Cloning, Over-Expression, and Purification of Carbonic Anhydrase from an Extremophilic Bacterium: An Introduction to Advanced Molecular Biology

The photosynthetic bioreactor research program is a training platform appropriate for introducing advanced molecular biology techniques to undergraduate students and advanced high school biology students. For this advanced molecular biology training exercise, the enzyme carbonic anhydrase was cloned, over-expressed, purified, and functionally characterized. Carbonic anhydrases are industrially important enzymes with potential use in carbon sequestration and biofuel production. Alpha and beta carbonic anhydrases from Photobacterium profundum, a psychrophilic, halotolerant bacterium, were characterized in this study. Carbonic anhydrases that can withstand high salinity and are active at low temperatures can be transformed into oleaginous marine microalgae to enhance biofuel production. Our research program started with a three-day boot camp with lectures in relevant topics of molecular biology, microbiology, and research methods. After the boot camp, the lab phase of the project involved training students to perform polymerase chain reaction, DNA gel electrophoresis, DNA ligation, and bacterial transformation. In the final phase of the project, students were trained in recombinant protein over-expression and protein purification techniques. Here we report successful cloning and over-expression by high school students of two novel carbonic anhydrases from a psychrohalophile with application in biofuel production.

Arizona State University has been involved in identifying the future Facility Manager (FM) paradigm which results in a sustainable FM profession. The $16M, 20-year international research program has identified, created and tested out a solution to the almost impossible task of replacing the aging FM professionals. The research has identified the future FM as one who is leadership based and leads the entire supply chain from inside the organization. This paper will cover the three year results of finding the future FM by accessing the top 10% of ASU’s 85,000 students through Barrett’s Honors Program, 7th – 10th graders through the Barrett’s Summer Honors Program, and culminating this year in placing the education in one of the top private schools in the state of Hawaii for high school students, testing the approach on 116 Brazilian engineering undergraduate students and getting approval for testing a 14 week program in the Tempe High School. The model is proposed as a prototype for future FM professionals and how the FM professional can become sustainable.

No presente trabalho, visamos ampliar discussões acerca das diferentes faces da interação Universidade-Escola, com base na apresentação de relatos de experiências sobre o Projeto Primeiros Passos na Ciência e em reflexões teóricas sobre o contexto universitário, motivadas pela prática vivenciada. O projeto em questão foi desenvolvido por alunos de pós-graduação em Ciências Biológicas (Biologia Celular e Molecular) da UNESP, Campus de Rio Claro-SP em parceria com a Escola Estadual Prof. Marciano de Toledo Piza, de ensino médio, da cidade de Rio Claro, estado de São Paulo. Este projeto teve o objetivo de introduzir o aluno do Ensino Médio no cotidiano da pesquisa universitária na área de Biologia Celular e Molecular, com a expectativa de promover a aproximação destes alunos com o desenvolvimento do conhecimento científico e o ambiente universitário. Com a realização desse projeto, a Universidade teve a chance de adentrar no contexto dos alunos de Educação Básica, mais especificamente do Ensino Médio público. Esses novos conhecimentos podem criar a possibilidade de um futuro diferente, com mais conhecimentos científicos e instigando a reflexão a partir da rotina da Universidade e seu contexto. A experiência resultou na divulgação científica, no contato do aluno do Ensino Médio com o método científico e toda a técnica e objetividade que dele derivam, ampliando os laços comunidade-universidade. Palavras-chave: Educação; Extensão; Vivência; Experimento Científico   Project First Steps in Science: breaking social barriers and strengthening ties between the academic community and public high schools Abstract: In the present study, we intend to expand the discussion about different approaches of the University-School interaction, based on the presentation of an account of experiences with the First Steps in Science Project and theoretical reflections on the University context motivated by the lived experience. The project in question was developed by graduate students in Biological Sciences (Cell and Molecular Biology) at UNESP, Rio Claro/SP (Brazil), in partnership with Escola Estadual Prof. Marciano de Toledo Piza a public high school in the city of Rio Claro, State of São Paulo. This project had the objective of introducing high school students to the routine of university research in the area of Cell and Molecular Biology, with the expectation of promoting the approximation of these students with the development of scientific knowledge and the university environment. With the realization of this project, the University had the opportunity to better understand the primary education context of the students, specifically within public high schools. This new knowledge can create the possibility of a different future, with more scientific properties and instigating the reflection from the routine of the University and its context. The experience resulted in scientific dissemination, high school students' contact with the scientific method, and all the technique and objectivity derived from it, expanding community-university ties. As advances in biotechnology and molecular biology rapidly expand in research settings, it is vital that we continue to prepare high school students to enter and thrive in those modern laboratories. This multistep, inquiry-based lab describes highly adaptable methods to teach students not only current molecular techniques and technologies, but also about proteomics and microorganisms. Students participate in protein extraction, gel electrophoresis, mass spectrometry, and data analysis to identify proteins present in microorganisms.

ABSTRAk   Generasi milenial yang sebagian besar merupakan pelajar SMP dan SMA lebih banyak menghabiskan waktunya dengan gadget. Banyak pelajar masa kini sulit memahami pelajaran yang dipelajari di sekolah sehingga sampai di rumah mereka kembali sibuk dengan gadget-nya masing-masing dan melupakan topik pelajaran yang telah dipelajari di sekolah. Oleh karena itu, kebanyakan orangtua menyadari hal tersebut sehingga memberikan pelajaran tambahan bagi anak-anaknya dengan mendaftarkan anak-anaknya ke lembaga-lembaga bimbingan belajar (bimbel) terkenal ataupun mencari guru-guru les privat. Dengan semakin berkembangnya m-learning, siswa tidak perlu lagi harus datang ke lokasi bimbel karena Mereka bisa belajar kapanpun mereka mau setiap mereka membawa laptop atau smartphone dengan mengakses bimbingan belajar online (bimbel online). Ruangguru hadir sebagai salah satu alternatif bimbel online yang dapat dengan mudah diakses melalui smartphone, laptop ataupun tablet. ABSTRACT   Millennial generations, mostly junior and senior high school students are spending much more of their time with gadgets. Nowadays, many students find it hard to understand their school lessons then neglect them at home. Parents realize this problem then help their children by enrolling them in tutoring agencies or seeking private tutors. With the development of m-learning, students no longer need to come to the location of guidance because they can learn whenever they want each they bring a laptop or smartphone by accessing online tutoring. Ruangguru presents as one of alternative online tutoring media can be easily accessed via smartphone, laptop or tablet.
