial and bactericidal effectiveness of essential oils and blends were evaluated through minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC). All of them showed sensitivity to the used antimicrobials. The EOs with the highest antibacterial activity were those belonging to the Lamiaceae family characterized by high concentrations of thymol, carvacrol and its precursor p-cymene, together with cinnamon EO, rich in cinnamaldehyde. In terms of both MIC and MBC values, the blend composed by Thymus capitatus EO 40%, Cinnamomum zeylanicum EO 20%, Thymus serpyllum EO 20% and Satureja montana EO 20% was found to be the most effective against all the isolates. Some essential oils appear to represent, at least in vitro, a valid tool against ovine mastitis pathogens. Some blends showed a remarkable effectiveness than the single oils, highlighting a synergistic effect in relation to the phytocomplex.

<p><strong>Objective: </strong>To evaluate the chemical composition, antibacterial and antioxidant properties of stem bark essential oil of <em>Litsea glutinosa </em>C. B. Rob.</p><p><strong>Methods: </strong>The essential oil isolated from stem bark of <em>L. glutinosa </em>and their chemical composition was analyzed by gas chromatography coupled with mass spectrometry detector. The <em>in vitro </em>antibacterial activity of the stem bark essential oil was investigated against eight human pathogenic bacterial clinical isolates using agar disc diffusion method and MIC value was determined by modified resazurin microtitre-plate assay. The antioxidant activity of essential oil was measured by 2, 2-diphenyl-1-picrylhydrazyl radical (DPPH), 2, 2-azinobis-3-ethylbenzothiazoline-6-sulphonate radical cation (ABTS) and β-carotene bleaching assay.</p><p><strong>Results: </strong>GC-MS analysis of stem bark essential oil resulted in the identification of 37 compounds, off which 9,12-octadecadienoic acid (62.57%), hexadecanoic acid (12.68%), stigmast-5-en-3-ol (6.87%) and vitamin E (2.51%) were the main constituents representing 84.63% of the oil. The determination of <em>in vitro</em> antibacterial activity of stem bark essential oil resulted in significant inhibition zone (15.00±0.57 mm) and MIC value (0.15±0.15×10<sup>-2</sup> mg/ml) against the pathogenic bacteria <em>Vibrio cholera</em> followed by <em>Pseudomonas aeruginosa</em> and <em>Salmonella typhi. </em>The results of DPPH radical scavenging (IC<sub>50</sub>:4.540±0.06 µg/ml), ABTS (IC<sub>50</sub>:256.02±0.06 µg/ml) and β-carotene bleaching assay (%I: 78.51±0.42 <strong>%</strong>) showed significant <em>in vitro</em> antioxidant property.</p><p><strong>Conclusion: </strong><em>L. glutinosa</em> stem bark essential oil showed potential antibacterial activity against the <em>Vibrio cholera</em>. The results of this investigation supported the ethnomedical claim of essential oil as a demulcent, antidiarrheal and antioxidant drug.</p>
