Marbling is one of primary factors influencing the palatability of beef. The AKIRIN2, EDG1 and TTN genes are considered as the three candidate genes responsible for high-marbling in beef cattle. This study aimed to investigate the genetic polymorphism of the SNPs of AKIRIN2:c.*188G>A, EDG1:c.-312A>G and TTN g.231054C>T in Bali cattle. A total of 125 DNA samples of Bali cattle were genotyped for AKIRIN2 and TTN gene and 82 DNA samples for EDG1 gene using PCR-RFLP method. In the present study, it was found that the A allele of AKIRIN2:c.*188G>A, the G allele of EDG1:c.-312A>G and the T allele of TTN g.231054C>T that considered as the favorable alleles were found to be very low in Bali cattle (0.004, 0.000, and 0.004, respectively). This results indicated that the polymorphism of AKIRIN2:c.*188G>A, EDG1:c.-312A>G and TTN g.231054C>T were very low and can be considered as monomorphic in Bali cattle. These findings suggested that Bali cattle may not genetically potential as a high-marbling cattle breed. However, the other specific genes for high-marbling still needs to be investigated in Bali cattle.

H-FABP, LEPR and MC5R genes were suggested as candidate genes for fat content in pig meat. The aim of this study was to detect genetic variation in the porcine H-FABP, LEPR and MC5R genes by PCR-RFLP method in a group of pigs. Genotyping of pigs was done by PCRRFLP methods. We identified three genotypes in the set of pigs, HH (0.504), Hh (0.412) and hh (0.084) for H-FABP (HinfI). Allele H showed higher frequency than allele h (0.710 vs. 0.290). Three genotypes were identified for the H-FABP (HaeIII) gene (DD - 0.194, Dd - 0.494, dd - 0.312). The allele D (0.441) showed slightly lower frequency than allele d (0.559). All three genotypes were identified for LEPR (HpaII) in the group of pigs (AA – 0.137, AB - 0.314, BB – 0.549). Higher frequency of LEPR gene was confirmed for allele B (0.706), as compared with allele A (0.294). We identified two genotypes for MC5R (BsaHI) in the group of pigs (AA - 0.348 and AG - 0.652), genotype GG was not found. As conforms with genotype structure, we recognize a higher frequency of allele A (0.674) as compared with allele G (0.326).

The ADIPOQ and EDG1 genes were responsible in intramuscular fat deposition and marbling scores. This study was aimed to identify polymorphism of indel g.81966364D>I in promoter region of ADIPOQ gene and SNP c.-312A>G in 5' UTR of EDG1 gene in Indonesian beef cattle. Blood samples were collected from 211 cattle, including Bali (44), Madura (20), Pesisir (18), Katingan (20), PO (22), Pasundan (20), SO (12), Brahman (20), Simmental (15) and Limousin (18). Polymorphism of ADIPOQ gene was analyzed using PCR and direct sequencingmethods, whereas EDG1 gene was analyzed using PCR-RFLP (MscI enzyme) and direct sequencingmethods. Results of genotyping indel g.81966364D>I was monomorphic (DD genotype). The SNP c.-312A>G was polymorphic (AA and AG genotype) in Madura, Pesisir, Pasundan, Brahman, and Limousine. The Frequencies of allele A and G were 0.95, 0.92, 0.98, 0.95, 0.94 and 0.05, 0.08, 0.02, 0.05, 0.06 respectively. The values of Ho and He were 0.05-0.17 and 0.05-0.15 respectively and in Hardy-Weinberg equilibrium (P>0.05). In Bali, Katingan, PO, SO and Simmental were monomorphic (GG genotype). In Bali cattle, two novel SNP candidates were found in position of c.-399C>T and c.-273C>G which were potential to be used as genetic markers of marbling score for Bali cattle. As result this study, it can be concluded that ADIPOQ gene was similar while EDG1 gene was different in Indonesian beef cattle. in addition, found two candidates potential SNP in Bali cattle.

The g.422C>T nucleotide variations in the 5’ untranslated region (5’UTR) of TG gene (called as TG5) has been reported to be associated with level in intramuscular fat (IMF) content or marbling in beef cattle. The objective of this study was to confirm genetic polymorphism of TG5 gene in Bali cattle populations from three different regions as the main resources of Bali cattle in Indonesia. A total of 200 head of Bali cattle have been performed genotyping on TG5 gene using polymerase chain reaction-restriction fragment lenght polymorphism (PCR-RFLP) method and sequence analysis. Results of the study confirmed that TG5 was monomorphic in Bali cattle wherever their origin regions. Moreover, nine candidate SNPs were detected within 5’UTR of TG gene in Bali cattle compared to Genbank reference sequences, although no SNP variations among Bali cattle sample studied. The new other genetic markers within an entire TG gene suggested to be explored and verified for their polymorphisms in Bali cattle. The nine candidate SNPs were also required further verification and validation in a larger sample to be regarded as new SNPs between Bali cattle and Genbank reference sequences.

The pituitary transcription factor (Pit-1) gene is one of the considered genes that responsible to growth in cattle. A specific Pit-1|HinfI gene polymorphism located within exon 6(g.1256G>A) has been shown to be associated with growth and milk traits in cattle. This study aimed to identify the Pit-1|Hinf1 gene polymorphism in Grati-Ongole Grade cattle (Grati-OG cattle). A total of 107 genomic DNA of Grati-OG cattle were used in this study. The detection of polymorphism was performed by PCR-RFLP method. Only AB and BB genotypes were succesfully identified with the frequency of 0.009 and 0.991, respectively. Whereas, the frequency of A and B alleles were 0.005 and 0.995, respectively. The observed genotype frequencies in this population do not deviated from HWE. The value of Ho, He and PIC were 0.009, 0.009 and 0.009, respectively.In conclusion, the Pit-1|HinfI gene polymorphism in Grati-OG cattle is monomorphic and hence it should not be used in further association studies.

A colorectalis rákok a második leggyakoribb halálokként szerepelnek a rosszindulatú betegségek között. A fej-nyak táji daganatok halálozása Magyarországon 265%-kal emelkedett az utóbbi 30 évben. Nem lehet eléggé hangsúlyozni e két daganatcsoport közegészségügyi jelentőségét. A colorectalis daganatok esetén a GSTM1, GSTT1 enzimek, valamint a p53 72-es kodonjának, fej-nyak táji tumorok esetén az XRCC1 Arg194Trp és Arg399Gln polimorfizmusainak hatását elemeztük. Intraoperatíve eltávolított daganatos és megfelelően illesztett daganatmentes mintákat válogattunk. A formalinban fixált mintákat deparaffinizáltuk és proteináz-K-emésztésnek vetettük alá. A genotipizálást PCR, illetve a fej-nyak táji tumorok esetén PCR-RFLP módszerrel végeztük. A vizsgált allélek gyakoriságában nem volt különbség a daganatos és a kontrollcsoport között. Túlélés tekintetében szignifikáns különbséget találtunk a GSTM1 és a p53 allélek között Dukes B stádiumú colorectalis daganatok esetén és az XRCC1 194 allélek között III-as stádiumú fej-nyak táji tumorokban. A fenti típusú genetikai különbségek szisztematikus vizsgálata a jövőben hozzájárulhat az egyéni rizikóbecslés és az individualizált kezelések megalapozásához.

The objective of the present study was to perform a genome-wide association study (GWAS) for growth curve parameters using nonlinear models that fit original weight–age records. In this study, data from 808 Chinese Simmental beef cattle that were weighed at 0, 6, 12, and 18 months of age were used to fit the growth curve. The Gompertz model showed the highest coefficient of determination (R2 = 0.954). The parameters’ mature body weight (A), time-scale parameter (b), and maturity rate (K) were treated as phenotypes for single-trait GWAS and multi-trait GWAS. In total, 9, 49, and 7 significant SNPs associated with A, b, and K were identified by single-trait GWAS; 22 significant single nucleotide polymorphisms (SNPs) were identified by multi-trait GWAS. Among them, we observed several candidate genes, including PLIN3, KCNS3, TMCO1, PRKAG3, ANGPTL2, IGF-1, SHISA9, and STK3, which were previously reported to associate with growth and development. Further research for these candidate genes may be useful for exploring the full genetic architecture underlying growth and development traits in livestock.

AbstractWater holding capacity (WHC) is an important sensory attribute that greatly influences meat quality. However, the molecular mechanism that regulates the beef WHC remains to be elucidated. In this study, the longissimus dorsi (LD) muscles of 49 Chinese Simmental beef cattle were measured for meat quality traits and subjected to RNA sequencing. WHC had significant correlation with 35 kg water loss (r = − 0.99, p < 0.01) and IMF content (r = 0.31, p < 0.05), but not with SF (r = − 0.20, p = 0.18) and pH (r = 0.11, p = 0.44). Eight individuals with the highest WHC (H-WHC) and the lowest WHC (L-WHC) were selected for transcriptome analysis. A total of 865 genes were identified as differentially expressed genes (DEGs) between two groups, of which 633 genes were up-regulated and 232 genes were down-regulated. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment revealed that DEGs were significantly enriched in 15 GO terms and 96 pathways. Additionally, based on protein–protein interaction (PPI) network, animal QTL database (QTLdb), and relevant literature, the study not only confirmed seven genes (HSPA12A, HSPA13, PPARγ, MYL2, MYPN, TPI, and ATP2A1) influenced WHC in accordance with previous studies, but also identified ATP2B4, ACTN1, ITGAV, TGFBR1, THBS1, and TEK as the most promising novel candidate genes affecting the WHC. These findings could offer important insight for exploring the molecular mechanism underlying the WHC trait and facilitate the improvement of beef quality.

The Wnt signaling pathway is mediated by a family of secreted glycoproteins through canonical and noncanonical mechanism. The signaling pathways are regulated by various modulators, which are classified into two classes on the basis of their interaction with either Wnt or its receptors. Secreted frizzled-related proteins (sFRPs) are the member of class that binds to Wnt protein and antagonizes Wnt signaling pathway. The other class consists of Dickkopf (DKK) proteins family that binds to Wnt receptor complex. The present review discusses the disease related association of various polymorphisms in Wnt signaling modulators. Furthermore, this review also highlights that some of the sFRPs and DKKs are unable to act as an antagonist for Wnt signaling pathway and thus their function needs to be explored more extensively.

Pottery is seen as creation of ornamentals, cooking and storing materials. Yet, while economic gains are often considered from producing these materials, the artistic and linguistic aspects have been ignored. This study discusses the factors influencing the culture of pottery, the processes of pottery making, and seeks to uncover the language used in processes of pottery making in Bari, Sibalom, Antique. A qualitative research employing ethnographic study with participant observation and face to face interviews using photo documentation, video recording and open-ended questions in gathering the data was employed. There were five manugdihon, or potters, purposively selected as key informants of the study. The study revealed that environmental factors influenced the culture of pottery making in the barangay. There were seven main processes in pottery making. These included gathering and preparing of materials, mixing the needed materials, cleaning the mixed clay, forming of desired shape, detaching, drying, and polishing and varnishing. Further findings indicate that, together the other processes, the language used in poterry making was archaic Kinaray-a, the language of the province. This language pattern suggests a specialized pottery making. Ultimately, the study suggest that the manugdihon should continue their artistic talents so that the language may be preserved. The educational institutions of the province may provide ways to include pottery making in the curriculum so that the art and language of pottery making will be preserved and promoted.
