Mathematical Modeling of Betanin Extraction from Red Beet (Beta vulgaris L.) by Solid–Liquid Method

Abstract Effective diffusion coefficient (D-value) is an important parameter for the extraction process. In this study, a cylindrical model to estimate the D-value of betanin extraction from red beet roots was developed. The influence of extraction temperature, pH, cylindrical thickness and type of acids on the D-value was also investigated. The optimal parameters of betanin extraction were 30°C, pH 4.0 adjusted with sulfurous acid or acetic acid. D-values under the above conditions were 18.95×10−11 m2/s and 17.87×10−11 m2/s, respectively. The result showed that D-value elevated with the increase of cylindrical thickness, which was consistent with earlier conclusion from spherical model. The modeling may be useful for the investigation into extraction process and practical application.

Turmeric has a bioactive compound namely curcuminoid. It has many pharmacology effects such as anticancer, antidiabetic, antioxidant, hypolipidemic, anti-inflammatory, antimicrobial, antifertility, anti-venom, anti-coagulant, anti-HIV hepatoprotective, nephroprotective, and anticoagulant properties. To increase the economic value of turmeric, it is necessary to develop a hydrothermal extraction process of turmeric’s active compound. The advantages of hydrothermal extraction were inexpensive, abundant availability, high purity, non-toxic, and easy to handle. This research aims to study the effect of operating conditions : temperature (130-150oC), time (10-40 minutes) and solid:liquid ratio (1:10 and 1:12) on the bioactive compounds of turmeric extracted from hydrothermal extraction process. Generally, high extraction yield was obtained at higher extraction temperature (140 and 150oC). Under these conditions, with a lower solid : liquid ratio (1:10), high concentration of curcumin is produced. Further, a higher solid : liquid ratio will likely produce the opposite result, except when it operates at low extraction temperature. The high temperature of the pressurized liquid water can reduce the viscosity and surface tension of water so it will increase the diffusion rates and absorption. The higher the solid:liquid ratio, the greater the different concentration between interior and exterior cell, which promote the high efficiency of diffusion process.

In order to study the the Chuanxiong polysaccharide ultrasonic extraction process, examine the ultrasonic time, ultrasonic power, solid-liquid ratio and extraction temperature on the Chuanxiong polysaccharide extract rate. The results show that the role of time at 30min Polysaccharide better, the extraction rate is 26.63%. when the ultrasonic power is 320W, Polysaccharide extraction rate is 26.15%.When the solid-liquid ratio is 1:20, polysaccharide extraction rate is 27.73%. When the extraction temperature is 60°C, the polysaccharide extraction rate is 27.73%.

Substances from Fructus Alpinia Oxyphylla (AOF) were extracted by using solvent extraction. In accordance with central composited design of response surface methodology system, the extracts were analyzed for the index which is the criteria formula of multiplying OD value by the yield. The optimum conditions obtained were extraction temperature of 60°C, liquid-solid ratio of 9mL/g, and solvent density of 80%. According to Ficks second law of diffusion, experimental parameters of K (rate constant), Ea (activation energy), Gs (effective diffusion coefficient) were gained, which represents a good agreement between the model simulation and the results of the actual process. The results can provide the valuable theory basis for the technical design and further research of extraction process. Antibacterial activities of the extraction were shown through MIC of which represents a significant effect.

Moringa leaves were used as raw materials, ethanol and ammonium sulfate were used to construct a aqueous two-phase system to extract the flavonoids from Moringa leaves. The solid-liquid ratio, ethanol content, ultrasonic extraction time, and ultrasonic extraction temperature were selected as single factors, and the extraction process of flavonoids was optimized through single-factor experiments and orthogonal experiments. The optimal process was: 1:110 of solid-liquid ratio, 45 % of ethanol concentration, 15 minutes of ultrasonic extraction time, 70 °C of ultrasonic extraction temperature. Under this condition, the extraction rate of Moringa leaf flavonoids reached 8.37 %.

Natural pigments have raised a growing interest due to their extensive range of colors, innocuous and beneficial health effects. In this paper, the extraction process and stability of strawberry red pigment were studied. An orthogonal test [L16(4)4] was applied to get the best extraction conditions. Results showed that the optimum conditions were that the extraction temperature was 65C, the solid-liquid ratio was 1:5 (g/mL), the ratio of 1.5mol/L hydrochloride to 97.5% ethanol was 3:17 (mL/mL), and the extraction time was 4.5 hours. The effects of heat treatment, oxidant, sugar and preservatives on the stability of strawberry red pigment were also discussed. The stability of strawberry red pigment was found to be affected by high temperature, hydrogen peroxide, and potassium sorbate. This study is expected to provide a basis for further research, development, and utilization of strawberry red pigment.

Taxus mairei is an important source for industrial extraction of taxol in China. However, the standard and steps of extraction are currently not uniform, which seriously affects the taxol yield. In the present study, the influence of four factors (methanol concentration, solid-liquid ratio, ultrasonic extraction temperature, and ultrasonic extraction time) on the taxol yield was successively explored in T. mairei. A response surface methodology (RSM) was used to optimize the extraction process based on the single-factor experiments above. The optimal conditions were as follows: methanol concentration was 90%, solid-liquid ratio was 1:15 (g/mL), ultrasonic extraction temperature was 40 °C and ultrasonic extraction time was 60 min. Moreover, the twigs and needles from T. mairei with different tree ages were treated by the optimum extraction process, which further revealed temporal and spatial distribution of taxol in the reproducible tissues. Interestingly, the taxol content was relatively higher in needles of T. ‘Jinxishan’ (a cultivar from T. mairei with yellow aril, FY), but was less in FY twigs. The accumulation of taxol in twigs and leaves of females (with red aril, FR) was significantly higher than that of males (M); however, the content showed a decreasing trend with the increasing tree ages. Therefore, it is suitable to increase the proportion of female trees especially the FY leaves as raw materials for the industrial production of taxol from T. mairei, and the tree ages should be better controlled at 3–7 years.

Banana peels which are the waste in abundance, are used to extract valuable pectin. The gelling ability of the pectin has gained attention in food and pharmaceutical industries. This research aims to select the best drying kinetic model for banana peels and also optimize the pectin extraction process using Box-Behnken response surface design (BBD). Determination of pectin gelling mechanism using degree of esterification (DE) is also focused in this research. In this study, oven drying with temperature 50°C was chosen as the best drying temperature due to highest extraction yield. Furthermore, Page-Two-term model was selected as the best model to describe the drying kinetics of banana peels due to highest R2 value (0.9991) and lowest RMSE value (0.001). The optimal extraction conditions given by BBD were 75°C extraction temperature, 23 min extraction time and 1:33.3 g/ml solid-liquid ratio. Likewise, the DE for both pectins extracted using unoptimised and optimised conditions were 71.92±1.38% and 76.1±2.07% respectively. Both of the pectins were classified as high-methoxyl pectins. The pectin with higher DE also indicated that the rate of gel formation is higher. The results showed that the pectin yield and gelling time has successfully improved after optimised the pectin extraction process.

Using Box-Behnken center composite design of three variables with three levels, the optimal extraction process for carotenoids from tamarillo fruits were determined by response surface methodology. The optimal extraction process was obtained as followed: solid-liquid ratio of 1:19.02(g/mL), extraction temperature of 44°C, and extraction time of 33.68 min. Under these conditions, the extraction of carotenoids content was 5.755[mg/100, similar to the theoretical prediction of 5.885[mg/100. This study confirms the efficiency of RSM as a simple and effective method and the great potential of tamarillo fruits as a health food and a resource of carotenoids.

Extraction of amygdalin by ethanol refluxing from waste loquat kernel was performed successfully. Amygdalin concentrations were determined by using ultraviolet spectrophotometry with ethanol as solvent at 210 nm. And the influences of various factors on extraction process were discussed. The result shows that the concentration of ethanol is the most influencing factor in this process, and then the solid-liquid volume ratio, the extraction temperature and the extraction time. Optimum conditions for this extraction process are: the solid-liquid volume ratio is 1/10, the concentration of ethanol is 65%, the extraction temperature is 60°C and the extraction time is 45min.

In these recent years, natural compound industry has been more and more attractive many interested. Although Persicaria pulchra (Bl.) Soják has been applied in Eastern medicine for long time, it has not researched deeply the chemical compounds and biological activities. In the experience, the presence of the polyphenol, flavonoid and alkaloid compounds with the value IC50 = 127,99 mg/mL proved that this plant owns potential anti-tyrosinase activity. To expand its using in the pharmaceutical and cosmetics industries, optimize the extraction conditions for tyrosinase inhibition from Persicaria pulchra (Bl.) Soják leaves using response surface methodology (RSM). The effect of the factors including ethanol concentration, temperature, solid/liquid ratio and extraction time on extraction of tyrosinase enzyme inhibitor from the Persicaria pulchra (Bl.) Soják leaves was investigated by experimental and Response Surface Methodology (RSM) methods. Results demonstrated that the mathematical modeling are compatible with experimental data and all four factors exhibit significant effect on tyrosinase inhibition activity of the extract (p<0,05). The optimal extraction conditions conjectured by the mathematical modeling as the content of solvent of 63% ethanol, the extraction temperature of 56.5 oC, the solid / liquid ratio 1: 8.18 g/mL, and the extraction time of 13.96 minutes. Under these conditions, the extract indicated that the best tyrosinase inhibition value of the extract for IC50 was 56,39 mg/mL, 2.5 times lower than the preliminary experiment value (127,99 mg/mL). Additionally, the experiment results of tyrosinase inhibition activity of optimal extraction showed that the results exported from mathematical modeling was equivalent with them.
